      • Association Study of Single-Nucleotide Polymorphisms of STAT2/STAT3/IFN-γ Genes in Cervical Cancer in Southern Chinese Han Women

        Objective: Interferon-${\gamma}$ (IFN-${\gamma}$) and signal transducers and activators of transcription (STATs) each play an important role in carcinogenesis associated with viral infection. Cervical cancer is almost invariably associated with infection by human papillomavirus (HPV), and previous studies suggested that dysregulation of the signal pathway involved in IFN-${\gamma}$ and STATs is associated. Our objective was to evaluate the association of SNPs in STAT2, STAT3, and IFN-${\gamma}$ with cervical cancer susceptibility in Chinese Han women in Hunan province. Materials and Methods: Genomic DNA was extracted from peripheral blood samples of 234 cervical cancer patients and 216 healthy female controls. STAT2 and STAT3 genotyping was performed using polymerase chain reaction-restriction enzyme (PCR-RE) analysis. IFN-${\gamma}$ genotyping was detected by PCR-amplification of specific allele (PASA). Results: For STAT2 rs2066807 polymorphisms, there was no significant difference of genotype distribution (P=0.827) and allele frequencies (P=0.830, OR=1.09, 95% CI: 0.51-2.31) between cases and controls. For STAT3 rs957970 polymorphisms, there was no significant difference of genotype distribution (P=0.455) and allele frequencies (P=0.560, OR=0.92, 95% CI: 0.71-1.20) between cases and controls. For IFN-${\gamma}$ +874A/T polymorphisms, there was no significant difference of genotype distribution (P=0.652) and allele frequencies (P=0.527, OR=1.12, 95% CI: 0.79-1.59) between cases and controls. Conclusion: These results suggest that polymorphisms in STAT2, STAT3 and IFN-${\gamma}$ genes are not likely to be strong predictors of cervical cancer in Han women in southern China.

        Correlations Between Fasciology and Yin Yang Doctrine

        The aim of this study is to explore the correlations between fasciology and yin yang doctrine. Professor Yuan developed fasciology by three-dimensional reconstruction of connective tissue (fascia) in the trunk and limbs of the human body and tracing back to tissue origins in light of biological evolution and developmental biology. Fasciology states that the human body can be divided into two systems: the supporting-storing system and the functional system. This article elaborates on the roles of the two systems and their mutual relationship. The two systems are used to analyze the yin,the yang, and their relationship. The two systems are promoted but also restricted in different contexts. The supporting-storing system is formed by undifferentiated connective tissue and provides undifferentiated cells and nutrients for differentiated cells of the functional system. Thus, the supporting-storing system could be classified as quiet, similar to yin. The functional system continuously maintains the various functional activities of the human body. Thus, the functional system could be classified as active, similar to yang. In interpreting the yin yang doctrine from the point of view of fasciology, yin can be compared with the supporting-storing system and yang can be compared with the functional system.

      • Expression Profile and Potential Roles of EVA1A in Normal and Neoplastic Pancreatic Tissues

        Background: EVA1A (eva-1 homolog A) is a novel gene that regulates programmed cell death through autophagy and apoptosis. Our objective was to investigate the expression profiles and potential role of EVA1A in normal and neoplastic human pancreatic tissues. Materials and Methods: The expression pattern of EVA1A in normal pancreatic tissue was examined by indirect immunofluorescence and confocal microscopy. Protein levels in paraffin-embedded specimens from normal and diseased pancreatic and matched non-tumor tissues were evaluated by immunohistochemistry. Results: EVA1A colocalized with glucagon but not with insulin, demonstrating production in islet alpha cells. Itwas strongly expressed in chronic pancreatitis, moderately or weakly expressed in the plasma membrane and cytoplasm in pancreatic acinar cell carcinoma, and absent in normal pancreatic acinar cells. Although the tissue architecture was deformed, EVA1A was absent in the alpha cells of pancreatic ductal adenocarcinomas, intraductal papillary mucinous neoplasms, mucinous cystadenomas, solid papillary tumors and pancreatic neuroendocrine tumors. Conclusions: EVA1A protein is specifically expressed in islet alpha cells, suggesting it may play an important role in regulating alpha-cell function. The ectopic expression of EVA1A in pancreatic neoplasms may contribute to their pathogenesis and warrants further investigation.

        Improvement of Fermentation and Nutritive Quality of Straw-grass Silage by Inclusion of Wet Hulless-barley Distillers' Grains in Tibet

        In order to develop methods that would enlarge the feed resources in Tibet, mixtures of hulless-barley straw and tall fescue were ensiled with four levels (0, 10%, 20%, and 30% of fresh weight) of wet hulless-barley distillers' grains (WHDG). The silos were opened after 7, 14 or 30 d of ensiling, and the fermentation characteristics and nutritive quality of the silages were analyzed. WHDG addition significantly improved fermentation quality, as indicated by the faster decline of pH, rapid accumulation of lactic acid (LA) (p<0.05), and lower butyric acid content and ammonia-N/total N (p<0.05) as compared with the control. These results indicated that WHDG additions not only effectively inhibited the activity of aerobic bacteria, but also resulted in faster and greatly enhanced LA production and pH value decline, which restricted activity of undesirable bacteria, resulting in more residual water soluble carbohydrates (WSC) in the silages. The protein content of WHDG-containing silages were significantly higher (p<0.05) higher than that of the control. In conclusion, the addition of WHDG increased the fermentation and nutritive quality of straw-grass silage, and this effect was more marked when the inclusion rate of WHDG was greater than 20%.

      • Expression of Osteopontin in Non-small Cell Lung Cancer and Correlative Relation with Microvascular Density

        Background and Objective: Lung cancer is one of the malignant diseases which most seriously threat humansurvival and development. This study aimed to assess osteopontin (OPN) expression in non-small cell lung cancer (NSCLC) and any relationship with clinicopathological features. Methods: Immunohistochemistry was used to determine OPN expression and microvascular density (MVD) in 120 cases of NSCLC also undergoing clinical assessment. Results: Moderately positive expression of OPN was found in 34.6% (41/120) and strong expression in 47.5% (57/120) of the NSCLCs; OPN expression in carcinomas was higher than in pericarcinoma tissues (P<0.05). While no obvious association was observed with NSCLC patient age, gender, maximum diameter of the tumor and pathological type, OPN expression was more commonly detected in poorly differentiated carcinoma tissue and lymph node metastasis as well as at advanced clinical stage (P<0.05); OPN expression in cancer tissue was positively correlated with MVD (r = 0.839, P = 0.000). Conclusion: OPN plays an important role in promoting tumor angiogenesis and progress of NSCLCs and has the possibility to become the new target for therapy.

      • A Novel Negative Selection Algorithm for Recognition Problems

        In this paper, a novel negative selection algorithm for recognition problems was given. Compared with the traditional negative selection algorithm, a co-stimulation signal was added to start the detectors, which a key factor in immune response. Co-stimulation signal was calculated by the techniques of the statistics and the sliding window, which not only reduced time complexity of algorithm but also improved accuracy of the algorithm. Entropy was adopted to evaluate the density of detectors for optimizing the coverage of nonself area. Experiment results proved high accuracy and efficiency of the proposed algorithm.

        Stretching Deformation Mechanism of Polyacrylonitrile-based Carbon Fiber Structure at High Temperatures

        In a high-temperature environment, polyacrylonitrile-based carbon fiber (PAN-CF) can be deformed by stretching, where the stretching deformation ability of PAN-CF is enhanced with the increase of the temperature. Further, the hightemperature stretching deformation of PAN-CF directly affects the control of the carbon crystalline orientation. Based on the techniques of high-resolution transmission electron microscopy (HRTEM), Raman spectroscopy, X-ray diffraction and in situ tension testing, the variation regularity and the intrinsic mechanism of high-temperature stretching deformation ability of PAN-CF obtained at different preparation temperatures were systematically studied in a high-temperature environment. The results indicated that the essence of PAN-CF high-temperature deformation was the relative motion of the carbon crystallite. Further, the main structural parameters that affected the high-temperature stretching deformation ability of PAN-CF were the degree of cross-linking between the carbon crystallites, the orientation angle(OA) of the carbon crystallite and the nitrogen content. When the testing temperature was lower than the preparation temperature, only physical structure changes were observed in the PAN-CF. For the PAN-CF tested undergoing physical structure changes, as the degree of cross-linking between the crystallites and the orientation angle decreased, the slipping of crystallites became easier. In the same environment, as the stretching tension decreased, the stretching deformation ability improved. When PAN-CF was tested under temperatures higher than the preparation temperature, the microcrystalline cross-linking in the PAN-CF was prone to fracture and slipping, and the high-temperature stretching deformation ability was enhanced. Also, for PAN-CF of lower preparation temperatures in PAN-CF containing no nitrogen (i.e., <0.15 wt%), the cross-linkages increased and the structures were more unstable, inducing an increase in the fracture of weak bonds and a reduction of the stretching tension. For nitrogen-containing PAN-CF, the removal of nitrogen led to severe shrinkage in the graphite layer and interlayer, and the fiber tension was thus increased, causing the high-temperature stretching deformation ability of the PAN-CF with less nitrogen content to be improved.

        A novel slippery liquid infused porous aluminum conductor with excellent corrosion resistance in simulated marine atmosphere

        The overhead aluminum conductors are prone to corrosion, especially in areas with marine climates,which interferes with the safe operation of the power system. Herein, slippery liquid infused porous surface(SLIPS) conductors were successfully prepared on pure aluminum (Bare) conductors by anodizingand infusion of silicone oil. The corrosion resistance of the conductors during 30 days of salt spray corrosionand 7 days of energized conditions was studied. The results indicated that the corrosion of theBare and anodic aluminum oxide (AAO) conductors was more severe under energized (alternating current(AC) and direct current (DC)) conditions. The exacerbation of corrosion by DC energization wasremarkably higher than that of AC energization. The AAO conductor showed better corrosion resistancethan the Bare conductor, which was attributed to the porous alumina retarding the medium intrusion. Due to the barrier effect provided by the lubricant layer, the SLIPS conductor always presented excellentcorrosion resistance than the Bare and AAO conductors. In addition, the mechanical and electrical propertiesof the SLIPS conductor were comparable to the Bare conductor and could satisfy the basic requirementsfor practical applications. The prepared SLIPS conductors offer great application prospects for thecorrosion protection of aluminum conductors.

        Effects of a traditional Chinese medicine formula and its extraction on muscle fiber characteristics in finishing pigs, porcine cell proliferation and isoforms of myosin heavy chain gene expression in myocytes

        Objective: This study evaluated the effects of a traditional Chinese medicine formula (TCMF) on muscle fiber characteristics in finishing pigs and the effects of the formula's extract (distilled water, ethyl acetate and petroleum ether extraction) on porcine cell proliferation and isoforms of myosin heavy chain (MyHC) gene expression in myocytes. Methods: In a completely randomized design, ninety pigs were assigned to three diets with five replications per treatment and six pigs per pen. The diets included the basal diet (control group), TCMF1 (basal diet+2.5 g/kg TCMF) and TCMF2 (basal diet+5 g/kg TCMF). The psoas major muscle was obtained from pigs at the end of the experiment. Muscle fiber characteristics in the psoas major muscle were analyzed using myosin ATPase staining. Cell proliferation was measured using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) dye and cytometry. Isoforms of MyHC gene expression were detected by real-time quantitative polymerase chain reaction. Results: The final body weight and carcass weight of finishing pigs were increased by TCMF1 (p<0.05), while the psoas major muscle cross-sectional area was increased by TCMF (p<0.05). The cross-sectional area and diameter of psoas major muscle fiber Ι, IIA, and IIB were increased by TCMF2 (p<0.05). The cross-sectional area and fiber diameter of psoas major muscle fiber IIA and IIB were increased by diet supplementation with TCMF1 (p<0.05). Psoas major muscle fiber IIA and IIB fiber density from the pigs fed the TCMF1 diet and the type IIB fiber density from the pigs fed the TCMF2 diet were lower compared to pigs fed the control diet (p<0.05). Pigs fed TCMF2 had a higher composition of type Ι fiber and a lower percentage of type IIB fiber in the psoas major muscle (p<0.05). The expression levels of MyHC Ι, MyHC IIa, and MyHC IIx mRNA increased and the amount of MyHC IIb mRNA decreased in the psoas major muscle from TCMF2, whereas MyHC Ι and MyHC IIx mRNA increased in the psoas major muscle from TCMF1 (p<0.05). Peroxisome proliferator-activated receptor ${\gamma}$ $coactivator-1{\alpha}$ and CaN mRNA expression in the psoas major muscle were up-regulated by TCMF (p<0.05). Porcine skeletal muscle satellite cell proliferation was promoted by $4{\mu}g/mL$ and $20{\mu}g/mL$ TCMF water extraction (p<0.05). Both $1{\mu}g/mL$ and $5{\mu}g/mL$ of TCMF water extraction increased MyHC IIa, MyHC IIb, and MyHC IIx mRNA expression in porcine myocytes (p<0.05), while MyHC Ι mRNA expression in porcine myocytes was decreased by $5{\mu}g/mL$ TCMF water extraction (p<0.05). Porcine myocyte MyHC Ι and MyHC IIx mRNA expression were increased, and MyHC IIa and MyHC IIb mRNA expression were down-regulated by $5{\mu}g/mL$ TCMF ethyl acetate extraction (p<0.05). MyHC Ι and MyHC IIa mRNA expression in porcine myocytes were increased, and the MyHC IIb mRNA expression was decreased by $1{\mu}g/mL$ TCMF ethyl acetate extraction (p<0.05). Four isoforms of MyHC mRNA expression in porcine myocytes were reduced by $5{\mu}g/mL$ TCMF petroleum ether extraction (p<0.05). MyHC IIa mRNA expression in porcine myocytes increased and MyHC IIb mRNA expression decreased by $1{\mu}g/mL$ in a TCMF petroleum ether extraction (p<0.05). Conclusion: These results indicated that TCMF amplified the psoas major muscle cross-sectional area through changing muscle fiber characteristics in finishing pigs. This effect was confirmed as TCMF extraction promoted porcine cell proliferation and affected isoforms of MyHC gene expression in myocytes.

        Flexural performance of fire damaged and rehabilitated two span reinforced concrete slabs and beams

        Five two-span reinforced concrete (RC) slabs and seven two-span RC beams were tested under the ISO 834 standard fire with different durations. CFRP strengthening was then applied to some of the specimens after the damaged concrete was removed from the specimens and replaced with polymer mortar. All the specimens were loaded to failure to investigate the influence of fire-damage and the effectiveness of strengthening methods. Test results indicated that the flexural capacities of specimens decrease with the fire duration increases. Moreover, fire exposure had more significant effect on the flexural rigidity than on the bearing capacity of the specimens. After rehabilitation, the bearing capacities of specimens reached or even exceeded that of the reference RC specimen, and the strengthening methods seemed to have limited effect on flexural rigidity recovery. From the analysis of moment redistribution of tested beams, elevated temperature is found having different impacts on sagging moment region and hogging moment region. The damage of RC continuous member is definitely a comprehensive response of different regions.
