        大豆(Glycine max. MERRILL, Lequminosae)는 전통적으로 많은 식품에 이용되고 있다. 일반성분으로 steroids, flavonoids, glyceolin, 외에도 필수인 지질, stigmasterol과 saponin등이 함유되어 있고, saponin ⅠㆍⅡㆍⅢㆍ은 혈중 과산화지질의 생성을 억제하므로 성인병에 방제로서의 개발이 기대된다는 보고와 더불어 大豆saponin의 性質및 生理作理에 미치는 영향을 연구하고자 우선 C.M.C.(the critical micelle concentration), bovine serum albumin과의 결합 및 Dipalmitoyl L-α-phosphatidyl choline(DPPC) liposome의 성질에 미치는 영향을 관찰하고 이에 보고하고자 한다. CMC, sap.-albumin binding and phase transition temperature of soyasaponin are following. 1. The critical micelle concentration(CMC) of soyasaponin(sap. ) was determined by fluorometry and surface tensiometry. The CMC values given from these two methods are 8×10??M and 7.4×10??M, respectively. The surface exces of the sap. (Γ) and the area (A) occupied by a sap. molecule at the monolayer absorbed at air and water interface were calculated employing Gibbs absorption equation. The values are Γ×10??=1.205mole/㎠, and A=132.7A??. The presence of buffer increased the surface activity of the sap, and decreased the CMC, the surface tension at the CMC. 2. The binding of soyasaponin to Bovine serum albumin(BSA) was studied by the fluorescence method, 1-anilinonaphthalene-8sulfonate(ANS) as fluorescent probes. The evaluations of the nature and degree of association of sap.-albumin complex are following: the number of binding sites of BSA for sap. is 2.2, and binding constant is 8.4×10₃M??. Competitive bindings between sap. and probe were observed. All of these are affected by pH and metal ions. 3. The differential scanning thermograms of the DPPC liposomes in the presence of soyasaponin showed that the phase transition temperature of the liposomes was significantly lowed and broadened by 0.1mM sap. This was ascribed to the possibility that incorporation of sap. into the lipid bilayer reduced the cooperativity unit of the layer and decreased the phase transition temperature. The order of lowering in the phase transition temperature by sap. were approximately indentical to the order of the pharmacological activity of sap.

        The anti-proliferating effects of some plants on the MCF-7 cells were studied by the 3-[4,5-Dimethylthiazol-2yl]-2,5-diphenyl-tetrazolium bromide(MTT assay), to investigate the anticancer effect with some polyphenols. YKK, Acacia catechu, BioB and pine oil of them are showed a good anticancer effect. We also examined morphological changes on the MCF-7 cells in this process. The cells included in medium were investigated to be aggregated and destroyed slowly by treatment with those above. In case of YKK, Acacia catechu and pine oil, the cells are almost disappeared after 12 days in culture bottle.

        I could take carboxyethylgermanium sesquioxide from ASAI Germanium Institute. It was experimented in cytotoxicity on carcinoma cell : Sarcoma180,L1210 and Molt-4, and in mitogenecity of lymphocytes in vitro. in vitro WBC contents were observed by Wright's staining under microscope after injected it, and the spleen and tumor size were measured by weight. Molt-4 is a suspension culture derived from the peripheral blood of acute lymphoblastic leukemia patient. L1210 is lymphocytic mouse leukemia and grows in suspension cultures or spinner cultures. Sarcoma 180 was derived from Swiss Webster Sarcoma 180 ascites. These cell lines have been used extensively for routine screening programs of chemical agents and natural products for cytotoxicity activity. These are also utilized in preliminary testing for antitumor activity in cancer chemotherapy screening studies. In order to test in vivo cytotoxicity of germanium compound, BALB/C mice were used. Sarcoma 180 cells were harvested by brief treatment with trypsin-EDTA and then inoculated in abdominal cavities of mice by intraperitoneal transplantation of 2×?cell/mouse. Germanium-30 ㎕ or nomal saline were injected intraperitoneally once a day for 6 weeks. The survival of treated sarcoma mice and the growth of tumor were observed during 6 weeks. At sixth week, sarcoma mice were decapitated and tumors and spleens were weighted. Into each well (0.2ml capacity) of 10³, 5×10³,2×10⁴,?cells/well were inoculated at Day-0. After culture during various time interval, the absorbances measured by MTT. At Day-2 or Day-3 after culture, and 5×10³and 10⁴cells/well, we can take the appropriate value in L1210 and Molt-4. Sarcoma cells were show the lower value compared with the other cells(Fig 1,2,3) To estimate the effect of germanium compound as a mitogen to lymphocytes, the absorbance of human lymphocytes cultured in medium containing germanium compound for 3days was measured by MTT method. Germanium compound increased it significantly at the concentration of 10㎍ and 30㎍/ml. When germanium compound compared with concanavalin A, that has higher ability of mitogenecity than concanavalinA on human lymphocytes.(Fig.4,5) The direct cytotoxicity of germanium compound was estimated by comparing absorbaned to the control which was cultivated without germanium compound. Fig 6,7 show that the absorbances of Sarcoma 180, L1210 of MOLT-4 cultured in medium containing germanium compound was decreased dose-dependently. It means that germanium compound exhibits direct cytotoxicity. However in the case of Molt-4, the absorbance was lower than the other, respectively of administered dose. Following injection of germanium compound of 2mg/kg/day for 2 weeks, whole blood was obtained in vivo. the leukocytes stained with wright's staining solution compared with the control to be injected normal saline instead of germanium compound. As shown in table 1, there was increase in lymphocytes, basophils and monocytes. At long-term treatment, LMP% was increased a little, but the tumor weight decreased or increased on individuality.

        We have studied that the effect on the inhibition of the cancer cell growth by the 3-[4,5-Dime wthylthiazol-2 yl]-2,5-diphenyl-tetrazolium bromide(MTT) assay, to investigate the anticancer effect with plants, observed around us, by culturing the human hepatoma cell-line. And we knew it was the best condition that the cell number was 4×10³ cells/well and the time of culture is 48 hours. We found that Equisetum arvense L. and Lactuca dentata Makino. var. Flaviflora Makino. exhibited a good anticancer effect and especially Capsicum annum L. angulosum Mill exhibited the best effect of them.

        The cells included in medium were invastigated to be aggregated and destroyed slowly by treatment with Lactuca dentata Makino. var. flaviflora Makino. On the other hand, the cells treated with Equisetum arvense L were aggregated to be coat-like. The case of Capsicum annuum L. var. angulosum Mill, the cells became vague after 2 days and then destroyed. This procedure was faster than other. The chromasome was more condensated after 37hr treated with Capsicum annuum L. var. angulosum Mill.

We observed the effect on the inhibition of the cuncer cell growth with plants, observed us and we can find that Capsicum annum L. angulosum Mill exhibited a good anticancer effect.

        Effects of Barbiturates on the Rotational Relaxation Time of 1, 6-Diphenyl-1, 3, 5-hexatriene in Native and Model Membranes

        Synaptosomal plasma membrane vesicles (SPMV) were isolated from fresh bovine cerebral cortex. The effects of barbiturates on the rotational relaxation time of 1.6-diphenyl-1, 3, 5-hexatriene (DPH) in intact SPMV and model membranes of total lipids (SPMVTL) and phosphlipids (SPMVPL) extracted from SPMV were examined. Barbiturates decreased the rotational relaxation time of DPH in intact SPMV in a dose-dependent manner. In contrast, they did not affect the rotational relaxation time of DPH in SPMVTL and even dose-dependently increased the rotational relaxation time of DPH in SPMVPL.

        Effects of n - alkanols on the rotational Relaxation Time of 1 , 6 - Diphenyl - 1 , 3 , 5 - hexatriene in the Model mebranes of Total Lipid Fraction Extracted from Brain Membranes

        The effects of n-alkanols on the rotational relaxation time of 1,6-diphenyl-1,3,5-hexatriene (DPH) were investigated in the liposomes of total lipids (SPMVTL) extracted from synaptosomal plasma membrane vesicles. n-Alkanols decreased the rotational relaxation time of DPH in the SPMVTL, and the potencies of n-alkanols up to 1-nonanol increased by 1 order of magnitude as the carbon chain length increased by two carbon atoms. The cut-off phenomenon was reached at 1-decanol, where further increase in hydrocarbon length resulted in an increase in the rotational relaxation time of DPH.
