Antibodies against human CD36 are responsible for several immune-mediated disorders. The detection of anti-CD36 antibodies using the standard monoclonal antibody (mAb) immobilization of platelet antigens (MAIPA) assay is hampered by a high frequency of false-negative results, most likely due to competitive inhibition of the mAb used as the capture antibody. We generated a panel of mouse mAbs against CD36 and seven hybridomas (GZ-3, GZ-13, GZ-70, GZ-143, GZ-413, GZ-507, and GZ-608), which were selected for MAIPA assays, as they reacted with mouse and human CD36. Fourteen anti-CD36 sera were assayed; all of which showed a positive reaction in a PakPlus (Immucor GTI Diagnostics, Inc., Waukesha, WI, USA) ELISA-based screening (optical density: 0.257–2.292). When the reference anti-CD36 mAb FA6-152 was used in the MAIPA assay, only 6/14 (42.9%) sera displayed a positive reaction. In contrast, anti-CD36 antibodies were detected in 13/14 (92.9%) sera when GZ-70 and GZ-608 mAbs were used. This significant improvement resulted in the identification of anti-CD36 antibodies by an antigen capture assay. Since patient’s platelets possibly carrying rare native antigens are used, this method will facilitate the identification of new platelet antibodies against CD36 that are involved in immune-mediated thrombocytopenia and other diseases, such as transfusion-related acute lung injury.

The current work fills a gap of a small-scale study on wave propagation behavior of symmetric, antisymmetric, and quasi-isotropic cross/angle-plies laminated composite nanoplates. The governing equations are derived through the Hamiltonian principle for four-variable refined shear deformation plate theory in conjunction with the assumption of a non-classical theory, and then size-dependent formulations are solved via an analytical solution procedure. This work provides information to accurately analyze the influence of lay-up numbers and sequences, geometry, fiber orientations, and wave numbers on the size-dependent wave propagation response of laminated composite nanoplates.

Osteoarthritis, characterized by articular cartilage degradation, is the leading cause of chronic disability in older adults. Studies have indicated that circular RNAs are crucial regulators of chondrocyte development and are involved in the progression of osteoarthritis. In this study, we investigated the function and mechanism of a circular RNA and its potential for osteoarthritis therapy. The expression levels of circCREBBP, screened by circular RNA sequencing during chondrogenic differentiation in adipose tissue-derived stem cells, and TGFβ2 were significantly increased in the cartilage of patients with osteoarthritis and IL-1β-induced chondrocytes. circCREBBP knockdown increased anabolism in the extracellular matrix and inhibited chondrocyte degeneration, whereas circCREBBP overexpression led to the opposite effects. Luciferase reporter assays, rescue experiments, RNA immunoprecipitation, and RNA pulldown assays confirmed that circCREBBP upregulated TGFβ2 expression by sponging miR-1208, resulting in significantly enhanced phosphorylation of Smad1/5 in chondrocytes. Moreover, intra-articular injection of adeno-associated virus-sh-circCrebbp alleviated osteoarthritis in a mouse model of destabilization of the medial meniscus. Our findings reveal a critical role for circCREBBP in the progression of osteoarthritis and provide a potential target for osteoarthritis therapy.

Serum alpha-fetoprotein (AFP) is a significant marker for clinical diagnosis and prognosis evaluation in hepatocellular carcinoma (HCC) patients. However, some proportion of liver cancer patients are AFP-negative (AFP ${\leq}$20ng/ml). In order to study the differences between clinicopathological factors and prognosis of alpha-fetoprotein negative and positive patients, a total of 114 cases (41 AFP-negative and 73 AFP-positive) were selected for our research. By systematically statistical analysis, the results demonstrated that compared with AFP-negative patients, AFP-positive examples were more likely to feature cirrhosis nodules, non-complete neoplasm capsules, and a poor Edmondson-steiner grade. Furthermore, AFP-negative patients demonstrated a favorable long-term prognosis. By univariate analysis and multivariate analysis with Cox's proportional hazards model, multiple tumors were found to be independent risk factors for worse survival of AFP negative patients; however, less tumor-free margins, multiple tumors and Edmondson-steiner grades III/IV, proved to be independent risk factors leading to a poor prognosis of AFP positive cases. Finally, we can infer that high levels of AFP signify a highly malignant tumor and unfavorable prognosis.

Purpose: Helicobacter pylori (HP)-infected gastric cancer (GC) is known to be a fatal malignant tumor, but the molecular mechanismsunderlying its proliferation, invasion, and migration remain far from being completely understood. Our aim in this studywas to explore miR-1915 expression and its molecular mechanisms in regulating proliferation, invasion, and migration of HP-infectedGC cells. Materials and Methods: Quantitative real-time PCR and western blot analysis were performed to determine miR-1915 and receptorfor advanced glycation end product (RAGE) expression in HP-infected GC tissues and gastritis tissues, as well as humangastric mucosal cell line GES-1 and human GC cell lines SGC-7901 and MKN45. CCK8 assay and transwell assay were performedto detect the proliferation, invasion, and migration capabilities. MiR-1915 mimics and miR-1915 inhibitor were transfected intoGC cells to determine the target relationship between miR-1915 and RAGE. Results: MiR-1915 was under-expressed, while RAGE was over-expressed in HP-infected GC tissues and GC cells. Over-expressedmiR-1915 could attenuate cellular proliferation, invasion, and migration capacities. RAGE was confirmed to be the target gene ofmiR-1915 by bioinformatics analysis and luciferase reporter assay. Moreover, HP-infected GC cellular proliferation, invasion, andmigration were inhibited after treatment with pcDNA-RAGE. Conclusion: MiR-1915 exerted tumor-suppressive effects on cellular proliferation, invasion, and migration of HP-infected GCcells via targeting RAGE, which provided an innovative target candidate for treatment of HP-infected GC.
Study on Settlement Calculation of the Long-Short Pile Composite Foundation

As a new foundation treatment technology, long-short pile composite’s design theory is still in primary phase, and there are no explicit settlement calculation methods in active codes. So it is necessary to study the working mechanism and the methods of settlement calculation. In this paper, the mechanics of long-short pile composite foundation are fully discussed. Meanwhile, based on the shear deformation method, the Mylonakis & Gazetas models about mutual action between two piles and the one between pile and soil are introduced, Considering the performance of cushion, the flexible factors of mutual actions are provided. Then the settlement calculation of long-short pile composite foundation which can consider the mutual actions between pile, soil and cap is deduced, and the correlated program is also developed. Finally, an engineering example is discussed with the method. A comparison shows that calculated results and measured data from a field test pile are in a good agreement, indicating that the presented approach is feasible and applicable in engineering practice.
Wave propagation analysis of porous functionally graded curved beams in the thermal environment

In the present paper, wave propagation behavior of porous temperature-dependent functionally graded curved beams within the thermal environment is analyzed for the first time. A recently-developed method is utilized which considers the reciprocal effect of mass density and Young’s modulus in order to explore the influence of porosity. Three different types of temperature variation (uniform temperature change (UTC), linear temperature change (LTC), sinusoidal temperature change (STC)) are employed to study the effect of various thermal loads. Euler-Bernoulli beam theory, also known as classic beam theory is implemented in order to derive kinetic and kinematic relations, and then Hamilton’s principle is used to obtain governing equations of porous functionally graded curved beams. The obtained governing equations are analytically solved. Eventually, the influences of various parameters such as wave number, porosity coefficient, various types of temperature change and power index are covered and indicated in a set of illustrations.
