Transcriptome analysis of the livers of ducklings hatched normally and with assistance

        Yali Liu,Shishan He,Tao Zeng,Xue Du,Junda Shen,Ayong Zhao,Lizhi Lu 아세아·태평양축산학회 2017 Animal Bioscience Vol.30 No.6

Objective: “Hatchability” is an important economic trait in domestic poultry. Studies on poultry hatchability focus mainly on the genetic background, egg quality, and incubation conditions, whereas the molecular mechanisms behind the phenomenon that some ducklings failed to break their eggshells are poorly understood. Methods: In this study, the transcriptional differences between the livers of normally hatched and assisted ducklings were systematically analyzed. Results: The results showed that the clean reads were de novo assembled into 161,804 and 159,083 unigenes (≥200-bp long) by using Trinity, with an average length of 1,206 bp and 882 bp, respectively. The defined criteria of the absolute value of log2 fold-change ≥1 and false discovery rate≤0.05 were differentially expressed and were significant. As a result, 1,629 unigenes were identified, the assisted ducklings showed 510 significantly upregulated and 1,119 significantly down-regulated unigenes. In general, the metabolic rate in the livers of the assisted ducklings was lower than that in the normal ducklings; however, compared to normal ducklings, glucose- 6-phosphatase and ATP synthase subunit alpha 1 associated with energy metabolism were significantly upregulated in the assisted group. The genes involved in immune defense such as major histocompatibility complex (MHC) class I antigen alpha chain and MHC class II beta chain 1 were downregulated in the assisted ducklings. Conclusion: These data provide abundant sequence resources for studying the functional genome of the livers in ducks and other poultry. In addition, our study provided insight into the molecular mechanism by which the phenomenon of weak embryos is regulated.

      • Image Denoising Method based on Threshold, Wavelet Transform and Genetic Algorithm

Yali Liu 보안공학연구지원센터 2015 International Journal of Signal Processing, Image Vol.8 No.2

In the process of image acquisition and transmission, noise is always contained inevitably. So it is necessary to image denoising processing to improve the quality of image. Generally speaking, each algorithm has some filtering and threshold parameters. Taking variety kinds of images into account, it is a key problem of how to set these parameters in denoising algorithms under different conditions to achieve better performance. There are many algorithms for the determination of the parameters, and each of them has its application field. Because the wavelet transform has good performance, therefore, it has been widely applied as a kind of signal and image processing tools. In this paper, wavelet transform is used in the image denoising, and the genetic algorithm is used to estimate the denoising results. Experimental results show the validity of the new algorithm.

Transcriptomic and proteomic profiling revealed global changes in Streptococcus thermophilus during pH-controlled batch fermentations

        Yali Qiao,Cong Leng,Gefei Liu,Yanjiao Zhang,Xuepeng Lv,Hongyu Chen,Jiahui Sun,Zhen Feng 한국미생물학회 2019 The journal of microbiology Vol.57 No.9

Understanding global changes of physiological processes at the molecular level during the growth of Streptococcus thermophilus is essential for the rational design of cultivation media and the optimization of bioprocesses. Transcriptomics and proteomics were combined to investigate the global changes at the transcript and protein level during the growth of S. thermophilus. The expression of 1396 genes (FDR ≤ 0.001) and 876 proteins (P < 0.05) changed significantly over time. The most remarkable growth phase dependent changes occurred in the late-lag phase and were related to heterofermentation, glycolysis, peptidoglycan biosynthesis, conversion between amino acids and stress response. The present results could provide theoretical guidance for high-cell-density culture, help design cultivation media, and help attain a high biomass of S. thermophilus.

Panduratin A Inhibits Cell Proliferation by Inducing G0/G1 Phase Cell Cycle Arrest and Induces Apoptosis in Breast Cancer Cells

        Liu, Qiuming,Cao, Yali,Zhou, Ping,Gui, Shimin,Wu, Xiaobo,Xia, Yong,Tu, Jianhong The Korean Society of Applied Pharmacology 2018 Biomolecules & Therapeutics(구 응용약물학회지) Vol.26 No.3

Because of the unsatisfactory treatment options for breast cancer (BC), there is a need to develop novel therapeutic approaches for this malignancy. One such strategy is chemotherapy using non-toxic dietary substances and botanical products. Studies have shown that Panduratin A (PA) possesses many health benefits, including anti-inflammatory, anti-bacterial, anti-oxidant and anticancer activities. In the present study, we provide evidence that PA treatment of MCF-7 BC cells resulted in a time- and dose-dependent inhibition of cell growth with an $IC_{50}$ of $15{\mu}M$ and no to little effect on normal human MCF-10A breast cells. To define the mechanism of these anti-proliferative effects of PA, we determined its effect critical molecular events known to regulate the cell cycle and apoptotic machinery. Immunofluorescence and flow cytometric analysis of Annexin V-FITC staining provided evidence for the induction of apoptosis. PA treatment of BC cells resulted in increased activity/expression of mitochondrial cytochrome C, caspases 7, 8 and 9 with a significant increase in the Bax:Bcl-2 ratio, suggesting the involvement of a mitochondrial-dependent apoptotic pathway. Furthermore, cell cycle analysis using flow cytometry showed that PA treatment of cells resulted in G0/G1 arrest in a dose-dependent manner. Immunoblot analysis data revealed that, in MCF-7 cell lines, PA treatment resulted in the dose-dependent (i) induction of $p21^{WAF1/Cip1}$ and p27Kip1, (ii) downregulation of Cyclin dependent kinase (CDK) 4 and (iii) decrease in cyclin D1. These findings suggest that PA may be an effective therapeutic agent against BC.

Fuzzy comprehensive performance evaluation method of rolling linear guide based on improved analytic hierarchy process

        Yali Ma,Jiayong Wei,Cancan Li,Chen Liang,Guochao Liu 대한기계학회 2020 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.34 No.7

Aiming at the problem that the multi-objective evaluation of performance and the weighting coefficients of indexes is difficult to find, a fuzzy comprehensive performance evaluation method of rolling linear guide (RLG) is proposed. Considering the static and dynamic indexes, a structure model containing nine weighting coefficients was constructed. The performance evaluation function was established by the compromise programming method and the average power method, while weighting coefficients were determined by the improved analytic hierarchy process (I-AHP); the mathematical model was obtained to evaluate the RLG in a certain application. The evaluation results show that the confidence of the static, dynamic and comprehensive performance of the RLG is 59.21 %, 81.97 %, and 63.34 %, respectively. The feasibility of the method was verified by numerical simulation. This method indicates the pros and cons of the performance of the RLG and also indicates the direction for its optimized design.

      • Calculation of PI Controller Stable Region Based on D-Partition Method

        Liu Jinggong,Xue Yali,Li Donghai 제어로봇시스템학회 2010 제어로봇시스템학회 국제학술대회 논문집 Vol.2010 No.10

Based on D-partition method, a fast calculation approach for PI controller stable region is introduced. The boundary of the PI stable region is calculated first using D-partition method, and then several sets of equations and inequalities are introduced sequentially to judge which side of the boundary is stable. The approach is simple, and can be applied to any self-stable systems with or without time-delays, while only the process frequency responses are needed. To illustrate its feasibility, some examples are given.

      • Caspofungin combined with hormones as preemptive therapy of chemotherapy-induced disseminated candidiasis in a patient

        Yali, Liang,Zhichao, Qiu,Yaohe, Li,Anping, Liu,Zhixiong, Chen,Huliwen, Huliwen,man, Luo,jing, He,Xiaoyang, Xiaoyang,Hai, Lan Techno-Press 2021 Advances in nano research Vol.10 No.6

Disseminated candidiasis (DC) arising from nosocomial fungal infection is a life-threatening complication in critically ill, nonneutropenic patients. The overall nosocomial fungal infection rate in United States hospitals doubled from 1980-1990. Until recently, amphotericin B was the only agent available for the treatment of life-threatening candidal infections, but its use is plagued by toxicities including nephrotoxicity and infusion-related reactions such as rigors and hypotension. The availability of fluconazole, which is regarded more much less toxic than amphotericin B, prompted a surge in research to determine if it is as efficacious in the management of candidemia and hematogenously disseminated candidiasis. Complicating the interpretation of studies is the broad range of infection severity, from candidemia that may be transient and self-limiting to life-threatening hematogenously disseminated candidiasis. This study has used the models of Artificial neural network (ANN) and Support Vector regression (SVR) to accurately assess the clinical trials comparing fluconazole and amphotericin B demonstrate the efficacy of fluconazole for catheter-associated candidemia in critically ill patients when the likely pathogen is Candida albicans. As a result, Amphotericin B should remain the first-line agent for the management of candidemia and hematogenously disseminated candidiasis in all other patients. Also, SVR could accurately assess the efficacy of fluconazole for catheter-associated candidemia in critically ill patients.

A Comparison of Eu-Doped -Fe2O3 Nanotubes and Nanowires for Acetone Sensing

        Yali Cheng,Yifang Wang,Jinbao Zhang,Haiying Li,Li Liu,Yu Lina,Liting Du,Haojie Duan 성균관대학교(자연과학캠퍼스) 성균나노과학기술원 2017 NANO Vol.12 No.11

Pure and Eu-doped (1.0, 3.0, 5.0 wt.%) α-Fe2O3 (PFO and EFO) nanotubes and nanowires have been successfully synthesized through the combination of electrospinning and calcination techniques. The structures, morphologies and chemical compositions of the as-obtained products were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), thermogravimetric and differential scanning calorimetry (TG-DSC) and energy dispersive spectrum (EDS), respectively. To demonstrate the superior gas sensing performance of the doped nanotubes, a contrastive gas sensing study between PFO (EFO) nanotubes and nanowires was performed. It turned out that Eu doping could magnify the impact of morphology on gas sensitivity. Specifically, at the optimum operating temperature of 240 ℃, the response value of PFO nanotubes to 100 ppm acetone is slightly higher than that of nanowires (3.59/2.20). EFO (3.0 wt.%) nanotubes have a response of 84.05, which is almost 2.7 times as high as that of nanowires (31.54). Moreover, they possess more rapid response/recovery time (11 s and 36 s, respectively) than nanowires (17 s and 40 s, respectively). The lowest detection limit for acetone is 0.1 ppm and its response is 2.15. In addition, both of EFO nanotubes and nanowires sensors have a good linearity (0.1–500 ppm) and favorable selectivity in acetone detection.
