Polo-like kinase 1 (Plk1) plays crucial roles in various stages of oocyte maturation. Recently, we demonstrated that synthesized peptidomimetics AB103-8, targeting the polo- box domain (PBD) of Plk1 disrupted oocyte meiotic maturation and meiosis resumption. However, as a peptidic character of AB103-8 appeared to be defective due to poor membrane permeability and protease stability. To overcome the drawbacks, we designed and synthesized a series of pyrrole-based small-molecule inhibitors and screened against porcine oocyte maturation rates. Among the synthesized compounds, the macrocyclic inhibitor compound 4 displayed the highest inhibition potential against the oocyte meiotic maturation and embryos blastocyst formation. Furthermore, the addition of this compound to culture medium efficiently blocked the maturation of porcine and mouse oocytes, indicating that the lead compound could penetrate zona pellucida and cell membrane. We also investigated the Plk1 inhibiting ability of this compound. Treated mouse oocytes confirmed the Plk1 inhibition by showing abnormal spindle formation. Collectively, these results suggest that the novel Pyrrole-based compounds could be used for the basis of developing contraceptive agents.
Necessity of adjuvant concurrent chemo-radiotherapy in D2-resected LN-positive gastric cancer

<P><B>Abstract</B></P> <P><B>Background and purpose</B></P> <P>To investigate the role of adjuvant concurrent chemo-radiotherapy (CCRT) by analyzing the outcomes of adjuvant CCRT versus chemotherapy alone (CA) in patients with D2-resected gastric cancer with lymph node (LN) metastasis.</P> <P><B>Materials and methods</B></P> <P>Patients with gastric cancer from the institutional registry who underwent curative D2 and R0 resection from December 2004 to January 2013 followed by adjuvant CCRT or CA and demonstrated pathologically confirmed LN metastasis without distant metastasis were included in the study.</P> <P><B>Results</B></P> <P>A total of 1633 patients were included (909 patients in the adjuvant CCRT group and 724 patients in the CA group), and median follow-up was 65.4 months (range, 3.9–141.7 months). There was a significant difference in age (<I>p</I> < 0.0001), Lauren’s classification (<I>p</I> = 0.02), number of LN metastases (<I>p</I> < 0.0001), and pN stage (<I>p</I> < 0.0001) between the CCRT and CA groups.</P> <P>During follow-up, recurrence was detected in 419 (25.7%) of patients overall, 236 (26.0%) in the CCRT group, and 183 (25.3%) in the CA group. Recurrence-free survival (RFS) was not significantly different between the CCRT and CA groups in univariable analysis (<I>p</I> = 0.92). After adjustment, pT/pN stage and perineural invasion showed statistical significance in multivariable Cox regression analysis; however, RFS was significantly higher in the CCRT group (<I>p</I> = 0.03, hazard ratio 0.801, 95% confidence interval 0.658–0.975).</P> <P><B>Conclusions</B></P> <P>The adjusted RFS was significantly higher in the CCRT group than the CA group in patients with D2 resected LN metastatic gastric cancer.</P>

Gate Driving Circuit With Active Pull-Down Function for a Solid-State Pulsed Power Modulator

<P>This paper proposes a new gate driving circuit for the solid-state pulsed power modulator (SSPPM) based on semiconductor devices. The proposed circuit can be easily implemented by applying a short gate signal to the bypass insulated gate bipolar transistor (IGBT), i.e., switches connected in parallel with the load instead of a pull-down resistor. The IGBT_BPs are turned ON at the end-point of the pulse. It can quickly discharge the pulse voltage applied to the load. In addition, it can operate as bypass diode, which is used to protect the semiconductor switches when the gate signal malfunctions, by using the antiparallel diode of the IGBT. Moreover, power loss in the pull-down resistor used to achieve fast falling pulses can be eliminated by turning OFF the IGBT while applying the pulse state. Thus, the proposed circuit can be used for implementing SSPPM with small values of falling time and high efficiency. The circuit configuration, operational principle, relevant analysis results, and a PSpice modeling are presented and verified by the 10-kV SSPPM.</P>

<P>The kinetics and mechanism by which tacrine is distributed in the rat brain were examined. Tacrine levels in plasma and striatal extracellular fluid were used to evaluate the pharmacokinetics of this process. The K(D,brain) was decreased with the dose for tacrine, indicating that the distribution to the brain is saturable. The uptake of organic cations such as choline, 1-methyl-4-phenylpyridinium (MPP), tetraethylammonium (TEA), and carnitine was inhibited by the addition of tacrine to cultures of mouse immortalized brain capillary endothelial cells. In addition, the apical to basal transport and basal to apical transport of tacrine were inhibited by the addition of organic cations to cultures of LLC-PK1 cells, suggesting that tacrine transport across the blood-brain barrier (BBB) is mediated by organic cation transport system(s). Consistent with the in vitro results, a standard reverse transcription-polymerase chain reaction procedure was able to amplify the message of mOCT2 and mOCTN2, but not mOCT1, in MBEC4 (mouse brain microvessel endothelial cell line 4) cells. Similarly, mRNAs for rOCT2 and rOCTN2 were present in representative rat brain samples. To determine whether OCT2 and/or OCTN2 transport tacrine, these transporters were cloned and then transfected in SK-HEP1 and HEK 293 cells. The uptake of choline, MPP, and TEA was inhibited by the presence of tacrine in rOCT2-expressing SK-HEP1 cells, whereas the uptake of carnitine was inhibited by the presence of tacrine in rOCTN2-expressing HEK 293 cells. Collectively, these observations suggest that the transport of tacrine across the BBB is mediated, at least in part, by multiple organic cation transport systems in rats.</P>

Background: Elderly patients visiting pain clinic may be at greater risk of misunderstanding the explanation because of age-related cognitive decline. Video instruction may provide a consistent from of teaching in a visual and realistic manner. We evaluated the effect of educational video on the patient understanding and satisfaction in a group of geriatric patients visiting pain clinic. Methods: Ninety two patients aged more than 60 years old who were scheduled for transforaminal epidural block were recruited. After exposure to either video or paper instruction process, each patient was asked 5-item comprehension questions, overall satisfaction and preference question. During follow-up period, number of outpatient referral-line call for further explanation was counted. Results: We observed significantly better comprehension in the video education compared with paper instruction (P<0.001). Patient satisfaction was also higher in the video group (P = 0.015), and patients visiting pain clinic were more preferred video instruction (P<0.001). Proportion of referral-line call for further explanation were similar (P = 0.302). Conclusions: Video approach to instruction process before consent improves treatment comprehension in geriatric patient visiting pain clinic.
A New Early Maturing, High Quality and Yielding Peanut, “Baekseon” for Vegetable Fruit

A new peanut variety “Baekseon” (Arachis hypogaea ssp. fastigiata L.) was developed at the Yeongnam Agricultural Research Institute, NICS, in Milyang in 2004. It was developed from the cross between the disease resistant cultivar “Suwon95” and the early maturing cultivar “Suwon94". “Beakseon” which has Shinpung plant type is less branch number with early flowering and long ellipse-shaped large kernel. Each pod which showed very shallow constriction had two grains with light brown testa and 100 seed weight was 96 g in the regional yield trials(RYT). This variety also contained higher free sugar and protein contents in seed quality, and had more resistant to early and late leaf spots and web blotch compared with check one. In the regional yield trials “Baekseon” was outyielded than check variety by 16% with 8.64 MT/ha for fresh pod and by 12% with 3.66 MT/ha for grain. It was evaluated to be suitable for vegetable peanut by its fresh pod yield and related characteristics.
