        Comparison of the Angiogenic Ability between SHED and DPSC in a Mice Model with Critical Limb Ischemic

        BACKGROUND: Regenerative medicine by using stem cells from dental pulp is promising for treating patients with critical limb ischemic (CLI). Here, we investigated the difference in the angiogenetic ability of stem cells from human exfoliated deciduous teeth (SHED) and human dental pulp stem cells (DPSC). METHODS: SHED and DPSC were harvested from dental pulp and analyzed in flow- cytometry for detecting the expression of surface markers. Levels of angiogenetic marker were examined by RT-PCR and Western-blot. Eighteen immunodeficient mice of critical limb ischemic model were divided into three groups: SHED, DPSC and saline, which was administered with SHED, DPSC or saline intramuscularly. Histological examination was performed to detect the regenerative results. RESULTS: A highly expression of CD146 was detected in SHED. Moreover, cells with negative expression of both CD146 and CD31inSHEDwere more in comparison with those inDPSC. Expression of angiogenesis factors includingCXCL12,CXCR4,Hif- 1a, CD31, VEGF and bFGFwere significant higher in SHED thanDPSC by the RT-PCR and Western-Blot results. SHED induced more CD31 expression and less fibrous tissue formation in the critical limb ischemic model as compare with DPSC and saline. CONCLUSION: Both SHED and DPSC possessed the ability of repairing CLI. With expressing more proangiogenesis factors, SHED may have the advantage of repairing CLI.

      • Experimental Study of Molecular Immune Mechanism of TGCO Anti-Adjurant Arthritis

        The total glycosides of cornus officinal (TGCO) is effective part of cornus officinals, TGCO suppress immune reactions has been reported in vivo or in vitro. In this paper, we researched effect of TGCO on rat adjuvant arthritis and its molecular immune mechanism. The results showed that TGCO had better action of curing to rat AA, suppressing joint swell of primary and secondary recreation to adjuvant, improving of pathological changes of joints and alleviating spain. On above basis, we forward investigated the molecular immune mechanism TGCO anti-AA, the main results showed such as: 1. TGCO could suppress T lymphocytes proliferation stimulated with ConA and DTH reaction induced by PPO in AA rats. 2. TGCO suppress production of IL-1, IL-6 and TNFα of peritoneal macrophages from AA rats, and PEG2 in plasma of AA rats.3. TGCO could suppress expression of iNOS mRNA gene and IL-lra mRNA gone of peritoneal macrophage of AA rats. 4. Cell-ELISA method was used to detect adhesion molecules(ICAM-1, CD44) and integrin (α4 β7) on ECV 304(It is an important endothelial cell line derived from HUVEC), TGCO could inhibit ECV304 expressing adhesion molecules (ICAM-1, CD44) and integrin(α4β7), with TNF-α and IL-1 stimulate in vitro. Summarizing above results, we infer that TGCO can cure rat AA is in relation to suppressing cell-mediated immune, anti inflammation cytokine(IL-1. IL-6 and TNF-α), and inhibiting adhesion molecule expression.

      • MiR-130a Overcomes Gefitinib Resistance by Targeting Met in Non-Small Cell Lung Cancer Cell Lines

        Non-small cell lung cancer (NSCLC) is the most common type of lung cancer and the most common cause of lung cancer death. Currently, the epidermal growth factor receptor inhibitor gefitinib is used for its treatment; however, drug resistance is a major obstacle. Expression of Met has been associated with both primary and acquired resistance to gefitinib, but the mechanisms regulating its expression are not fully understood. Recently, miRNAs such as miR-130a have been shown to play a role in gefitinib resistance, but importance in NSCLC and relationships with Met have not been fully explored. Here we show that miR-130a is over-expressed in gefitinibsensitive NSCLC cell lines, but is low in gefitinib-resistant NSCLC cell lines. Moreover, miR-130a expression was negatively correlated with that of Met. Further analysis revealed that over-expression of miR-130a increased cell apoptosis and inhibited proliferation of NSCLC cells treated with gefitinib, whereas lowering the expression of miR-130a decreased cell apoptosis and promoted cell proliferation after treatment with gefitinib in both gefitinib-sensitive and -resistant NSCLC cell lines, suggesting that miR-130a overcomes gefitinib resistance. We also demonstrated that miR-130a binds to the 3'-UTR of Met and significantly suppresses its expression. Finally, our results showed that over-expressing Met could "rescue" the functions of miR-130a regarding cell apoptosis and proliferation after cells are treated with gefitinib. These findings indicate that the miR-130a/Met axis plays an important role in gefitinib resistance in NSCLC. Thus, the miR-130a/Met axis may be an effective therapeutic target in gefitinib-resistant lung cancer patients.

        Knockdown of LncRNAZFAS1 suppresses cell proliferation and metastasis in nonsmall cell lung cancer

        To evaluate the effects of LncRNAZFAS1 on cell proliferation and tumor metastasis in non-small cell lung cancer (NSCLC), we detected the expression level of LncRNAZFAS1 in NSCLC-related tissues and cells. qRT-PCR results revealed that LncRNAZFAS1 in tumor tissues was significantly higher than that in normal lung tissue, especially significantly up-regulated in stage III / IV and in metastatic NSCLC tissues. LncRNAZFAS1 expression was dramatically up-regulated in 4 NSCLCrelated cells (A549, SPC-A1, SK-MES-1, and NCI-H1299), with having the highest expression level in A549 cells. Furthermore, we implemented a knockdown of LncRNAZFAS1 in A549 cells, and the results of CCK8 and Transwell assays suggested that knockdown of LncRNAZFAS1 significantly inhibited NSCLC cell proliferation and metastasis. Next, we constructed a tumor xenograft model to evaluate the effect of LncRNAZFAS1 on the NSCLC cell proliferation in vivo. The results indicated that knockdown of LncRNAZFAS1 dramatically inhibited A549 cells proliferation and repressed tumor growth. Additionally, knockdown of LncRNAZFAS1 drastically weakened the expressions of MMP2, MMP9 and Bcl-2 proteins, whereas noticeably strengthened the expression of BAX protein. Our results altogether suggest that knockdown of LncRNAZFAS1 has a negative effect on the proliferation and metastasis of NSCLC cell, which implying LncRNAZFAS1 is a potential unfavorable biomarker in patients with NSCLC.

        One Metal-Organic Framework Showing Two-Dimensional Sheet Structure: Synthesis, Structure, and Magnetic Property

        Under hydrothermal condition, the reaction of 2,4-dichlorophenoxyacetic acid (2,4-DH) and 1,3-bis(4-pyridyl)propane with MnCl2·2H2O gave a novel metal-organic framework (MOF), [Mn(2,4-D)2 (H2O)2] n (1). This complex was characterized by IR, elemental analysis, powder X-ray diffraction, and thermogravimetry analysis. X-Ray single-crystal diffraction shows that the Mn(II) ions are bidendate bridged by carboxyl groups in the syn–anti mode, giving a two-dimensional (2D) network. Temperature-dependent magnetic studies reveal that there are weak antiferromagnetic exchange interactions between the Mn(II) ions transmitted by carboxyl groups.

        Microstructure and property evolution of diamond-like carbon films co-doped by Al and Ti with different ratios

        <P><B>Abstract</B></P> <P>Diamond-like carbon (DLC) films with weak carbide metal Al and carbide metal Ti co-doping (Al/Ti-DLC) were prepared by a hybrid ion beam deposition system. The atomic ratios of doped Al to Ti were tailored via designing the special Al/Ti combined sputtering target. The composition, microstructure, roughness, residual stress, hardness, toughness, and tribological behaviors of the deposited films were systematically evaluated to explore the dependence of structural properties on Al/Ti ratios. Results indicated that the high-throughput preparation of DLC films with different Al/Ti atomic ratios was achieved by tailoring the sputtering target and process parameters without the difference in both the film thickness and total Al/Ti content. With the Al/Ti ratios in the films decreased from 8.8 to 3.0, the residual stress, hardness, and toughness of Al/Ti-DLC films increased obviously, originating from the increased fraction of titanium carbide and the reduced Al content. However, the coefficient of friction and wear rate with decreasing the Al/Ti ratio were obviously improved, which was related with the transformation of underlying friction mechanism from the sliding interface graphitization to dangling bond-passivation. The present results not only suggest a high-throughput method to fabricate co-doped DLC films, but also promote the scientific understanding and engineering application of DLC films with high performance.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Ti/Al co-doped diamond-like carbon films were fabricated by a hybrid ion beam method. </LI> <LI> Different Al/Ti ratios were successfully achieved at one time using designed target. </LI> <LI> Al/Ti ratios had no effect on the chemical state of co-doped Ti and Al atoms. </LI> <LI> The mechanical and tribological properties were strongly dependent on Ti/Al ratios. </LI> <LI> Different friction mechanisms were observed with Al/Ti ratios ranged from 8.8 to 3.0. </LI> </UL> </P>

        Optimization of preparation and properties of Gardenia yellow pigment-loaded alginate beads

        Gardenia yellow pigment (GYP) loaded alginate beads were prepared by the ionic gelation technique, and the preparation parameters were optimized by response surface methodology for high encapsulation efficiency. The optimized parameters were alginate concentration of 3.3%, CaCl2 concentration of 2.4%, and GYP concentration of 3.2mg/mL, under which the encapsulation efficiency was 73.61%. The surface morphology and bead size analysis showed that the GYP-loaded alginate beads had a roughly spherical morphology with a wrinkled surface, and their average diameter was 0.87 mm. In vitro release test revealed that the GYP release had a pH-dependent release profile and a two-step release process. The Rigter-Peppas model was the most proper model to assess the GYP release from alginate beads. The release mechanism of GYP at pH 1.2 and 7.4 was non-Fickian transport and case-II transport, respectively. The 2,2-diphenyl-1-picrylhydrazyl assay indicated that the encapsulated GYP had effectively maintained 82.56% of the antioxidant activity.

        Classification and existence of nonoscillatory solutions of higher order nonlinear neutral difference equations

        In this paper, we consider the higher order nonlinear neutral delay difference equation of the form r(xn + pxn−) + f(n, xn−1(n), xn−2(n), . . . ,xn−m(n)) = 0. We give an integrated classification of nonoscillatory solutions of the above equation according to their asymptotic behaviours. Necessary and suffi- cient conditions for the existence of nonoscillatory solutions with designated asymptotic properties are also established.
