        Quantitative analyses of ricinoleic acid and ricinine in Ricinus communis extracts and its biopesticides

        The quantitative analytical method for the bioactive substance, 3-cyano-4-methoxy-N-methyl-2-pyridone (ricinine) and an index compound, ricinoleic acid in castor plant (Ricinus communis) extract or oil was developed. For the determination of a pyridone alkaloid compound, ricinine, successive cartridge cleanup method combined with ultra-performance liquid chromatography was set up with ENVI-CarbTM (0.5 g) and C18 SPE cartridges. Accuracy and precision were evaluated through fortification studies of one biopesticide (PE) at 10 and 100 mg kg.1. Mean recoveries of ricinine were 98.7 and 96.0 % associated with less than 10 % RSD, respectively. For the determination of ricinoleic acid in castor extract and oil, saponification and methylation were optimized using gas chromatography-time of flight mass spectrometry. Recovery was more than 84.8 % associated with 6.2 % RSD after derivatization procedure. Both methodologies developed were applied to analyze real samples including three castor oil products and six commercially available biopesticides containing R. communis, collected at Korean market. The contents of ricinine and ricinoleic acid in most commercial biopesticides were less than the oil or extract contents indicated by label.

        <P>Telmisartan is an angiotensin II receptor blocker and partial peroxisome proliferator-activated receptor gamma agonist that modulates the renin-angiotensin-aldosterone system. It is used primarily to manage hypertension, diabetic nephropathy, and congestive heart failure. Recent studies have reported that myocardial infarction (MI) has occurred in telmisartan-treated patients. The purpose of the study was to investigate the specific conditions and underlying mechanisms that may result in telmisartan-induced MI. We evaluated the effect of telmisartan on whole hearts, cardiomyocytes, and cardiac sarcolemmal ion channels. Hearts of 8-week-old male Sprague-Dawley rats were perfused with 3, 10, 30, or 100?μM telmisartan or losartan or with normal Tyrode's solution (control) for 3?h. We found that telmisartan induced myocardial infarction, with an infarct size of 21?% of the total at 30?μM (P?<?0.0001) and 63?% of the total area at 100?μM (P?<?0.001). Telmisartan also induced cardiac dysfunction (e.g., decreased heart rate, diminished coronary flow, hypercontracture, and arrhythmia). Confocal microscopy demonstrated that 30?μM telmisartan significantly elevated the intracellular Ca(2+) level, leading to hypercontracture and cell death. Patch clamp analysis of isolated cardiomyocytes revealed that telmisartan induced Na(+) overload by slowing the inactivation of voltage-gated Na(+) current (I (Na)), activating the reverse mode of Na(+)-Ca(2+) exchanger activity, and causing Ca(2+) overload. Telmisartan significantly delayed the inactivation of the voltage-gated Na(+) channel, causing cytosolic Na(+) overload, prolonged action potential duration, and subsequent Ca(2+) overload. Above 30?μM, telmisartan may potentially cause cardiac cell death and MI.</P>

        Tetrahydrobiopterin enhances mitochondrial biogenesis and cardiac contractility via stimulation of PGC1α signaling

        <P><B>Abstract</B></P> <P>Tetrahydrobiopterin (BH4) shows therapeutic potential as an endogenous target in cardiovascular diseases. Although it is involved in cardiovascular metabolism and mitochondrial biology, its mechanisms of action are unclear. We investigated how BH4 regulates cardiovascular metabolism using an unbiased multiple proteomics approach with a sepiapterin reductase knock-out (<I>Spr</I> <SUP>−/−</SUP>) mouse as a model of BH4 deficiency. <I>Spr</I> <SUP>−/−</SUP> mice exhibited a shortened life span, cardiac contractile dysfunction, and morphological changes. Multiple proteomics and systems-based data-integrative analyses showed that BH4 deficiency altered cardiac mitochondrial oxidative phosphorylation. Along with decreased transcription of major mitochondrial biogenesis regulatory genes, including <I>Ppargc1a</I>, <I>Ppara</I>, <I>Esrra</I>, and <I>Tfam</I>, <I>Spr</I> <SUP>−/−</SUP> mice exhibited lower mitochondrial mass and severe oxidative phosphorylation defects. Exogenous BH4 supplementation, but not nitric oxide supplementation or inhibition, rescued these cardiac and mitochondrial defects. BH4 supplementation also recovered mRNA and protein levels of PGC1α and its target proteins involved in mitochondrial biogenesis (mtTFA and ERRα), antioxidation (Prx3 and SOD2), and fatty acid utilization (CD36 and CPTI-M) in <I>Spr</I> <SUP>−/−</SUP> hearts.       • Visualization of resin impregnation through opaque reinforcement textiles during the vacuum-assisted resin transfer molding process using ultrasound

        <P>In this study, an ultrasound visualization system has been set up for in-situ monitoring of the resin flow impregnating through opaque carbon fiber reinforcements during the vacuum-assisted resin transfer molding. The flow front advancement through the carbon fabrics covered by a bagging film can be identified and visualized by the high frequency B (brightness) mode ultrasound imaging technique. The resin advancement in the opaque carbon fabrics has turned out to form a non-uniform plug flow along the pressure gradient direction and the potential void formation can be observed from the mesoscopic resin flow behavior: the inter-tow regions have been preferentially filled by the resin fluid and the fiber tow region behaves as a sink that probably remain as a void defect. The local unsaturated transient velocity of the resin flow has been also evaluated, which is particularly important in understanding saturation behaviors in dual-scale fabrics and is hardly measurable by other means. In spite of the hardware limitations on the resolution, the proposed ultrasound visualization system can provide a less expensive and portable visualization tool to understand the microstructure of opaque reinforcements and monitor the resin flow behavior during the vacuum-assisted resin transfer molding process in the industrial composite manufacturing environment.</P>

        친숙도가 선호도, 위험지각, 관광활동에 미치는 영향

        본 연구는 중국을 방문하는 한국인 관광객들의 중국에 대한 친숙도가 관광지로서 중국에 대한 선호도, 위험지각, 중국 방문시 선호하는 관광활동에 미치는 영향을 조사하였다. 본 연구에서 제시한 연구과제들을 검증하기 위해 중국으로 여행을 떠나기 전 인천국제공항에서 대기하고 있던 한국인 관광객들을 대상으로 설문을 진행하였다. 설문조사 결과 총305개의 유효한 설문지가 최종적으로 수집되었다. 분석 결과에 따르면, 중국에 대한 친숙도는 관광지로서 중국에 대한 선호도에 긍정적인 영향을 미치는 것으로 나타났다. 또한, 친숙도는 중국에 대한 일부 위험지각영역과 부정적인 관계를 가지는 것으로 나타났다. 마지막으로, 친숙도는 중국 방문시 선호하는 관광활동 중 역사?자연지역 방문과 영향관계에 있는 것으로 밝혀졌다. 본 연구의 결과는 친숙도가 관광객들의 다른 지각변수들을 이해하기 위한 중요요인이라는 점을 입증하고 있다. This study examines Korean tourists visiting China as to their perceived familiarity, and its impacts on the degree of preference toward China as a tourism destination, perceived risk, and the preferred tourist activities. A survey was conducted to the Korean tourists departing to China at the In-Cheon international airport, which results in collecting 305 usable questionnaires. The results show that the perceived familiarity toward China positively affects the degree of preference to China as a tourism destination. As to the impacts of familarity on the perceived risks, the results reveal that the degree of familarity is negatively associated with several risk perceptions. Lastly, the degree of perceived familiarity is positively related to the one type of preferred tourist activity, visiting nature?historical sites. The results of this study support the role of familiarity as a significant factor in understanding other tourist perceptions.

        <P>A yellow-pigmented, Gram-negative, short rod-shaped, non-motile and non-spore-forming bacterial strain, designated HU1-AH51(T), was isolated from freshwater sediment and was characterized using a polyphasic approach, in order to determine its taxonomic position. On the basis of 16S rRNA gene sequence similarity, strain HU1-AH51(T) was shown to belong to the genus Novosphingobium, showing the highest level of sequence similarity with respect to Novosphingobium resinovorum NCIMB 8767(T) (96.0???%), Novosphingobium naphthalenivorans TUT562(T) (96.0???%) and Novosphingobium panipatense SM16(T) (96.0???%). Strain HU1-AH51(T) had a genomic DNA G+C content of 62.6 mol% and Q-10 as the predominant respiratory quinone. Furthermore, the major polyamine component (spermidine) in the cytoplasm and the presence of sphingoglycolipids suggested that strain HU1-AH51(T) belongs to the family Sphingomonadaceae. On the basis of its phenotypic properties and phylogenetic distinctiveness, strain HU1-AH51(T) represents a novel species of the genus Novosphingobium, for which the name Novosphingobium sediminicola sp. nov. is proposed. The type strain is HU1-AH51(T) (???=???LMG 24320(T) ???=???KCTC 22311(T)).</P>

      • Relati ve importance of factors affecting text reading time and preference(II): Focusing on non-square form letter

        Effectiveness of information conveyance in reading is affected by several factors such as line length, letter size, lines pacing arrangement as well as typeface itself. This study examined relative importance of these factors by asking people to read the texts that was constituted with non-square form letter and rank the preference of texts through conjoint analysis. In the case of reading time, justification was the most important factor, followed by leading, line spacing, letter width, line length, font size, font type in their order of importance. And in the case of preference decision, letter width was the most important factor, followed by font size, justification, line spacing, leading, line length, font type. The result will be useful in understanding how to consider human preference in the Hangul typography.
