        치주인대 세포의 교원질 생성에 대한 Substance P의 효과

        Substance P는 교정력이 가해진 치아의 치주인대 중 인장력을 받는 부위에 많이 분포하는 neuropeptide 중의 하나이며, 또한 여러 조직에서 neurogenic inflammation을 야기하는 neuropeptide 중의 하나로도 알려져 있다. 그러나 중요한 세포의 단백기질인 교원질의 생성에 대한 Substance P의 효과는 잘 알려져 있지 않다. 따라서 이 연구의 목적은 배양 치주인대 세포에서 교원질 생성에 대한 Substance P의 효과를 평가하는 것이었다. collagenase-digestion method로 교원질 생성을 평가하였고 mRNA 수준에서 작용효과를 평가하기 위하여 Northern blot hybridization을 시행하였다. 이 연구는 또한 교원질 생성에 대한 prostaglandin과 gelatinase 생성도 포함하였으며 변성된 교원질의 분해를 평가하기 위하여 Zymography를 이용하였다. 비교원성 단백질, 교원성 단백질, 상대교원질에 대한 dose-dependent effect를 보면 Substance P는 비교원성 단백질 합성을 증가시켰으나 교원성 단백질 합성은 감소시켰다. 그리하여 총 단백합성에 대한 상대적인 교원질 생성을 나타내는 상대교원질은 7%에서 3.6%로 감소시켰다. 세포를 indomethacin과 동시에 처리할 때 substance P의 교원질 합성 억제효과는 나타나지 않았다. 이것은 Substance P의 교원질 합성 억제효과가 prostaglandin의 생성 때문이라는 것을 의미한다. Substance P의 교원질 합성 억제효과가 procollagen mRNA의 정상(steady-state)수준에 부합하는가를 평가하기 위하여 northern blot hybridization을 시행한 결과 Substance P는 α1(1) procollagen mRNA의 양적 변동을 일으키지 않았다. Substance P의 교원질 생성 억제효과는 전사이후의 어떤 단계에서 이루어지는 현상임을 나타낸다. 치주인대세포에서 gelatinase 생성에 대한 Substance P의 효과를 알아보기 위하여 zymography를 이용하였다. zymogram을 보면 Substance P는 치주인대세포에서 gelatinase 생성에는 아무 효과도 나타내지 않음을 알 수 있다. Substance P의 교원질 생성 억제효과가 치주인대세포에 대해 선택적인가 아닌가를 알아보기 위하여 MC3T3-E1세포를 이용하였는데 Substance P는 MC3T3-E1세포의 교원질 합성에는 영향을 미치지 않았다. 이상에서 Substance P는 인간의 치주인대세포에서 교원질 합성을 억제하였다. 이 효과는 procollagen mRNA와 gelatinase 생성의 정상(steady-state) 수준의 변화 때문이 아니라 prostaglandin 생성과 연관이 있음을 알았다. Substance P is one of the neuropeptide which presents highly in tension site of periodontal ligament during the orthodontic tooth movement. It has been also known as one of the neuropeptides which cause neurogenic inflammation in various tissues and organs. However, there is no report about the effect of substance P on major extracellualar matrix protein, collagen production. The purpose of this study was to evaluate the collagen production by substance P in human periodontal ligament cell, The collagenase-digestion method was used to evaluate collagen production and also used Northern blot hybridization for the evaluation of collagen mRNA level. This study also included in terms of prostanglandins and gelatinase production with respect to collagen production. For the collagen degradation, zymography was used to estimate denatured collagen degradation. Dose-dependent effect of substance P on noncollagen protein, collagen, and percent collagen was that substance P increased noncollagen protein synthesis, but decreased collagen systnisis. So the percent collagen, which determined by relative collagen production against total protein production, was decreased from 7% to 3.6%. This inhibitory effect of substance P on collagen production was disappeared when cells were treated concomitantly with indomethacin. It means that substance P-induced inhibitory effect on collagen production was due at least in part to the production of prostaglandins. To evaluate whether substance P-induced inhibitory effect on collagen production is correspond to the steady-state levels of procollagen mRNA, Northern blot hybridizartion was performed and it showed that substance P has no effect on the steady-state level of α1(1) procollagen mRNA. It means that the inhibitory effect of substance P on collagen production was due to the change of a certain mechanism after posttranscription. In this context, gelatinase production by substance P in periodontal ligament cells was evaluated by zymography. Zymogram showed that substance P has no effect on gelatinase production in periodontal ligament cells. To explore wheter substance P-induced inhibitory effect on collagen production is selevtive in periodontal ligament cells or not, MC3T3-31 cells which originated from mouse calvaria was used. It showed that substance P has no effect on collagen production in MCDTD-E1 cells. Taken together, substance P inhibits collagen production in human periodontal ligament cells. This effect was not due to the change of the steady-state level of procollagen mRNA and gelatinase production, but due at least in part to the change of prostaglandins production.

        In this paper, we examine if the lexical information of the verb and its noun object affects the pitch accent patterns of the verb phrase focus. Three types of verb-object dominations with different semantic weights are discussed: when the verbs have optional direct objects, when the objects have the greater semantic weight elative to verbs, and when the verbs and the objects have equal semantic weight. Argument-structure-based works note that the pith accent location in a focused phrase is closely related to the argument structure and contextual information. For example, it has been agued that contextually new noun objects receive accent while given noun objects don't. Contrary to nouns, verbs can be accented or not in verb phrase focus regardless of whether they are given information or new information (Selkirk 1984, 1992). However, the production experiment in this paper how that the accenting of verbs is not fully optional, but influenced by the lexical semantic information of the verbs. The accenting of noun objects with given information is possible and the deaccenting of new noun objects also occurs depending on the lexical information of the noun objects. The results demonstrate that in addition to argument structure and information by means of context sentences, the lexical semantic information of words influences the pitch accent location in focused phrase.

      • 스포츠센터 소비자가 지도자 서비스품질과 구매 후 행동에 미치는 영향 분석

        With the improvement of the living standard, people pursued a better quality of life, keeping a healthy balance between the three dimensions - mentally, physically and socially. They further aspired to fulfill their dreams and make waves around the world. In keeping abreast of these changes, leisure sports that could be enjoyed by general public came to the forefront. As the number of participants in the sports increased sharply, the pressing need for places for sports activities became apparent. Well-aware of the increasing need, this researcher attempted to analyze the influences of commercial sports and fitness center customers on service quality and post-purchase actions. This study limited its targets to customers of sports and fitness centers in Seoul, 2003. A total of 500 customers were selected using stratified random sampling; however, those who repeated the same answers for several questions or provided insufficient answers were eliminated from the study. After this screening process, data gathered from 480 subjects were used for the analysis. The collected data were analyzed statistically by the following SPSS programs: First, Descriptive Analysis was used to analyze variables of population statistics and the distribution of answers to each question. Second, the reliability of the questionnaires were examined by Cronbach'a. Third, the hypothesis was verified thoroughly by T-test, one-way ANOVA, correlation analysis, and multiple regression analysis. For post-verification, Ducan test was performed. The statistically significant level was set to p<. .05 for verification. The following results were obtained through the analysis: First, customers' age, sex, and occupations seemed to affect service quality factors (i.e., reliability, empathy, responsiveness, tangibles and assurance) as differences were found in them. However, no difference was found in the service quality in relation to the customers' educational level and monthly average income. Second, regarding customers' post-purchase actions, differences were found in sex and age. With respect to sex, differences were found in a factor such as recommendation intention to others. In case of age, differences were found in a factor such as purchase intention. However, no difference was found in customers' post-purchase actions in terms of their educational level, income, and occupations. Third, the sports and fitness center customers did not seem to affect service quality and post-purchase action factors such as purchase and recommendation intentions to others.

        In this paper, a temperature controller for simiconductor test equipment is developed using LQG/LTR methodology. The developed controller is applied to a plate designed for temperature control of semiconductor chip. Control accuracy within ±1℃ and control time within 5minutes are attained successfully.

        지혈대 마비에 대한 임상적 고찰

        As the society has been industrialized, the number if industrial accident has increased. The hand surgeries have increased as the result of hand injuries in case of handing machine, postburn scar contractures of the hand and congenital anomalies of hand. The tourniquets used in these operations are essential instruments. The cause of tourniquet paralyses are carelessness in using touriquets and disorder of tourniquet. The causes of tourniquet paralysis seem to be related to tourniquet pressure and inchemic time. We experienced 5 cases of tourniquet induced paralysis of arm after the use of tourniquet. We surveyed similar cases of other surgeons and presented the cause of this serious complication. As a result, we recommend for the prevention of tourniquet induced paralysis as follows. 1. Check the pressure gauze before the use of s tourniquet. 2. Removal of the tourniquet cuff after deflation 3. Removal of the soft bandage after removal of the cuff 4. Optimum pressure for obtaining the bloodless field Adult : 250/300㎜Hg(upper/Lower extremity) Child : 200/250㎜Hg(Upper/Lower extremity) 5. Recommendable tourniquet time is within 2 hours. 6. Tourniquet should be placed 10 ㎝ above the epicondyle in the upper extremity. 7. Tourniquet be released in case of dissection of major artery due to prevention of postoperation hematoma

        To improve administration of ketolorac tromethamine, we prepared matrix tablets with Eudragit (RS 100 or 30D), diluents (avicel:lactose=l:l or Ca phosphate) and Mg stearate using wet-granulation method. Sustained release matrix tablets using Eudragit® RS 100 7 and 10% decreased dissolution rate at pH 1.2 and 68 Ca. phosphate as diluents showed more decreased dissolution rate than that of avicel:lactose (1:l). Sustained release matrix tablets using Eudragit® RS 30D also showed very effective sustained release of KT.

        Ketorolac (KA) and ketorolac tromethamine (KT) is the nonsteroidal anti-inflammatory drugs (NSAIDs) and has been used widely. To improve its frequent administration by control of drug release. we prepared SLM of KA and KT using Mirj 45 and 52 as surfactants and tripalmitin and tristearin as lipids by spry drying method SLM, as a disperse system for injection, were evaluated by measurement of particle sizes. SEM and drug release pattern As a result. spherical SLM were prepared and their particle size were in the range of 25-125 μm, which were dependant on the types and contents of lipid and sururfactants. Their dissolution rate were decreased as increasing lipid contents.

        This study has been performed to experiment of plastic metallic mold KP-4M on the variation of spindle revolution, feedrate, polishing times, grain size, and the pressure adding to tool.The results obtained from this study are as follows : 1. Surface roughness(Ra) grows worse, as the velocity is lower in the area of low revolution, and the velocity highter in the area of high revolution. 2. Above 1500rpm at the spindle revolution, Ra grows worse if polishing times (N) is less than 2 because of the preceding process, and the surface turn brillant if polishing times more than 4 because of loading. 3. As the higher pressure which adding to a tool is better when we expect more manufacturing quantity, and we need low pressure when surface roughness take precedence of quantity. 4. On the basic of the experimental findings, it is possible to be automatic of mirror like finishing with the machining center.
