      • Structural characterization of the pulmonary innate immune protein SPLUNC1 and identification of lipid ligands

        Ning, Fangkun,Wang, Chao,Berry, Karin Zemski,Kandasamy, Pitchaimani,Liu, Haolin,Murphy, Robert C.,Voelker, Dennis R.,Nho, Chu Won,Pan, Choel-Ho,Dai, Shaodong,Niu, Liwen,Chu, Hong-Wei,Zhang, Gongyi The Federation of American Societies for Experimen 2014 The FASEB Journal Vol.28 No.12

<P>The short palate, lung and nasal epithelial clone 1 (SPLUNC1) protein is a member of the palate, lung, and nasal epithelium clone (PLUNC) family, also known as bactericidal/permeability-increasing (BPI) fold-containing protein, family A, member 1 (BPIFA1). SPLUNC1 is an abundant protein in human airways, but its function remains poorly understood. The lipid ligands of SPLUNC1 as well as other PLUNC family members are largely unknown, although some reports provide evidence that lipopolysaccharide (LPS) could be a lipid ligand. Unlike previous hypotheses, we found significant structural differences between SPLUNC1 and BPI. Recombinant SPLUNC1 produced in HEK 293 cells harbored several molecular species of sphingomyelin and phosphatidylcholine as its ligands. Significantly, <I>in vitro</I> lipid-binding studies failed to demonstrate interactions between SPLUNC1 and LPS, lipoteichoic acid, or polymyxin B. Instead, one of the major and most important pulmonary surfactant phospholipids, dipalmitoylphosphatidylcholine (DPPC), bound to SPLUNC1 with high affinity and specificity. We found that SPLUNC1 could be the first protein receptor for DPPC. These discoveries provide insight into the specific determinants governing the interaction between SPLUNC1 and lipids and also shed light on novel functions that SPLUNC1 and other PLUNC family members perform in host defense.—Ning, F., Wang, C., Berry, K. Toxoplasma gondii induces autophagy and apoptosis in human umbilical cord mesenchymal stem cells via downregulation of Mcl−1

        Chu, Jia-Qi,Jing, Kai-Peng,Gao, Xiang,Li, Peng,Huang, Rui,Niu, Yan-Ru,Yan, Shou-Quan,Kong, Jun-Chao,Yu, Cai-Yuan,Shi, Ge,Fan, Yi-Ming,Lee, Young-Ha,Zhou, Yu,Quan, Juan-Hua Landes Bioscience 2017 Cell Cycle Vol.16 No.5

<P>Autophagy and apoptosis are critical for controlling Toxoplasma gondii (T. gondii) infection. T. gondii infection during pregnancy can damage the fetus and cause birth defects; however, the molecular mechanisms of this process are poorly understood. This study aims to determine the activities of autophagy and apoptosis as well as their regulatory mechanisms during T. gondii infection by using human umbilical cord mesenchymal stem cells (hUC-MSCs) as a model of congenital diseases. LC3B, a hallmark protein of autophagy was incrementally upregulated with the infection duration, whereas p62 was downregulated in T. gondii-infected hUC-MSCs. Concurrent to this result, the invasion of T. gondii into hUC-MSCs increased in a time-dependent manner. The expression levels of Bcl-2 family proteins including Bcl-2, Bcl-xL, Bim, Bax, Bid and Bak were not altered; however, Mcl-1 levels in hUC-MSCs were dramatically decreased upon T. gondii infection. In addition, at 24h post-infection, cleaved PARP and cleaved caspase-3 protein levels were elevated in hUC-MSCs. Importantly, Mcl-1 overexpression reduced the levels of autophagy- and apoptosis-related proteins in T. gondii-infected hUC-MSCs. Mcl-1 proteins were primarily expressed in the fraction containing mitochondria and strongly interacted with Beclin-1 under normal conditions; however, these interactions were remarkably attenuated by T. gondii infection. These results suggest that mitochondrial Mcl-1 is an essential signaling mediator regulating the activation of autophagy and apoptosis during T. gondii infection.</P>

Analysis of Local Surface Plasmon Resonance in Multilayered Au/Ag/Graphene Nanoshells

        Chao Liu,Zhaoting Liu,Jingwei Lv,Tao Sun,Qiang Liu,Haiwei Mu,Paul K. Chu 성균관대학교(자연과학캠퍼스) 성균나노과학기술원 2017 NANO Vol.12 No.5

"The localized surface plasmon resonance (LSPR) properties of Au/Ag/graphene nanoshells are studied by discrete dipole approximation (DDA). The coupled resonance wavelengths show a remarkable dependence on the graphene thickness as well as refractive index of the surrounding medium. The resonance wavelengths of Au/Ag/graphene nanoshells red-shift as the thickness of the graphene layer is increased, when the radii of the Au core and Ag interlayer are 40 nm and 45nm, respectively. Specifically, the longer wavelength red-shifts from 540 nm to 740 nm when the refractive index varies from 1.25 to 2.05. "

Flow-induced Orientations of Fibers and Their Influences on Warpage and Mechanical Property in Injection Fiber Reinforced Plastic (FRP) Parts

        Chao-Tsai Huang,Jia-Hao Chu,Wei-Wen Fu,Chia Hsu,Sheng-Jye Hwang 한국정밀공학회 2021 International Journal of Precision Engineering and Vol.8 No.3

During the past two centuries, due to too fast growth of the human population, the pollution made by human has seriously impacts on our environment, particularly, for the CO2 emission. To diminish the CO2 emission problem, one of the effective solutions is applying lightweight material, such as the fiber-reinforced plastics (FRP), to replace metal in the manufacturing of transportation vehicles. However, since the reinforced function of the fibers inside plastic matrix is very complex, it is not easy to be visualized and managed. Specifically, the connection from microstructures of the fibers to the physical properties of the final product is far from our understanding. In this study, we have proposed a benchmark with three standard specimens based on ASTM D638 with different gate designs. This system is used to study the fiber microstructures and associated mechanical properties using numerical simulation and experimental studies. Results showed that the tensile properties (including tensile modulus and tensile stress) of all three ASTM standard specimens can be improved significantly in the appearance of the fibers. Moreover, the tensile properties variation of the finished parts associated with the microstructures of the short fibers based on the gate design have been also investigated. Specifically, the tensile modulus and the strength of the Model I are greater than that of Model II, while Model III is much less than others because of its double gate effect. The reason why the tensile modulus and the strength of the Model I is greater than that of Model II is due to some entrance effect. That entrance effect will further provide flow-induced fiber orientation to melt and then enhance the tensile properties of Model I. To confirm the observation, a series simulation and experimental studies have been performed. Specifically, the fiber orientation distribution is predicted using CAE, and verified using micro-CT scan and image analysis by Avizo software. Hence, the correlation from fiber microstructure feature (particularly in fiber orientation) to tensile modulus and tensile stress for fiber reinforced thermoplastic (FRP) in injection molding process can be validated.

Analysis of Localized Surface Plasmon Resonance in Ag/ITO/CdS/SiO2 Multilayered Nanostructured Composite

        Chao Liu,Jingwei Lv,Famei Wang,Qiang Liu,Haiwei Mu,Tao Sun,Qiang Liu,Paul K. Chu 성균관대학교(자연과학캠퍼스) 성균나노과학기술원 2015 NANO Vol.10 No.8

Multilayered nanoshells have attracted much attention due to their unique optical, electronic and magnetic properties. In this work, numerical calculation using discrete dipole approximation (DDA) is conducted to investigate the quad-layered metal nanoshell consisting of a particle with a dielectric silica (SiO2) core, inner cadium sulfide (CdS) shell, middle indium tin oxide (ITO) shell and outer metal silver (Ag) shell. The phenomenon is interpreted by plasmon hybridization theory and the Ag–ITO–CdS–SiO2 multilayered nanoshells are studied by extinction spectra of localized surface plasmon resonance. The variation in the spectrum peak with nanoparticle thickness and refractive index of the surrounding medium is derived. The electric field enhancement contour around the nanoparticles under illumination is analyzed at the plasmon resonance wavelength. The |ω-->, |ω+->, and |ω-+> modes red-shift with the refractive index of the surrounding medium and increase in the layer thickness causes either blue-shift or red-shift as shown by the extinction spectra. The mechanism of the red-shift or blue-shift is discussed. The |ω--> blue-shifts and furthermore, the |ω-+> and |ω+-> modes of the Ag coated multilayered nanostructure are noticeable by comparing the extinction efficiency spectra of the Au–ITO–CdS–SiO2 and Ag–ITO–CdS–SiO2 multilayered nanoshells.

A novel illumination system design for application in the integrated screen 3D display

        Chu-Hsun Lin,Chun-Chuan Lin,Hsin-Hsiang Lo,Shuang-Chao Chung,Tian-Yuan Chen,Chy-Lin Wang 한국정보디스플레이학회 2010 Journal of information display Vol.11 No.1

A mini-projector prototype employing a LED light source, a nontelecentric structure, and an LCOS panel for application in the integrated 3D display was fabricated. A seamless image was obtained by tilting an array of mini-projectors. Seamless quality was created by the excellent uniformity of the projection intensity on the mini-projector’s screen, which was simulated as 98.34%. Great uniformity can be realized by optimizing the design of the light source and the optics configuration, which is the key to such realization.

      • Construction as a Theoretical Entity: An Argument Based on MandarinExistential Sentences

( Chao Ran Chen ),( Chu Ren Huang ),( Kathleen Ahrens ) 한국언어정보학회 1995 국제 워크샵 Vol.1995 No.-

The role that constructions play in a linguistic theory has changed throughout the evolution of generative theories. Construction specific rules are common when transformations are envisioned as tree to tree operations in classical TG. On the other extreme, constructions, as well as all structural properties, are regarded as derived linguistic properties predictable from various principles in recent GB theories. Since whether a construction is an autonomous linguistic entity or not has great implications for either a formal or a computational linguistic theory, we will examine the status of Mandarin existential construction based on the theory of Construction Grammar [1,2]. We will show that the Mandarin existential construction represents all unique structure-meaning pair that cannot be captured in a grammar unless the pairing is regarded as a theoretical entity in linguistics ([3]). Since constructions are shown to exist in Mandarin Chinese, we support the theoretical claims of Construction Grammar as well as the position that constructions must be taken into account in NLP.

Accurate delivery of pristimerin and paclitaxel by folic acid-linked nano-micelles for enhancing chemosensitivity in cancer therapy

        Chen Chao,Du Shiyu,Zhong Wu,Liu Kunguo,Qu Lihua,Chu Feiyi,Yang Jingjing,Han Xin 나노기술연구협의회 2022 Nano Convergence Vol.9 No.52

Chemoresistance remains a huge challenge for effective treatment of non-small cell lung cancer (NSCLC). Previous studies have shown Chinese herbal extracts possess great potential in ameliorating tumor chemoresistance, however, the efficacy is clinically limited mainly because of the poor tumor-targeting and in vivo stability. The construction of nano-delivery systems for herbal extracts has been shown to improve drug targeting, enhance therapeutic efficacy and reduce toxic and side effects. In this study, a folic acid (FA)-modified nano-herb micelle was developed for codelivery of pristimerin (PRI) and paclitaxel (PTX) to enhance chemosensitivity of NSCLC, in which PRI could synergistically enhance PTX-induced growth inhibition of A549 cancer cell. PTX was firstly grafted with the FA-linked polyethylene glycol (PEG) and then encapsulated with PRI to construct the PRI@FA-PEG-PTX (P@FPP) nano-micelles (NMs), which exhibited improved tumor-targeting and in vivo stability. This active-targeting P@FPP NMs displayed excellent tumor-targeting characteristics without obvious toxicity. Moreover, inhibition of tumor growth and metastasis induced by P@FPP NMs were significantly enhanced compared with the combined effects of the two drugs (PRI in combination of PTX), which associated with epithelial mesenchymal transition inhibition to some extent. Overall, this active-targeting NMs provides a versatile nano-herb strategy for improving tumor-targeting of Chinese herbal extracts, which may help in the promotion of enhancing chemosensitivity of NSCLC in clinical applications.

The EGF/hnRNP Q1 axis is involved in tumorigenesis via the regulation of cell cycle-related genes

        Yu-Chu Wang,Kung-Chao Chang,Bo-Wen Lin,Jenq-Chang Lee,Chien-Hsien Lai,Li-Jyuan Lin,Yun Yen,Chang-Shen Lin,Shiang-Jie Yang,Peng-Chan Lin,Chung-Ta Lee,Liang-Yi Hung 생화학분자생물학회 2018 Experimental and molecular medicine Vol.50 No.-

Heterogeneous nuclear ribonucleoprotein (hnRNP) Q1, an RNA-binding protein, has been implicated in many posttranscriptional processes, including RNA metabolism and mRNA splicing and translation. However, the role of hnRNP Q1 in tumorigenesis remains unclear. We previously performed RNA immunoprecipitation (RIP)-seq analysis to identify hnRNP Q1-interacting mRNAs and found that hnRNP Q1 targets a group of genes that are involved in mitotic regulation, including Aurora-A. Here, we demonstrate that altering the hnRNP Q1 level influences the expression of the Aurora-A protein, but not its mRNA. Stimulation with epidermal growth factor (EGF) enhances both binding between hnRNP Q1 and Aurora-A mRNA as well as the efficacy of the hnRNP Q1-induced translation of Aurora-A mRNA. The EGF/hnRNP Q1-induced translation of Aurora-A mRNA is mediated by the mTOR and ERK pathways. In addition, we show that hnRNP Q1 up-regulates the translation of a group of spindle assembly checkpoint (SAC) genes. hnRNP Q1 overexpression is positively correlated with the levels of Aurora-A and the SAC genes in human colorectal cancer tissues. In summary, our data suggest that hnRNP Q1 plays an important role in regulating the expression of a group of cell cycle-related genes. Therefore, it may contribute to tumorigenesis by up-regulating the translation of these genes in colorectal cancer.
