Antihypertensive and Antihyperlipemic Actions of Chitosan

The effect of dietary fibers on the hypertensive action of NaCl was examined by administration of a high salt diet containing alginic acid, which readily absorbs cations or chitosan, which readily absorbs anions, to normotensive rats and SHRSP (Stroke-prone spontaneously hypertensive rats) for 40 days. Addition of alginic acid to the high salt diet increased the amount of sodium and the addition of chitosan increased the amount of chloride in the feces of normotensive rats. Addition of chitosan to the high salt diet resulted in a significantly lower systolic blood pressure than addition of alginic acid in both groups. Serum ACE(angiotensin converting enzyme)was significantly reduced in SHRSP fed with the high salt diet containing chitosan. Serum chloride ion was lower in the normotensive rats fed with the high salt diet containing chitosan than alginic acid. In humans, the high salt diet increased the systolic blood pressure and serum ACE activity and chloride concentration after 1 h and oral administration of chitosan inhibited these increases. It also reduced the serum bicarbonate level after 1 h, but did not affect the sodium concentration. Serum ACE in humans was found to be stimulated by chloride ion. These results suggest that chitosan prevents increase in the systolic blood pressure of humans induced by high salt intake by inhibiting intestinal absorption of chloride, an activator of ACE. Based on these results, the relationship between serum ACE and chloride concentration was discussed. Chitosan was found to inhibit strongly hydrolysis of triolein emulsified with soybean phosphatidylcholine by pancreatic lipases at its dose of 10yg/ml. Chitosan, however, failed to inhibit the hydrolysis of triolein emulsified with gum arabic. In vivo, oral administration of chitosan to rats reduced the plasma triglyceride concentration. Based on these results, antihyperlipemic action of chitosan was discussed.
Nepal산 Pseudo Ginseng 성분이 암독소 호르몬-L의 체지방 분해작용에 미치는 영향

Nepal産인 Nepal pseudo ginseng(NPG)의 大根과 小根 2종에 있어서 물추출 성분과 ethanol 침전성분이 암독소(Toxohormone-L)가 유도하는 지방분해에 미치는 영향을 관찰하고자 본 연구를 시도하였다. 지방질 분해인자로 알려진 독소호르몬-L은 Sarcoma180痛생쥐의 복수증액으로부터 부분정제하여 이용하였다. 암독소가 유도하는 지방분해 저해능은 반응의 최종농도가 500 및 1,000 dalton 일 때 大根의 NPG은 ethanol 침전성분(평균 55.5%)이 물추출 성분보다 평균 1.37배 더 높았으나 小根의 NPG는 반대로 물추출 성분(평균 55.5%)이 평균 1.14배 더 높았다. 한편 대근의 NPG의 ethanol 침전성분이 지방분해에 미치는 저해능(47.6%)을 최종 반응농도가 1,000㎍/㎖일 때 고려홍삼의 ethanol 침전성분(79.8%)에 비교하면 약 60%에 불과하였다. This study was divised to observe an inhibitory effect toward a lipolytic action of toxohormone-L from large root and small root Nepal pseudo ginseng(NPG: Nepal products) components by water extract and ethanol precipitate in vitro. Toxohormone-L is known to be a lipolytic factor that was partially purified from the ascites fluid of Sarcoma 180-bearing mice and of patients with hepatoma. The inhibitory effect that inhibited the lipolytic action of toxohormone-L by ethanol precipitate component of large root NPG (mean 55.5%) was higher (mean 1.37 times) than that of water extract component in final reaction concentration of 500 and l,000㎍/㎖, on the other side inhibitory effect of water extract component in small root NPG(mean 55.5%) was higer(mean 1.14 times) than that of ethanol precipitate component. In a way inhibitory effect of precipitate component in large root NPG (47.6%), when final reaction concentration of sample were l,000㎍/㎖, was about 40% lower than that of Korean red ginseng.
高麗人蔘 中 酸性多糖 成分이 選擇的 調整劑 및 食鹽不振 改善 役割에 미치는 硏究

Korean red ginseng powder was found to contain adenosine and an acidic peptide which inhibited epinephrine - induced lipolysis and stimulated insulin- mediated lipogenesis from add glucose. We isolated a toxic substance named "Toxohormone-L" from ascites fluid of patients with various malignant tumors. The toxohormone-L stimulated lipolysis in rat adipocytes and induced anorexia in rats. Both the lipolytic and the anorexigenic actions of toxohormone-L were found to be inhibited by ginsenoside Rb₂ in Korean red ginseng ; The ginsenoside Rb₂inhibits toxohormone-L induced lipolysis in fat cells, while it stimulates adrenocorticotropic hormone (ACTH) stimulated lipolysis. This, all these substances extracted from Korean red ginseng exhibited selective modulations toward the opposite metabolic pathways in rat adipocyte . We call these substances "Selective Modulators". This study was divised to abserve the role of selective modulator and anti-anorexigenic agents in acidic polysaccharide of Korean red ginseng. A substance that inhibited the lipolytic action of toxohormone-L was isolated and purified from Korean red ginseng powder. This substance had a pectin-like α-1,4-polygalacturonan backbone with some acetoxyl group, and so was an acidic polysaccharide. It inhibited toxohormone-L induced lipolysis in a dose dependent manner at concentrations higher than 10 ㎍/㎖. And the anorexigenic action of toxohormone-L was found to be inhibited by red ginseng in patients with malignant tumor in digestive tract. These results that both the lipolytic and the anorexigenic actions of toxohormone-L were found to be inhibited by acidic polysaccharide in Korean red ginseng. And suggest that Panax ginseng may protect debilitation of cancer patients through inhibiting the lipolytic and the anorexigenic actions of toxohormone-L.

The objective of this study was to investigate the inhibitory reaction toward an lipolytic action of toxohormone-L from Anaphalis Sinica Hance powder. Toxohormone-L is known to be a lipolytic factor that was purified from the ascites fluid of sarcoma 180-bearing mice and of patients with hepatoma. Anaphalis Sinica Hance powder was found to 50%up inhibit significantly toxohormone-L-induced lipolysis at its concentration of 0.50 ㎎/㎖(by AS-1) and 0.25㎎/㎖(by AS-2).

This study was divised to observe the inhibitory effect of ethanol treatment precipitate from Nepalese pseudo ginseng(NPG-2 kinds ; large root and small root) on a lipolytic action of Toxohormone-L and angiotensin converting enzyme(ACE) which has been known as blood pres sure control action. In vitro, at the concentration of 1,000ug/㎖, Inhibitory activity of lipolysis by the large root and small root of NPG in 3,500∼1,000 Dalton were each 58.7 and 61.9% higher than other molecular weight group of active fraction. In a way, at the concentration of 1 ㎎/R.V.. ACE Inhibitory activity by the large root and small root of NPG in 1,000 Dalton below were each 87.4 and 91.9% higher than other reaction concentration groups, and that of NPG In 1,000 Dalton below were increase by reaction concentration but small root groups were higher than large root groups, respectively.

This study was devised to observe an inhibitory reaction toward an lipolytic action of toxohormone-L from KEUMSUNYEON powder. Toxohormone-L is known to be a lipolytic factor that was purified from the ascites fluid of sarcoma 180-bearing mice and of patients with hepatoma. KEUMSUNYEON powder was found to inhibit toxohormone-L induced lipolysis at its concentration of 0.5㎎/㎖.
