Although the possible cellular roles of several ubiquitin-specific proteases (UBPs) were identified in Arabidopsis, almost nothing is known about UBP homologs in rice, a monocot model plant. In this report, we searched the rice genome database (http://signal.salk.edu/cgi-bin/RiceGE) and identified 21 putative UBP family members (OsUBPs) in the rice genome. These OsUBP genes each contain a ubiquitin carboxyl-terminal hydrolase (UCH) domain with highly conserved Cys and His boxes and were subdivided into 9 groups based on their sequence identities and do-main structures. RT-PCR analysis indicated that rice OsUBP genes are expressed at varying degrees in differ-ent rice tissues. We isolated a full-length cDNA clone for OsUBP6, which possesses not only a UCH domain, but also an N-terminal ubiquitin motif. Bacterially expressed OsUBP6 was capable of dismantling K48-linked tetra-ubiquitin chains in vitro. Quantitative real-time RT-PCR indicated that OsUBP6 is constitutively expressed in dif-ferent tissues of rice plants. An in vivo targeting experi-ment showed that OsUBP6 is predominantly localized to the nucleus in onion epidermal cells. We also examined how knock-out of OsUBP6 affects developmental growth of rice plants. Although homozygous T3 osubp6 T-DNA insertion mutant seedlings displayed slower growth rela-tive to wild type seedlings, mature mutant plants appeared to be normal. These results raise the possibility that loss of OsUBP6 is functionally compensated for by an as-yet unknown OsUBP homolog during later stages of devel-opment in rice plants.
초등학교 2학년 슬기로운 생활에서 창의적 과학글쓰기 프로그램이 창의성과 과학적 태도에 미치는 영향

A creative science writing program was developed, and applied to 2nd grade elementary students. Two 2nd grade elementary school classes in Seoul district were selected for the current study. A Pretest-Posttest Control Group Design was applied. The experimental group adopted the creative science writing program and the control group maintained traditional lectures for 12 classes. TTCT (Torrance Test of Creative Thinking) figural test and a scientific attitude test were conducted on both groups. According to the analysis, TTCT score of the experimental group was significantly higher than that of the control group (p<.01). Among several subscales of TTCT, elaboration and resistance to premature closure was especially improved. Also, scientific attitude scale of the experimental group was significantly higher than that of the control group (p<.01). Among subscales of scientific attitude, the creative science writing program was effective on improving students`` open-mindedness, critical mindedness, cooperation, voluntariness, and endurance. Gender was not a factor in the improvement of both students`` creativity and scientific attitude.
Endophthalmitis due to Streptococcus agalactiae of a serotype V strain in a healthy adult

Background: Streptococcus agalactiae, a group B streptococcus (GBS), is a normal flora. Endogenous endophthalmitis caused by GBS is an extremly rare but visually devastating disease. Here, we describe the first-reported Korean case of GBS infective endocarditis presenting as endogenous endophthalmitis and skin and soft tissue infection. Case Report: A 43-year old female without any underlying medical condition visited our emergency room complaining of a 10-hour history of decreased visual acuity with ocular pain on the right eye. Physical examination revealed a 3 cm-sized skin swelling with purulent discharge on right buttock area which was tender and erythematous. Ophthalmic examination revealed endogenous endophthalmitis on the right eye. Penicillin-susceptible S. agalactiae, sequence type 1 associated with a serotype V, was identified as the causative organism on the specimens from the eyeball, blood, and skin pus of right buttock. For the first 12 days of hospitalization, intravenous ceftriaxone (2 g/day) was prescribed with the regimen of intravitreal vancomycin (1 mg/0.1 mL) and ceftazidime (2.25mg/0.1 mL). However, her visual acuity rapidly deteriorated to light perception within a day. On the third day of hospitalization, she underwent a therapeutic vitrectomy of right eye. Ten days after initially normal transthoracic echocardiography, transesophageal echocardiography revealed a 5 mm-sized vegetation attached to anterior mitral leaflet. The patient was finally diagnosed as invasive GBS infection with infective endocarditis presented as endogenous endophthalmitis and skin and soft tissue infection. At the 6-month follow-up visit after completion of antibiotic therapy for 40 days, the patient remained free of any complications, except phthisis bulbi of the right eye without perception to light and visual acuity. Conclusions: In conclusion, this case suggests that GBSinfections should be added to the list for differential diagnosis of endogenous endophthalmitis even in healthy adults. Our report also highlights the requirement of a high index of suspicion for infective endocarditis among them.
Mitoribosome insufficiency in β cells is associated with type 2 diabetes-like islet failure

Genetic variations in mitoribosomal subunits and mitochondrial transcription factors are related to type 2 diabetes. However, the role of islet mitoribosomes in the development of type 2 diabetes has not been determined. We investigated the effects of the mitoribosomal gene on β-cell function and glucose homeostasis. Mitoribosomal gene expression was analyzed in datasets from the NCBI GEO website (GSE25724, GSE76894, and GSE76895) and the European Nucleotide Archive (ERP017126), which contain the transcriptomes of type 2 diabetic and nondiabetic organ donors. We found deregulation of most mitoribosomal genes in islets from individuals with type 2 diabetes, including partial downregulation of CRIF1. The phenotypes of haploinsufficiency in a single mitoribosomal gene were examined using β-cell-specific Crif1 (Mrpl59) heterozygous-deficient mice. Crif1beta+/− mice had normal glucose tolerance, but their islets showed a loss of first-phase glucose-stimulated insulin secretion. They also showed increased β-cell mass associated with higher expression of Reg family genes. However, Crif1beta+/− mice showed earlier islet failure in response to high-fat feeding, which was exacerbated by aging. Haploinsufficiency of a single mitoribosomal gene predisposes rodents to glucose intolerance, which resembles the early stages of type 2 diabetes in humans.
