      • Molecular Analyses of the Metallothionein Gene Family in Rice (Oryza sativa L.)

        Metallothioneins are a group of low molecular mass and cysteine-rich metal-binding proteins, ubiquitously found in most living organisms. They play an important role in maintaining intracellular metal homeostasis, eliminating metal toxification and protecting against intracellular oxidative damages. Analysis of complete rice genome sequences revealed eleven genes encoding putative metallothionein (OsMT), indicating that OsMTs constitute a small gene family in rice. Expression profiling revealed that each member of the OsMT gene family differs not only in sequence but also in their tissue expression patterns, suggesting that these isoforms may have different functions they perform in specific tissues. On the basis of OsMT structural and phylogenetic analysis, the OsMT family was classified as two classes and class I was subdivided into four types. Additionally, in this paper we also present a complete overview of this family, describing the gene structure, genome localization, upstream regulatory element, and exon/intron organization of each member in order to provide valuable insight into this OsMT gene family.

        The buried Daxing Uplift connects the Beijing Sag and Dachang Sag, which is the critical tectonic unit for understanding the structural evolution of the Beijing Sub-plain and its linkages to regional tectonics. This study combines paleomagnetic, 26Al-10Be isochron dates and sedimentary analyses from a new borehole (Yx02) in the northeastern Beijing Sub-plain to shed fresh light on regional tectonic processes and the sedimentary history since the Pliocene. The main findings are as follows: (1) the Daxing Uplift had formed three depositional episodes: alluvial-fan, lacustrine-delta, and alluvial-fan sedimentation since the Cenozoic; (2) The borehole records the Brunhes, Gauss normal chron, and the Matuyama, Gilbert reversed chron in the Daxing Uplift, the corresponding depths are 0–148.9 m, 148.9–315.0 m, 315.0–520.5 m and 520.5–650 m; (3) An important tectonic event started in ~4.3 Ma, disintegrated NE-trending basin and range terrain, reactivated the NW-trending Nankou-Sunhe fault, and formed Shunyi fault in Beijing Sub-plain. Sedimentation occurs exclusively in the Beijing Sag and the northeastern Daxing Uplift during ~4.3–1.77 Ma; (4) During ~1.77–1.07 Ma, a transition from an extension to a strike-slip occurred, and is described not only in a progressively higher increase of the overall deposition rate, but also in a decrease of the relative deposition disparity between sags and uplifts. Since then, the Beijing Sub-plain has been shaped, associated with an overall subsidence depression in the Bohai Bay Basin. This work enhances our understanding of the formation and evolution of the Beijing Sub-plain, the division of tectonic episodes, and the initiation time of the latest tectonic movement in the break-up region, North China Craton, since the Neogene.

        Intrinsic memory characteristic of polycrystalline ZnTe film originating from Mott variable range hopping conduction

        The resistive switching characteristics of Au/ZnTe/ITO structure with polycrystalline ZnTe film as resistive switching layer is investigated. Macroscopically, 100 bipolar switching cycles under the direct current (dc) voltages were carried out and the conduction states can retain for several hours. Microscopically, reading and writing operations can be achieved on ZnTe film with Au top electrode replaced by conductive Atomic Force Microscopy (c-AFM) tip. The I–V characteristic in low resistance state (LRS) is linear in the whole range of voltage. The I–V characteristic in high resistance state (HRS) is linear in the low voltage while it obeys Schottky emission in the high voltage, and Schottky barrier height is symmetric in the positive and negative voltage. During linear I–V characteristic voltage range, the electrons transport between adjacent point defects via Mott variable range hopping. The higher hopping distance and higher activation energy in HRS contribute to the higher resistance value in HRS compared with LRS. Impedance spectroscopy in HRS and LRS both behave as a semicircle, which accords with the semiconductor-like characteristic of conductive point defects. Photoluminescence (PL) spectroscopy indicates the decisive role of deep level defects in conduction. This study confirms the intrinsic resistive switching characteristic of ZnTe film and provides a new choice for intrinsic nonoxides material in nonvolatile memory application.

        Synthesis and Biological Evaluation of 3-(1H-indol-3-yl)pyrazole- 5-carboxylic Acid Derivatives

        A series of novel compounds bearing a 3-(1H-indol-3-yl)pyrazole-5-carboxylic acid nucleus were synthesized. Analytical and spectral data confirmed the structures of the new compounds. The structures of the regioisomers in this series were determined by 1H-NMR spectra. The title compounds were evaluated for their endothelin-1 antagonist activities. In the in vitro functional assay, compounds 23, 24, 28 and 29 exhibited significant efficacy at the concentration of 1 μg/mL, and compounds 5b, 5c, 26 and 28 were as potent as the positive control bosentan at high concentration. In the experiment to assess prevention of endothelin-1-induced sudden death in mice, compound 5b showed comparable activity to bosentan, and 30 was more potent than bosentan. The final compounds were also screened for antibacterial activity against four Gram-positive and -negative bacteria. Some of the tested compounds showed weak antibacterial activity.

        Upregulation of adiponectin by Ginsenoside Rb1 contributes to amelioration of hepatic steatosis induced by high fat diet

        Background: Ginsenoside Rb1 (GRb1) is capable of regulating lipid and glucose metabolism through itsaction on adipocytes. However, the beneficial role of GRb1-induced up-regulation of adiponectin in liversteatosis remains unelucidated. Thus, we tested whether GRb1 ameliorates liver steatosis and insulinresistance by promoting the expression of adiponectin. Methods: 3T3-L1 adipocytes and hepatocytes were used to investigate GRb1's action on adiponectinexpression and triglyceride (TG) accumulation. Wild type (WT) mice and adiponectin knockout (KO)mice fed high fat diet were treated with GRb1 for 2 weeks. Hepatic fat accumulation and function as wellas insulin sensitivity was measured. The activation of AMPK was also detected in the liver andhepatocytes. Results: GRb1 reversed the reduction of adiponectin secretion in adipocytes. The conditioned medium(CM) from adipocytes treated with GRb1 reduced TG accumulation in hepatocytes, which was partlyattenuated by the adiponectin antibody. In the KO mice, the GRb1-induced significant decrease of TGcontent, ALT and AST was blocked by the deletion of adiponectin. The elevations of GRb1-induced insulinsensitivity indicated by OGTT, ITT and HOMA-IR were also weakened in the KO mice. The CM treatmentsignificantly enhanced the phosphorylation of AMPK in hepatocytes, but not GRb1 treatment. Likewise,the phosphorylation of AMPK in liver of the WT mice was increased by GRb1, but not in the KO mice. Conclusions: The up-regulation of adiponectin by GRb1 contributes to the amelioration of liver steatosisand insulin resistance, which further elucidates a new mechanism underlying the beneficial effects ofGRb1 on obesity
