Assessment of the Severity of Coronavirus Disease: Quantitative Computed Tomography Parameters versus Semiquantitative Visual Score

        Yin Xi,Min Xiangde,Nan Yan,Feng Zhaoyan,Li Basen,Cai Wei,Xi Xiaoqing,Wang Liang 대한영상의학회 2020 Korean Journal of Radiology Vol.21 No.8

Objective: To compare the accuracies of quantitative computed tomography (CT) parameters and semiquantitative visual score in evaluating clinical classification of severity of coronavirus disease (COVID-19). Materials and Methods: We retrospectively enrolled 187 patients with COVID-19 treated at Tongji Hospital of Tongji Medical College from February 15, 2020, to February 29, 2020. Demographic data, imaging characteristics, and clinical data were collected, and based on the clinical classification of severity, patients were divided into groups 1 (mild) and 2 (severe/ critical). A semiquantitative visual score was used to estimate the lesion extent. A three-dimensional slicer was used to precisely quantify the volume and CT value of the lung and lesions. Correlation coefficients of the quantitative CT parameters, semiquantitative visual score, and clinical classification were calculated using Spearman’s correlation. A receiver operating characteristic curve was used to compare the accuracies of quantitative and semi-quantitative methods. Results: There were 59 patients in group 1 and 128 patients in group 2. The mean age and sex distribution of the two groups were not significantly different. The lesions were primarily located in the subpleural area. Compared to group 1, group 2 had larger values for all volume-dependent parameters (p < 0.001). The percentage of lesions had the strongest correlation with disease severity with a correlation coefficient of 0.495. In comparison, the correlation coefficient of semiquantitative score was 0.349. To classify the severity of COVID-19, area under the curve of the percentage of lesions was the highest (0.807; 95% confidence interval, 0.744–0.861: p < 0.001) and that of the quantitative CT parameters was significantly higher than that of the semiquantitative visual score (p = 0.001). Conclusion: The classification accuracy of quantitative CT parameters was significantly superior to that of semiquantitative visual score in terms of evaluating the severity of COVID-19.

F-box Protein Arabidillo-1 Promotes Lateral Root Development by Depressing the Functioning of GA3 in Arabidopsis

        Changjun Mu,Ni Chen,Xiaofeng Li,Pengfei Jia,Zhaoyan Wang,Heng Liu 한국식물학회 2010 Journal of Plant Biology Vol.54 No.5

In Arabidopsis, Arabidillo-1 and Arabidillo-2have great sequence homology to Dictyostelium and metazoan β–catenin/Armadillo, which are important to animal and Dictyostelium development. Arabidillo-1 and Arabidillo-2 promote lateral root formation redundantly in Arabidopsis. Here, we showed that gibberellins (GA3) has a greater inhibitory effect on lateral root growth from the null mutant arabidillo-1 than from the wild type, suggesting that the mechanism for Arabidillo-1-regulated modulation of lateral root proliferation is associated with GA3-metabolic or signaling pathways. Our yeast two-hybrid analysis demonstrated that Arabidillo-1 interacts with ASK2 and ASK11, and that ASK2 can bind with the F-box domain of Arabidillo-1. Therefore, Arabidillo-1 is involved in the ubiquitin/26S proteasome-mediated proteolytic pathway. Based on these results, we conclude that Arabidillo-1 can degrade some positive regulator of the GA3 signaling pathway through selective protein degradation of ubiquitin/26S. Moreover, that process is believed to be the mechanism for Arabidillo-1 promotion of lateral root development in Arabidopsis.

Synthesis of a Novel Anthraquinone Diamino-Bridged Bis(β-cyclodextrin) and Its Cooperative Binding toward Guest Molecules

Zhao, Yan,Yang, Zi Ming,Chi, Shao Ming,Gu, Juan,Yang, Yong Cun,Huang, Rong,Wang, Bang Jin,Zhu, Hong You Korean Chemical Society 2008 Bulletin of the Korean Chemical Society Vol.29 No.5

A novel anthraquinone diamino-bridged bis($\beta$ -cyclodextrin) 2 was synthesized. The inclusion complexation behaviors of the native $\beta$ -cyclodextrin 1 and the novel bis($\beta$ -cyclodextrin) 2 with guests, such as acridine red (AR), neutral red (NR), ammonium 8-anilino-1-naphthalenesulfonate (ANS), sodium 2-(p-toluidinyl) naphthalenesulfonate (TNS) and rhodamine B (RhB) were investigation by fluorescence, circular dichroism and 2D NMR spectroscopy. The spectral titrations were performed in phosphate buffer (pH 7.20) at 25 ${^{\circ}C}$ to give the complex stability constants (Ks) and Gibbs free energy changes (−${\Delta}G^0$) for the stoichiometric 1:1 inclusion complexation of host 1 and 2 with guests. The results indicated that the novel bis($\beta$ -cyclodextrin) 2 greatly enhanced the original binding affinity of the native $\beta$ -cyclodextrin 1. Typically, bis($\beta$ -cyclodextrin) 2 showed the highest binding constant towards ANS up to 34.8 times higher than that of 1. The 2D NMR spectra of bis($\beta$ -cyclodextrin) 2 with RhB and TNS were performed to confirm the binding mode. The increased binding affinity and molecular selectivity of guests by bis($\beta$ -cyclodextrin) 2 were discussed from the viewpoint of the size/shape-fit concept and multipoint recognition mechanism.
