Structural and functional properties of Maillard-reacted casein phosphopeptides with different carbohydrates

        Meng Yuan,Yu Cao,Haoyang Zheng,Kunlin Chen,Yuping Lu,Jing Wang,Liqin Zhu,Ming Chen,Zhipeng Cai,Yonggen Shen 한국식품과학회 2024 Food Science and Biotechnology Vol.33 No.7

This study used glucose, fructose, maltose and dextran to explore the effects of different carbohydrates on the Maillard reaction of casein phosphopeptides (CPP). The color parameter results showed that heating time from 1 to 5 h led to brown color, which was consistent with the observed increased in browning intensity. Fourier transform infrared spectroscopy results verified that four carbohydrates reacted with CPP to produce Maillard conjugates. Fluorescence spectroscopy showed that the Maillard reaction changed the tertiary structure of CPP by decreasing the intrinsic fluorescence intensity and surface hydrophobicity compared with the CPP-carbohydrate mixture. At the same time, the Maillard reaction effectively improved the emulsifying properties, reducing power and DPPH radical scavenging activity of CPP. Furthermore, this study also found that glucose and fructose improved CPP more than maltose and dextran. Therefore, monosaccharides have good potential in modifying CPP via the Maillard reaction.

Single nucleotide polymorphism-based analysis of the genetic structure of the Min pig conserved population

        Meng Fanbing,Cai Jiancheng,Wang Chunan,Fu Dechang,Di Shengwei,Wang Xibiao,Chang Yang,Xu Chunzhu 아세아·태평양축산학회 2022 Animal Bioscience Vol.35 No.12

Objective: The study aims to uncover the genetic diversity and unique genetic structure of the Min pig conserved population, divide the nucleus conservation population, and construct the molecular pedigree. Methods: We used KPS Porcine Breeding Chip v1 50K for SNP detection of 94 samples (31♂, 63♀) in the Min pig conserved population from Lanxi breeding Farm. Results: The polymorphic marker ratio (PN), the observed heterozygosity (Ho), and the expected heterozygosity (He) were 0.663, 0.335, and 0.330, respectively. The pedigreebased inbreeding coefficients (FPED) was significantly different from those estimated from runs of homozygosity (FROH) and single nucleotide polymorphism (FSNP) based on genome. The Pearson correlation coefficient between FROH and FSNP was significant (p<0.05). The effective population content (Ne) showed a continuously decreasing trend. The rate of decline was the slowest from 200 to 50 generations ago (r = 0.95), then accelerated slightly from 50 to 5 generations ago (1.40<r<1.50) and increased significantly in the last 5 generations (r = 2.6). According to the composition of Chinese lineage, we separated the nucleus conservation population (81 individuals) and the candidate conservation population (13 individuals) of Min pig, then the nucleus conservation population of Min pig was divided into 9 families by genomic information matrix. Conclusion: Our study indicated that the genetic diversity of the Min pig conserved population was inadequate. Due to the introgression of European commercial pig breeds and the unscientific breeding process, it is necessary to construct the molecular pedigree of the nucleus conservation population for the Min pig. Creep performance of concrete-filled steel tubular (CFST) columns and applications to a CFST arch bridge

        Meng-Gang Yang,C. S. Cai,Yong Chen 국제구조공학회 2015 Steel and Composite Structures, An International J Vol.19 No.1

This paper first presents an experimental study of twelve specimens for their creep performance, including nine concrete-filled steel tubular (CFST) columns and three plain concrete columns, subjected to three levels of sustained axial loads for 1710 days. Then, the creep strain curves are predicted from the existing creep models including the ACI 209 model, the MC 78 model, and the MC 90 model, and further a fitted creep model is obtained by experimental data. Finally, the creep effects of a CFST arch bridge are analyzed to compare the accuracy of the existing creep models. The experimental results show that the creep strains in CFST specimens are far less than in the plain concrete specimens and still increase after two years. The ACI 209 model outperforms the MC 78 model and the MC 90 model when predicting the creep behavior of the CFST specimens. Analysis results indicate that the creep effects in the CFST arch bridge are significant. The deflections and stresses calculated by the ACI 209 model are the closest to the fitted model in the three existing models, demonstrating that the ACI 209 model can be used for creep analysis of CFST arch bridges and can meet the engineering accuracy requirement when lack of experimental data.

A Novel Zero-Voltage-Switching Push-Pull Forward Converter with a Parallel Resonant Network

Cai, Chunwei,Shi, Chunyu,Guo, Yuxing,Yang, Zi,Meng, Fangang The Korean Institute of Power Electronics 2017 JOURNAL OF POWER ELECTRONICS Vol.17 No.1

A novel zero-voltage-switching (ZVS) push-pull forward converter with a parallel resonant network is presented in this paper. The novel topology can provide a releasing loop for the energy storage in a leakage inductor for the duration of the power switching by the resonant capacitors paralleled with the primary windings of the transformer. Then the transformer leakage inductor is utilized to be resonant with the parallel capacitor, and the ZVS operation is achieved. This converter exhibits many advantages such as lower duty-cycle losses, limited peak voltage across the rectifier diodes and a higher efficiency. Furthermore, the operating principles and key problems of the converter design are analyzed in detail, and the ZVS conditions are derived. A 500W experimental converter prototype has been built to verify the effectiveness of the proposed converter, and its maximum efficiency reaches 94.8%.

Alfalfa xenomiR-162 targets G protein subunit gamma 11 to regulate milk protein synthesis in bovine mammary epithelial cells

        Meng Guizhi,Duan Hongjuan,Jia Jingying,Liu Baobao,Ma Yun,Cai Xiaoyan 아세아·태평양축산학회 2024 Animal Bioscience Vol.37 No.3

Objective: It was shown that microRNAs (miRNAs) play an important role in milk protein synthesis. However, the post-transcriptional regulation of casein expression by exogenous miRNA (xeno-miRNAs) in ruminants remains unclear. This study explores the regulatory roles of alfalfa xeno-miR162 on casein synthesis in bovine mammary epithelial cells (bMECs). Methods: The effects of alfalfa xenomiR-162 and G protein subunit gamma 11 (GNG11) on proliferation and milk protein metabolism of bMECs were detected by 5-Ethynyl-2′- Deoxyuridine (EdU) staining, flow cytometry, cell counting kit-8 (CCK-8), enzyme-linked immunosorbent assay, quantitative real-time polymerase chain reaction (qRT-PCR), and Western blot. Dual-luciferase reporter assay was used to verify the targeting relationship between GNG11 and xenomiR-162. Results: Results showed that over-expression of xenomiR-162 inhibited cell proliferation but promoted apoptosis, which also up-regulated the expression of several casein coding genes, including CSN1S1, CSN1S2, and CSN3, while decreasing the expression of CSN2. Furthermore, the targeting relationship between GNG11 and xenomiR-162 was determined, and it was confirmed that GNG11 silencing also inhibited cell proliferation but promoted apoptosis and reduced the expression of casein coding genes and genes related to the mammalian target of rapamycin (mTOR) pathway. Conclusion: Alfalfa xenomiR-162 appears to regulate bMECs proliferation and milk protein synthesis via GNG11 in the mTOR pathway, suggesting that this xeno-miRNA could be harnessed to modulate CSN3 expression in dairy cows, and increase κ-casein contents in milk.

Three-Phase Four-Wire Inverter Topology with Neutral Point Voltage Stable Module for Unbalanced Load Inhibition

Cai, Chunwei,An, Pufeng,Guo, Yuxing,Meng, Fangang The Korean Institute of Power Electronics 2018 JOURNAL OF POWER ELECTRONICS Vol.18 No.5

A novel three-phase four-wire inverter topology is presented in this paper. This topology is equipped with a special capacitor balance grid without magnetic saturation. In response to unbalanced load and unequal split DC-link capacitors problems, a qusi-full-bridge DC/DC topology is applied in the balance grid. By using a high-frequency transformer, the energy transfer within the two split dc-link capacitors is realized. The novel topology makes the voltage across two split dc-link capacitors balanced so that the neutral point voltage ripple is inhibited. Under the condition of a stable neutral point voltage, the three-phase four-wire inverter can be equivalent to three independent single phase inverters. As a result, the three-phase inverter can produce symmetrical voltage waves with an unbalanced load. To avoid forward transformer magnetic saturation, the voltages of the primary and secondary windings are controlled to reverse once during each switching period. Furthermore, an improved mode chosen operating principle for this novel topology is designed and analyzed in detail. The simulated results verified the feasibility of this topology and an experimental inverter has been built to test the power quality produced by this topology. Finally, simulation results verify that the novel topology can effectively improve the inhibition of an inverter with a three-phase unbalanced load while decreasing the value of the split capacitor.
