        Phenylboronic acid-functionalized gelatin–oleic acid nanoparticles for high loading and efficient transdermal delivery of Celastrol towards the treatment of psoriasis

        Systemic toxicity, poor aqueous solubility, and poorly cell permeable ability hindered the clinical application of Celastrol. In this study, we aimed to design and synthesize an amphiphilic conjugate to encapsulate Celastrol into micelles to improve its water solubility, cellular membrane penetration, improving the clinic translation potential of Celastrol for the treatment of psoriasis. For this purpose, we first synthesized gelatin and oleic acid conjugate (GOC-1), and then covalently bonded 4-(3-boronophenylamino)-4-oxobutanoic acid (BPOA) with GOC-1 to form a stable GOC-2 conjugate which can self-assemble into micelles in aqueous solution. Celastrol (Cel) was physically encapsulated into the core of GOCs micelles. The dynamic stability, particle size, drug release, zeta potential, drug-loading efficiency, and surface morphology of Cel/loaded GOCs nano-micelles were determined. In addition, cell viability, cellular uptake of Cel/loaded GOC-2, and skin permeation and in vivo anti-psoriasis effect of Cel-loaded GOC-2 were investigated. Our results have shown that Cel/loaded GOC-1 and Cel/loaded GOC-2 have spherical shapes with diameters of around 200–300 nm. Compared to GOC-1, GOC-2 micelles showed higher drug-loading efficiency and excellent permeation ability in vitro. Moreover, Cel/GOC-2 micelles reduced erythema and white scales on the dorsal skin of psoriatic mice. In conclusion, BPOA attached GOC nanoparticles as a Celastrol carrier not only increase its water solubility but also improve drug-loading efficiency and cell permeation ability, exhibiting superior anti-psoriatic effect than the commercially available tacrolimus. Our work is expected to provide a facile approach to prepare nanocarrier for Celastrol to improve the clinic translation potential of Celastrol.

        Dual stimuli-responsive polymeric prodrug consisting of reversible covalent bonded celastrol for tumor targeted delivery

        In this work, pH/GSH-responsive amphiphilic polymeric prodrug (EDA-GLA/CE/2-FPBA) was successfully prepared and could self-assembled into micelles in an aqueous solution. The EDA-GLA/CE/2-FPBA micelles possessed high stability in physiological condition and were pH and GSH sensitive due to the reversible borate ester bonds and disulfide bonds within the prodrug polymer. The structures of the prodrug polymers were characterized by NMR, FTIR, UV–vis spectroscopy. Transmission electron microscopy and dynamic light scattering measurement indicated that the resulting micelles have desirable size distribution and regular spherical shape. Free active Celastrol can be released under low pH and high GSH environment; In vitro cellular uptake and growth inhibition assays suggested that the blank polymer micelles showed good biocompatibility. EDA-GLA/CE/2-FPBA micelles were more efficiently internalized by monolayer tumor cells and demonstrated superior tumor targeting effects as compared to free Celastrol control. These results demonstrated that the novel prodrug self-assembled dual-responsive nano-delivery platform was able to improve the bioavailability and tumor targeting activity of Celastrol, which provides a basis for further clinical applications of Celastrol and its derivatives.

        Cone-beam computed tomography-guided threedimensional evaluation of treatment effectiveness of the Frog appliance

        Objective: To evaluate the effectiveness of the Frog appliance in three dimensions by using cone-beam computed tomography (CBCT) images. Methods: Forty patients (21 boys and 19 girls), averaged 11.7 years old, with an Angle Class II division 1 malocclusion were included in our study. They had either late mixed dentition or early permanent dentition, and the maxillary second molars had not yet erupted. All patients underwent CBCT before and after the treatment for measuring changes in the maxillary first molars, second premolars, central incisors, and profile. Paired-samples t-test was used to compare the mean difference in each variable before treatment and after the first phase of treatment. Results: The maxillary first molars were effectively distalized by 4.25 mm (p < 0.001) and 3.53 mm (p < 0.05) in the dental crown and root apex, respectively. The tipping increased by 2.25o, but the difference was not significant. Moreover the teeth moved buccally by 0.84 mm (p < 0.05) and 2.87 mm (p < 0.01) in the mesiobuccal and distobuccal cusps, respectively, whereas no significant changes occurred in the root apex. Regarding the anchorage parts, the angle of the maxillary central incisor’s long axis to the sella-nasion plane increased by 2.76o (p < 0.05) and the distance from the upper lip to the esthetic plane decreased by 0.52 mm (p = 0.01). Conclusions: The Frog appliance effectively distalized the maxillary molars with an acceptable degree of tipping, distobuccal rotation, and buccal crown torque, with only slight anchorage loss. Furthermore, CBCT image demonstrated that it is a simple and reliable method for three-dimensional analysis.

        Characterization of a New Isolate of Chilli ringspot virus in Yunnan, China

        Virus diseases are the major limiting factors of chilli production in China. A virus isolate of chilli ringspot virus-Yunnan (ChiRSV-YN) from chilli (Capsicum annuum L.) showing mild mosaic, leaf crinkling, ringspot, and vein banding symptoms was recently identified in Yunnan, China. The electron microscopy confirmed the virus as a typical potyvirus having flexuous filamentous particles ca. 780 nm in length. Several chilli and tobacco varieties were tested through mechanical inoculation and found to be susceptible to the virus isolate ChiRSV-YN. The viral genome was sequenced through RNA sequencing. The complete genomic RNA of the isolate (GenBank Acc. No. KX258620) consisted of 9,652 nucleotides (nt) excluding the poly (A) tail at the 3ʹ end. The isolate contains a large ORF which encodes a polyprotein of 3,086 amino acids (aa) with Mr. of 349.21 kDa. It had a typical genomic organization of potyviruses. The complete nucleotide and polyprotein sequences of ChiRSV-YN had 87.6 and 93.2% identities with the ChiRSV-HN/14, respectively. The result showed ChiRSV-YN is a new isolate of ChiRSV and it is the first full-length genome characterization of ChiRSV in Yunnan, China. Understanding of the characteristics of this virus isolate might provide the basis for disease diagnosis and control in the future.

        Production of Cyanocarboxylic Acid by Acidovorax facilis 72W Nitrilase Displayed on the Spore Surface of Bacillus subtilis

        Nitrilase is a valuable hydrolase that catalyzes nitriles into carboxylic acid and ammonia. Its applications, however, are severely restricted by the harsh conditions of industrial reaction processes. To solve this problem, a nitrilase from Acidovorax facilis 72W was inserted into an Escherichia coli-Bacillus subtilis shuttle vector for spore surface display. Western blot, enzyme activity measurements and flow cytometric analysis results all indicated a successful spore surface display of the CotB-nit fusion protein. In addition, the optimal catalytic pH value and temperature of the displayed nitrilase were determined to be 7.0 and 50°C, respectively. Moreover, results of reusability tests revealed that 64% of the initial activity of the displayed nitrilase was still retained at the 10th cycle. Furthermore, hydrolysis efficiency of upscale production of cyanocarboxylic acid was significantly higher in the displayed nitrilase-treated group than in the free group expressed by E. coli (pET-28a-nit). Generally, the display of A. facilis 72W nitrilase on the spore surface of Bacillus subtilis may be a useful method for immobilization of enzyme and consequent biocatalytic stabilization.

        Nematicidal activity against Aphelenchoides besseyi and Ditylenchus destructor of three biflavonoids, isolated from roots of Stellera chamaejasme

        Aphelenchoides besseyi and Ditylenchus destructor can cause serious problems for a number of important agricultural crops and vegetables. In this study, the ethanol extract of Stellera chamaejasme L. roots showed strong nematicidal activity against Aphelenchoides besseyi and Ditylenchus destructor. By using a bioactivity-driven fractionation, three biflavonoids were isolated from the extract and their structures were identified by mass and nuclear magnetic resonance spectral data. Nematicidal activity bioassays revealed that isoneochamaejasmin A had the strongest nematicidal activity against A. besseyi and D. destructor with LC 50 values of 2.32 and 0.18 mM at 72 h, respectively. Chamaejasmenin B displayed weaker nematicidal activity against A. besseyi with an LC 50 value of 3.94 mM at 72 h. Neochamaejasmin B induced the lowest mortality against D. destructor with an LC 50 values of 15.6 mM at 72 h. These results suggested that the kind and position of substitutions and the relative configuration of 2-H/3-H and 2”-H/3”-H could be considered as important factors responsible for the nematicidal activity of these purified C-3/C-3″ biflavonoids.

        한·중 연어 대조를 위한 연어 개념 구축과 범주 한정

        본 연구는 한국어 '연어'(collocation)와 중국어 '搭配'를 논의함으로써 한·중 연어 대조를 위한 연어 개념을 구축하는 데 목적을 둔다. 최근 한·중 연어 대조 연구가 활발해진 것에 비하면 한국어 연어에 상응하는 중국어 개념에 대한 이론적 논의가 많이 부족하다. 영어 'collocation'에 대하여 일반적으로 한국어에서는 연어라고, 중국어에서는 '搭配'라고 한다. 본고는 한국어 연어와 중국어 搭配에 관한 논의를 살펴봄으로써 두 가지 개념은 일치하지 않는다고 지적하였다. 그리고 搭配의 유형을 분석한 결과 特定搭配/有限组合/受限搭配/限制性搭配 등을 한국어 연어에 상응하는 개념으로 볼 수 있으며, 이 중 본고에서 주장하는 연어 판별 기준을 고려해 '特定搭配'가 가장 적합하다고 보았다. 또한 일차적으로 '의미 해석의 투명성'을, 이차적으로 '어휘 선택 제약'을 연어와 特定搭配를 판별하는 기준으로 제시하고 그들의 범주를 규정하였다. The purpose of this study was to establish the concept of collocation for the Korean-Chinese collocation contrast by discussing Korean "collocation" and Chinese "搭配". Compared to recent active studies on Korean-Chinese collocation, there is a lack of theoretical discussion on Chinese concepts corresponding to Korean collocation. This paper points out that these two concepts do not match, looking at discussions on Korean collocation and Chinese 搭配. After analyzing the type of 搭配, "特定搭配/有限组合/受限搭配/限制性搭配" can be considered as a corresponding concept to Korean collocation, among which "特定搭配" is the most appropriate one considering the collocation discrimination criteria proposed in this paper. In addition, in terms of the criteria for collocation or 特定搭配, this study proposed "meaning interpretation of transparency" as the primary criterion and "lexical selection restrictions" as the secondary standard. This study also specified their category.
