Anti-proliferative, anti-inflammatory and antioxidant effects of curcumin analogue A2

        Zhi-Yun Du,Xingchuan Wei,Mou-Tuan Huang,Xi Zheng,Yue Liu,Allan H. Conney,Kun Zhang 대한약학회 2013 Archives of Pharmacal Research Vol.36 No.10

In the present study, we determined the antiproliferative,anti-inflammatory and antioxidant effects of acurcumin analogue, 2,6-bis(3,4-dihydroxybenzylidene) cyclohexanone(designated as A2). In vitro studies showed that A2had a stronger inhibitory effect on the growth of mousemacrophage RAW 264.7 cells than curcumin. A2 also showeda stronger inhibitory effect than curcumin on 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced increases inNF-jB activation and IL-1b expression as well as in aldosereductase activity. A2 was a stronger antioxidant than curcuminas determined by inhibition of lipid peroxidation, inhibition of 1,1-diphenyl-2-picryl-hydrazyl free radical formation, and inhibition of 2,20-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) radical formation. In vivo studiesindicated that A2 was more potent than curcumin for inhibitingTPA-induced ear edema and TPA-induced increases inIL-1b. In addition, oral administration of A2 at a dose of2,000 mg/kg body weight did not cause acute toxicity inmice. Taken together, the results of our study indicate that thecurcumin analogue A2 has stronger anti-proliferative, antiinflammatoryand antioxidant activities than curcumin.

      • Shortest Path Analyses in the Protein-Protein Interaction Network of NGAL (Neutrophil Gelatinase-associated Lipocalin) Overexpression in Esophageal Squamous Cell Carcinoma

        Du, Ze-Peng,Wu, Bing-Li,Wang, Shao-Hong,Shen, Jin-Hui,Lin, Xuan-Hao,Zheng, Chun-Peng,Wu, Zhi-Yong,Qiu, Xiao-Yang,Zhan, Xiao-Fen,Xu, Li-Yan,Li, En-Min Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.16

NGAL (neutrophil gelatinase-associated lipocalin) is a novel cancer-related protein involves multiple functions in many cancers and other diseases. We previously overexpressed NGAL to analyze its role in esophageal squamous cell carcinoma (ESCC). In this study, a protein-protein interaction (PPI) was constructed and the shortest paths from NGAL to transcription factors in the network were analyzed. We found 28 shortest paths from NGAL to RELA, most of them obeying the principle of extracellular to cytoplasm, then nucleus. These shortest paths were also prioritized according to their normalized intensity from the microarray by the order of interaction cascades. A systems approach was developed in this study by linking differentially expressed genes with publicly available PPI data, Gene Ontology and subcellular localizaton for the integrated analyses. These shortest paths from NGAL to DEG transcription factors or other transcription factors in the PPI network provide important clues for future experimental identification of new pathways.

      • Green synthesis of silver nanoparticles to the microbiological corrosion deterrence of oil and gas pipelines buried in the soil

        Zhi Zhang,Jingguo Du,Tayebeh Mahmoudi Techno-Press 2023 Advances in nano research Vol.15 No.4

Biological corrosion, a crucial aspect of metal degradation, has received limited attention despite its significance. It involves the deterioration of metals due to corrosion processes influenced by living organisms, including bacteria. Soil represents a substantial threat to pipeline corrosion as it contains chemical and microbial factors that cause severe damage to water, oil, and gas transmission projects. To combat fouling and corrosion, corrosion inhibitors are commonly used; however, their production often involves expensive and hazardous chemicals. Consequently, researchers are exploring natural and eco-friendly alternatives, specifically nano-sized products, as potent corrosion inhibitors. This study aims to environmentally synthesize silver nanoparticles using an extract from Lagoecia cuminoides L and evaluate their effectiveness in preventing biological corrosion of buried pipes in soil. The optimal experimental conditions were determined as follows: a volume of 4 ml for the extract, a volume of 4 ml for silver nitrate (AgNO3), pH 9, a duration of 60 minutes, and a temperature of 60 degrees Celsius. Analysis using transmission electron microscopy confirmed the formation of nanoparticles with an average size of approximately 28 nm, while X-ray diffraction patterns exhibited suitable peak intensities. By employing the Scherer equation, the average particle size was estimated to be around 30 nm. Furthermore, antibacterial studies revealed the potent antibacterial activity of the synthesized silver nanoparticles against both aerobic and anaerobic bacteria. This property effectively mitigates the biological corrosion caused by bacteria in steel pipes buried in soil.

      • Tumor-derived CD4+CD25+ Tregs Inhibit the Maturation and Antigen-Presenting Function of Dendritic Cells

Du, Yong,Chen, Xin,Lin, Xiu-Qing,Wu, Wei,Huang, Zhi-Ming Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.7

CD4+CD25+regulatory T cells (Tregs) play a key role in regulation of immnue response and maintenance of self-tolerance. Studies have found Tregs could suppress tumor-specific T cell-mediated immune response and promote cancer progression. Depletion of Tregs can enhance antitumor immunity. Dendritic cells (DCs) are professional antigen-presenting cells and capable of activating antigen-specific immune responses, which make them ideal candidate for cancer immunotherapy. Now various DC vaccines are considered as effective treatment for cancers. The aim of this study was to evaluate variation of Tregs in BALB/C mice with hepatocellular carcinoma and investigate the interaction between tumor-derived Tregs, effector T cells (Teff) and splenic DCs. We found the percentages of Tregs/CD4+ in the peripheral blood of tumor-bearing mice were higher than in normal mice. Tumor-derived Tregs diminished the up-regulation of costimulatory molecule expression on splenic DCs, even in the presence of Teff cells and simultaneously inhibited IL-12 and $TNF-{\alpha}$ secretion by DCs.

      • Decreased Expression of FADS1 Predicts a Poor Prognosis in Patients with Esophageal Squamous Cell Carcinoma

        Du, Yong,Yan, Shu-Mei,Gu, Wan-Yi,He, Fan,Huang, Li-Yun,Li, Mei,Yuan, Yan,Chen, Ren-Hui,Zhong, Qian,Li, Man-Zhi,Li, Yong,Zeng, Mu-Sheng Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.12

FADS1 (fatty acid desaturase 1) plays a crucial role in fatty acid metabolism, and it was recently reported to be involved in tumorigenesis. However, the role of FADS1 expression in esophageal squamous cell carcinoma (ESCC) remains unknown. In the current study, we investigated the expression and clinical pathologic and prognostic significance of FADS1 in ESCC. Immunohistochemical analyses revealed that 58.2% (146/251) of the ESCC tissues had low levels of FADS1 expression, whereas 41.8% (105/251) exhibited high levels of FADS1 expression. In positive cases, FADS1 expression was detected in the cytoplasm of cells. Correlation analyses demonstrated that FADS1 expression was significantly correlated with tumor location (p=0.025) but not with age, gender, histological grade, tumor status, nodal status or TNM staging. Furthermore, patients with tumors expressing high levels of FADS1had a longer disease-free survival time (p<0.001) and overall survival time (p <0.001). Univariate and multivariate analyses revealed that, along with nodal status, FADS1 expression was an independent and significant predictive factor (p<0.001). In conclusion, our study suggested that FADS1 might be a valuable biomarker and potential therapeutic target for ESCC.

Thermal error analysis, modeling and compensation of five-axis machine tools

Zhi Huang,Yongchao Liu,Li Du,Han Yang 대한기계학회 2020 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.34 No.10

The role of five-axis CNC machine tools (FAMT) in the manufacturing industry is becoming more and more important, but due to the large number of heat sources of FAMT, the thermal error caused by them will be more complicated. To simplify the complicated thermal error model, this paper presents a new modelling method for compensation of the thermal errors on a cradle-type FAMT. This method uses artificial neural network (ANN) and shark smell optimization (SSO) algorithm to evaluate the performance of FAMT, and developing the thermal error compensation system, the compensation model is verified by machining experiments. Generally, the thermal sensitive point screening is performed by a method in which a large number of temperature sensors are arranged randomly, it increases the workload and may cause omission of the heat sensitive point. In this paper, the thermal imager is used to screen out the temperature sensitive points of the machine tool (MT), then the temperature sensor is placed at the position of the heat sensitive point of the FAMT, and the collected thermal characteristic data is used for thermal error modeling. The C-axis heating test, spindle heating test, and the combined movement test are applied in this work, and the results show that the shark smell optimization artificial neural network (SSO-ANN) model was compared to the other two models and verified better performance than back propagation artificial neural network (BP-ANN) model and particle swarm optimization neural network (PSO) model with the same training samples. Finally, a compensation experiment is carried out. The compensation values, which was calculated by the SSO-ANN model are sent to the real-time error compensation controller. The compensation effect of the model is then tested by machining the ‘S’-shaped test piece. Test results show that the 32 % reduction in machining error is achieved after compensation, which means this method improves the accuracy and robustness of the thermal error compensation system.

      • Personalized Trusted Service Recommendation Method based on Social Work

Du Ruizhong,Pei Zhi,Tian Junfeng 보안공학연구지원센터 2016 International Journal of Security and Its Applicat Vol.10 No.9

In order to improve credibility and precision of the personalized service recommendation, we propose a personalized trusted services recommendation method based on social network. Firstly, according to the similarity of user's preference, the method build new relations combined with direct trust and indirect trust of users between them. Secondly, form a set of similar services which includes services invoked by target users. Eventually, get user’s indirect trustrank to the recommended services through the weighted average of the target users’ indirect trustrank to the recommended service and thus form a recommendation list. The experiment take precision rate, recall rate and F1_Measure as evaluation indexes. Comparing with traditional method which based on no confidence and users’ trusted network, this approach demonstrates its effectiveness and feasibility through the result of the experiment.

      • Influence of Serum VEGF Levels on Therapeutic Outcome and Diagnosis/Prognostic Value in Patients with Cervical Cancer

Du, Ke,Gong, Hong-Ying,Gong, Zhi-Min Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.20

Objective: To explore the influence of serum vascular endothelial growth factor (VEGF) level on therapeutic outcome and diagnosis/prognostic value in patients with cervical cancer. Materials and Methods: A total of 37 patients diagnosed with cervical cancer by biopsy were selected and treated with concurrent chemoradiotherapy. Double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) was adopted before treatment to assess VEGF levels, and its relationships with clinicopathological features and short-term therapeutic effects were analyzed. Results: The median VEGF level in 37 patients before treatment was 647.15 (393.35~1125.16) pg/mL. Serum VEGF levels in patients aged <50 years, in International Federation of Gynecology and Obstetrics (FIGO) stage IIIa~IVa, with lymph node metastasis and tumor size >4 cm were significantly increased (P<0.05). The complete remission (CR) rate was 48.7% (18/37), partial remission (PR) rate was 35.1% (13/37), stable disease (SD) rate was 13.5% (5/37) and progressive disease (PD) rate was 2.70% (1/37), so the objective remission rate (ORR) after treatment was 83.8% (31/37). Logistic regression analysis showed that tumor size and serum VEGF level before treatment were independent risk factors affecting the therapeutic outcome, and the higher the level of serum VEGF, the worse the prognosis when tumor size>4 cm. Some 56.8% of patients manifested with myelosuppression, 37.8% with leucopenia, 24.3% with thrombocytopenia, 5.41% with diarrhea, 46.0% with nausea and vomiting, 21.6% with hair loss and 8.11% with hepatic and renal injury during the treatment. Conclusions: Serum VEGF level may reflect the degree of malignancy of cervical cancer and predict therapeutic effect, which is of great importance to cancer diagnosis and prognosis.
