      • Molecular Cloning and Bioinformatic Analysis of SPATA4 Gene

        Full-length cDNA sequences of four novel SPATA4 genes in chimpanzee, cow, chicken and ascidian were identified by bioinformatic analysis using mouse or human SPATA4 cDNA fragment as electronic probe. All these genes have 6 exons and have similar protein molecular weight and do not localize in sex chromosome. The mouse SPATA4 sequence is identified as significantly changed in cryptorchidism, which shares no significant homology with any known protein in swissprot databases except for the homologous genes in various vertebrates. Our searching results showed that all SPATA4 proteins have a putative conserved domain DUF1042. The percentages of putative SPATA4 protein sequence identity ranging from 30% to 99%. The high similarity was also found in 1 kb promoter regions of human, mouse and rat SPATA4 gene. The similarities of the sequences upstream of SPATA4 promoter also have a high proportion. The results of searching SymAtlas (http://symatlas.gnf.org/SymAtlas/) showed that human SPATA4 has a high expression in testis, especially in testis interstitial, leydig cell, seminiferous tubule and germ cell. Mouse SPATA4 was observed exclusively in adult mouse testis and almost no signal was detected in other tissues. The pI values of the protein are negative, ranging from 9.44 to 10.15. The subcellular location of the protein is usually in the nucleus. And the signal peptide possibilities for SPATA4 are always zero. Using the SNPs data in NCBI, we found 33 SNPs in human SPATA4 gene genomic DNA region, with the distribution of 29 SNPs in the introns. CpG island searching gives the data about CpG island, which shows that the regions of the CpG island have a high similarity with each other, though the length of the CpG island is different from each other.This research is a fundamental work in the fields of the bioinformational analysis, and also put forward a new way for the bioinformatic analysis of other genes.

      • Supplementing lactation diets with herbal extract mixture during summer improves the performance of sows and nursing piglets

        <P>A total of 45 Landrace x Yorkshire multiparous sows were used to evaluate the effect of dietary herbal extract mixture (Scutellaria baicalensis and Lonicera japonica, HEM) supplementation in lactating sows under heat stress. Sows were randomly allotted to 1 of 3 dietary treatments: 1) CON, basal diet; 2) TRT 1, basal diet with 5 g/d HEM; 3) TRT 2, basal diet with 10 g/d HEM. During lactation, dietary HEM supplementation linearly increased (P<0.05) the average daily feed intake (ADFI) and linearly decreased (P<0.05) backfat loss. The digestibility of dry matter (DM) was increased after farrowing (linear, P<0.05; quadratic, P<0.05) and weaning (linear, P<0.05) by HEM supplementation. Furthermore, HEM treatment led to a lower (linear, P<0.01) serum cortisol level. In addition, administration of HEM improved (linear, P<0.05) the piglets weaning weight and overall average daily gain (ADG) during suckling period. Meanwhile, on day 7 and 14 after birth, the fecal score of piglets was decreased (linear, P<0.01) by HEM supplementation. Taken together, under high ambient temperatures, inclusion of HEM to lactation diets could improve the feed intake, digestibility of DM, piglets weaning weight and ADG, while decreasing backfat loss, serum cortisol level, as well as the diarrhea of piglets.</P>

      • Effects of different dietary n-6:n-3 PUFA ratios on growth performance, blood lipid profiles, fatty acid composition of pork, carcass traits and meat quality in finishing pigs

        <P>A total of 72 crossbred pigs [(Landrace x Yorkshire) x Duroc] with an average initial BW of 51.59 +/- 1.59 kg were used in this 10-wk feeding trial to investigate the effects of different dietary n-6: n-3 polyunsaturated fatty acids (PUFA) ratios in finishing pigs. Pigs were randomly allotted to 3 dietary treatments (each n=24) with 6 replications and 4 pigs per pen (2 barrows and 2 gilts). Pigs were fed a corn-soybean meal-based diets formulated by replacing soybean oil with linseed oil to achieve n-6: n-3 close to 5: 1, 10: 1 and 15: 1, respectively. The growth performance, carcass traits and meat quality parameters (meat color, sensory evaluation, cooking loss, drip loss, pH, longissimus muscle area and water holding capacity) were not influenced (P>0.10) by various dietary n-6: n-3 ratios. Pigs fed dietary n-6: n-3 PUFA ratios of 5: 1 had a lower (P<0.05) serum total cholesterol, LDL cholesterol and triglyceride levels. The concentrations of n-3 PUFA, including C18: 3n-3, C22: 5n-3 and C22: 6n-3 were improved (P<0.05) in the longissimus dorsi muscles of pigs fed dietary n-6: n-3 PUFA ratios of 5: 1. Furthermore, pigs fed dietary n-6: n-3 PUFA ratio of 5: 1 decreased (P<0.05) the n-6 concentrations (C18: 2n-6 and C20: 4n-6) of longissimus dorsi muscles. In conclusion, lowering the dietary n-6: n-3 ratios to 5: 1 could be beneficial for the blood lipid profiles, and improve the nutritional value of pork, without adverse effect on growth performance and meat quality parameters that are related to the consumer acceptance.</P>

      • Application of Complex Probiotics in Swine Nutrition - A Review

        <P>The use of probiotics as alternatives to antibiotics for farm animals is gaining more and more interest during recent years. Probiotics are living microorganisms that provide a wide variety of health benefits to the host when ingested in adequate amounts. The bacterial strains most frequently used as probiotic agents are Bacillus, lactic acid bacteria, Enterococcus and Saccharomyces cerevisiae. It has been suggested that multi-strain probiotics might be more effective than mono-strain probiotics due to the additive and synergistic effects, and many previous studies demonstrated that dietary complex probiotics supplementation had growth promoting effects on pigs. However, the effect of complex probiotics in practice is not always consistent, the effect of probiotic could be affected by strain composition, dosage, feed formula, and the age of animals. In this review, we will give an overview on the current use of complex probiotics for weaning, growing and finishing pigs and sows.</P>

        Effects of Variants in Proopiomelanocortin and Neuropeptide Y Genes on Growth, Carcass, and Meat Quality Traits in Rabbits

        Appetite-related neuropeptides proopiomelanocortin (POMC) and Neuropeptide Y (NPY) are essential for regulating feeding behavior and energy homeostasis. The objective of this study was to evaluate the effects of variants in POMC and NPY genes on growth, carcass and meat quality traits in rabbits. A total of six SNPs were identified for POMC (n = 2) and NPY (n = 4) genes by direct sequencing. Three SNPs were subsequently genotyped by using MassArray system (Sequenom iPLEXassay) in 235 individuals, which belong to three meat rabbit breeds, including 93 Ira rabbits; 81 Champagne rabbits and 61 Tianfu black rabbits. The SNP c.112-12G>T was in intron-exon boundaries (intron 1) of POMC gene, and the association analysis showed that individuals with TT genotype had a greater 84 d body weight (BW84), eviscerated weight and semi-eviscerated weight than those with GT genotype (p<0.05); the TT individuals were also higher than those GG in the ripe meat ratio (RMR) (p<0.05). The g.1778G>C SNP, which was in complete linkage with other three SNPs (g.1491G>A, g.1525G>T and g.1530C>T) in intron 1 of NPY gene, was significantly correlated with eviscerated slaughter percentage and semi-eviscerated slaughter percentage in rabbits, and the individuals with CC genotype had a better performance than CG genotype (p<0.05). These findings would provide primary clues for the biological roles of POMC and NPY underlying the rabbit growth-related traits.

        RESEARCH : Open Access ; A synonymous mutation of uncoupling protein 2 (UCP2) gene is associated with growth performance, carcass characteristics and meat quality in rabbits

        Background: Uncoupling proteins 2 (UCP2) plays an important role in energy regulation, previous studies suggested that UCP2 is an excellent candidate gene for human obesity and growth-related traits in cattle and chicks. The current study was designed to detect the genetic variation of UCP2 gene, and to explore the association between polymorphism of UCP2 gene and growth, carcass and meat quality traits in rabbits. Results: A synonymous mutation in exon 1 and four variants in the first intron of the UCP2 gene were identified by using PCR-sequencing. The synonymous mutation c.72G>A was subsequently genotyped by MassArray system (Sequenom iPLEXassay) in 248 samples from three meat rabbit breeds (94 Ira rabbits, 83 Champagne rabbits, and 71 Tianfu black rabbits). Association analysis suggested that the individuals with AA and AG genotypes showed greater 70 d body weight (P < 0.05), 84 d body weight (P < 0.01), ADG from 28 to 84 days of age (P < 0.05), eviscerated weight (P < 0.01), semi-eviscerated weight (P < 0.01) and semi-eviscerated slaughter percentage (P < 0.05), respectively. Additionally, the individuals with AA and AG genotype had a lower pH value of longissimus muscle (P < 0.01) and hind leg muscle (P < 0.05) after slaughter 24 h. Conclusions: These findings indicated that UCP2 could be a candidate gene that associated with growth performance, body composition and meat quality in rabbits, and this would contribute to advancements in meat rabbit breeding practice.

      • Dietary Lactobacillus plantarum GB805 supplementation improves growth performance and nutrient digestibility in weaning pigs

        <P>A total of 60 weaned pigs with initial body weight (BW) of 8.51 ± 0.42 kg (28 d of age) were randomly divided into 2 groups of 30 each consisted of 6 pens with 2 gilts and 3 borrows and supplemented (treatment) 5.3×108 cfu/kg Lactobacillus plantarum GB805 in addition to basal diet offered during d 0 to 14 and d 15 to 42 (control). During the total trial period or different phases of trial, pigs fed L. plantarum diet had a higher (P less than 0.05) ADG, ADFI and feed efficiency. At the end of experiment, dietary L. plantarum supplementation improved (P less than 0.05) the apparent total tract digestibility of dry matter, nitrogen and the fecal Lactobacillus populations (P less than 0.05) while the fecal E.coli populations were decreased (P less than 0.05). In conclusion, dietary supplementation of L. plantarum GB805 had positive influence on growth rate, nutrient digestibility and gut microbial balance and its supplementation recommended to weaning pigs.</P>

        Effects of dietary xylanase supplementation on performance and functional digestive parameters in broilers fed wheat-based diets

        <P>This experiment was conducted to evaluate dietary xylanase supplementation in broilers wheat-based diets on performance and functional digestive parameters including ileal digesta viscosity, apparent ileal digestibility, intestinal morphology and microflora, digestive enzyme activities, and excreta odor content. A total of 600 1-day-old Ross 308 male broilers with an initial average BW of 45 +/- 0.6 g were randomly allotted into 4 treatments with 10 replicate pens per treatment and 15 broilers in each pen for 35 d. The 4 dietary treatments were wheat-based diets and supplemented with 0, 1,875, 3,750, and 5,625 XU/kg xylanase. Xylanase supplementation improved (linear, P < 0.05) the body weight gain and decreased (linear, P < 0.05) the feed conversion ratio during d 1 to 18 and for the duration of the experiment. Dietary supplementation of xylanase led to a decrease (linear, P < 0.01) in ileal digesta viscosity. The apparent ileal digestibility of dry matter (DM), crude protein (CP), gross energy, and most amino acids (with the exception of Ile, Phe, Asp, Glu, and Pro) were increased (linear, P < 0.05) by xylanase supplementation. Increasing the dietary xylanase levels improved (linear, P < 0.05) the villus height and the ratio of villus height to crypt depth of the duodenum, jejunum, and ileum. In addition, inclusion of xylanase increased (linear, P < 0.05) the Lactobacillus numbers in the ileum and cecum, while decreased the ileal E. coli counts (linear, P < 0.01) and cecal E. coli populations (linear, P < 0.01; quadratic, P < 0.05). There were no significant differences (P > 0.05) in trypsin, amylase, and protease activities of small intestine among dietary treatments. Furthermore, xylanase supplementation reduced excreta NH3 (linear, P < 0.05; quadratic, P < 0.05) and total mercaptan (R. SH) (linear, P < 0.01) concentration. Taken together, dietary xylanase supplementation in broilers wheat-based diets had beneficial effects on growth performance, apparent ileal digestibility of nutrients, intestinal morphology, and microflora balance. Furthermore, the xylanase could reduce the ileal digesta viscosity and excreta odor contents.</P>
