The telescope to be onboard SPICA (Space Infrared Telescope for Cosmology and Astrophysics) has an aperture diameter of 2.5 m and its imaging performance is to be diffraction-limited at a wavelength of 20 μm at the operating temperature of < 8 K. Because manufacturing precise autocollimating flat mirrors (ACFs) with sizes comparable to the SPICA telescope is not technically feasible, we plan to use sub-aperture stitching interferometry through ACFs for optical testing of the telescope. We have verified the applicability of the sub-aperture stitching technique to the SPICA telescope by performing stitching experiments in a vacuum at a room temperature, using the 800-mm telescope and a 300-mm ACF. We have also developed a new method to reduce uncertainties possibly caused by cryogenic and gravitational deformations of ACFs.
Influence Of Ectomycorrhizal Colonization On Cesium Uptake By Pinus densiflora Seedlings

Radionuclides were deposited at forest areas in eastern parts of Japan following the Fukushima Daiichi Nuclear Power Plant incident in March 2011. Ectomycorrhizal (EM) fungi have important effects on radiocaesium dynamics in forest ecosystems. We examined the effect of colonization by the EM fungus Astraeus hygrometricus on the uptake of cesium (Cs) and potassium (K) by Pinus densiflora seedlings. Pine seedlings exhibited enhanced growth after the EM formation due to the colonization by A. hygrometricus. Additionally, the shoot Cs concentration increased after the EM formation when Cs was not added to the medium. This suggests that A. hygrometricus might be able to solubilize Cs fixed to soil particles. Moreover, the shoot K concentration increased significantly after the EM formation when Cs was added. However, there were no significant differences in the root K concentration between EM and non-EM seedlings. These results suggest that different mechanisms control the transfer of Cs and K from the root to the shoot of pine seedlings.

When the nucleus in G0/G1 phase is transferred to an enucleated oocyte by nuclear transfer (NT), its nuclear envelope is broken, followed by condensation of chromosome structure by maturation promoting factor (MPF). This morphological remodeling of the transferred interphase nucleus seems to be essential for subsequent development of NT embryos. In this study, we treated porcine NT embryos with caffeine, which has been reported to increase MPF activity, to keep their MPF level high during NT. When 2.5 mM caffeine was added to the handling medium, the proportion of NT embryos showing condensed chromosome increased significantly (P < 0.05). In NT embryos treated with caffeine, the activity of p34(cdc2) kinase was significantly (P < 0.05) higher than in those without caffeine at 3 h post-injection. In addition, the rate of development to the blastocyst stage after activation was significantly (P < 0.05) higher in NT embryos treated with caffeine. These results indicate that caffeine treatment can increase not only the rate of chromosome condensation but also the developmental rate to the blastocyst stage of porcine NT embryos. This action is most likely due to the support/increase of MPF activity throughout the process of NT.

Background/Aims: Application of polyglycolic acid (PGA) sheets using fibrin glue in post-endoscopic submucosal dissection (ESD)ulcers to prevent bleeding has been reported to be difficult with the conventional delivery method because of gravity. This studyassessed the usefulness of the envelope-based delivery system with and against gravity in living pigs. Methods: PGA sheets were applied on post-ESD ulcers with and against gravity six times each using the conventional and envelopemethods, respectively. The PGA sheet delivery time and the endoscopic and histological findings of the treated ulcer floors werecompared. Results: With gravity, the median PGA sheet application time was 1.00 (0.68–1.30) min/cm2 and 0.32 (0.18–0.52) min/cm2 with theconventional and envelope techniques (p=0.002), respectively, and against gravity, it was 1.20 (1.13–1.63) min/cm2 and 0.50 (0.39–0.58) min/cm2 (p=0.002), respectively. Against gravity, the endoscopic and histological findings revealed that the conventional grouphad insufficient fixation of the PGA sheets, but the envelope groups had sufficient fixation. The results with gravity were similarbetween the groups. Conclusions: The envelope method makes it possible to deliver PGA sheets to the stomach quickly and cover ulcers appropriatelyboth with and against gravity in living pigs.
