        Exosomal CircPRRX1 Enhances Doxorubicin Resistance in Gastric Cancer by Regulating MiR-3064-5p/PTPN14 Signaling

        Purpose: Gastric cancer (GC) is a malignant tumor with a high mortality rate. Drug resistance is a major obstacle to GC therapy. This study aimed to investigate the role and mechanism of exosomal circPRRX1 in doxorubicin resistance in GC. Materials and Methods: HGC-27 and AGS cells were exposed to different doses of doxorubicin to construct doxorubicin-resistantcell lines. Levels of circPRRX1, miR-3064-5p, and nonreceptor tyrosine phosphatase 14 (PTPN14) were detected by quantitativereal-time PCR or Western blot assay. Then, 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide, transwell,and Western blot assays were used to explore the function of circPRRX1 in GC cells. Interactions among circPRRX1, miR-3064-5p,and PTPN14 were confirmed by dual-luciferase reporter assay. The in vivo function of circPRRX1 was analyzed in a xenograft tumormodel. Results: CircPRRX1 was highly expressed in doxorubicin-resistant GC cell lines. Knockdown of circPRRX1 reversed doxorubicinresistance in doxorubicin-resistant GC cells. Additionally, extracellular circPRRX1 was carried by exosomes to spread doxorubicinresistance. CircPRRX1 silencing reduced doxorubicin resistance by targeting miR-3064-5p or regulating PTPN14. In GC patients,high levels of circPRRX1 in serum exosomes were associated with poor responses to doxorubicin treatment. Moreover, depletionof circPRRX1 reduced doxorubicin resistance in vivo. Conclusion: CircPRRX1 strengthened doxorubicin resistance by modulating miR-3064-5p/PTPN14 signaling and might be atherapeutic target for GC patients.

        Long-term and stable correction of uremic anemia by intramuscular injection of plasmids containing hypoxia-regulated system of erythropoietin expression

        Relative deficiency in production of glycoprotein hormone erythropoietin (Epo) is a major cause of renal anemia. This study planned to investigate whether the hypoxia-regulated system of Epo expression, constructed by fusing Epo gene to the chimeric phosphoglycerate kinase (PGK) hypoxia response elements (HRE) in combination with cytomegalovirus immediate-early (CMV IE) basal gene promoter and delivered by plasmid intramuscular injection, might provide a long-term physiologically regulated Epo secretion expression to correct the anemia in adenine-induced uremic rats. Plasmid vectors (pHRE-Epo) were synthesized by fusing human Epo cDNA to the HRE/CMV promoter. Hypoxia-inducible activity of this promoter was evaluated first in vitro and then in vivo in healthy and uremic rats (n = 30 per group). The vectors (pCMV-Epo) in which Epo expression was directed by a constitutive CMV gene promoter served as control. ANOVA and Student’s t-test were used to analyze between-group differences. A high-level expression of Epo was induced by hypoxia in vitro and in vivo. Though both pHRE-Epo and pCMV-Epo corrected anemia,the hematocrit of the pCMV-Epo-treated rats exceeded the normal (P < 0.05), but that of the pHRE-Epo-treated rats didn’t. Hypoxia-regulated system of Epo gene expression constructed by fusing Epo to the HRE/CMV promoter and delivered by plasmid intramuscular injection may provide a long-term and stable Epo expression and secretion in vivo to correct the anemia in adenine-induced uremic rats.

        Comparative Evaluation of Hepatoprotective Activities of Geniposide, Crocins and Crocetin by CCl<sub>4</sub>-Induced liver Injury in Mice

        Iridoid glycosides (mainly geniposide) and crocetin derivatives (crocins) are the two major active constituents in Gardenia jasminoides Ellis. In the present study, geniposide, crocins, crocin-1 and crocetin were separated from gardenia chromatographically. Then, mice were orally administrated with geniposide (400 mg/kg b.w.), crocins (400 mg/kg b.w.), crocin-1 (400 mg/kg b.w.) and crocetin (140 mg/kg b.w.) once daily for 7 days with $CCl_4$. Hepatoprotective properties were evaluated by biochemical parameters: Administration of geniposide, crocins, crocin-1and crocetin significantly lowered serum alanine transaminase (ALT), aspartate transaminase (AST) and alkaline phosphatase (ALP) levels in $CCl_4$-treated mice. The reduced glutathione (GSH) levels and antioxidant enzymes (SOD and CAT) activities were also increased by geniposide, crocins, crocin-1 and crocetin. Histopathological examination of livers showed that these components reduced deformability, irregular arrangement and rupture of hepatocyte in $CCl_4$-treated mice. These biochemical results and liver histopathological assessment demonstrated that geniposide, crocetin derivatives and crocetin show comparative beneficial effects on $CCl_4$-induced liver damage via induction of antioxidant defense. Therefore, contents of geniposide and crocetin derivatives should be both considered for hepatoprotective efficacy of Gardenia jasminoides Ellis.

        Anti-Reactive Jamming Technology Based on Jamming Utilization

        Since the existing anti-jamming methods, including intelligent methods, have difficulty against high-speed reactive jamming, we studied a new methodology for jamming utilization instead of avoiding jamming. Different from the existing jamming utilization techniques that harvest energy from the jamming signal as a power supply, our proposed method can take the jamming signal as a favorable factor for frequency detection. Specifically, we design an intelligent differential frequency hopping communication framework (IDFH), which contains two stages of training and communication. We first adopt supervised learning to get the jamming rule during the training stage when the synchronizing sequence is sent. And then, we utilize the jamming rule to improve the frequency detection during the communication stage when the real payload is sent. Simulation results show that the proposed method successfully combated high-speed reactive jamming with different parameters. And the communication performance increases as the power of the jamming signal increase, hence the jamming signal can help users communicate in a low signal-to-noise ratio (SNR) environment.

        PREPARATION AND CHARACTERIZATION OF SILVER NANOPARTICLES/CARBON NANOFIBERS VIA ELECTROSPINNING WITH RESEARCH ON THEIR CATALYTIC PROPERTIES

        Ag nanoparticles (Ag NPs) embedded carbon nano¯bers (CNFs) were prepared by a new routewhich included chemical reaction process, electrospinning and calcination technique. The mor-phology and structure of the composite nano¯bers were investigated by scanning electron mi-croscopy, energy dispersive X-ray spectroscopy, transmission electron microscopy and X-raydi®raction. It indicated that Ag NPs were uniformly distributed in the CNFs. This e®ectivesynthesis method can be used to prepare other composite nano¯bers with functionality. The AgNPs/CNFs that served as supported catalysts were used in the styrene epoxidation by TBHP. The Ag NPs/CNFs catalyst showed its highly catalytic activity for the epoxidation of styrene(conversion: 40.6%, SO selectivity: 35.9%). This kind of composite nano¯ber membrane wasproved e®ectively catalytic activity and recycled easily in the styrene epoxidation.
