        LncRNA A2M-AS1 lessens the injury of cardiomyocytes caused by hypoxia and reoxygenation via regulating IL1R2

        Background: Myocardial ischemia and reperfusion injury (MI/RI) is a complex pathophysiological process, which can lead to severe myocardial injury. The long noncoding RNA alpha-2-macroglobulin antisense RNA 1 (A2M-AS1) has been revealed to be abnormally expressed in MI, However, its function in MI and the potential mechanism are still unclear. Objective: To evaluate the functional role of A2M-AS1 in hypoxia/reoxygenation (H/R)-induced neonatal cardiomyocytes and its potential molecular mechanism. Methods: Dataset GSE66360 was obtained from GEO database for analyzing the RNA expression of A2M-AS1 and interleukin 1 receptor type 2 (IL1R2). KEGG pathway enrichment analysis of the genes that co-expressed with A2M-AS1 was performed. Human neonatal cardiomyocytes were subjected to H/R to construct in vitro models. QRT-PCR and Western blot were adopted to test the levels of mRNA and protein. The viability and apoptosis of cardiomyocytes were tested by CCK-8 and flow cytometry assays, respectively. Results: The expression of A2M-AS1 was notably downregulated in H/R-treated cardiomyocytes. Overexpression of A2M-AS1 can notably enhance the cell viability of H/R-damaged cardiomyocytes, whereas knockdown of A2M-AS1 showed the opposite outcomes. Besides, a negative correlation was showed between A2M-AS1 and IL1R2 expression. In H/R-treated cardiomyocytes, overexpression of IL1R2 weakened the promoting proliferation and anti-apoptosis effects caused by overexpressing A2M-AS1, however, IL1R2-knockdown abolished the anti-proliferation and pro-apoptosis effects caused by silencing A2M-AS1. Conclusion: This study demonstrates the potential regulatory role of A2M-AS1/ IL1R2 axis in cardiomyocytes suffered from H/R, and provides insight into the protection of MI/RI.

        Fever in cancer patients is often due to the following causes: evil qi and toxity stagnancy, disorders of qi and blood, deficiencies of zang and fu organs, and the disorder of yin and yang. The treatments given to cancer patients with a fever are according to five: (a) Excessive inner heat and toxicants: remove heat and the toxicant, induce purgation. We use Cheng-Qi-Tang plus Qing-Wen-Bai-Du-Yin. (b) Tangle of damp and heat, and qi stagnancy: remove damp and heat, smooth the qi channel. We use Gan-Lu-Xiao-Du-Dan or San-Ren-Tang. (c) Obvious blood and heat stagnancy: remove heat and blood stasis. We use Xue-Fu-Zhu-Yu-Tang. (d) Deficiency of spleen qi, inner heat caused by a yin deficiency: nourish spleen qi and yin to remove the inner heat. We use Bu-Zhong-Yi-Qi-Tang or Xiao-Jian-Zhong-Tang. (e) Prominent yin deficiency and hectic fever: replenish yin and remove inner heat. We use Qing-Hao-Bie-Jia-Tang or Chai-Qian-Mei-Lian-San. The pathogenesis of fever in cancer patients is complicated. We can see both deficiency and excess in one differentiation. Therefore, we must make sure of it, then we can get the most effective treatment.

        The Influence of Immaterial Incentives on Job Performance of Knowledge Workers : Focusing on Psychological Contract as Mediating Variable

        본 논문의 목적은 지식근로자의 직무성과에 영향을 미치는 비물질적 인센티브가 심리계약으로 서 어떠한 매개역할을 하는 지에 관해 연구하기 위한 것이다. 본 논문의 연구대상은 중국 허베이성 지역에 기반을 둔 지식근로자를 대상으로 하였으며 설문조사에서는 총 560개의 설문지 가 배포되었고 52개의 설문지를 제외한 508개의 유효설문지가 최종적으로 회수되어 분석의 필요를 기본적으로 충족시켰다. 본 연구에서는 Guo Wei(2015)의 비물질적인 인센티브 척도를 바탕으로 연구에 적합하게 수정 및 보완해 실시하였으며 개별 인센티브, 직무 인센티브, 조직환경 인센티브의 3가지 차원으로 구분하였다. 또한 직무성과의 측정척도는 주로 Goodman&Svyantek (1999년)에서 설계한 측정 방법을 위주로 하였으며, 상황별 성과척도 선정에서는 Smith, Organ, Near(1983)가 제안한 척도를 주로 활용하였다. 심리계약은 Rousseau(1998,2000)와 같은 학자에 의해 설계되거나 사용된 척도로서 인지된 조직책임과 종업원책임의 두 가지 차원으로 구분하였다. 이러한 척도는 Lee Tinsley(1999), Robinson Turnley(1995), Chen California 등에서도 많이 활용된 바 있다. 따라서 선행연구 사용된 심리계약 척도를 기초로 적절한 조정을 거쳐 최종 측정척도로 사용되었다. 설문지는 네 부분으로 구성되어 있다. 첫 번째 부분은 성별, 연령, 혼인 여부, 교육 수준, 직급 등 7개 질문을 포함해 설문조사와 응답자 기본정보에 대한 설문조사 내용을 간략하게 설명한 것이며, 두 번째 부분은 14개의 질문을 포함하는 비물질적 인센티브에 관한 내용을 주로 하였다. 세 번째 부분은 심리계약에 관한 것으로써 20개의 설문으로 구성되어 있다. 네 번째 부분은 8개의 설문이 포함된 직무성과와 관련한 설문으로 구성되었다. 본 연구의 분석방법은 상관관계분석, 구조 방정식 및 기타 통계 기법을 사용하여 측정된 데이터를 처리함으로써 비물질적 인센티브, 심리계약 및 직무성과 간의 관계를 파악하고 모델에서 심 계약의 매개 효과를 검증하였다. 분석의 결과에 의하면 심리계약은 직무성과에 대한 비물질적 인센티브의 영향을 중재하는 역할을 한다는 것으로 나타났다. 중재영향분석 결과에 의하면 심리계약 및 조직책임, 종업원책임의 두 가지 차원이 직무성과에 미치는 비물질적 인센티브의 영향에서 중재역할을 하고 있었다. 마지막으로 본 연구에서는 다음과 같은 몇 가지를 제안해 보았다. (1) 조직은 공정하고 정의로운 급여관리체계 를 구축하고 유연한 복지제도를 시행할 필요가 있다. (2) 조직은 종업원의 경력계획을 수립해야 한다. (3) 조직은 종업원들에 대한 인간관계를 중시하는 관리를 강화하고 조화로운 관계를 구축해야 한다. (4) 조직은 종업원들에게 자율성을 부여하고 업무 재설계를 강화해야 한다. (5) 종업원은 조직에 대한 책임감을 높이고 동시에 심리적 기대를 맞출 필요가 있다. This article aims to study the mediating role of psychological contract in the process of immaterial incentives influencing job performance. This paper investigates knowledge workers in Hebei province by network questionnaire. In this survey, a total of 560 questionnaires were issued, 52 invalid questionnaires were excluded, and 508 valid questionnaires were finally recovered. In this study, the Chinese scholar Guo Wei (2015) immaterial incentive scale was adopted and modified on this basis. The measurement scale of job performance mainly refers to the measurement method designed by Goodman & Svyantek (1999), and it is adjusted according to the actual situation. The psychological contract scales designed or used by scholars like Rousseau (1998, 2000), Lee Tinsley (1999) and Wang Pei (2018) were referred to in the selection of the scales. The questionnaire consists of four parts. The first part is a brief description of the survey and a survey of the basic information of the respondents, including seven questions, such as gender, age, marital status, education level and job position. The second part is the immaterial incentives questionnaire. The third part is the psychological contract questionnaire. The fourth part is the job performance questionnaire. Correlation analysis, structural equation and other statistical techniques were used to process the measured data, so as to verify the mediating effect of psychological contract in the model. According to the results of the mediating influence test, some discussions are made : (1) The organization needs to establish a fair and just salary management system and implement flexible welfare system. (2) The organization shall develop career plans for employees. (3) The organization should strengthen humanistic care for employees and build a harmonious relationship. (4) The organization should give employees autonomy and strengthen work redesign. (5) Employees should enhance their sense of responsibility and adjust their psychological expectations of the organization.

      • Autographa californica Multiple Nucleopolyhedrovirus ac11 is Required for Virus Infection

        ORF11 (ac11) of Autographa californica multiple nucleopolyhedrovirus (AcMNPV) is a highly conserved baculovirus gene whose homologs are found in all lepidoteran Group I NPV, but its function is unknown so far. To determine the role of ac11 in baculovirus life cycle, ac11 knock-out mutant, Ac11KO, was constructed using the plasmid capture system (PCS). Real-Time PCR analysis showed that ac11 transcript was first detected at 6 h post-infection (p.i.) and accumulated to maximum at 48 h p.i., indicating that ac11 is belong to late gene. When the genomic DNA of Ac11KO was transfected into Sf9 cells, viral replication was restricted to a cell transfected originally. While viral transmission of the Ac11KO was not observed in Sf9 cells, production of budded virus (BV) in Sf9 cells transfected with Ac11KO was observed by transmission electron microscopy (TEM). These results suggest that the ac11 is essential for AcMNPV to produce infective BV.

        ORF11 (ac11) of Autographa californica multiple nucleopolyhedrovirus (AcMNPV) is a highly conserved gene of unknown function. To determine the role of ac11 in baculovirus life cycle, an ac11-knockout mutant AcMNPV, Ac11KO, was constructed. qPCR analysis revealed that ac11 is an early gene in the life cycle. After transfection into Spodoptera frugiperda cells, Ac11KO produced a single cell infection phenotype indicating that no infectious budded viruses (BVs) were produced. The defection in BV production was confirmed by both viral titration and Western blot. However, viral DNA replication is unaffected. Electron microscopy showed that ac11 is required for nucleocapsids envelopment to form ODV and their subsequent embedding into OB. This study therefore demonstrates that ac11 is an early gene which is essential for the viral life cycle.

        Split addition of enzymes in enzymatic hydrolysis at high solids concentration to increase sugar concentration for bioethanol production

        One challenge in making bioethanol production economical is to increase total solids in hydrolysis system while maintaining sugar conversion efficiency. Because the removal of excess water from hydrolysate requires enormous amounts of heat, large volume of reaction towers and high capital expenditure (CAPEX) for equipment, a lengthy operating time, and high operating costs. When solids loading in hydrolysis system increased from 5% to 20% with no mixing strategies, final sugar conversion decreased markedly. If cellulase is mixed with pulp at 5% solids and pressed to 20% solids, then 80% of the cellulase retained in the pulp thinned down the pulp mixture in 2 h. This thinning effect enabled additional cellulase, xylanase, and b-glucosidase to be mixed into the slurry. Sugar concentration was significantly improved; from 26 g/L to 121 g/L, while sugar conversion was remained as enzymatic hydrolysis with 5% total solids enzymatic hydrolysis. A US patent has been granted to NCSU for this concept and licenses have been granted to various companies.

      • Functional analysis of Autographa californica multiple nucleopolyhedrovirus ac78 and ac79

        Among 154 putative ORFs of Autographa californica multiple nucleopolyhedrovirus (AcMNPV), ac78 and ac79 are highly conserved genes in baculovirus, but their functions in the virus life cycle have been unknown so far. To determine their roles in AcMNPV replication, knockout mutants, ac78KO and ac79KO, were constructed using the plasmid capture system (PCS). Real-Time PCR analysis showed that both of ac78 and ac79 transcripts were first detected at 6 hours post-infection, and accumulated to maximum at 24 hours post-infection, suggesting that both of ac78 and ac79 are belong to late gene. When the genomic DNA of ac78KO was transfected into Sf9 cells, viral replication was restricted to a single cell infection. These results demonstrated that the ac78 play an important role in BV production, and therefore is essential for AcMNPV to mount a successful infection. Whereas Sf9 cells infected with the ac79KO showed normal viral symptoms such as rounding and swelling, OBs were not observed from majority of infected cells. These results suggested that the ac79 might play an important role in OB production.

        ORF78 (ac78) of Autographa californica multiple nucleopolyhedrovirus (AcMNPV) is a baculovirus core gene of unknown function. To determine the role of ac78 in baculovirus life cycle, an ac78-deleted mutant AcMNPV, Ac78KO, was constructed. Quantitative PCR analysis revealed that ac78 is a late gene in the viral life cycle. After transfection into Spodoptera frugiperda cells, Ac78KO produced a single-cell infection phenotype indicating that no infectious budded viruses (BVs) were produced. The defection in BV production was also confirmed by both viral titration and Western blot. However, viral DNA replication is unaffected. Analysis of BV and occlusion derived virus (ODV) revealed that AC78 is associated with both forms of the virions and is a structural protein located to viral envelope. Electron microscopy showed that ac78 also plays an important role in embedding of ODV into occlusion body. This study therefore demonstrates that AC78 is a late virion associated protein and is essential for the viral life cycle.

        Autographa californica nucleopolyhedrovirus (AcMNPV) has a large doublestrand DNA genome of approximately 134 kbp and harbors 156 open reading frames (ORFs). To elucidate DNA replication cascade of AcMNPV, we developed a novel baculovirus genome that can be maintained in Escherichia coli as a plasmid and can infect susceptible lepidopteran insect cells. This genome, named bAc-MK, contains a mini-F replicon and a kanamycin resistance marker. Using a convenient Tn7 transposon-based system, pPCS-S, 55 single ORF-truncated mutants were generated by random insertion into bAc-MK genome. These single ORF-truncated mutants were independently transfected into Sf9 cells, 16 of them were found affecting viral replication since they defected in producing polyhedra. Furthermore, to verify the pathogenicity of the single ORF-truncated mutants, the remaining 39 mutants were subjected to bioassay to Spodoptera exigua 3rd instar larvae. Among them, ac9-, ac49-, ac103- and ac105-knockout mutants showed higher mortality compared to that of bAc-MK. These results suggested that these ORFs could be related to pathogenicity of AcMNPV.
