      • Binding Mode Analysis of Bacillus subtilis Obg with Ribosomal Protein L13 through Computational Docking Study

        Lee, Yu-No,Bang, Woo-Young,Kim, Song-Mi,Lazar, Prettina,Bahk, Jeong-Dong,Lee, Keun-Woo Korean Society for Bioinformatics 2009 Interdisciplinary Bio Central (IBC) Vol.1 No.1

Introduction: GTPases known as translation factor play a vital role as ribosomal subunit assembly chaperone. The bacterial Obg proteins ($Spo{\underline{0B}}$-associated ${\underline{G}}TP$-binding protein) belong to the subfamily of P-loop GTPase proteins and now it is considered as one of the new target for antibacterial drug. The majority of bacterial Obgs have been commonly found to be associated with ribosome, implying that these proteins may play a fundamental role in ribosome assembly or maturation. In addition, one of the experimental evidences suggested that Bacillus subtilis Obg (BsObg) protein binds to the L13 ribosomal protein (BsL13) which is known to be one of the early assembly proteins of the 50S ribosomal subunit in Escherichia coli. In order to investigate binding mode between the BsObg and the BsL13, protein-protein docking simulation was carried out after generating 3D structure of the BsL13 structure using homology modeling method. Materials and Methods: Homology model structure of BsL13 was generated using the EcL13 crystal structure as a template. Protein-protein docking of BsObg protein with ribosomal protein BsL13 was performed by DOT, a macro-molecular docking software, in order to predict a reasonable binding mode. The solvated energy minimization calculation of the docked conformation was carried out to refine the structure. Results and Discussion: The possible binding conformation of BsL13 along with activated Obg fold in BsObg was predicted by computational docking study. The final structure is obtained from the solvated energy minimization. From the analysis, three important H-bond interactions between the Obg fold and the L13 were detected: Obg:Tyr27-L13:Glu32, Obg:Asn76-L13:Glu139, and Obg:Ala136-L13:Glu142. The interaction between the BsObg and BsL13 structures were also analyzed by electrostatic potential calculations to examine the interface surfaces. From the results, the key residues for hydrogen bonding and hydrophobic interaction between the two proteins were predicted. Conclusion and Prospects: In this study, we have focused on the binding mode of the BsObg protein with the ribosomal BsL13 protein. The interaction between the activated Obg and target protein was investigated with protein-protein docking calculations. The binding pattern can be further used as a base for structure-based drug design to find a novel antibacterial drug.

      • Vapor transport deposited tin monosulfide for thin-film solar cells: effect of deposition temperature and duration

        Lee, Dajeong,Cho, Jae Yu,Yun, Hee-Sun,Lee, Doh-Kwon,Kim, Taehoon,Bang, Kijoon,Lee, Yun Seog,Kim, Ho-Young,Heo, Jaeyeong Royal Society of Chemistry 2019 Journal of Materials Chemistry A Vol.7 No.12

<P>The influence of the vapor transport deposition (VTD) conditions of tin sulfide (SnS) on the formation of secondary phases, preferred orientation, and solar cell performance is investigated in this study. It is concluded that 600 °C is the optimal growth temperature for the formation of pure SnS absorbers. When the growth temperature was 550 °C, the formation of secondary Sn2S3 and SnS2 phases was detected by Raman analysis. When the growth temperature was 625 °C, a noticeable change in morphology was observed with the plate-shaped grains aligned vertically to the substrate, which is detrimental to solar cell performance. The duration of growth also affected the morphology of the SnS absorber. Thin absorbers exhibited strong (120) preferred orientation. With increased duration of growth, (101) and (111) orientations increased. Such a variation in preferred orientation influenced the SnS/CdS solar cell’s performance. When the absorber thickness was as thin as ∼0.7 μm, the cell was prone to shunting and severe series resistance. When the absorber was as thick as ∼3.0 μm, physical shunting prevailed, worsening the performance of the cell. The highest efficiency of 3.93% with good cell-to-cell uniformity was achieved when the absorber thickness was ∼1.2 μm. The good stability of the best device was also confirmed under continuous illumination and damp-heat conditions for 100 h.</P>

<P><B>Background</B></P><P>For effective bone regeneration, it is necessary to implant a biocompatible scaffold that is capable of inducing cell growth and continuous osteogenic stimulation at the defected site. Here, we suggest an injectable hydrogel system using enzymatic cross-linkable gelatin (Gel) and functionalized gold nanoparticles (GNPs).</P><P><B>Methods</B></P><P>In this work, tyramine (Ty) was synthesized on the gelatin backbone (Gel-Ty) to enable a phenol crosslinking reaction with horseradish peroxidase (HRP). N-acetyl cysteine (NAC) was attached to the GNPs surface (G-NAC) for promoting osteodifferentiation.</P><P><B>Results</B></P><P>The Gel-Ty hydrogels containing G-NAC (Gel-Ty/G-NAC) had suitable mechanical strength and biocompatibility to embed and support the growth of human adipose derived stem cells (hASCs) during a proliferation test for three days. In addition, G-NAC promoted osteodifferentiation both when it was included in Gel-Ty and when it was used directly in hASCs. Based on our results, we suggest that Gel-Ty/G-NAC hydrogels can serve both as a biodegradable graft material for bone defect treatment and as a good template for tissue engineering applications such as drug delivery, cell delivery, and various tissue regeneration uses.</P>

      • Hidradenitis suppurativa in association with Keratitis-ichthyosis-deafness syndrome treated by adalimumab

( Yu Mee Song ),( Hyun Ji Lee ),( Seung Ah Yoo ),( Jae Yeon Ryu ),( Yeong Ho Kim ),( Ju Hee Han ),( Chul Hwan Bang ),( Young Min Park ),( Jun Young Lee ),( Ji Hyun Lee ) 대한피부과학회 2018 대한피부과학회 학술발표대회집 Vol.70 No.1

A 43-year-old man came to our clinic suffering from a malodorous fistulating tract of the lower extremities and a purulent draining sinus of the groin embedded in keloidal tissue. He was diagnosed with Keratitis-ichthyosis-deafness (KID) syndrome via sequencing analysis 11 years prior. A punch biopsy of lesion on the lower extremities was done. Clinically and histologically, Hidradenitis suppurativa (HS) was diagnosed in association with KID syndrome. Previous therapies with antibiotics and isotretinoin did not produce satisfactory improvements. We started adalimumab with an induction dose of 160 mg followed by 80mg and a maintenance dose of 40 mg every weeks. After 8 weeks on adalimumab therapy, the lesion showed marked improvement. Modified Sartorius scores showed progressive improvement during treatment, from 43 at the first visit to 17 after 8 weeks of treatment with adalimumab. Hurley stage was also improved from 3 to 2. Despite appropriate treatment, HS recurs frequently. KID syndrome is a rare congenital disorder of the ectoderm that gives rise to keratitis, erythrokeratoderma and neurosensory deafness. In addition to xerosis, HS has rarely been reported as a skin symptom accompanying KID. Antibiotics, acitretin, retinoids and anti-TNF drugs have been reported to treat it, but cases of severe HS are usually refractory to treatment. Herein, we report a case of HS in association with KID syndrome treated with adalimumab.

      • Two cases of lymphomatoid papulosis type D

( Yu Mee Song ),( Hyo Jung Kim ),( Hyun Ji Lee ),( Ju Hee Han ),( Chul Hwan Bang ),( Ji Hyun Lee ),( Young Min Park ),( Junyoung Lee ) 대한피부과학회 2018 대한피부과학회 학술발표대회집 Vol.70 No.1

A 29-year-old woman presented with 3-years history of erythematous papules on the right shoulder and left arm. The patient had no history of underlying disease. A 36-year-old woman presented with several erythematous to brownish macules and papules on the trunk and extremities. The patient had a history of thyroid cancer. Skin biopsy taken from these two patient showed wedge-shaped infiltration of atypical lymphocytes admixed with histiocytes, neutrophils and extravasated red blood cells, along with coexpression of CD 3, CD 8 and CD 30 in T lymphocytes associated with prominent epidermotropism. Based on these clinical and histological findings, two patients were diagnosed with lymphomatoid papulosis (LyP) type D. During a 4-year follow-up period, patients reported spontaneous occurrence and disappearance of erythematous papules. LyP subtype D is characterized by medium to large atypical cells with marked epidermotropism and coexpression of CD8 and CD30 in a scant inflammatory background with no known risk of progression to lymphoma. It is a rare subtype of LyP, which has only one reported case in Korea. Herein, we report two rare cases of lymphomatoid papulosis type D.

      • Low-Dose Persistent Organic Pollutants Impair Insulin Secretory Function of Pancreatic β-Cells: Human and In Vitro Evidence

        Lee, Yu-Mi,Ha, Chae-Myeong,Kim, Se-A,Thoudam, Themis,Yoon, Young-Ran,Kim, Dae-Jung,Kim, Hyeon-Chang,Moon, Hyo-Bang,Park, Sungmi,Lee, In-Kyu,Lee, Duk-Hee American Diabetes Association 2017 Diabetes Vol.66 No.10

<P>Low-dose persistent organic pollutants (POPs), especially organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs), have emerged as a new risk factor for type 2 diabetes. We evaluated whether chronic exposure to low-dose POPs affects insulin secretory function of beta-cells in humans and in vitro cells. Serum concentrations of OCPs and PCBs were measured in 200 adults without diabetes. Mathematical model-based insulin secretion indices were estimated by using a 2-h seven-sample oral glucose tolerance test. Insulin secretion by INS-1E beta-cells was measured after 48 h of treatment with three OCPs or one PCB mixture. Static second-phase insulin secretion significantly decreased with increasing serum concentrations of OCPs. Adjusted means were 63.2, 39.3, 44.1, 39.3, 39.7, and 22.3 across six categories of a summary measure of OCPs (P-trend = 0.02). Dynamic first-phase insulin secretion remarkably decreased with increasing concentrations of OCPs among only insulin-sensitive individuals (P-trend = 0.02); the insulin levels among individuals with high OCPs were approximate to 30% of those with low OCPs. Compared with OCPs, PCBs showed weaker associations. The decreased insulin secretion by INS-1E beta-cells was observed for even 1 pmol/L OCP. The data from human and in vitro cell experiments suggest that chronic exposure to low-dose POPs, especially OCPs, can induce pancreatic beta-cell dysfunction.</P>

Anemia and activities of daily living in the Korean urban elderly population: results from the Korean Longitudinal Study on Health and Aging (KLoSHA).

        Bang, Soo-Mee,Lee, Jeong-Ok,Kim, Yu Jung,Lee, Keun-Wook,Lim, Soo,Kim, Jee Hyun,Park, Young Joo,Chin, Ho Jun,Kim, Ki Woong,Jang, Hak-Chul,Lee, Jong Seok Springer International 2013 Annals of hematology Vol.92 No.1

<P>This study was planned to investigate the prevalence and risk factors of anemia and its impact on health-related quality of life and activities of daily living (ADL) in elderly Koreans. Of the 1,118 randomly sampled elderly Koreans aged 65?years or older living in Seongnam, Korea, on Aug. 1, 2005, we estimated the prevalence of anemia from 695 responders. We investigated the risk factors of anemia using a merged sample of this random sample and 270 volunteers enrolled from Seongnam residents aged 85?years or older. We diagnosed anemia according to the World Health Organization criteria. The estimated age- and gender-standardized prevalence of anemia was 8.33?% for the overall random sample (95?% confidence intervals (CI) 6.28-10.39), 10.58?% in men (95?% CI 7.09-14.07), and 6.85?% in women (95?% CI 4.37-9.34). The identified risk factors were age 80?years, male, iron deficiency, history of stroke, renal dysfunction, and metabolic syndrome. Anemia was associated with impairment in physical functioning (p?=?0.031) and instrumental ADL (p?<?0.001). This is the first report about anemia's prevalence in community-dwelling Korean elders, adjusted and standardized according to the city's and nation's population. Timely diagnosis of anemia and correction of its treatable cause may improve QOL and ADL in elderly individuals.</P>

Case Reports : Two Cases of Interstitial Pneumonitis Caused by Rituximab Therapy

Yu Na Lee,Sun Young Kyung,Soo Jin Choi,Soo Mee Bang,Seong Hwan Jeong,Dong Bok Shin,Jae Hoon Lee 대한내과학회 2006 The Korean Journal of Internal Medicine Vol.21 No.3

Rituximab, a chimeric monoclonal antibody directed against CD20, has become a part of the standard therapy for patients with non-Hodgkin`s lymphoma either in combination with other drugs or as a single agent. The CD20 antigen is expressed on 95% of B-cell lymphoma cells and normal B-cells but, is not found on precursor B-cells or stem cells. Rituximab is now approved for patients with diffuse large B-cell lymphoma when combined with standard CHOP chemotherapy (cyclophosphamide, doxorubicin, vincristine and prednisone) or patients with follicular lymphoma who have failed first line chemotherapy. The monoclonal antibody is generally well tolerated. Most of the adverse events are infusion-associated, mild to moderate non-hematological toxicities. Severe respiratory adverse events have been infrequent. Here, we report two patients with non-Hodgkin`s lymphoma in whom interstitial pneumonitis developed with rituximab therapy.
