      • Microarray를 이용한 카드뮴처리 인간 폐세포(A549 cell line)에서의 유전자발현연구

        The recent development of DNA microarrays provides a strong tool for the standard toxicity analysis through genome-wide assessments of gene regulation. In these study, we used cDNA microarrays to evaluate the gene expression pattern in A549 cells derived from human epithelial cell when cadmium was treated. A549 cells were cultured in 5% CO2 incubator till confluent monolayer was formed and then metal cadmium was treated for 6 hours. Total RNA was extracted and purified by guanidine thiocyanate method and the Cy3- and Cy5-labelled cDNA produced by reverse transcription of the RNA was hybridized to microarray. As results, we found that cadmium markedly up-regulated metallothionein, heat shock protein, hemeoxigenase and down-regulated several other genes including general transcription factors.

        α, α-Disubstituted amino acids have two alkyl substituents at the α-position of α-amino acids and are conformational restricted. We synthesized 4-amino-tetrahydropyran-4-carboxylic acid as an unusual cyclic α, α-disubstituted amino acid mimicking the natural threonine residue. We have chosen the efficient synthetic route via hydantoin formation from tetrahydropryan-4-one by a Bucherer-Bergs reaction and mild hydrolysis of hydantoin.

      • 캔디다 질염에 대한 호중구의 역할

        In this study, role of neutrophils was examined in the protection by MAb B6.1 against vaginal infection. Mice were given intravenously anti-neutrophile antibody prior to intraperitoneal administration of the MAb B6.1 to these mice. The mice were examined for antibody presence in their reproductive tract. This neutropenia was induced by a single dose of the anti-neutrophil antibody, but lymphocytes were also partially depleted. By ELISA analysis, MAb B6.1 was found in the vaginal homogenates. The protective effect was reduced when mice that were pre-treated with MAb RB6-8C5 were given MAb B6.1 before challenge intravaginally with C. albicans. These results show that neutrophils are involved in the protection by the B6.1 to Candida vaginal infection.

      • Ginsenosides Re와 Rg1 혼합성분의 캔디다감염에 대한 면역조절효과

        Immunomodulatory activity of combination of ginsenosides Re and Rg1 against systemic infection caused by Candida albicans. By agar diffusion susceptibility test, the ginsenosides had no growth-inhibitory activity on C. albicans yeast cells. On the other hand, under the in-vivo condition, BALB/c mice treated with the ginsenosides had longer survival times than control mice groups that were given diluent (buffer solution), instead of the Re and Rg1. In experiments 60% percent of the ginsenosides-treated mice survived the entire duration of the observation. Treatment of a RAW 264.7 macrophage/monocyte cell line with the Re and Rg1 mixture induced nitric oxide (NO) production. Microscopical observation displayed that the ginsenosides-treated macrophages changed their morphology by producing pseudopods, whereas morphology of macrophages that received diluent remained unchanged. These data suggest that immunostimulation by the ginsenosides Re and Rg1 would be responsible for the protection against the systemic candidiasis.

      • 비파괴 및 신속 분석을 위한 나프록센 정제의 정량분석법 개발

        Near-infrared (NIR) spectroscopy has been widely applied in various field, since it is nondestructive and no sample preparation is required. In this paper, NIR spectroscopy was used for the determination of naproxen in a commercial pharmaceutical preparation. NIR spectroscopy was used to determine the content of naproxen in intact naproxen tablets containing 250㎎ (65.8% nominal concentration) by collecting NIR spectra in the range of 1100~1150 ㎚. The laboratory-made samples had 46.1~85.5% nominal naproxen concentration. The measurements were made by reflection using a fiber-optic probe and calibration was carried out by partial least square regression (PLSR). Model validation was performed by randomly splitting the data set into calibration and validation data set(63 samples as a calibration data set and 42 samples as a validation data set). The developed NIR calibration gave results comparable to the known values of tablets in a laboratorial manufacturing process with standard error of calibration(SEC) and standard error of prediction(SEP) of 1.06% and 1.04%, respectively. The NIR method showed good accuracy and repeatability. NIR spectroscopic determination in intact tablets allowed the potential use of real time monitoring for a running production process.

      • 사람 혈장 중 트라마돌의 HPLC 분석 및 단회 경구 투여시의 약물 동태

        Validated high-performance liquid chromatographic determination of tramadol in human plasma was developed using fluorescence detection, and bioavailability study of Tridol Capsules (reference drug: Yuhan Corp.) was conducted. Human plasma samples (1.0 mL) spiked with known concentrations of tramadol hydrochloride and 5.0 ㎍ atenolol as an internal standard are alkalinized with 200 ㎕ of 0.5 mol/L NaOH solution and extracted with 7 mL of tert-butylmethyl ether for 5 min. Extracts were centrifuged and 6 mL-portions of organic layer were back-extracted with 200 ㎕ of 0.1 mol/L HCI for 3 min. Thirty mieroliters of centrifuged aqueous layer were injected into the reversed-phase C18 column and eluted with a mixture of 0.01 mol/L KH_(2)PO_(4) buffer, acetonitrile and triethylamine (70 : 30 : 0.1, v/v, pH 5.9 with phosphoric acid) at a flow rate of 1.0 mL/min. Fluorescence detector was set at 270 nm (excitation) and 300 nm (emission). The precision and accuracy were satisfactory over the concentration range 20 - 500 ng/mL. The calibration curve showed a good linearity (R2 = 1). The recovery of tramadol from quality control samples was 98% at 50 ng/mL. The applicability of the method was demonstrated by analysis of plasma after oral administration of a single 100-mg dose (2 capsules) to healthy eight subjects. From the plasma tramadol concentration vs. time curves, the AUC, C_(max) and T_(max) values were found to be 3634.6 ± 1151.9, 411.8 ± 151.5 ng/mL and 2.8 ± 0.9 hour, respectively. The elimination phase followed first order kinetic and the mean elimination rate constant was 0.11 ± 0.03 hr^(-1). The t_(1/2β) was calculated to be 6.8 ± 1.6 hour. Based on the results, this simple and validated assay could be readily applied to any pharmackinetic and/or bioequivalence studies using humans.

      • 흰쥐에 있어 Dopamine 길항약인 Veralipride의 심혈관에 대한 영향

        Cardiovascular effects of veralipride, dopamine antagonist, on blood pressure and vascular response were evaluated in rats. Veralipride did not directly changed on the blood pressure response and vascular tone in rats. But veralipride completely blocked the hypotensive response induced by bromocriptine, dopamine agonist, in consious rats. Veralipride did not affect on the bromocriptine induced vasorelaxation which was precontracted by norepinephrine in isolated rat thoracic aorta. These reaults suggested that mechanisms of action of veralipride on the menopausal hot flush would be related cental dopamine antagonistic action rather than peripheral vasomoter action or estrogen-like action.

      • 은행추출물 EGb 761에서 Ginkgolides 분리 및 검색

        The major active components of EGb 761, extract of Ginkgo biloba leaves, include falconoid glycosides and unique diterpenes known as ginkgolides. In this study, we attempted to find a method to isolate ginkgolides from commercially available EGb 761. The EGb 761 was such as bilobol. The chloroform-treated EGb 761 was then sequentially extracted methanol and water. The water-soluble part was dried by heat. This powder form of final extract was analyzed by TLC and HPLC methods. As an indicator component, ginkgolide B purchased from Sigma was used. The analyses revealed the final extract contained ginkgolides A, b, C, and J as compared with HPLC profiles that were reported from other`s observations.

      • 녹농균에서 생산되는 Pyocin의 구조검색 : examination of the pyocin structure

        Pyocin-producing Pseudomonas aeruginosa strains isolated from water samples were typed. Three isolates of the five pyocin producers (D5, M3, P4, R1, and M4) produced R-type pyocins and the remaining isolates produced S-type pyocins against indicator strains C2 and W2. The R-type pyocin producer, D5 and S-type pyocin producer R1 were examined by electron-microscopy. The R-type pyocin resembling the tail of a phage was rod shaped. The rods were hollow, closed at an end and opend at the other end. S-type pyocin produced by the R1 strain resemble a nail. A basal plate-like structure was visible at a distal end of this S-type pyocin. Electron microscopic observation of sensitive strains with active R-type or S-type pyocin is under investigation.
