        Isolation and characterization of a novel gossypol-degrading bacteria Bacillus subtilis strain Rumen Bacillus Subtilis

        Objective: The aim of the study was to isolate gossypol-degrading bacteria and to assess its potential for gossypol degradation. Methods: Rumen liquid was collected from fistulated cows grazing the experimental pasture. Approximately 1 mL of the rumen liquid was spread onto basal medium plates containing 2 g/L gossypol as the only source of carbon and was then cultured at 39°C to isolate gossypol-degrading bacteria. The isolated colonies were cultured for 6 h and then their size and shape observed by microscope and scanning electron microscope. The 16S rRNA gene of isolated colonies was sequenced and aligned using National Center for Biotechnology Information-Basic Local Alignment Search Tool. The various fermentation conditions, initial pH, incubation temperature, inoculum level and fermentationperiod were analyzed in cottonseed meal (CSM). The crude protein (CP), total gossypol (TG), and free gossypol (FG) were determined in CSM after fermentation with isolated strain at 39°C for 72 h. Results: Screening results showed that a single bacterial isolate, named Rumen Bacillus Subtilis (RBS), could use gossypol as a carbon source. The bacterium was identified by 16S rDNA sequencing as being 98% homologous to the sequence of Bacillus subtilis strain GH38. The optimum fermentation conditions were found to be 72 h, 39°C, pH 6.5, moisture 50%, inoculum level 107 cell/g. In the optimum fermentation conditions, the FG and TG content in fermented CSM decreased 78.86% and 49% relative to the control. The content of CP and the essential amino acids of the fermented CSM increased respectively, compared with the control. Conclusion: The isolation of a gossypol-degrading bacterium from the cow rumen is of great importance for gossypol biodegradation and may be a valuable potential source for gossypol-degradation of CSM.

        Expression of Arabidopsis HOMEODOMAIN GLABROUS 11 Enhances Tolerance to Drought Stress in Transgenic Sweet Potato Plants

        Sweet potato (Ipomoea batatas L. cv. Lizixiang)is a nutritious arable crop with a low drought tolerance during growth and maturation. The Arabidopsis HOMEODOMAIN GLABROUS 11 (HDG11) gene can increase drought tolerance in tobacco and tall fescue plants. To determine the effect of HDG11 in the sweet potato, transgenic plants that expressed the HDG11 gene were generated by Agrobacterium-mediated transformation. Expression of the transgene was confirmed using Southern blotting, reverse transcription-polymerase chain reaction (RT-PCR) and DNA sequencing. Two independent HDG11 transgenic lines were evaluated and increased drought stress tolerance was observed in both lines, compared to wild-type (WT) plants. Under drought stress conditions, net photosynthesis rate (Pn), the efficiency of excitation energy captured by open PSII reaction centers (Fv/Fm) and water use efficiency (WUE) increased, and transpiration rate (Tr) decreased in HDG11 transgenic plants compared to WT. HDG11 transgenic plants also had decreased lipid membrane oxidative damage, reduced H2O2 accumulation and increased ROSscavenging enzyme activity during drought stress treatment. This study indicates that overexpression of the Arabidopsis HDG11 gene improved drought tolerance in the sweet potato.

      • 3D Model Compression For Collaborative Design

        The compression of CAD models is a key technology for realizing Internet-based collaborative product development because big model sizes often prohibit us to achieve a rapid product information transmission. Although there exist some algorithms for compressing discrete CAD models, original precise CAD models are focused on in this paper. Here, the characteristics of hierarchical structures in CAD models and the distribution of their redundant data are exploited for developing a novel data encoding method. In the method, different encoding rules are applied to different types of data. Geometric data is a major concern for reducing model sizes. For geometric data, the control points of B-spline curves and surfaces are compressed with the second-order predictions in a local coordinate system. Based on analysis to the distortion induced by quantization, an efficient method for computation of the distortion is provided. The results indicate that the data size of CAD models can be decreased efficiently after compressed with the proposed method.

        Efficient Geographical Information-Based En-route Filtering Scheme in Wireless Sensor Networks

        The existing en-route filtering schemes only consider some simple false data injection attacks, which results in lower safety performance. In this paper, we propose an efficient geographical information-based en-route filtering scheme (EGEFS), in which each forwarding node verifies not only the message authentication codes (MACs), but also the report identifier and the legitimacy and authenticity of locations carried in a data report. Thus, EGEFS can defend against not only the simple false data injection attacks and the replay attack, but also the collusion attack with forged locations proposed in this paper. In addition, we propose a new method for electing the center-of-stimulus (CoS) node, which can ensure that only one detecting node will be elected as the CoS node to generate one data report for an event. The simulation results show that, compared to the existing en-route filtering schemes, EGEFS has higher safety performance, because it can resist more types of false data injection attacks, and it also has higher filtering efficiency and lower energy expenditure.

        A Mild Strategy to Strengthen Three Dimensional Graphene Aerogel for Supporting Sulfur as a Free‐standing Cathode in Lithium–Sulfur Batteries

        Recently, three dimensional graphene aerogel (3DGA) supported sulfur microparticles was used as a cathode material for lithium?sulfur batteries, which was considered as one of the most promising next generation rechargeable batteries due to its ultra?high theoretical specific capacity (1675 mAh/g). However, the mechanical strength of 3DGA remains an issue for further application. Herein, a strengthened 3DGA (S3DGA) was achieved by soaking in a low concentration ammonia solution at a relative low temperature. Then the S3DGA loaded sulfur (S3DGA?S) was cut into a round piece and directly used as a cathode without additional binders or conductive additives in Li?S batteries. The mechanical strength, microstructure, and electrochemical properties were investigated by compare with a 3DGA prepared without strengthen. The S3DGA?S presented good mechanical strength, excellent capacity retention, and lower electrochemical impedance.

        Phenolic features and anthocyanin profiles in winemaking pomace and fresh berries of grapes with different pedigrees

        The total contents and antioxidant activities of phenolic compounds as well as anthocyanin profiles were analyzed and compared in fresh berries and fermented pomace of three grape cultivars with different pedigrees. The phenolic contents and antioxidant activities decreased significantly in skins (p < 0.05), while relatively large amounts of them were retained in seeds after fermentative maceration. Fermentative maceration also had a significant impact on the anthocyanin compositions. The proportions of anthocyanins with more stable structures, such as malvidin derivatives, methylated, diglucosides and nonacylated anthocyanins, increased significantly in the pomace skins (p < 0.05). There were obvious differences in phenolic features and anthocyanin profiles among the three cultivars. ‘NW196’, a wine hybrid of Vitis vinifera and V. quinquangularis, was characterized by the highest total anthocyanin contents and degree of diglucosylation. The results obtained in this study could contribute to the primary data for the development and utilization of winemaking pomace, especially from local non-Vitis vinifera grapes.
