      • Tax is Involved in Up-regulation of HMGB1 Expression Levels by Interaction with C/EBP

        The high mobility group box 1 (HMGB1) protein is a multifunctional cytokine-like molecule that plays an important role in the pathogenesis of tumors. In this study, real-time polymerase chain reactions and Western blot assays indicated that HMGB1 transcriptional activity and protein level are increased in $Tax^+$-T cells (TaxP). To clarify the mechanisms, a series of HMGB1 deletion reporter plasmids (pHLuc1 to pHLuc6) were transfected into $Tax^-$-T cells (TaxN, Jurkat) and $Tax^+$-T cells (TaxP). We found that promoter activity in $Tax^+$-T cells to be higher than that in $Tax^-$-T cells, indicating a significant increase in pHLuc6. Bay11-7082 (NF-${\kappa}B$ inhibitor) treatment did not block the enhancing effect. Chromatin immunoprecipitation assays revealed that Tax was retained on a HMGB1 promoter fragment encompassing -1163 to -975. Bioinformatics analysis showed six characteristic cis-elements for CdxA, AP-1, AML-1a, USF, v-Myb, and C/EBP in the fragment in question. Mutation of cis-elements for C/EBP reduced significant HMGB1 promoter activity induced by Tax. These findings indicate that Tax enhances the expression of HMGB1 gene at the transcriptional level, possibly by interacting with C/EBP.

        ACCURACY OF LAMOST DR1 STELLAR PARAMETERS

        We adopt the PASTEL catalog combined with SIMBAD radial velocities as a testing standard to validate the stellar parameters (effective temperature $T_{eff}$, surface gravity log g, metallicity [Fe/H] and radial velocity $V_r$) from the first data release (DR1) of The Large Sky Area Multi-Object Fiber Spectroscopic Telescope (LAMOST) survey. After applying data reduction and temperature constraints to the sample obtained by cross-identification, we compare the stellar parameters from DR1 and PASTEL. The results show that the DR1 results are reliable under certain conditions. We derive a dispersion of 110 K, 0.19 dex, 0.11 dex and $4.91kms^{-1}$ in specified effective temperature ranges, for $T_{eff}$, log g, [Fe/H] and $V_r$ respectively. Systematic errors are negligible except for those of $V_r$. In addition, for stars with PASTEL [Fe/H] < -1:5, the metallicities in DR1 are systematically higher than those in PASTEL.

        Microstructure Characteristics, Mechanical Properties and Strain Hardening Behavior of Nitrogen-Bearing Fe-0.2 C-5Mn TRIP Steel

        As a strong and low-priced austenite stabilizer, nitrogen (0.085 wt%) was deliberately added into a hot-rolled Fe-0.2C-5Mn TRIP steel for optimizing the strength–ductility balance. Nitrogen addition can not only increase austenite fraction (> 50%), but also favor the improvement of austenite stability due to its partition behavior from ferrite to austenite during intercritical annealing. Besides, the low annealing temperature (635 °C) and nitrogen addition result in the simultaneous precipitation of cementite (size, 21–396 nm) and carbonitride (size, 7–193 nm) in ferrite and at grain boundaries, which can effectively increase the yield strength through precipitation strengthening (33–63 and 41–61 MPa, respectively). Furthermore, the sufficient interstitial solute originated from N addition as well as the discontinuous TRIP effect benefits the strong serrated flow behavior and multi-peak strain hardening behavior. As a result, the strong TRIP effect together with precipitation strengthening is believed to be responsible for the excellent strength–ductility balance of present steel annealed for 3 h, demonstrating a high tensile strength of 1043 MPa, outstanding total elongation of 43.3% and superior product of strength and elongation (PSE) of 44.8 GPa%.

      • A simulant study of the Network of Five Elements by using the computer

        The theory of Chinese medicine is a based theory to research the paranomarl function of human body including ESP and PK etc. The theory of the traditional Chinese medicine indictes that the life prosesses can be discribed by the system of five elements (SFE), which are: wood, fire, earth, metal and water. We take the SFE for a network consisted of this five elements (NFE), and study the characteries of the structure of this NFE. they include multi-cross character; directiong and irreversible character; mesomerism character; "open" character and Fuzzy character etc. Then, based on these characters, we simulate the partial function of NFE by using computer. The results of simulation show that the NFE is a very useful network to explain the physiological and pathological phenomenan of human body according to the theory of traditional Chinese medicine.

      • Disruption of endothelial barrier function is linked with hyposecretion and lymphocytic infiltration in salivary glands of Sjögren's syndrome

        <P><B>Abstract</B></P> <P>Sjögren's syndrome (SS) is an inflammatory autoimmune disease that causes hyposecretion in salivary glands. Endothelial tight junctions (TJs) play crucial roles in salivation and barrier function of blood vessels. However, whether the alteration of endothelial TJs were involved in pathogenesis of SS was still unknown. Here, the ultrastructure and function of endothelial TJs in submandibular glands (SMGs) were detected by transmission electron microscopy and in vivo paracellular permeability assay in different aged NOD mouse model for SS. CFSE-labeled lymphocytes were injected into tail vein to trace the infiltration, while claudin-5 expression and distribution were detected by immunofluorescence, qRT-PCR, and western blot. Results showed that the stimulated salivary flow rate was gradually decreased and lymphocytic infiltration was found as age increased in 12- and 21-week-old NOD mice, but not 7-week-old NOD mice. Blood vessels were dilated, while endothelial TJ width and paracellular tracer transport were increased in 12-week-old NOD mice. Moreover, the injected CFSE-labeled lymphocytes were observed in SMGs of 12-week-old NOD mice. Claudin-5 level was increased and relocalized from the apical portion of neighboring endothelial cells to lateral membranes and cytoplasm in 12-week-old NOD mice. Additionally, the alteration of claudin-5 expression and distribution was further confirmed in labial salivary glands and bilateral parotid glands from SS patients. In cultured human microvessel endothelial cell line (HMEC-1), IFN-γ stimulation significantly increased claudin-5 expression.         Analysis of three-dimensional thermal gradients for arch bridge girders using long-term monitoring data

        Thermal loads, especially thermal gradients, have a considerable effect on the behaviors of large-scale bridges throughout their lifecycles. Bridge design specifications provide minimal guidance regarding thermal gradients for simple bridge girders and do not consider transversal thermal gradients in wide girder cross-sections. This paper investigates the three-dimensional thermal gradients of arch bridge girders by integrating long-term field monitoring data recorded by a structural health monitoring system, with emphasis on the vertical and transversal thermal gradients of wide concrete-steel composite girders. Based on field monitoring data for one year, the time-dependent characteristics of temperature and three-dimensional thermal gradients in girder cross-sections are explored. A statistical analysis of thermal gradients is conducted, and the probability density functions of transversal and vertical thermal gradients are estimated. The extreme thermal gradients are predicted with a specific return period by employing an extreme value analysis, and the profiles of the vertical thermal gradient are established for bridge design. The transversal and vertical thermal gradients are developed to help engineers understand the thermal behaviors of concrete-steel composite girders during their service periods.

        Optimal sensor placement under uncertainties using a nondirective movement glowworm swarm optimization algorithm

        Optimal sensor placement (OSP) is a critical issue in construction and implementation of a sophisticated structural health monitoring (SHM) system. The uncertainties in the identified structural parameters based on the measured data may dramatically reduce the reliability of the condition evaluation results. In this paper, the information entropy, which provides an uncertainty metric for the identified structural parameters, is adopted as the performance measure for a sensor configuration, and the OSP problem is formulated as the multi-objective optimization problem of extracting the Pareto optimal sensor configurations that simultaneously minimize the appropriately defined information entropy indices. The nondirective movement glowworm swarm optimization (NMGSO) algorithm (based on the basic glowworm swarm optimization (GSO) algorithm) is proposed for identifying the effective Pareto optimal sensor configurations. The one-dimensional binary coding system is introduced to code the glowworms instead of the real vector coding method. The Hamming distance is employed to describe the divergence of different glowworms. The luciferin level of the glowworm is defined as a function of the rank value (RV) and the crowding distance (CD), which are deduced by non-dominated sorting. In addition, nondirective movement is developed to relocate the glowworms. A numerical simulation of a long-span suspension bridge is performed to demonstrate the effectiveness of the NMGSO algorithm. The results indicate that the NMGSO algorithm is capable of capturing the Pareto optimal sensor configurations with high accuracy and efficiency.

        A monophosphonic group-functionalized ion-imprinted polymer for a removal of Fe3+ from highly concentrated basic chromium sulfate solution

        An ion-imprinted polymer (IIP) with monophosphonic groups was prepared by thermal copolymerization. Bis(2-methacryloxyethyl) phosphate (BMAOP) was used as functional monomer to react with Fe3+ in dimethyl sulfoxide (DMSO). Ethylene glycol dimethacrylate (EGDMA) was used as cross-linker during polymerization process. A suitable molar ratio of BMAOP to Fe3+ was investigated by UV/Vis/NIR Spectrometer and ICP-OES. The obtained results showed that the monophosphonic groups could be selectively combined with Fe3+ in solutions containing other coexisting ions, and the selectivity could be further enhanced by ion-imprinted process. The prepared IIP was used for removing trace Fe3+ from high concentration basic chromium sulfate solutions. After adsorption process, the concentration of Fe3+ could be reduced from 4.486mg L1 to 0.171mg L1, which was much lower than the concentration in the solution treated by non-imprinted polymer (NIP). Moreover, the IIP exhibited excellent recyclability after six adsorption-desorption cycles.
