      • DMNQ S-64 Induces Apoptosis via Caspase Activation and Cyclooxygenase-2 Inhibition in Human Nonsmall Lung Cancer Cells

        <P>Shikonin has been reported to induce apoptosis and inhibit angiogenesis in vivo and in vitro. 6-(1-propoxyiminoalkyl)-5,8-dimethoxyoxy 1,4-naphtoquinone S-64 (DMNQ S-64) was synthesized as a shikonin derivative. In this article, the underlying apoptotic mechanism of DMNQ S-64 was examined. DMNQ S-64 exerted cytotoxicity against A549 lung carcinoma cells with IC(50) of 27.3 microM. Apoptotic bodies were observed in DMNQ S-64-treated A549 cells by 4'-6-diamidino-2-phenylindole (DAPI) staining assay. DMNQ S-64 also increased sub-G1 DNA portion in a concentration-dependent manner by flow cytometric analysis. Western blotting has revealed that DMNQ S-64 effectively activates the expression of caspase 8, 9, and 3, cleaves poly (ADP-ribose) polymerase, and increases the ratio of Bax/Bcl-2. Furthermore, cytochrome c was released in a concentration-dependent manner by DMNQ S-64. Similarly, DMNQ S-64 significantly increased caspase 3 activity by enzyme-linked immunosorbent assay (ELISA). It also significantly inhibited the level of prostaglandin E2 (PGE(2)) by ELISA and downregulated the expression of cyclooxygenase-2 (COX-2) in a concentration-dependent manner. Taken together, DMNQ S-64 may exhibit cytotoxicity against A549 cells via caspase activation and COX-2 inhibition.</P>

        Introduction The purpose of this study is to gain nutritional control and that of Fat data through the measurement of the Skinfold thickness. In a group of the subjects of study 54 ball game player's (soccer player's ; 18, basketball player's ; 9, baseball player's ; 16, rugby-football player's ; 11) Classification of the Measurement For the convenience of the measurement, the researcher of this study classify the measurement is as following ; A. Physical Constitution 1. Body Height 2. Body Weight B. Skinfold Thickness 1. Back 2. Waist 3. Abdomen 4. Arm Summary The gesults of this study pointed out as follwing ; 1. The average of Height was 175.1cm for total average of ball game player's, 173.1cm for soccer player's, 184.7cm for basketball player's, 173.1cm for baseball player's and 173.5cm for rugby-football player's. 2. The average of Weight was 69.6 Kg for total average for ball game player's, 67.6 Kg for soccer player's, 76.3 Kg for basketball player's, 68.1 Kg for baseball player's and 69.8 Kg for rugby-football player's. 3. The Fat of accumulation a portion in order waist, back, abdomen and arm for soccer, basketball and baseball player's but rugby-football player's order waist, abdomen, back and arm. 4. The average of Skinfold Thickness was 6.3mm for soccer player's, 6.3mm for basketball player's, 7.3mm baseball player's and 7.5mm for rugby-football player's. 5. The ball game player's average of skinfold thickness was 6.8mm (back; 7.5mm, waist; 8.8mm, abdomen; 6.2mm, arm; 5.7mm) 6. The average of Fat Weight was 8.0 Kg for soccer Player's, 8.0 Kg for basketball player's, 8.7 Kg for baseball player's, 8.8 Kg for rugby-football player's and total average was 8.4 for ball game player's. 7. The average of Fat Free Weight was 59.6 Kg for soccer player's, 68.3 Kg for basketball player's, 59.4 Kg for baseball player's and 61.0 Kg for rugby-football player's. 8. The average of % Fat was 11.8% for soccer player's, 10.5% for basketball player's, 12.8% for baseball player's, 12.7% for rugy-football player's and total average 12.1% for ball game player's.

        벼 육묘트레이를 이용한 어린 잎 채소 생육에 미치는 광 조절 영향

        벼 육묘 트레이를 활용한 어린잎 채소 재배 가능성을 탐색하고자 재배 기간 중 차광 처리하여 다채, 청경채, 알파인배추, 청치마 상추 등 4종을 17일간 재배하였다. 차광 처리는 무차광 17일(NS), 차광 8일+무차광 9일 (S8NS9), 무차광 8일+차광 9일(NS8S9), 차광 17일(S17) 하였다. 처리에 따른 누적 광량은 NS>S8NS9>NS8S9>S17처리구 순으로 높았으나, 누적 온도는 처리에 따른 차이가 크지 않았다 차광 8일 째 어린잎채소 4종의 생육과 엽록소함량은 감소하였다. 광 조절 17일 째 어린잎채소 4종의 초장, 엽장, 엽폭, 경경, 생체중, 엽록소 함량 등의 생육은 무차광 NS 처리구에서 가장 높았다. 생육 초기 광조절에 따른 생육 후기 초장과 생체중 증가율이 어린잎채소 4종 모두 NS 또는 S8NS9 처리구에서 높았다. 따라서 벼 육묘장 다층 시스템에서의 공간 활용을 고려한 어린잎채소 재배시 차광이 많은 하부 공간은 생육초기에 활용하고, 수확기에는 광 확보가 이루어지는 상부에서 재배가 이루어짐이 적합하리라 본다. This study was conducted to investigate the effect of light control on growth of baby leaf vegetables (tatsoi, pak-choi, chinese cabbage ‘alpine’ and leaf lettuce ‘cheongchima’) in rice seedling tray. Light treatment controlled in non-shading (NS), shading for 8 days+non-shading for 9 days (S8NS9), non-shading for 8 days+shading for 9 days (NS8S9), and shading for 17 days (S17). Integrated solar radiation in NS was highest, and in S8NS9>NS8S9>S17 order. Integrated temperature did not shown difference among the treatments for growing periods. Growth and chlorophyll content of baby leaf vegetables were lowest in shading treatment for 8days. The highest growth characteristics (plant height, leaf length, leaf width, stem diameters, and fresh weight) and chlorophyll content of baby leaf vegetables was shown in NS. Increment rate of plant height and fresh weight of baby leaf vegetables were high in NS or S8NS9 as affected by light control for 8 days in early growth periods. These results suggest that we can utilize in low layer for early period and in high layer for harvest periods for baby vegetable production in rice seedling factory.

        S6K1 Phosphorylation of H2B Mediates EZH2 Trimethylation of H3: A Determinant of Early Adipogenesis

        S6K1 has been implicated in a number of key metabolic responses, which contribute to obesity. Critical among these is the control of a transcriptional program required for the commitment of mesenchymal stem cells to the adipocytic lineage. However, in contrast to its role in the cytosol, the functions and targets of nuclear S6K1 are unknown. Here, we show that adipogenic stimuli trigger nuclear translocation of S6K1, leading to H2BS36 phosphorylation and recruitment of EZH2 to H3, which mediates H3K27 trimethylation. This blocks Wnt gene expression, inducing the upregulation of PPARγ and Cebpa and driving increased adipogenesis. Consistent with this finding, white adipose tissue from S6K1-deficient mice exhibits no detectable H2BS36 phosphorylation or H3K27 trimethylation, whereas both responses are highly elevated in obese humans or in mice fed a high-fat diet. These findings define an S6K1-dependent mechanism in early adipogenesis, contributing to the promotion of obesity.

        Detection of Genotype associated with Disease Activity and Development of Probe

        질환성과 관련된 세균의 분포 및 유전자형을 탐색하고자 구강농양 및 골수염의 급성감염 혼자와 진료실 및 실험실의 정상인을 대상으로 시료를 채취하여 포도상구균을 분리 및 동정을 시행하고, 특성을 규명하였으며, plasmid 및 염색유전자를 분리하여 제한효소를 처리후 전기영동을 실시하고 분리된 plasmid로 탐색자를 제작하여 dot blot을 시행하였다. 대부분의 급성환자에서 분리된 포도상구균을 S. lugdunensis와 S. aureus이었으나, 진료실 및 실험실에서는 coagulase 음성 staphylococci가 분리되었다. 급성환자에서 분리된 포도상구균은 ampicillin과 penicillin에 내성을 보였다. 분리된 S. lugdunensis균주중 네 균주는 δ형의 용혈소를 생산하였다. Plasmid를 분리한 결과 S. lugdunensis균주중 세 균주는 약 6.5 kilobases이었으나 S. aureus는 약 4.3 kilobases 정도 크기의 band를 보였다. S. lugdunensis에서 분리된 plasmid로 제작한 탐색자로 dot blot를 시행한 결과 치과 영역에서 분리한 plasmid를 갖는 균주는 양성반응을 보였다. 염색체유전자의 유전자형을 분석한 결과 δ형의 용혈소를 생산한 네 균주의 S. lugdunensis는 유사한 유전자형을 보였다. 이러한 연구결과 질환의 진행에 S. lugdunensis가 중요한 역할을 하는 것으로 생각되고, 치과영역에 존재하는 plasmid는 공통적인 유전자 서열을 갖는 것으로 추정된다.

      • 야구경기에서 타격순서별 출루와 득점에 관한 연구 : 추계대학 야구연맹전을 중심으로

        In order to examine not only the details how batter's advanced by hitting a safety, or four ball, or deall ball but also score in each battering order, Yungnam, Kunkook, Donga, Inha, Hanyang, and Yonsei Universities were selected as subjects for this study, which were successful in pushing into the final league match in Fall Season Nation-Wide College Base-ball Championship held from September 14 till 18, 1977. The results obtained by the test being conducted carefully to those teams are as follows; 1. The details of batter's frequency in advancing to another base, which was sinvestigated on the percentage basis. (1)The ratio, having made it possible for the batters in each team to advance to another was revealed to be 35.6% and the ratio of opposite case was exposed to be 64.4%. Accordingly the ratio between the former and the latter was proved to be 1:2. (2)The ratio of each batter's hit was as follows; 1st batter's hit (40.2%), 4th batter's hit (42.7%), 7th batter's hit (39.8%), 9th batter's hit(39.1%). Generally, those batter's abve mentioned are in the state of being good and high level in hitting. 8th batter's hit(21.3%), 2nd batter's hit (28/3%). These batters were not good in the ratio of hitting. In battings per one game, the first batter hit 4.4 times. The nineth; 3.5 times, there-fore there was a noticeable difference being revealed in the ability of batting between 1st and 9th batter. However no distinct difference in battings was being found between 3rd and fourth batters. (3)The more detailed ratio of batters' activity throughout the whole games are recorded as follows; one base hit (16.2%), four ball (7%) two base hit (3.9%), home run (1.2%). Grounder out (24.3%), flyout(22.4%), strike out(13.3%), sacrifice batting(fly & bunt) (2.3%) can be regarded as the direct cause of their having been prevented from advancing to the 2nd base. (4) The ratio of the 4th batter's strike out was 12.9%, which was fourth in low level. (5) The ratio of batters'advancing frequency to 2nd base in each team are as follows; Kunkook(41.5%), Hanyang(39.5%), Yungnam(38.2%), Inha(32.4%), Yonsei(32.4%), Donga(29.5%) II. Score (1) Score being made in each battering order can be described as follows; 1st & 9th batter (17 points) 4th & 9th batter (11 points) 3rd batter (1Opoints) 8th batter (7 points) 2nd batter (5 points) (2) In the aspect of score being recorded throughout the whole games, we were able to get such results as follows. In such a case as a base hit(a one timer), the teams got 38 points (36.5%). In such a case as home run hit, 27 points (26%). In such a case as a two-base hit(a two-beggar), 18 points(17.3%), and by errors and a sacrifice batting, they got merely 6 points(5.85%). No score, being made by a three-base hit (a triple). (3) Mutual relation between the batting order and the position of fielders was recorded as follows; 1st batter; short stop (15 times) 2nd " ; 2nd base man (15 times) 3rd " ; left fielder (11 times) 4th " ; 1st base-man (12 " ) 5th " ; catcher (10 " ) 6th " ; pitcher (6 " ) 7th " ; 2nd baseman (7 " ) 8th " ; Pitcher (19 " ) 9th " ; center fielder(8 " ) The above data show most popular frequency of each batter's position being changed while they are defending.

        Carbon nanotube-based biosensor for detection of matrix metallopeptidase-9 and S-100B

        We fabricated carbon nanotube (CNT)-based biosensor with field effect transistor structure for detection of matrix metallopeptidase-9 (MMP-9) and S-100B. To minimize contact effects, the metal electrodes were passivated with SiO<SUB>x</SUB> film, and anti-MMP-9 and S-100B antibody were immobilized on the CNT surface using a chemical linker. When MMP-9 or S-100B antigen was added to bind anti-MMP-9 or S-100B antibody, the decrease in electrical conductance was observed. We have also investigated the dependence of MMP-9 and S-100B concentration on the conductance change. A linear relationship was found between the conductance and the concentration in semi-logarithm scale.

      • Application of genetically engineered Salmonella typhimurium for interferon-gamma-induced therapy against melanoma

        Salmonella have been experimentally used as anti-cancer agents, because they show selective growth in tumours. In this study, we genetically modified attenuated Salmonella typhimurium to express and secrete interferon-gamma (IFN-γ) as a tumouricidal agent to enhance the therapeutic efficacy of Salmonella. IFN-γ was fused to the N-terminal region (residues 1-160) of SipB (SipB160) for secretion from bacterial cells. Attenuated S. typhimurium expressing recombinant IFN-γ (S. typhimurium (IFN-γ)) invaded the melanoma cells and induced cytotoxicity. Subcutaneous administration of S. typhimurium (IFN-γ) also efficiently inhibited tumour growth and prolonged the survival of C57BL/6 mice bearing B16F10 melanoma compared with administration of phosphate-buffered saline (PBS), unmodified S. typhimurium or S. typhimurium expressing empty vector (S. typhimurium [Vec]) in a natural killer (NK) cell-dependent manner. Moreover, genetically modified Salmonella, including S. typhimurium (IFN-γ), showed little toxicity to normal tissues with no observable adverse effects. However, S. typhimurium (IFN-γ)-mediated tumour suppression was attributed to direct killing of tumour cells rather than to stable anti-tumour immunity. Collectively, these results suggest that tumour-targeted therapy using S. typhimurium (IFN-γ) has potential for melanoma treatment.

        Progress in the development of heating systems towards long pulse operation for KSTAR

        <P>Construction of the Korea superconducting tokamak advanced research (KSTAR) tokamak is in its final phase. For the long-pulse KSTAR discharges, the ion cyclotron range of frequencies (ICRF) and neutral beam injection (NBI) heating systems are expected to play important roles through a selective heating of ions and electrons, control of the plasma pressure and current profiles, a core fuelling and beam diagnostics for the KSTAR. In addition, the ICRF system is expected to be used for possible discharge cleaning and assisting in the tokamak startup. In this paper, the recent progress in the development of the ICRF and the NBI heating systems is described. The four-strap ICRF antenna has been successfully tested for a voltage up to 41 kV for a pulse length of 300 s (to 46 kV for 20 s) in a test chamber. A prototype KSTAR NBI system has been developed. At present, the system has successfully produced a 1 MW beam power for 200 s and a 3.5 MW output beam power for 4 s.</P>
