        Probabilistic Tensile Strength Model of Q345 Steel Plates with Random Pitting Corrosion for Fragility Analysis

        To evaluate the fracture behavior of steel structures under some severe corrosion environments, experiments on eight artificially pitting corroded Q345 steel plates and numerical investigations on 200 finite element models of randomly pitting damaged steel were conducted. The effects of the distribution, diameter and depth of the corrosion pits on the tensile behavior were investigated and a probability density function of randomly pitting corroded steel strength was developed. The results indicate that both the yield strength and the ultimate strength of steel plates with random pitting corrosion were lower than those of uniformly corroded steels with the same corrosion ratios. The effects of pitting depth on the yield strength and the ultimate strength were more significant than those of pitting diameter. The influences of pitting distribution on ultimate strength were greater than that on yield strength. The residual tensile strength of steel plates with random pitting corrosion was found to well conform to the Gamma distribution, and thus the probability density functions of the residual yield strength and the ultimate strength were proposed based on the Gamma distribution.

        Circular RNA CREBBP modulates cartilage degradation by activating the Smad1/5 pathway through the TGFβ2/ALK1 axis

        Osteoarthritis, characterized by articular cartilage degradation, is the leading cause of chronic disability in older adults. Studies have indicated that circular RNAs are crucial regulators of chondrocyte development and are involved in the progression of osteoarthritis. In this study, we investigated the function and mechanism of a circular RNA and its potential for osteoarthritis therapy. The expression levels of circCREBBP, screened by circular RNA sequencing during chondrogenic differentiation in adipose tissue-derived stem cells, and TGFβ2 were significantly increased in the cartilage of patients with osteoarthritis and IL-1β-induced chondrocytes. circCREBBP knockdown increased anabolism in the extracellular matrix and inhibited chondrocyte degeneration, whereas circCREBBP overexpression led to the opposite effects. Luciferase reporter assays, rescue experiments, RNA immunoprecipitation, and RNA pulldown assays confirmed that circCREBBP upregulated TGFβ2 expression by sponging miR-1208, resulting in significantly enhanced phosphorylation of Smad1/5 in chondrocytes. Moreover, intra-articular injection of adeno-associated virus-sh-circCrebbp alleviated osteoarthritis in a mouse model of destabilization of the medial meniscus. Our findings reveal a critical role for circCREBBP in the progression of osteoarthritis and provide a potential target for osteoarthritis therapy.

        Study on Anti-Proliferative Activity in Cancer Cells and Preliminary Structure–Activity Relationship of Pseudo-Peptide Chiral Thioureas

        In our previous studies, we have shown that thiourea compounds containing phosphate esters have potent antitumor activity and can be used as a novel strategy for the development of antitumor agents. Herein, a series of novel phosphonate thioureas 5–38 have been synthesized, which were fully characterized by 1H NMR, 13C NMR spectrum, elemental analysis. Three human cancer cell lines (Bcap-37, BGC-823, and PC-3) have been used to investigate these compounds’ antitumor activities. After the summarization of the structure–activity relationships, we found that the variation of R, R1, and R2 in these novel phosphonate thioureas contribute to the antitumor activities. All these SAR-guided efforts may lead to novel antitumor drugs in the market in the near future.

        Bioinformatic and integrated analysis identifies an lncRNA–miRNA–mRNA interaction mechanism in gastric adenocarcinoma

        Background lncRNAs–miRNAs–mRNAs networks play an important role in Gastric adenocarcinoma (GA). Identifcation of these networks provide new insight into the role of these RNAs in gastric cancer. Objectives Biological information databases were screened to characterize and examine the regulatory networks and to further investigate the potential prognostic relationship this regulation has in GA. Methods By mining The Cancer Genome Atlas (TCGA) database, we gathered information on GA-related lncRNAs, miRNAs, and mRNAs. We identifed diferentially expressed (DE) lncRNAs, miRNAs, and mRNAs using R software. The lncRNA–miRNA–mRNA interaction network was constructed and subsequent survival examination was performed. Representative genes were selected out using The Biological Networks Gene Ontology plug-in tool on Cytoscape. Additional analysis of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) terms were used to screen representative genes for functional enrichment. Reverse transcription quantitative polymerase chain reaction (RT-qPCR) were used to identify the expression of fve candidate diferential expressed RNAs. Results Information of samples from 375 cases of gastric cancer and 32 healthy cases (normal tissues) were downloaded from the TCGA database. A total of 1632 DE-mRNAs, 1008 DE-lncRNAs and 104 DE-miRNAs were identifed and screened. Among them, 65 DE-lncRNAs, 10 DE-miRNAs, and 10 DE-mRNAs form lncRNAs–miRNAs–mRNAs regulatory network. Additionally, 10 lncRNAs and 2 mRNAs were associated with the prognosis of GA. Multivariable COX analysis revealed that AC018781.1 and VCAN-AS1 were independent risk factors for GA. GO functional enrichment analysis found DE-mRNA was signifcantly enriched TERM (P<0.05). The KEGG signal regulatory network analysis found 11 signifcantly enrichment networks, the most prevailing was for the AGE-RAGE signaling pathway associated with Diabetic complications. Results of RT-qPCR was consistent with the in silico results. Conclusions The results of the present study represent a view of GA from a analysis of lncRNA, miRNA and mRNA. The network of lncRNA–miRNA–mRNA interactions revealed here may potentially further experimental studies and may help biomarker development for GA.

        Integrated proteomic and metabolomic analyses reveal significant changes in chloroplasts and mitochondria of pepper (Capsicum annuum L.) during Sclerotium rolfsii infection

        Infection by Sclerotium rolfsii will cause serious disease and lead to significant economic losses in chili pepper. In this study, the response of pepper during S. rolfsii infection was explored by electron microscopy, physiological determination and integrated proteome and metabolome analyses. Our results showed that the stomata of pepper stems were important portals for S. rolfsii infection. The plant cell morphology was significantly changed at the time of the fungal hyphae just contacting (T1) or surrounding (T2) the pepper. The chlorophyll, carotenoid, and MDA contents and the activities of POD, SOD, and CAT were markedly upregulated at T1 and T2. Approximately 4129 proteins and 823 metabolites were clearly identified in proteome and metabolome analyses, respectively. A change in 396 proteins and 54 metabolites in pepper stem tissues was observed at T1 compared with 438 proteins and 53 metabolites at T2. The proteins and metabolites related to photosynthesis and antioxidant systems in chloroplasts and mitochondria were disproportionally affected by S. rolfsii infection, impacting carbohydrate and amino acid metabolism. This study provided new insights into the response mechanism in pepper stems during S. rolfsii infection, which can guide future work on fungal disease resistance breeding in pepper.

        BIFURCATION ANALYSIS OF A DELAYED EPIDEMIC MODEL WITH DIFFUSION

        In this paper, a class of delayed epidemic model with diffusion is investigated. By analyzing the associated characteristic transcendental equation, its linear stability is investigated and Hopf bifurcation is demonstrated. Some explicit formulae determining the stability and the direction of the Hopf bifurcation periodic solutions bifurcating from Hopf bifurcations are obtained by using the normal form theory and center manifold theory. Some numerical simulation are also carried out to support our analytical findings. Finally, biological explanations and main conclusions are given.

        In this paper, a Lotka-Volterra model with time delays is considered. A set of sufficient conditions for the existence of Hopf bifurcation are obtained via analyzing the associated characteristic transcendental equation. Some explicit formulae for determining the stability and the direction of the Hopf bifurcation periodic solutions bifurcating from Hopf bifurcations are obtained by applying the normal form method and center manifold theory. Finally, the main results are illustrated by some numerical simulations.

      • Structural Conservation and Food Habit-related Liver Expression of Uncoupling Protein 2 Gene in Five Major Chinese Carps

        The full-length cDNA of grass carp (Ctenopharyngodon idellus) and silver carp (Hypophthalmichthys molitrix) uncoupling protein 2 (UCP2) was obtained from liver. The grass carp UCP2 cDNA was determined to be 1152 bp in length with an open reading frame that encodes 310 amino acids. Five introns (Intron 3, 4, 5, 6 and 7) in the translated region, and partial sequence of Intron 2 in the untranslated region of grass carp UCP2 gene were also obtained. Gene structure comparison between grass carp and mammalian (human and mouse) UCP2 gene shows that, the UCP2 gene structure of grass carp is much similar to that of human and mouse. Partial UCP2 cDNA sequences of bighead carp (Aristichthys nobilis) and mud carp (Cirrhinus molitorella), were further determined. Together with the common carp (Cyprinus carpio) UCP2 sequence from GenBank (AJ243486), multiple alignment result shows that the nucleotide and amino acid sequences of the UCP2 gene, were highly conserved among the five major Chinese carps that belong to four subfamilies. Using beta-actin as control, the ratio UCP2/beta-actin mRNA (%) was determined to be $149.4{\pm}15.6$ (common carp), $127.4{\pm}22.1$ (mud carp), $96.7{\pm}12.7$ (silver carp), $94.1{\pm}26.8$ (bighead carp) and $63.7{\pm}16.2$ (grass carp). The relative liver UCP2 expression of the five major Chinese carps, shows a close relationship with their food habit: benthos and detrituseating fish (common carp and mud carp) > planktivorious fish (silver carp and bighead carp) > herbivorious fish (grass carp). We suggest that liver UCP2 might be important for Chinese carps to detoxify cyanotoxins and bacteria in debris and plankton food.

        Deficit irrigation and silicate spray applied since blooming period improved strawberry fruit quality without reducing fruit size

        The drought stress created by defi cit irrigation has been widely introduced into production for fruit quality improvement, whilst silicon is considered as a functional element in plant antistress process. To explore the infl uence of defi cit irrigation and silicon application on the strawberry fruit quality and the plant’s response to drought stress, a split-plot experiment was conducted. The impacts of the treatments start-time was also evaluated. The plants were subjected to two irrigation regimes, defi cit irrigation (DI) or full irrigation, and two foliar sprays, 0.01 mol L − 1 Na 2 SiO 3 (Si) or water, with two treatment start-time, as after fi rst bloom (Fr) or after transplanting. Therefore, there were 6 diff erent treatments (consider ing treatment start-time) and a control group, which were DI, FrDI, Si, FrSi, DISi, FrDISi and CK. As a result, the DI signifi cantly increased the leaf ascorbate, leaf anthocyanins and specifi c leaf weight by 93.5%, 109.9% and 24.5%, but it also reduced fruit reducing sugars concentration and single fruit mass by 25.1% and 12.6%, respectively. Whilst the FrDI increased specifi c leaf weight by 27.9% and had no negative eff ect on fruit quality. The timing of starting silicate spray didn’t infl uence leaf morphology or fruit quality, but the FrSi signifi cantly increased the percentage of leaf sucrose to total sugars by 9.0%, and the Si didn’t. The DISi and FrDISi tended to increase the concentration of fruit sugars, glutamate, polyphenols, and organic acids. Although the DISi showed more obvious eff ect on improving the fruit nutritive values, the FrDISi tended to have lighter negative impact on fruit size, compared to CK. We concluded that the combination of defi cit irrigation and silicate spray started after the fi rst bloom could eff ectively improve the fruit sugars, glutamate and polyphenols, without reducing the fruit size, due to the mitigation of plant antistress response.

By analyzing the associated characteristic transcendental equation, its linear stability is investigated and Hopf bifurcation is demonstrated. Some explicit formulae for determining the stability and the direction of the Hopf bifurcation periodic solutions bifurcating from Hopf bifurcations are obtained by using the normal form theory and center manifold theory. Some numerical simulations for justifying the theoretical analysis are also provided. Finally, biological explanations and main conclusions are given.
