        Non-circular gear transmission mechanism with variable-involute and variable-cycloid composite tooth profile

        Non-circular gears with small curvature radius pitch curve are often used in the mechanical transmission of special motion path, but this kind of non-circular gear produces serious undercutting. Therefore, this research presented a kind of variable-involute and variable-cycloid composite tooth profile (VIVC-CTF). A transmission ratio curve of non-circular gear was designed by using Fourier function. The tooth profile equation of VIVC-CTF and its conjugate tooth profile equation were constructed on the basis of the normal and envelope methods. The influence of slip coefficient and ellipse eccentricity on the VIVC-CTF was investigated, and the variation of slip coefficient and eccentricity on the meshing line length, theoretical contact ratio, and pressure angle at pitch curve was analyzed. The meshing experiment of the VIVCCTF non-circular gear transmission mechanism was carried out on the basis of high-speed camera technology. The theoretical results of the transmission ratio were consistent with the experimental results.

        Numerical Model about the Influence of Cyclic Load on the Hydraulic Conductivity of Soft Clay

        The hydraulic conductivity k is one of the most important engineering properties of soft clay, and it plays an important role during the whole life cycle of underwater tunnel. Therefore, it is necessary to systematically study the response of k to dynamic load. In this study, a series of seepage tests were conducted on remolded Hangzhou soft clay using dynamic triaxial seepage apparatus after cyclic loading. The results show that the hydraulic conductivitys of all samples decrease after cyclic loading, and the maximum drop of the hydraulic conductivity can reach 29.1%. Also, the hydraulic conductivity of soil samples is significantly affected by cyclic stress ratio (CSR). With the increase of CSR, the decrease range of hydraulic conductivity under cyclic loading is between 22.3% and 24.3%. The dynamic load reconstructed the microstructure of clay and made the number of large pores getting decreased and the small pores increased. This resulted in a decrease in the smaller hydraulic conductivity (k) of the specimens. Futhermore, based on the nuclear magnetic resonance (NMR) scanning on the microstructure of soil samples under cyclic loading and the microscopic pore channel theory, a prediction model for the k of soft clay is proposed. The validity of the model is verified based on test data. It is believed that the proposed model could provide some theoretical support for the design and long-term performance prediction in practical engineering issues.

        Mannich Reaction as a Key Strategy for the Synthesis of Trifluoroethyl Derived Tertiary and Secondary Amine

        A Mannich reaction of 2,2,2-trifluoroethylamine hydrochloride, paraformaldehyde and acetophenone was described both in oil bath and microwave-heating conditions to give main products varied from trifluoroethyl derived tertiary to secondary amine. In the microwave-heating manner, the reaction was carried out in a commercial available microwave oven and took only 10 min to complete this transformation. After a simple post-treatment including washing and recrystallization, the product of trifluoroethyl derived secondary amine was obtained in high yield (71.4%).

      • Impact of AhR, CYP1A1 and GSTM1 Genetic Polymorphisms on TP53 R273G Mutations in Individuals Exposed to Polycyclic Aromatic Hydrocarbons

        This study was to undertaken to investigate the impacts of AhR, CYP1A1, GSTM1 genetic polymorphisms on the R273G mutation in exon 8 of the tumor suppressor p53 gene (TP53) among polycyclic aromatic hydrocarbons (PAHs) exposed to coke-oven workers. One hundred thirteen workers exposed to PAH and 82 control workers were recruited. We genotyped for polymorphisms in the AhR, CYP1A1, GSTM1, and TP53 R273G mutation in blood by PCR methods, and determined the levels of 1-hydroxypyrene as PAH exposure marker in urine using the high pressure liquid chromatography assay. We found that the distribution of alcohol users and the urinary excretion of 1-OHP in the exposed workers were significantly higher than that of the control workers (p=0.004, p<0.001, respectively). Significant differences were observed in the p53 genotype distributions of smoking subjects (p=0.01, 95%CI: 1.23-6.01) and PAH exposure (p=0.008, 95%CI: 1.24-4.48), respectively. Further, significant differences were observed in the p53 exon 8 mutations for the genetic polymorphisms of Lys/Arg for AhR (p=0.02, 95%CI: 0.70-15.86), Val/Val for CYP1A1 (p=0.04, 95%CI: 0.98-19.09) and null for GSTM1 (p=0.02, 95%CI: 1.19-6.26), respectively. Our findings indicated that polymorphisms of PAH metabolic genes, such as AhR, CYP1A1, GSTM1 polymorphisms may interact with p53 genetic variants and may contribute to PAH related cancers.

        Molecular analysis of gastric cancer identifies subtypes associated with distinct clinical outcomes

        Gastric cancer, a leading cause of cancer-related deaths, is a heterogeneous disease. We aim to establish clinically relevant molecular subtypes that would encompass this heterogeneity and provide useful clinical information. We use gene expression data to describe four molecular subtypes linked to distinct patterns of molecular alterations, disease progression and prognosis. The mesenchymal-like type includes diffuse-subtype tumors with the worst prognosis, the tendency to occur at an earlier age and the highest recurrence frequency (63%) of the four subtypes. Microsatellite-unstable tumors are hyper-mutated intestinal-subtype tumors occurring in the antrum; these have the best overall prognosis and the lowest frequency of recurrence (22%) of the four subtypes. The tumor protein 53 (TP53)-active and TP53-inactive types include patients with intermediate prognosis and recurrence rates (with respect to the other two subtypes), with the TP53-active group showing better prognosis. We describe key molecular alterations in each of the four subtypes using targeted sequencing and genome-wide copy number microarrays. We validate these subtypes in independent cohorts in order to provide a consistent and unified framework for further clinical and preclinical translational research.
