Trust, Risk Perception, and Policy-making in Drinking Water Problems

The purpose of this study is to investigate how environ-mental risks associated with water problems are identified and assessed among Korean elites and general population. By doing so, this study is expected to introduce how the Koreans focus on different dimensions of the risks in Korea. Although Korea has improved its quality of life sig-nificantly as the country has developed, people do not trust the quality of tap water that they drink everyday. Despite efforts made by the gov-ernment, the public hasnt shown any confidence in water quality, due to a series of environmental accidents in the 1990s that included carci-nogen detection in tap water and phenol flows into Nak-Tong river in South Kyong-Sang Province. Using Q method, this study finds different environmental risk perceptions among elites and citizens and provides rich exploratory information about existing values in tap water prob-lems, which will be significant information to policy makers. The study finds three different types among Korean elites and citizens. First group is Anti-Capitalist, Techno-Skepticist, Critical Environmentalists. Second group is pro-technology, Optimistic Modernists, and finally, Political ecologists and Distrustful Modernists are the last group.

<P>X-linked Adrenoleukodystrophy (X-ALD) is a genetic disease that caused bymutations in adenosine triphosphate [ ATP]-binding-cassette transporter superfamily D member 1 (ABCD1) gene. We generated an induced pluripotent stem cell (iPSC) line from a 21-year-old male X-ALD patient-derived fibroblasts by Sendai virus mediated reprogramming. Established iPSCs stably expanded while maintaining immunoreactivity for various pluripotency markers and alkaline phosphatase, as well as normal 44+ XY karyotype. This study shows systematic and empirical findings of the determinants of foreign direct investment in case of Korea, which has experienced various political, social, and economic phenomena. This study especially investigates the determinants of U.S. foreign direct investment flows in Korea before the country was democratized. Six variables of political instability: They inclide riots (protest demonstration); labor strikes; external threat; data from domestic violence; assassination; and coup d’etat. They are categorized with regard to the data from the the Far Eastern Economic Review weekly magazine(1977 to 1991). This study finds that there is no significant relationship between political instability and FDI flows in Korea, unless the political instability is severe enough to affect the FDI policy and economic conditions, which was the case of 1980. Although investors normally say that political instability is important to their investment decisions, their actions do not confirm this. In making their decision, we found economic conditions and economic policy matters more in the case of Korea.

<P>A small 29 nm monodispersed silica nanoparticle <B>1a</B> was synthesized as a diarylethene-based reversible fluorescence photoswitch by copolymerizing silane precursors in one-pot including <B>3a</B> and <B>4</B>. Objective: Previous studies have shown that amniotic fluid-derived mesenchymal stem cells (AF-MSCs) exhibited the immunomodulatory properties in LPS-inducing preterm birth model. To analyze the mechanism of this therapeutic effect, we focused on the secretory profiles of AF-MSCs which were contained in conditioned media (AF-MSC-CM). In this study, we hypothesize that the AF-MSC-CM treatment reduces the inflammatory reaction of uterus and placenta in pregnant females and ameliorates the preterm birth caused by inflammation. Methods: Day 25 gestation ICR mice were used for preterm birth model. For induction of preterm birth, lipopolysaccharide (LPS, 1mg/kg) was injected into peritoneum. AF-MSC-CM was collected from 5.0 x 105 cells/100 for 72 h and concentrated 10 folds for injection. 4.0 x 105 amniotic fluid mesenchymal stem cells were applied for positive control. All of the experiments groups were executed more than 5 times (n5). All of mice were sacrificed after LPS and materials (AF-MSCs, AF-MSC-CM) treatment. Uterus, placenta, and blood were harvested to analyze the inflammatory reaction by qRT-PCR, ELISA, and immunofluorescence staining. Results: Intraperitoneal injection of AF-MSCs into LPS-induced preterm birth mice reduced the incidence of preterm birth up to less than 30%. Additionally, the expression of inflammatory markers of uterus and placenta (TNF-, IL6, IL10, IL11) was significantly decreased by AF-MSC-CM injection. On immunofluorescence staining, marcrophage markers expression (MHC II, CD80) was diminished in AF-MSC-CM group. Conclusion: Although not all of preterm birth were inhibited by AF-MSC-CM, large portion of MSC effects on anti-inflammation accounted for the paracrine factors secreted from AF-MSCs. This research suggests that AF-MSC-CM is comprehensive therapeutic approach for the preterm birth by immunomodulation effects.

<P><B>Abstract</B></P> <P>4-Methylbenzylidene-camphor (4-MBC) is an estrogenic compound used in a variety of personal care products and is associated with water pollution. In this study, we verified that exposure to 4-MBC suppresses the proliferation and invasiveness of the HTR8/SVneo human trophoblast cell line. Moreover, HTR8/SVneo cells treated with 4-MBC underwent apoptosis with increased DNA fragmentation. 4-MBC also activated the PI3K/AKT and ERK1/2 signaling pathways in HTR8/SVneo cells. Furthermore, 4-MBC induced oxidative stress mediated by reactive oxygen species production, which was associated with HTR8/SVneo cell death. 4-MBC promoted lipid peroxidation and loss of mitochondrial membrane potential in HTR8/SVneo cells and activated the expression of genes encoding a protein expressed on the surface of human trophoblast cells, including the EPH receptor B4 and G protein-coupled receptor 56 genes. Therefore, 4-MBC may retard the normal growth and survival of human trophoblast cells and may hamper normal placental formation during early pregnancy.</P> <P><B>Highlights</B></P> <P> <UL> <LI> 4-MBC induces abnormal placental formation during early pregnancy. </LI> <LI> 4-MBC inhibits the normal growth and survival of human trophoblast cells. </LI> <LI> 4-MBC-induced oxidative stress results in mitochondrial membrane disruption. </LI> </UL> </P>

This study was performed to determine the developmental potentials of nuclear transfer (NT) embryos using life-span extended cells transfected with a foreign gene as donor cells. A life-span extended bovine embryonic fibroblast cell line was transfected with an expression vector in which the human type II collagen (BOMAR) and ear fibroblasts were used as a donor cell. Cytogenetic analysis was performed to analyze the chromosomal abnormality of donor cells. The fusion rate of 1.8 kV/cm for $15{\mu}sec$ given twice was significantly higher than that of other groups (p<0.05) and the embryos lysed were significantly higher after 1.8 kV/cm for $20{\mu}sec$ given once compared to other groups (p<0.01). The blastocyst development in the ear cell group was statistically significant compared to both BOMAR groups (p<0.01). Both BOMAR groups cultured more than 40 passages (>40 passages) had a lower number of chromosomes; however, fresh granulosa cell (GC) and BOMAR groups cultured less than 20 passages had normal chromosome numbers. Both >40 passages BOMAR groups had numerous obscure debris in metaphase spreads. The transfected foreign gene was expressed in all BOMAR groups, but not in the GC group. Based on these results, the lower developmental potential of NT embryos using life-span extended donor cells transfected with a foreign gene might be a cause of chromosomal abnormality in donor cells.
