Three new compounds named penicitor A,aculene E and penicitor B, as well as four known compounds,were isolated from the fermentation broth ofPenicillium sp. SCS-KFD08 associated with a marineanimal Sipunculus nudus from the Haikou bay of China. Their planar structures and absolute configurations wereunambiguously elucidated by spectroscopic data, Mosher’smethod, CD spectrum analysis along with quantumECD calculation. Among them, compounds 2–7 showedquorum sensing inhibitory activity against Chromobacteriumviolaceum CV026, and could significantly reduceviolacein production in N-hexanoyl-l-homoserine lactone(C6-HSL) induced C. violaceum CV026 cultures at subinhibitoryconcentrations.
Synthesis and Cytotoxicity of Novel 3-Amino-4-indolylmaleimide Derivatives

In an attempt to develop potent and selective antitumor agents, a series of novel 3-amino-4-indolylmaleimides were designed and synthesized. The reaction showed high regioselectivity. The structure of compound 7a was determined by an X-ray single crystal diffraction method. The cytotoxicities of the title compounds were evaluated against HeLa, SMMC 7721 and HL 60 cancer cell lines by a standard MTT assay in vitro. The pharmacological results showed that some of the title compounds displayed moderate or high cytotoxic activity against the tested cell lines. Compound 7d was the most promising compound against the tested cancer cell lines. Structure-activity relationships are discussed based on the experimental data obtained. A hydroxyethylamino group at the 3-position in the side chain of indolylmaleimide is associated with an increase in cytotoxicity.

In this study, we report the identification and characterization of a novel C2H2 zinc finger protein, ZNF552, from a human embryonic heart cDNA library. ZNF552 is composed of three exons and two introns and maps to chromosome 19q13.43. The cDNA of ZNF552 is 2.3 kb, encoding 407 amino acids with an amino-terminal KRAB domain and seven carboxyl-terminal C2H2 zinc finger motifs in the nucleus and cytoplasm. Northern blotting analysis indicated that a 2.3 kb transcript specific for ZNF552 was expressed in liver, lung, spleen, testis and kidney, especially with a higher level in the lung and testis in human adult tissues. Reporter gene assays showed that ZNF552 was a transcriptional repressor, and overexpression of ZNF552 in the COS-7 cells inhibited the transcriptional activities of AP-1 and SRE, which could be relieved through RNAi analysis. Deletion studies showed that the KRAB domain of ZNF552 may be involved in this inhibition. [BMB reports 2010; 43(3): 193-198]

Flavonoid glycosides are metabolized byintestinal bacteria, giving rise to a wide range of phenolicacids that may exert systemic effects in the body. Themicrobial metabolism of flavonoids extracted from theleaves of Diospyros kaki (FLDK) by intestinal bacteria wasinvestigated in vitro. High-performance liquid chromatography/linear trap quadrupole orbitrap mass spectrometrywas performed to analyze the metabolites of flavonoidsin vivo using Xcalibur2.1 software. The results showed thatthe levels of flavonoid glycosides and flavonoid aglyconesdecreased rapidly in the process of microbial metabolismby intestinal bacteria in vitro, and the metabolic rate maybe related to the concentration of intestinal bacteria in theculture solution. In vivo metabolites of FLDK weredetected in rat plasma and urine after oral administration ofFLDK. Eight flavonoids were identified in the urine, andthree were identified in the plasma; however, flavonoidaglycones were not found in the plasma.
Profiling Gene Expression During Gland Morphogenesis of a Glanded and a Glandless Upland Cotton

The pigment gland is an important character of the Gossypium plant. With the aim of identifying genes involved in pigment gland morphogenesis in cotton, gene expression during pigment gland morphogenesis in Chuan 2802, which is glanded both in seed and plant, and a glandless line N5 was profiled using Affymetrix Cotton microarray. The results showed that there were 564 differentially expressed genes greater than twofold during gland morphogenesis. About 60.2% of these genes shares similarity with known genes on GenBank and about 39.8% with no functional description in the database. These described genes may play roles in defense response, response to oxidative stress, peroxidase activity, and the other metabolic pathways. The KEGG Orthology-Based Annotation System indicated that these above twofold expressed genes involved seven biochemical pathways on KEGG. These findings suggest that a complicated regulation is associated with pigment gland morphogenesis and the associated defense response including gossypol biosynthesis in cotton.

Background: Although Becker`s nevus (BN) is a relatively common disease, the systematic studies of clinicopathological and immunohistochemical results are poorly reported. Objective: To investigate the clinicopathological features and immunohistochemical alterations of keratinocyte proliferation, melanocyte density, smooth muscle hyperplasia and nerve fiber distribution in BN. Methods: Clinical and pathological data were collected in 60 newly-diagnosed BN cases. Immunohistochemical stain of Ki-67, Melan-A, keratin 15, smooth muscle actin and protein gene product 9.5 was performed in 21 cases. Results: The median diagnostic and onset age was 17 and 12 years, respectively. Skin lesions usually appeared on the upper trunk and upper limbs. The pathological features included the rete ridge elongation and fusion and basal hyperpigmentation. Epidermal Ki-67, Melan-A and keratin 15 expression and dermal nerve fiber length were significantly higher in lesional and perilesional skin than in normal skin (p<0.05∼0.01), while smooth muscle actin expression was upregulated only in skin lesion (p<0.05). Conclusion: Although the clinical diagnosis of BN is often straightforward, histopathology is helpful to differentiate from other pigmentary disorders. The hyperproliferation of keratinocytes, melanocytes, arrector pili muscle and dermal nerve fibers could be involved in the patho-genesis of BN. (Ann Dermatol 28(6) 697∼703, 2016)

A cDNA encoding a novel cysteine proteinase inhibitor (CPI) was isolated from a gland mutant Xiangmian-18 of upland cotton during the pigments gland forming stage. The cDNA comprises 378 bp and encodes 125 amino acid residues with molecular mass of 13.8 kDa. It contains the conserved motif of cysteine protease inhibitors and belongs to the cystatin superfamily (Gln- Val-Val-Ala-Gly). The deduced amino acid sequences of the domains are highly similar to the normal upland cotton (96.8%). SDS-PAGE and western hybridization analysis showed that the expressed recombinant protein was recombinant CPI. The inhibitory activity of recombinant CPI was 46 u/μg which was measured by inhibiting the protease activity of papain. RT-PCR results indicated that the expression level of developing gland stage was higher than that of undeveloped gland stage.

This paper addresses the formation tracking control problem of multiple underactuated unmanned surface vehicles. Considering many actual situations, a practical formation control scheme, which is performed by using a leader-follower approach, minimum learning parameter technique, adaptive technology and so on. Firstly, a virtual unmanned surface vehicle is designed according to the location information of the leader unmanned surface vehicle to estimate the leader’s speed information while reducing the communication bandwidth. Secondly, a formationcontrol law is designed to make the follower underactuated unmanned surface vehicles track the leader. Unknown dynamics and external disturbances are regarded as a whole and compensated by the minimum learning parameter technique instead of multi-layer neural network and the neural shunt model can handle multiple derivation problems of virtual control laws. Meanwhile, the robustness of the controlled system is improved through adaptive technology. Besides, an auxiliary design system is employed to constrain the output range of the control law. Finally, numerical simulations are implemented to prove the feasibility of the formation tracking control strategy.

This paper addresses two interrelated problems concerning the tracking control of podded propulsion unmanned surface vehicle (USV), namely, the modeling of podded propulsion USV, and trajectory tracking controller design. Based on the force analysis, the separation model of podded propulsion USV is established. Furthermore, a practical adaptive neural tracking controller is proposed by backstepping technique, neural network minimum parameter learning method, neural shunting model and auxiliary dynamic system without the exact information of hydrodynamic damping structure and the sea disturbances. Using Lyapunov stability analysis theory, it is proven that all error signals in the system are uniformly ultimately bounded. The advantages of the paper are that first, the underactuated characteristic of podded propulsion USV is demonstrated; then, neural shunting model and neural network minimum parameter learning method are introduced to cope with the problem of "explosion of complexity" and uncertainty factors, respectively; third, auxiliary dynamic system is introduced into controller design to reduce the risk of actuator saturation. Taking into account the above practical problems is helpful to engineering implementation in the marine practice. Finally, numerical simulation has been given to demonstrate the effectiveness of the proposed scheme.

This paper investigates the path following control problem for underactuated unmanned surface vehicle(USV) in the presence of unmodeled dynamics, external disturbances and input saturation. A novel adaptive robustpath following control scheme is proposed by employing trajectory linearization control (TLC) technology andfinite-time disturbance observer, which is composed of a concise yaw rate controller and a surge speed controller. The salient features of the proposed scheme include: a path following guidance law is designed to ensure USVeffectively converging to and following the desired path; TLC is introduced into the field of USV motion controlas new effective technique, and it is the first time used to design path following controller for underactuated USV;a finite-time nonlinear tracking differentiator is constructed not only to avoid the signal jump caused by derivation,but also to filter noise and high frequency interference. A finite-time disturbance observer (FDO) is devised toexactly observe the uncertain dynamics and unknown external disturbances, which improves the tracking accuracyand precise disturbance rejection of the proposed controller; then, an auxiliary dynamic system that is governed bysmooth switching function is developed to compensate for the saturation constraint on actuator. Stability analysisverifies that all signals in the closed-loop system are uniformly ultimately bounded. Finally, simulation results andcomparisons illustrate the superiority of the proposed control scheme.
