When acetaminophen (25mM) was introduced into the perfused rat liver, the hepatic O_2 uptake was rapidly inhibited first and then later slow-down. The rapid inhibition was found to be due to mitochondrial blokade, whereas the so-called "slow inhibition" was associated with microcirculatory aberrations as evidenced by inhomogeneous staining of the liver tissue by trypan blue infusion(0.1%). NaCN(0.5mM) also caused rapid and slow respiratory inhibitions, giving heterogeneous trypan blue staining. Similarly inhomogeneous trypan blue staining was noted when the liver was perfused with anoxic medium. Accompanying the slow inhibition and heterogeneous trypan blue staining appeared to be a release of lactate dehydrogenase from the liver into the effluent.

DETERMINATION OF WOUND HEALING EFFECT OF EPIDERMAL GROWTH FACTOR ON THE FIRST DEGREE BURN SKIN

To investigate the effect of EGF on the wound healing process of burn skin, histological study has been performed. Under strictly regulated experimental burn condition, the burn skin was prepared and the cytological observation was performed. Also micro auto-radiograph study of [2-^14C] thymidine uptake study has been conducted. EGF in 20 % gelatin gel film was applied on the burn skin area. At pre-determined post burn time, mice were sacrificied and the treated area was excised and processed for the cytological observation. The optimum burn condition (70℃, 10 sec burn) and the post-burn time (24hr) to observe the wound healing effect of EGF have been established. Labeling index of ^14C-thymidine incorporation into the nucleus of the cell can provide us the clear understanding of the wound healing effect of EGF.
No Role of Protected Region B of Human Cytochrome P4501A2 Gene (CYP1A2) As an AP-1 Response Element

Cytochrome P4501A2 (CYP1A2) is a member of the cytochrome P450 family of isozymes involved in the phase I drug metabolism of vertebrates. CYP1A2 is responsible for the activation of a number of aromatic amines to mutagenic and carcinogenic forms. Thus, the level of CYP1A2, which varies among different populations, may determine an individual's susceptibility to these chemicals. We have previously reported on the importance of a cis element named PRB (protected region B) in the regulation of human Cytochrome P4501A2 (CYP1A2) gene, sequence(-2218 to -2187 bp) revealed a putative AP-1 binding site, TGACTAA, at -2212 bp(Chung and Brenick, 1997). To elucidate the role of AP-1 in CYP1A2 regulation, we transiently overexpressed c-Jun and c-Fos transcription factors in human hepatoma HepG2 cells, and examined their influence on the CYP1A2 promoter activity by reporter gene assays. Cotransfection of the c-Jun and the c-Fos expression vectors increased the induced transactivation by five to six fold from the CYP1A2 promoter constructs. However, deletion of the PRB element did not affect the degree of activation by the c-Jun and the c-Fos. Therefore, it is unlikely that the c-Jun and the c-fos activate the CYP1A2 promoter through this AP-1 consensus-like sequence in the PRB region.

A 2-month-old female Holstein Friesian calf was noted to leak urine from the umbilicus during urination. She urinated through both the umbilicus and the urethra. A conduit from the umbilicus to the bladder was identified with ultrasonography. Uroperitoneum was ruled out because blood urea nitrogen and creatinine were within reference ranges. Surgery was performed to resect the patent urachus. Urachitis was identified during surgery. Dexamethasone and an antibiotic were used for the inflammation and postoperative cares. Meloxicam was substituted for dexamethasone since the calf showed side effects to dexamethasone. The calf recovered completely and there was no urine leakage from the umbilicus.
