Analysis of TRRAP as a Potential Molecular Marker and Therapeutic Target for Breast Cancer

        Ji Wang,Ming Shan,Tong Liu,Qingyu Shi,Zhenbin Zhong,Wei Wei,Da Pang 한국유방암학회 2016 Journal of breast cancer Vol.19 No.1

Purpose: This study was designed to assess the protein levels of transformation/transcription domain-associated protein (TRRAP) in invasive ductal breast carcinomas, and investigated the association between TRRAP and the clinicopathological features of breast cancer. Methods: We examined TRRAP protein expression in 470 breast cancer tissues and normal breast tissues by tissue microarray to study the correlation between TRRAP expression and clinicopathological features. This was analyzed using the chi-square test. Kaplan-Meier survival curves and log-rank tests were applied to analyze the survival status. Cox regression was applied for multivariate analysis of prognosis. Results: The data demonstrated that expression of TRRAP was significantly lower in breast carcinomas (36.6%) than in corresponding normal breast tissues (50.8%). In addition, TRRAP protein levels negatively correlated with tumor size, and indicated poor differentiation, increased nodal involvement, and low p53-positive rates. Analysis of survival revealed that lower TRRAP expression correlated with shorter survival time. Univariate analyses identified TRRAP and progesterone receptor as independent protective factors for breast cancer prognosis. However, Ki-67, tumor size, and nodal involvement appeared to be independent risk factors. Conclusion: The findings indicate a significant correlation between TRRAP protein levels and adverse prognosis in breast cancer. Therefore, TRRAP could be a prognostic biomarker for breast cancer. In addition, TRRAP is also a predictive biomarker of breast cancer treatment.

      • Synthesis and Antibacterial Evaluation of (S,Z)-4-methyl-2-(4-oxo-5-((5-substituted phenylfuran-2-yl) methylene)-2-thioxothiazolidin-3-yl)Pentanoic Acids

        Song, Ming-Xia,Deng, Xian-Qing,Wei, Zhi-Yu,Zheng, Chang-Ji,Wu, Yan,An, Chang-Shan,Piao, Hu-Ri Shaheed Beheshti University of Medical Sciences 2015 Iranian journal of pharmaceutical research Vol.14 No.1

<P>The microbial resistance has become a global hazard with the irrational use of antibiotics. Infection of drug-resistant bacteria seriously threatens human health. Currently, there is an urgent need for the development of novel antimicrobial agents with new mechanisms and lower levels of toxicity. In this paper, a series of (<I>S</I><I>,Z</I>)-4-methyl-2-(4-oxo-5-((5-substitutedphenylfuran-2-yl) methylene)-2-thioxothiazolidin-3-yl)pentanoic acids via a Knoevenagel condensation were synthesized and evaluated for their antibacterial activity <I>in</I><I>-</I><I>vitro</I>. The synthesized compounds were characterized by IR, 1H NMR and MS. The antibacterial test<I> in</I><I>-</I><I>vitro</I> showed that all of the synthesized compounds had good antibacterial activity against several Gram-positive bacteria (including multidrug-resistant clinical isolates) with minimum inhibitory concentration (MIC) values in the range of 2–4 µg/mL. Especially compounds 4c, 4d, 4e and 4f were the most potent, with MIC values of 2 µg/mL against four multidrug-resistant Gram-positive bacterial strains.</P>

Influence of internal pores and graphene platelets on vibration of non-uniform functionally graded columns

        Wen-qi Liu,Shan-jun Liu,Ming-yu Fan,Wei Tian,Ji-peng Wang,Vahid Tahouneh 국제구조공학회 2020 Steel and Composite Structures, An International J Vol.35 No.2

This paper deals with free vibration analysis of non-uniform column resting on elastic foundations and subjected to follower force at its free end. The internal pores and graphene platelets (GPLs) are distributed in the matrix according to different patterns. The model is proposed with material parameters varying in the thickness of column to achieve graded distributions in both porosity and nanofillers. The elastic modulus of the nanocomposite is obtained by using Halpin-Tsai micromechanics model. The differential quadrature method as an efficient and accurate numerical approach is used to discretize the governing equations and to implement the boundary conditions. It is observed that the maximum vibration frequency obtained in the case of symmetric porosity and GPL distribution, while the minimum vibration frequency is obtained using uniform porosity distribution. Results show that for better understanding of mechanical behavior of nanocomposite column, it is crucial to consider porosities inside the material structure.

The effects of paeoniflorin injection on soluble triggering receptor expressed on myeloid-1 (sTREM-1) levels in severe septic rats

        Liu, Xiao-Rong,Xu, Jie,Wang, Yi-Min,Ji, Ming-Suo,Liu, Fu-Shan The Korean Society of Pharmacology 2016 The Korean Journal of Physiology & Pharmacology Vol.20 No.6

Paeoniflorin (PAE) is the most abundant compound in Xuebijing injection widely used to treat sepsis. We aimed to investigate effect of PAE on expression of soluble triggering receptor expressed on myeloid cells-1 (sTREM-1) in a rat model of sepsis. Wistar rats were divided into Normal, Model, and PAE groups (n=20 each). Endotoxin was administrated at 5 mg/ml/kg in Model and PAE rats to establish rat sepsis model. 1 h after endotoxin administration, PAE was administrated at 4 ml/kg in PAE group once per day for 3 days. Routine blood tests and biochemical indexes were assessed, including aspartate aminotransferase (AST) and creatine kinase-MB (CK-MB). The plasma sTREM-1 level was measured using quantitative ELISA. At the end of experiment, the small intestine, liver, kidney and lung were subjected to pathological examinations. A rat model of sepsis-induced multiple organ dysfunction syndrome (MODS) was established successfully with endotoxin administration (5 mg/ml/kg), evidenced by histo-pathological examinations, routine blood tests and biochemical indexes: platelet count decreased and white blood cell count increased (p<0.05), CK-MB and AST increased (p<0.05). PAE treatment significantly reduced the plasma levels of AST, CK-MB, and sTREM-1, compared to Model group (p<0.05). Meanwhile, sepsis-induced damages in the liver, lung, stomach and intestinal mucosa were also markedly ameliorated by PAE treatment. PAE demonstrated a significantly protective effect in a rat model of sepsis by decreasing plasma sTREM-1 level, reducing inflammation, preventing MODS and protecting organ functions.
