        The complete mitochondrial genome of Mahanta tanyae compared with other zygaenoid moths (Lepidoptera: Zygaenoidea)

        The complete mitochondrial genome (mitogenome) of Mahanta tanyae was sequenced and extensively compared with all seven additionally reported zygaenoid mitogenomes. The M. tanyae mitogenome is circular, doublestranded, and 15,323 bp long. Gene content, gene order, and orientation are all typical of Lepidoptera, despite the existence of gene rearrangements for some other zygaenoid mitogenomes. Comparative analyses further showed that the incomplete termination codon T is consistently recognized in the mitochondrial cox1, cox2 and nad4 genes of all zygaenoid species, as well as in the nad5 gene in two limacodid species. Among 13 proteincoding genes, nad6 exhibits the highest evolutionary rate. The structure for each tRNA is highly conserved, including loss of the dihydorouidine (DHU) arm in trnS1 (AGN), but remarkable nucleotide variation exists, primarily in the pseudouridine (TψC) loops. Interestingly, in four species of Zygaenidae, the anticodons for trnS1 (AGN) are consistently UCU, instead of the routinely used codon GCU, in all three species of Limacodidae. In the intergenic region between trnS2 and nad1, a short sequence before the motif “ATACTAA” is present in the M. tanyae mitogenome that is unique among reported zygaenoid mitogenomes. In the A + T-rich region between the motif “ATTTA” and the microsatellite (AT) n element, some nucleotides were present for most zygaenoid mitogenomes, which is, to our knowledge, rare even in reported lepidopteran mitogenomes. Phylogenetic analyses based on the combined 37 mitochondrial genes confirmed the position of M. tanyae in Limacodidae of the Zygaenoidea.

        The unascertained clustering is a new clustering method, which combines unascertained theory and clustering theory to construct the unascertained measure, and uses the unascertained measure as set membership to indicate the membership relation between the samples with the different classes. It overcomes the disadvantage of means clustering algorithm, that a sample definitely belongs to a class, which made greater progress than -means clustering. There are complex non-linear relationship between the coal industry competitiveness and various factors. The article established the evaluation influencing factors system of coal industry international competitiveness. 6 unascertained clustering method to cluster competitiveness. It found out each class center, and gave the membership degree of the samples belong to each class, which better resolved the problem of classifying the coal industry international competitiveness.

        Four new species of the genus Rhagonycha (Coleoptera: Cantharidae) from Southeast China

        Four new species of Rhagonycha Eschscholtz, 1830 are described, including Rh. hainanensis sp. nov. (China, Hainan), Rh. nigroimpressimima sp. nov. (China, Fujian), Rh. tumida sp. nov. and Rh. nigrotibia sp. nov. (China, Guangxi). These new species are illustrated with habitus, aedeagi, abdominal sternites VIII and internal genitalia of females. Besides, the female internal genitalia of Rh. nigroimpressa (Pic, 1922a) is illustrated for the first time to make comparison with the new species. An identification key and a distribution map of those Rhagonycha species with yellowish elytra from southeast China are present.

        Complete mitochondrial genome of the hemp borer, Grapholita delineana (Lepidoptera: Tortricidae): Gene variability and phylogeny among Grapholita

        The mitochondrial genome (mitogenome) has been extensively used in phylogenetics and species-level evolu tionary investigations. The lepidopteran family Tortricidae (leaf-roller moths), including the genus Grapholita, contains numerous species of economic importance, but for the majority of Grapholita species, their mitogenomes remain poorly studied. Here, we sequence and annotate the full mitogenome of Grapholita delineana, an important pest of hemp worldwide and compare it with the mitogenomes of two congeneric species available from GenBank. The G. delineana mitogenome is 15,599 bp long, including 37 typical mitochondrial genes and an A + T-rich region. Gene content, order and orientation are identical to other reported tortricid mitogenomes. Analyses of nucleotide diversity, Ka/Ks, genetic distance and number of variable sites together suggest that nad6 is the fastest-evolving gene among the mitochondrial PCGs of Grapholita. Our analyses indicate that Grapholita, as presently defined, is not monophyletic, confirming previous morphological and multiple-gene studies, using mitogenomic evidence. Our study provides information on comparative mitogenomics of Tortricidae especially Grapholita.

        Five new species of Stenothemus Bourgeois from Guangxi, China (Coleoptera, Cantharidae)

        Five new species of Stenothemus Bourgeois, 1907 from Guangxi, China are described, S. normalis sp. nov., S. flavicans sp. nov., S. nigripennis sp. nov., S. elongaticeps sp. nov. and S. maculaticollis sp. nov.. They are illus trated with habitus photos and aedeagi of males, also abdominal sternites VIII and internal genitalia of females. A key for the identification of the above species is provided.

        The effects of an innovative pulping technique of synchronously pulping and gelatinizing treatment on raw materials properties, oenological parameters, fermentation process, and flavor characteristics of glutinous rice wine

        Liquid-state fermentation has been increasingly applied in the industrial glutinous rice wine (GRW) production. However, products brewed by this emerging technique possess some deficiencies in flavor quality. Therefore, this study firstly developed and optimized an innovative pulping technique by the synchronously pulping and gelatinizing treatment (Process I) to improve GRW flavor quality, and then revealed the influences of Process I on raw materials properties, oenological parameters, fermentation process, and flavor characteristics of GRW. Results show that Process I significantly (p < 0.05) enriched the soluble solid and crude protein content of glutinous rice milk by improving gelatinization degree and pulping efficiency, which consequently enhanced the microbial growth, glycolysis, and protein decomposition during the GRW fermentation process. GC–MS analysis shows that Process I sequentially significantly (p < 0.05) enhanced the esterification and Ehrlich or Harrison pathway during the fermentation process. This contributed to a higher content of key ester and alcohol compounds.

        Insect cytokine growth-blocking peptide may regulate density-dependent phase trait of cuticular melanization in the larval armyworm, Mythimna separata

        Growth-blocking peptide (GBP) is a 25 amino acid insect cytokine found in lepidopteran insects that has diverse biological activities such as larval growth regulation, paralysis induction, cell proliferation and stimulation of immune cells. Density-dependent phase polyphenism is a phenomenon of phenotypic plasticity in which the expression of a variety of traits can be affected by local population density. In the present study, the armyworm Mythimna separata larvae with four rearing densities (1 larva/vial, 2 larvae/vial, 4 larvae/vial and 6 larvae/vial) were tested for cuticular melanization and body weight throughout the third-fifth instar, and the functional role of GBP in regulating the changes was investigated. The results indicated that when reared at high densities, the larvae exhibited less body weight and more degree of cuticular melanization than larvae reared at low densities. The gene expression of GBP in armyworm larvae showed an initial rise and then decline trend with increased rearing densities in the third to fifth instar. Compared with control, more degree of cuticular melanization was observed in GBP-injected larvae (500 ng/larva in volume 50 μL) than that in Ringer’s solution-injected counterparts. Furthermore, the gene expression level of dopa decarboxylase and prophenoloxidase increased significantly in GBP-injected fifth instar larvae from 6 h to 12 h after injection, suggesting the role of GBP in modulating density-dependent phase trait of armyworm cuticular melanization.

        ATP6V0d2 Suppresses Alveoli Macrophage Alternative Polarization and Allergic Asthma via Degradation of PU.1

        Purpose Macrophages are important regulators of environmental allergen-induced airway inflammation and asthma. ATP6V0d2 is a subunit of vacuolar ATPase highly expressed in macrophages. However, the functions of ATP6V0d2 in the regulation of pathogenesis of allergic asthma remain unclear. The aim of this study is to determine the function and related molecular mechanisms of macrophage protein ATP6V0d2 in allergic asthma. Methods We compared the disease severity between female C57BL/6 wild-type and ATP6V0d2−/− mice in an ovalbumin (OVA)-induced asthma model. We also investigated the association of expression of ATP6V0d2, PU.1 and CCL17 with disease severity among asthmatic patients. Results The expression of ATP6V0d2 in sputum cells of asthmatic patients and in the lungs of OVA-challenged mice was enhanced compared to healthy subjects and their counterparts, respectively. However, ATP6V0d2-deficient mice exaggerated inflammatory cell infiltration as well as enhanced alternative activated macrophage (AAM) polarization and mucus production in an OVA-induced asthma model. Furthermore, we found that Atp6v0d2 promoted lysosomal degradation of Pu.1, which induced AAM polarization and Ccl17 production. Among asthma patients, ATP6V0d2 expression was inversely associated with disease severity, whereas PU.1 and CCL17 expression was positively associated with disease severity. Conclusions Our results identify macrophage Atp6v0d2, as an induced feedback inhibitor of asthma disease severity by promoting Pu.1 lysosomal degradation, which may in turn leads to reduced AAM polarization and Ccl17 production.

        Prescribed Performance-tangent Barrier Lyapunov Function for Adaptive Neural Backstepping Control of Variable Stiffness Actuator with Input and Output Constraints

        Due to the complexity of modeling and the strong transmission coupling, the rich background of rigid actuator control has not been transferred to variable stiffness actuator (VSA). Therefore, most model-based control techniques developed for VSA require feedback linearization first. Alternatively, VSA can use non-model-based control techniques such as PD control, but it does not show strong robustness under disturbances. This paper is concerned with designing a novel adaptive neural network backstepping control scheme without using feedback linearization for a special VSA with saturation inputs, output constraints, and disturbances. Firstly, for ensuring the VSA with lower tracking error and higher security, the prescribed performance-tangent barrier Lyapunov function (PP-TBLF) is introduced to handle the prescribed output performance constraints. Subsequently, the Chebyshev neural network and the Nussbaum-type function are exploited to approximate the unknown nonlinearities and unknown gains. Meanwhile, the inverse hyperbolic sine function tracking differentiator is utilized to solve the “explosion of complexity” caused by the differentiation of virtual inputs and also approximate the complex partial derivatives caused by the auxiliary control signals. Finally, the stability of the whole scheme is proved by the Lyapunov criterion. The simulation results illustrate the raised control scheme’s feasibility and show a better closed-loop behavior relative to that obtained using a classic PD controller.
