The prevalence of BRCA1 gene mutations in breast cancer differs between diverse ethnic groups. Relatively little information is known about patterns of BRCA1 mutations in early-onset breast cancer in women of Uighur or Han descent, the major ethnic populations of the Xinjiang region in China. The aim of this study was to identify BRCA1 mutations in Uighur and Han patients with early-onset (age <35 years), and sporadic breast cancer for genetic predisposition to breast cancer. For detection of BRCA1 mutations, we used a polymerase chain reaction single-stranded conformation polymorphism approach, followed by direct DNA sequencing in 22 Uighur and 13 Han women with early-onset sporadic breast cancer, and 32 women with benign breast diseases. The prevalence of BRCA1 mutations in this population was 22.9% (8/35) among early-onset sporadic breast cancer cases. Of these, 31.8% (7/22) of Uighur patients and 7.69% (1/13) of Han patients were found to have BRCA1 mutations. In 7 Uighur patients with BRCA1 mutations, there were 11 unique sequence alterations in the BRCA1 gene, including 4 clearly disease-associated mutations on exon 11 and 3 variants of uncertain clinical significance on exon 11, meanwhile 4 neutral variants on intron 20 or 2. None of the 11 BRCA1 mutations identified have been previously reported in the Breast Cancer Information Core database. These findings reflect the prevalence of BRCA1 mutations in Uighur women with early-onset and sporadic breast cancer, which will allow for provision of appropriate genetic counseling and treatment for Uighur patients in the Xinjiang region.

Objectives: To explore the expression of astrocyte elevated gene-1 (AEG-1) in cervical cancer and analyze its correlation with microvascular density (MVD), nuclear factor kappaB (NF-kB p65) and vascular endothelial growth factor (VEGF). Materials and Methods: Immunohistochemical MaxVision method was adopted to detect the expression level of AEG-1, NF-kB p65 and VEGF in 45 samples of invading cervical cancer and 12 samples of cervicitis from The First Affiliated Hospital of Wenzhou Medical University. Tumor microvascular endothelial marker CD34 combined with Weidner was used to determine the MVD in cervical cancer tissue. The positive expression and staining conditions of AEG-1, NF-kB p65 and VEGF in cervical cancer tissues were observed under a light microscope. Correlations between expression of AEG-1 protein and those of NF-Kb p65 and VEGF, as well as MVD, were analyzed using Pearson correlation. Results: The expression levels of AEG-1 were $0.186{\pm}0.043$ in cervical cancer and $0.051{\pm}0.002$ in chronic cervicitis (p<0.01). Moreover, expression of AEG-1 was related to vascular invasion and lymphatic metastasis of cervical cancer (p<0.01), but not with age of the patients, differentiation degree, tumour size, pathological type and parametrial infiltration (p>0.05). Pearson correlation analysis showed that the expression of AEG-1 was linked with NF-kB p65 (r=0.501, p=0.000), VEGF (r=0.718, p=0.000) as well as MVD in cervical cancer tissue (r=0.815, p=0.000). Conclusions: AEG-1 is highly expressed in cervical cancer and promotes angiogenesis, which might be related to the fact that AEG-1 activating the signal pathway of NF-kB could up-regulate the level of VEGF expression.

The main pathological mechanism of intervertebral disc degeneration (IVDD) is the programmed apoptosis of nucleus pulposus (NP) cells. Oxidative stress is a significant cause of IVDD. Whether mitophagy is induced by strong oxidative stress in IVDD remains to be determined. This study aimed to investigate the relationship between oxidative stress and mitophagy and to better understand the mechanism of IVDD in vivo and in vitro. To this end, we obtained primary NP cells from the human NP and subsequently exposed them to TBHP. We observed that oxidative stress induced mitophagy to cause apoptosis in NP cells, and we suppressed mitophagy and found that NP cells were protected against apoptosis. Interestingly, TBHP resulted in mitophagy through the inhibition of the HIF-1α/NDUFA4L2 pathway. Therefore, the upregulation of mitochondrial NDUFA4L2 restricted mitophagy induced by oxidative stress. Furthermore, the expression levels of HIF-1α and NDUFA4L2 were decreased in human IVDD. In conclusion, these results demonstrated that the upregulation of NDUFA4L2 ameliorated the apoptosis of NP cells by repressing excessive mitophagy, which ultimately alleviated IVDD. These findings show for the first time that NDUFA4L2 and mitophagy may be potential therapeutic targets for IVDD.
TAZ as a novel regulator of oxidative damage in decidualization via Nrf2/ARE/Foxo1 pathway

TAZ, as a crucial effector of Hippo pathway, is required for spermatogenesis and fertilization, but little is known regarding its physiological function in uterine decidualization. In this study, we showed that TAZ was localized in the decidua, where it promoted stromal cell proliferation followed by accelerated G1/S phase transition via Ccnd3 and Cdk4 and induced the expression or activity of stromal differentiation markers Prl8a2, Prl3c1 and ALP, indicating the importance of TAZ in decidualization. Knockdown of TAZ impeded HB-EGF induction of stromal cell proliferation and differentiation. Under oxidative stress, TAZ protected stromal differentiation against oxidative damage by reducing intracellular ROS and enhancing cellular antioxidant capacity dependent on the Nrf2/ARE/Foxo1 pathway. TAZ strengthened the transcriptional activity of Nrf2 which directly bound to the antioxidant response element (ARE) of Foxo1 promoter region. Additionally, silencing TAZ caused accumulation of intracellular ROS through heightening NOX activity whose blockade by APO reversed the disruption in stromal differentiation. Further analysis revealed that TAZ might restore mitochondrial function, as indicated by the increase in ATP level, mtDNA copy number and mitochondrial membrane potential with the reduction in mitochondrial superoxide. Additionally, TAZ modulated the activities of mitochondrial respiratory chain complexes I and III whose suppression by ROT and AA resulted in the inability of TAZ to defend against oxidative damage to stromal differentiation. Moreover, TAZ prevented stromal cell apoptosis by upregulating Bcl2 expression and inhibiting Casp3 activity and Bax expression. In summary, TAZ might mediate HB-EGF function in uterine decidualization through Ccnd3 and ameliorate oxidative damage to stromal cell differentiation via Nrf2/ARE/Foxo1 pathway.

<P><B>Abstract</B></P> <P>x%Pt-Naf-CV (Pt-Nafion-Cyclic Voltammetry) catalysts with homogeneously distributed platinum nanoparticles and ultra-low Pt loading are successfully synthesized by using a facile potential cycling approach. The as-synthesized 0.8%Pt-Naf-CV catalyst exhibits an enhanced electrocatalytic activity for hydrogen evolution reaction (HER) in 0.5 M H<SUB>2</SUB>SO<SUB>4</SUB> solution, which obtains a low overpotential of 34 mV at 10 mA cm<SUP>−2</SUP>. The linear sweep voltammetry (LSV) curve of 0.8%Pt-Naf-CV catalyst is almost consistent with that of commercial Pt/C. However, the 0.8%Pt-Naf-CV catalyst displays a more excellent stability and durability in comparison with commercial Pt/C. Besides, the Pt loading of Pt/C (Pt–10 wt%) is about 10 times that of 0.8%Pt-Naf-CV catalyst. The improved electrocatalytic performances are derived from the synergistic effects of Pt and Nafion. The Nafion plays a significant role as a dispersant, carrier and structure directing agent on the morphology and size of the Pt catalyst. An experimental investigation on chloride diffusion in structural concrete under the cyclic impact loading is conducted. Effects of magnitudes and loading times are studied. Relationship between damage coefficients and chloride diffusion coefficients is obtained. The cyclic impact loading causes the initiation of new cracks and the propagation of existing cracks, which brings an obvious promotion for chloride diffusion. There should be a damage threshold. If the magnitude of the external loading is large enough to make the damage exceed the threshold, initiation and propagation of cracks become quite significant and chloride diffusion can be greatly promoted. With the increase of the damage coefficient, the chloride diffusion coefficient increases. The increasing rates decrease with long immersion times. Relationship curves of damage coefficients and chloride diffusion coefficients show a good correlation and can be well described with power functions.

The intrinsic relationships between deforming parameters and microstructural mechanisms for Ti-6Al-4V alloy were analyzed by processing maps. A series of thermal compression tests were carried out in the temperatures range of 1023~1323 K (across β-transus) and strain rates range of 0.01~10 s-1 on a Gleeble-3500 thermo-mechanical simulator. Based on the stress-strain data collected from compression tests, a back-propagation artificial neural network (BP-ANN) model was developed, which presents reliable performance in tracking and predicting strain-stress data. By utilizing this model, the volume of stress-strain data was expanded. According to the intensive stress-strain data, the apparent activation energy was calculated to be 564.05 kJ mol-1 and 300.20 kJ mol-1 for α+β-phase field and single β-phase field, respectively. Moreover, the processing maps were constructed at finer intervals of temperature, from which, the stable regions with higher power dissipation efficiency (η > 0.3) and unstable regions with negative instability parameter (ξ < 0) were clarified clearly. By combining processing map with microstructure observations, two main stable softening mechanisms, i.e., globularization and dynamic recovery (DRV) were identified, and globularization-predominant (0.3 < η < 0.55) parameter domain ( < 0.1 s-1) in α+β-phase field and DRV-predominant (0.25 < η < 0.41) parameter domain (0.032 s-1< <1 s-1) in β- phase field were recommended. The aim of this study was to explore the angiographic diagnosis and embolization therapy for renal artery pseudoaneurysms due to acute urinary tract hemorrhage after conservative medical management failed. Seven out of ten cases had fever symptoms after the kidney surgery. The pseudoaneurysms were treated with gelatin sponge and (or) spring coil and the majority demonstrated rapid blockage of hemorrhage. Angiography diagnosis and trans catheter embolization are rapid, safe and effective methods for diagnosis and treatment of renal artery pseudoaneurysms.
