        <i>Yersinia pseudotuberculosis</i> Exploits CD209 Receptors for Promoting Host Dissemination and Infection

        <P><I>Yersinia pseudotuberculosis</I> is a Gram-negative enteropathogen and causes gastrointestinal infections. It disseminates from gut to mesenteric lymph nodes (MLNs), spleen, and liver of infected humans and animals.</P><P><I>Yersinia pseudotuberculosis</I> is a Gram-negative enteropathogen and causes gastrointestinal infections. It disseminates from gut to mesenteric lymph nodes (MLNs), spleen, and liver of infected humans and animals. Although the molecular mechanisms for dissemination and infection are unclear, many Gram-negative enteropathogens presumably invade the small intestine via Peyer’s patches to initiate dissemination. In this study, we demonstrate that <I>Y. pseudotuberculosis</I> utilizes its lipopolysaccharide (LPS) core to interact with CD209 receptors, leading to invasion of human dendritic cells (DCs) and murine macrophages. These <I>Y. pseudotuberculosis</I>-CD209 interactions result in bacterial dissemination to MLNs, spleens, and livers of both wild-type and Peyer’s patch-deficient mice. The blocking of the <I>Y. pseudotuberculosis</I>-CD209 interactions by expression of O-antigen and with oligosaccharides reduces infectivity. Based on the well-documented studies in which HIV-CD209 interaction leads to viral dissemination, we therefore propose an infection route for <I>Y. pseudotuberculosis</I> where this pathogen, after penetrating the intestinal mucosal membrane, hijacks the <I>Y. pseudotuberculosis</I>-CD209 interaction antigen-presenting cells to reach their target destinations, MLNs, spleens, and livers.</P>

        Prediction of postoperative pancreatic fistula using a nomogram based on the updated definition

        Purpose: The International Study Group on Pancreatic Fistula’s definition of postoperative pancreatic fistula (POPF) has recently been updated. This study aimed to identify risk factors for POPF in patients having pancreaticoduodenectomy (PD) and to generate a nomogram to predict POPF. Methods: Data on 298 patients who underwent PD from March 2012 to October 2017 was retrospectively reviewed and POPF statuses were redefined. A nomogram was constructed using data from 220 patients and validated using the remaining 78 patients. Independent risk factors for POPF were identified using univariate and multivariate analyses. A predictive nomogram was established based on the independent risk factors and was compared with existing models. Results: Texture of the pancreas, size of the main pancreatic duct, portal vein invasion, and definitive pathology were the identified risk factors. The nomogram had a C-index of 0.793 and was internally validated. The nomogram performed better (C-index of 0.816) than the other most cited models (C-indexes of 0.728 and 0.735) in the validation cohort. In addition, the nomogram can assign patients into low- (less than 10%), intermediate- (10% to 30%), and high-risk (equal or higher than 30%) groups to facilitate personalized management. Conclusion: The nomogram accurately predicted POPF in patients having PD.

        Optimization of the microemulsion formulation of curcuma oil and evaluation of its acaricidal efficacy against Tetranychus cinnabarinus (Boisduval) (Acari: Tetranychidae)

        The microemulsion formulation (hereafter formulation) of curcuma oil and its acaricidal efficacy against Tetranychus cinnabarinus Boisduval (Acari: Tetranychidae) were optimized in the laboratory to evaluate their spray effectiveness of oviposition inhibition and repellence. Ethovision XT6 was used to analyse the effects of the sublethal concentrations (LC 20 ) of curcuma oil and the formulation on the behaviors of T. cinnabarinus. The results showed that Tween-80 was the best surfactant, Isopropanol was the best co-surfactant and K m = 2:1 was the best condition for the formulation. The prepared microemulsions are stable under conditions of centrifugation and incubation for extended periods. The results showed that the effect of the spray bioassays of the formulation against T. cinnabarinus continuously increased during the experiment, but for curcuma oil almost no longer increase observed when the exposure time went beyond 24 h. Moreover, compared with curcuma oil (LC 50 = 0.716%), the spray bioassay of the formulation (LC 50 = 0.035%) was stronger against T. cinnabarinus. The repellency of the formulation to T. cinnabarinus was stronger with increasing exposure time, but that of curcuma oil declined after 12 h of exposure. The mobile distance of T. cinnabarinus treated with the formulation continuously declined during the experiment but that due to the curcuma oil almost no longer declined when the treatment time reached 12 h. The maximum mobile frequency of T. cinnabarinus treated by curcuma oil and the formulation was recorded at 6 h and 12 h, respectively. Thus, the formulation is a promising candidate as a botanical acaricide of green vegetables.

        Generation of Selenoprotein with Glutathione Peroxidase Activity by Chemical Modification of the Single-chain Variable Fragment Expressed in a Single-protein Production System and Its Antioxidant Ability

        Glutathione peroxidase (GPX) is one of the important members of the antioxidant enzyme family. It can catalyze the reduction of hydroperoxides with glutathione to protect cells against oxidative damage. Single-chain variable fragment (scFv) can be converted into seleniumcontaining single-chain variable fragment (Se-scFv) by chemical modification of the hydroxyl groups in scFv, thus Se-scFv possesses GPX activity and becomes a prodrug. To improve the expression of scFv and simplify its purification steps, Single-protein production (SPP) system was used to express scFv and chemical modification was used to synthesize Se-scFv. Therefore, we must construct a new scFv-WCD1-lessACA gene, which can express its mRNA not containing any ACA sequences and express its amino acid sequence of target protein (scFv) being same to scFv-WCD1. In this way, the scFv-WCD1-lessACA can be only expressed in SPP system and no other background proteins in the cells could be expressed. The expression results showed that high level of scFv-WCD1-lessACA synthesis was at least sustained for 96 h in the virtual absence of background protein synthesis. Then, selenocysteine (Sec) was incorporated into the scFv-WCD1-lessACA by chemical modification and resulted in Se-scFv-WCD1-lessACA. The enzymatic characteristics of Se-scFv-WCD1-lessACA were determined. GPX activity was 2,563 U/μmol,its binding constant for GSH was 0.687 ×105/mol. Moreover,Se-scFv-WCD1-lessACA was confirmed to have a strong antioxidant ability to protect mitochondria against oxidative damage induced by Vc/Fe2+ (mitochondrial damage model),suggesting that Se-scFv-WCD1-lessACA has potential application for protection of mitochondrial damage induced by reactive oxygen species (ROS).

        Development of human IgE biosensor using Sezawa-mode SAW devices

        This paper reports Sezawa-mode surface acoustic wave (SAW) devices with via-isolated cavity to construct the allergy biosensor. To fabricate Sezawa-mode SAW devices, the RF magnetron sputtering method for the growth of piezoelectric ZnO thin films are adopted and influences of the sputtering parameters are investigated. The optimal substrate temperature of 300 C, RF power of 120 W and sputtering pressure of 2 Pa were used to deposit piezoelectric ZnO films with a smooth surface, uniform grain size and strongly c-axis-orientated crystallization. A back-etched SAW resonator is used in this study. The wet etching of (100)-oriented silicon wafers is used to form a back-side cavity which is critical to the formation of a hopper cavity for holding bio-analytes. The remaining membrane structure silicon thickness was 25 mm. In this report, the chrome (Cr, 12 nm)/gold (Au, 66 nm) layer was initially deposited onto the sensing area of SAW devices as the binding layer for biochemical sensor. The resonance frequency of the Sezawa-mode SAW device is 1.497 GHz. The maximum sensitivity of the Sezawa-mode is calculated to be 4.44 106 cm2/g for human immunoglobulin-E (IgE) detection. The stability for human IgE detection is calculated to be 80% and the variation of the stability 3% was obtained after several tests.

        Clinicopathological Features and Immunohistochemical Alterations of Keratinocyte Proliferation, Melanocyte Density, Smooth Muscle Hyperplasia and Nerve Fiber Distribution in Becker`s Nevus

        Background: Although Becker`s nevus (BN) is a relatively common disease, the systematic studies of clinicopathological and immunohistochemical results are poorly reported. Objective: To investigate the clinicopathological features and immunohistochemical alterations of keratinocyte proliferation, melanocyte density, smooth muscle hyperplasia and nerve fiber distribution in BN. Methods: Clinical and pathological data were collected in 60 newly-diagnosed BN cases. Immunohistochemical stain of Ki-67, Melan-A, keratin 15, smooth muscle actin and protein gene product 9.5 was performed in 21 cases. Results: The median diagnostic and onset age was 17 and 12 years, respectively. Skin lesions usually appeared on the upper trunk and upper limbs. The pathological features included the rete ridge elongation and fusion and basal hyperpigmentation. Epidermal Ki-67, Melan-A and keratin 15 expression and dermal nerve fiber length were significantly higher in lesional and perilesional skin than in normal skin (p<0.05∼0.01), while smooth muscle actin expression was upregulated only in skin lesion (p<0.05). Conclusion: Although the clinical diagnosis of BN is often straightforward, histopathology is helpful to differentiate from other pigmentary disorders. The hyperproliferation of keratinocytes, melanocytes, arrector pili muscle and dermal nerve fibers could be involved in the patho-genesis of BN. (Ann Dermatol 28(6) 697∼703, 2016)

        Investigation of Proteomic Responses of Streptomyces lydicus to Pitching Ratios for Improving Streptolydigin Production

        Streptomyces lydicus has been reported to produce antibiotic streptolydigin. Pitching ratios play crucial roles in primary and secondary metabolism of Streptomyces bacteria. The higher pitching ratio (30%, v/v)significantly enhanced the levels of streptolydigin products in S. lydicus. Proteome analysis revealed that betaglucosidase and UTP-glucose-1-phosphate uridylyltransferase were up-regulated to accelerate the starch hydrolyzation at the high pitching ratios. Enhancement in the levels of UDPN-acetylmuramoylalanyl-D-glutamate-2, 6-diaminopimelate ligase and glycine cleavage system aminomethyltransferase were involved in the conversion of amino acids into secondary metabolites. Additionally, the expression levels of PfkA2, PfkA3, Zwf2, SucD, GalE1, GatB, TktA1 and ThcA, associated with glycolysis, pentose phosphate pathway, TCA cycle and amino acid metabolism, were dramatically elevated at high pitching ratios, which play important roles in the enhanced streptolydigin production in S. lydicus E9. Interestingly, the levels of proteins (glutamine synthetase I, glutamate synthase subunit beta and glutamine synthetase) were down-regulated with the increases of pitching ratios and fermentation progress,revealing that pitching ratio altered the glutamine synthetase levels and consequently regulated the streptolydigin production of S. lydicus E9. The up-regulation of proteins (eg, aldehyde dehydrogenase and alkyl hydroperoxide reductase) was involved in the redox-based regulation network triggered by an imbalance of the intracellular cell redox homeostasis and by crosstalk with secondary metabolism at the higher pitching ratio. These results settle new insights into physiological facts of S. lydicus E9 in responses to pitching ratios and will eventually improve the antibiotic production schemes in industry.

        Cis-3-O-p-hydroxycinnamoyl Ursolic Acid Induced ROS-Dependent p53-Mediated Mitochondrial Apoptosis in Oral Cancer Cells

        Cis-3-O-p-hydroxycinnamoyl ursolic acid (HCUA), a triterpenoid compound, was purified from Elaeagnus oldhamii Maxim. This traditional medicinal plant has been used for treating rheumatoid arthritis and lung disorders as well as for its anti-inflammation and anticancer activities. This study aimed to investigate the anti-proliferative and apoptotic-inducing activities of HCUA in oral cancer cells. HCUA exhibited anti-proliferative activity in oral cancer cell lines (Ca9-22 and SAS cells), but not in normal oral fibroblasts. The inhibitory concentration of HCUA that resulted in 50% viability was 24.0 μM and 17.8 μM for Ca9-22 and SAS cells, respectively. Moreover, HCUA increased the number of cells in the sub-G1 arrest phase and apoptosis in a concentrationdependent manner in both oral cancer cell lines, but not in normal oral fibroblasts. Importantly, HCUA induced p53-mediated transcriptional regulation of pro-apoptotic proteins (Bax, Bak, Bim, Noxa, and PUMA), which are associated with mitochondrial apoptosis in oral cancer cells via the loss of mitochondrial membrane potential. HCUA triggered the production of intracellular reactive oxygen species (ROS) that was ascertained to be involved in HCUA-induced apoptosis by the ROS inhibitors YCG063 and N-acetyl-L-cysteine. As a result, HCUA had potential antitumor activity to oral cancer cells through eliciting ROS-dependent and p53-mediated mitochondrial apoptosis. Overall, HCUA could be applicable for the development of anticancer agents against human oral cancer.

        Transgenic Rice Overexperessing a Tomato Mitochondrial Phosphate Transporter, SlMPT3;1, Promotes Phosphate Uptake and Increases Grain Yield

        Mitochondrial phosphate transporter plays animportant regulatory role in promoting the uptake andtransport of phosphate in plants. In this study, the SlMPT3;1gene, a member of mitochondrial phosphate transporterfamily in tomato, was isolated and transformed into the riceOryza sativa L. ssp. japonica cultivar Kitaake. The SlMPT3;1 islocalized to the mitochondrial membrane and functions incompensating the phosphate uptake in yeast MB192 mutantthat is defective in phosphate transport under Pi deficiency. RT-qPCR showed that the SlMPT3;1 is expressed in all oftomato tissues, but highly accumulated in the young leavesand stems under Pi deficiency. The data demonstrated that atleast two copies of the SlMPT3;1 gene are inserted into therice genome, and the transcripts of the SlMPT3;1 mRNA arehighly accumulated in the roots of the transgenic rice. Theoverexpression of the SlMPT3;1 gene not only promotesphosphate uptake by the roots, but also increases thetranslocation of phosphate from the roots to the shoots in thetransgenic rice. The transgenic rice accumulated morechlorophyll and soluble sugar in the shoots than the wildtype under Pi deficiency. Microassay sequencing showedthat the differentially expressed genes in the transgenic riceare mainly involved in the regulations of biological processand molecular function under Pi deficiency. Further RTqPCRanalyses revealed that the differentially expressedgenes, which are involved in the regulations of the biologicalprocess, cell component, and molecular function, are upregulatedunder Pi deficiency, and exhibit similar expressiontrends to the relative expression folds of these partial differentiallyexpressed genes in the transcriptomic analyses. This studysuggests that the overexpression of the SlMPT3;1 gene promotedthe uptake and transport of phosphate in rice, thus leading to anenhanced increase in tiller number and effective panicle of perplant, and increasing grain yield under Pi deficiency.

        A phase II trial of cytoreductive surgery combined with niraparib maintenance in platinum-sensitive, secondary recurrent ovarian cancer: SGOG SOC-3 study

        Background: In China, secondary cytoreductive surgery (SCR) has been widely used in ovarian cancer (OC) over the past two decades. Although Gynecologic Oncology Group-0213 trial did not show its overall survival benefit in first relapsed patients, the questions on patient selection and effect of subsequent targeting therapy are still open. The preliminary data from our pre-SOC1 phase II study showed that selected patients with second relapse who never received SCR at recurrence may still benefit from surgery. Moreover, poly(ADP-ribose) polymerase inhibitors (PARPi) maintenance now has been a standard care for platinum sensitive relapsed OC. To our knowledge, no published or ongoing trial is trying to answer the question if patient can benefit from a potentially complete resection combined with PARPi maintenance in OC patients with secondary recurrence. Methods: SOC-3 is a multi-center, open, randomized, controlled, phase II trial of SCR followed by chemotherapy and niraparib maintenance vs chemotherapy and niraparib maintenance in patients with platinum-sensitive second relapsed OC who never received SCR at recurrence. To guarantee surgical quality, if the sites had no experience of participating in any OC-related surgical trials, the number of recurrent lesions evaluated by central-reviewed positron emission tomography–computed tomography image shouldn't be more than 3. Eligible patients are randomly assigned in a 1:1 ratio to receive either SCR followed by 6 cycles of platinum-based chemotherapy and niraparib maintenance or 6 cycles of platinum-based chemotherapy and niraparib maintenance alone. Patients who undergo at least 4 cycles of chemotherapy and must be, in the opinion of the investigator, without disease progression, will be assigned niraparib maintenance. Major inclusion criteria are secondary relapsed OC with a platinum-free interval of no less than 6 months and a possibly complete resection. Major exclusion criteria are borderline tumors and non-epithelial ovarian malignancies, received debulking surgery at recurrence and impossible to complete resection. The sample size is 96 patients. Primary endpoint is 12-month non-progression rate. Trial Registration: ClinicalTrials.gov Identifier: NCT03983226
