        XIST Induced by JPX Suppresses Hepatocellular Carcinoma by Sponging miR-155-5p

        Purpose: The influence of X-inactive specific transcript (XIST) and X-chromosome inactivation associated long non-coding RNAs (lncRNAs) just proximal to XIST (JPX) on hepatocellular carcinoma (HCC) remains controversial in light of previous reports, which the present study aimed to verify. Materials and Methods: The DIANA lncRNA-microRNA (miRNA) interaction database was used to explore miRNA interactions with JPX or XIST. JPX, XIST, and miR-155-5p expression levels in paired HCC specimens and adjacent normal tissue were analyzedby RT-qPCR. Interaction between XIST and miR-155-5p was verified by dual luciferase reporter assay. Expression levels of miR-155-5p and its known target genes, SOX6 and PTEN, were verified by RT-qPCR and Western blot in HepG2 cells with or withoutXIST knock-in. The potential suppressive role of XIST and JPX on HCC was verified by cell functional assays and tumor formationassay using a xenograft model. Results: JPX and XIST expression was significantly decreased in HCC pathologic specimens, compared to adjacent tissue, which correlated with HCC progression and increased miR-155-5p expression. Dual luciferase reporter assay revealed XIST as a direct target of miR-155-5p. XIST knock-in significantly reduced miR-155-5p expression level and increased that of SOX6 and PTEN, while significantly inhibiting HepG2 cell growth in vitro, which was partially reversed by miR-155-5p mimic transfection. JPX knock-in significantly increased XIST expression and inhibited HepG2 cell growth in vitro or tumor formation in vivo in a XIST dependent manner. Conclusion: JPX and XIST play a suppressive role in HCC. JPX increases expression levels of XIST in HCC cells, which suppresses HCC development by sponging the cancer promoting miR-155-5p.

        Simulation of the Transmission Characteristics of a Hexagonal Photonic Crystal Slab Etched into an Ion-implanted LiNbO_3 Waveguide

        A photonic crystal slab that was constructed as a 2D hexagonal photonic crystal with a finite depth and was etched into an ion-implanted LiNbO_3 waveguide was simulated by using the finitedifference time-domain (FDTD) method. Both the ΓK-oriented and ΓM-oriented propagations were simulated with the fundamental TE mode of the LiNbO_3 waveguide as the incident beam. Despite the weak vertical confinement, the transmittance spectra show that they both have quite deep stop gaps. These gaps show a good overlap. At wavelengths within the overlap region, both the line defects that we created in the ΓK and the ΓM directions worked very well as channel waveguides.

        Optimization of preparation and properties of Gardenia yellow pigment-loaded alginate beads

        Gardenia yellow pigment (GYP) loaded alginate beads were prepared by the ionic gelation technique, and the preparation parameters were optimized by response surface methodology for high encapsulation efficiency. The optimized parameters were alginate concentration of 3.3%, CaCl2 concentration of 2.4%, and GYP concentration of 3.2mg/mL, under which the encapsulation efficiency was 73.61%. The surface morphology and bead size analysis showed that the GYP-loaded alginate beads had a roughly spherical morphology with a wrinkled surface, and their average diameter was 0.87 mm. In vitro release test revealed that the GYP release had a pH-dependent release profile and a two-step release process. The Rigter-Peppas model was the most proper model to assess the GYP release from alginate beads. The release mechanism of GYP at pH 1.2 and 7.4 was non-Fickian transport and case-II transport, respectively. The 2,2-diphenyl-1-picrylhydrazyl assay indicated that the encapsulated GYP had effectively maintained 82.56% of the antioxidant activity.

      • Tumor-derived CD4+CD25+ Tregs Inhibit the Maturation and Antigen-Presenting Function of Dendritic Cells

        CD4+CD25+regulatory T cells (Tregs) play a key role in regulation of immnue response and maintenance of self-tolerance. Studies have found Tregs could suppress tumor-specific T cell-mediated immune response and promote cancer progression. Depletion of Tregs can enhance antitumor immunity. Dendritic cells (DCs) are professional antigen-presenting cells and capable of activating antigen-specific immune responses, which make them ideal candidate for cancer immunotherapy. Now various DC vaccines are considered as effective treatment for cancers. The aim of this study was to evaluate variation of Tregs in BALB/C mice with hepatocellular carcinoma and investigate the interaction between tumor-derived Tregs, effector T cells (Teff) and splenic DCs. We found the percentages of Tregs/CD4+ in the peripheral blood of tumor-bearing mice were higher than in normal mice. Tumor-derived Tregs diminished the up-regulation of costimulatory molecule expression on splenic DCs, even in the presence of Teff cells and simultaneously inhibited IL-12 and $TNF-{\alpha}$ secretion by DCs.

      • A New Cell Counting Method to Evaluate Anti-tumor Compound Activity

        Determining cell quantity is a common problem in cytology research and anti-tumor drug development. A simple and low-cost method was developed to determine monolayer and adherent-growth cell quantities. The cell nucleus is located in the cytoplasm, and is independent. Thus, the nucleus cannot make contact even if the cell density is heavy. This phenomenon is the foundation of accurate cell-nucleus recognition. The cell nucleus is easily recognizable in images after fluorescent staining because it is independent. A one-to-one relationship exists between the nucleus and the cell; therefore, this method can be used to determine the quantity of proliferating cells. Results indicated that the activity of the histone deacetylase inhibitor Z1 was effective after this method was used. The nude-mouse xenograft model also revealed the potent anti-tumor activity of Z1. This research presents a new anti-tumor-drug evaluation method.

      • Historical Long-term Exposure to Pentachlorophenol Causing Risk of Cancer - A Community Study

        Background: Pervious studies suggested occupational workers exposure to pentachlorophenol (PCP) might contribute to increased risk of cancer. However, few studies have focused on associations between PCP and cancer risk at the community level. Objective: The present study was to explore the cancer risk for the community population living long-term in a PCP contaminated area. Methods: All the cancer cases diagnosed in 2009-2011 in Tongling City were collected. The cancer patients' residencies were geo-referenced in each district. The historical PCP usage for each district of Tongling was calculated as the PCP pollution index, which was further used to divide into PCP exposure categories. Standardized rate ratios (SRRs) of cancer incidence were applied to detect the cancer risk as exposure grade elevated. Correlation analysis was performed to analyze the relationship between PCP pollution and cancer incidence. Results: A total of 5,288 cancer cases (3,451 male and 1,837 female) were identified. PCP usage was correlated with the incidence of leukemia (r=0.88, P=0.002) for males, and with cancer of the esophagus for males (r=0.83, P=0.008) and females (r=0.71, P=0.020). Compared with the low exposure category, significant SRRs for total cancer sites was obtained for high PCP exposure category (SRR=1.61, 95%CI=1.59-1.62). Most SRR values of the cancer sites were significantly increased as exposure grade elevated and exposure time extended. Conclusion: The present study found that community residents living in the PCP contaminated area had increased risk of cancers. Leukemias, lymphomas and nasopharyngeal and esophageal cancers are most possibly associated with PCP exposure.

        Angelica sinensis Supercritical Fluid CO2 Extract Attenuates D-Galactose-Induced Liver and Kidney Impairment in Mice by Suppressing Oxidative Stress and Inflammation

        Angelica sinensis (AS, Danggui in Chinese) is an important herbal component of various traditional formulae for the management of asthenia and its tonic effects. Although AS has been shown to ameliorate cognitive damage and nerve toxicity in D-galactose (D-gal)-elicited senescent mice brain, its effects on liver and kidney injury have not yet been explored. In this work, mice were subjected to hypodermic injection with D-gal (200 mg/kg) and orally gavaged with AS (20, 40, or 80 mg/kg) once a day for 8 successive weeks. Results revealed that AS significantly improved liver and kidney function as assessed by organ index and functional parameters. In addition, AS pretreatment effectively ameliorated the histological deterioration. AS attenuated the MDA level and markedly enhanced the activities and gene expressions of antioxidative enzymes, namely Cu, Zn-SOD, CAT, and GPx. Furthermore, AS markedly inhibited the D-gal-mediated increment of expressions of inflammatory cytokines iNOS, COX-2, IκBα, p-IκBα, and p65 and promoted the IκBα expression level in both hepatic and renal tissues. In sum, AS pretreatment could effectively guard the liver and kidney of mice from D-gal-induced injury, and the underlying mechanism was deemed to be intimately related to attenuating oxidative response and inflammatory stress.

        Studies of the microbial metabolism of flavonoids extracted from the leaves of Diospyros kaki by intestinal bacteria

        Flavonoid glycosides are metabolized byintestinal bacteria, giving rise to a wide range of phenolicacids that may exert systemic effects in the body. Themicrobial metabolism of flavonoids extracted from theleaves of Diospyros kaki (FLDK) by intestinal bacteria wasinvestigated in vitro. High-performance liquid chromatography/linear trap quadrupole orbitrap mass spectrometrywas performed to analyze the metabolites of flavonoidsin vivo using Xcalibur2.1 software. The results showed thatthe levels of flavonoid glycosides and flavonoid aglyconesdecreased rapidly in the process of microbial metabolismby intestinal bacteria in vitro, and the metabolic rate maybe related to the concentration of intestinal bacteria in theculture solution. In vivo metabolites of FLDK weredetected in rat plasma and urine after oral administration ofFLDK. Eight flavonoids were identified in the urine, andthree were identified in the plasma; however, flavonoidaglycones were not found in the plasma.

      • Prognostic Values of Various Clinical Factors and Genetic Subtypes for Diffuse Large B-cell lymphoma Patients: A Retrospective Analysis of 227 Cases

        Aim: To analyze the significance of different clinical factors for prognostic prediction in diffuse large B-cell lymphoma (DLBCL) patients. Methods: Two hundred and twenty-seven DLBCL patients were retrospectively reviewed. Patients were managed with cyclophosphamide, doxorubicin, vincristine, and prednisone (CHOP) regimen or rituximab plus the CHOP (RCHOP) regimen. Results: Lactate dehydrogenase (LDH), ${\beta}2$-microglobulin (${\beta}2$-M), B symptoms, Ann Arbor stage and genetic subtypes were statistically relevant in predicting the prognosis of the overall survival (OS). In the CHOP group, the OS in patients with germinal center B-cell-like (GCB)(76.2%) was significantly higher than that of the non-GCB group (51.9%, P=0.032). With RCHOP management, there was no statistical difference in OS between the GCB (88.4%) and non-GCB groups (81.9%, P=0.288). Conclusion: Elevated LDH and ${\beta}2$-M levels, positive B symptoms, Ann Arbor stage III/IV, and primary nodal lymphoma indicate an unfavorable prognosis of DLBCL patients. Patients with GCB-like DLBCL have a better prognosis than those with non-GCB when treated with the CHOP regimen. The RCHOP treatment with the addition of rituximab can improve the prognosis of patients with DLBCL.
