Two new isoflavanones, (3R) 5,7,3',4'-tetrahydroxy-2'-methoxyisoflavanone (1) and (3R) 5',8-di-(γ,γ- dimethylallyl)-2',5-dihydroxyl-4',7-dimethoxyl-isoflavanone (2), were isolated from Uraria clarkei, together with two known compounds dalbergioidin (3), 5,7-dihydroxy-2',4'-dimethoxyisoflavanone (4). The structures involving the absolute configuration of the new compounds were well elucidated by MS, IR, UV, CD, 1D and 2D NMR analyses. Cytotoxicity of the four compounds were assessed, results suggested that compound 2 possessed well cytotoxic activity, against the Hela, K562, and HL60 cell lines with IC50 values of 28.0, 40.6 and 35.1 μM, respectively.
Bayesian Prediction Model Based on Attribute Weighting and Kernel Density Estimations

<P>Although naïve Bayes learner has been proven to show reasonable performance in machine learning, it often suffers from a few problems with handling real world data. First problem is conditional independence; the second problem is the usage of frequency estimator. Therefore, we have proposed methods to solve these two problems revolving around naïve Bayes algorithms. By using an attribute weighting method, we have been able to handle conditional independence assumption issue, whereas, for the case of the frequency estimators, we have found a way to weaken the negative effects through our proposed smooth kernel method. In this paper, we have proposed a compact Bayes model, in which a smooth kernel augments weights on likelihood estimation. We have also chosen an attribute weighting method which employs mutual information metric to cooperate with the framework. Experiments have been conducted on UCI benchmark datasets and the accuracy of our proposed learner has been compared with that of standard naïve Bayes. The experimental results have demonstrated the effectiveness and efficiency of our proposed learning algorithm.</P>
Screening of the Antigen Epitopes of Basic Fibroblast Growth Factor by Phage Display

In order to investigate the epitope of basic fibroblast growth factor (bFGF) and its immunogenicity, the epitopes of bFGF were screened from the phage display library with monoclonal antibody GF22, which can neutralize the bio-activity of bFGF. By three rounds of screening, the positive phage clones with bFGF epitopes were selected, which can effectively block the bFGF to bind with GF22. Sequence analysis showed that the epitopes shared a highly conservative sequence (Leu-Pro-Pro/Leu-Gly-His-Phe/Ile-Lys). The sequence of PPGHFK was located at 22-27 of the bFGF. The specific immuno-response of mouse could be highly induced by phage clones with the epitopes. And the anti-bFGF activity induced by LPGHFK was 3 times higher than the original sequence, which showed that the mimetic peptide LPLGHIK might be used as a tumor vaccine in the prevention and treatment of tumor.

Cyclin-dependent kinase 2 (CDK2) is a member of serine/threonine protein kinases, which initiates the principal transitions of the eukaryotic cell cycle and is a promising target for cancer therapy. The present study was designed to inhibit cdk2 gene expression to induce cell cycle arrest and cell proliferation suppression. Here, we constructed a series of RNA interference (RNAi) plasmids which can successfully express small interference RNA (siRNA) in the transfected human cells. The results showed that the RNAi plasmids containing the coding sequences for siRNAs down-regulated the cdk2 gene expression in human cancer cells at the mRNA and the protein levels. Furthermore, we found that the cell cycle was arrested at G0G1 phases and the cell proliferation was inhibited by different siRNAs. These results demonstrate that suppression of CDK2 activity by RNAi may be an effective strategy for gene therapy in human cancers. [BMB reports 2010; 43(4): 291-296]

Meta-analysis of Gene Expression Data Identifies Causal Genes for Prostate Cancer

Prostate cancer is a leading cause of death in male populations across the globe. With the advent of gene expression arrays, many microarray studies have been conducted in prostate cancer, but the results have varied across different studies. To better understand the genetic and biologic mechanisms of prostate cancer, we conducted a meta-analysis of two studies on prostate cancer. Eight key genes were identified to be differentially expressed with progression. After gene co-expression analysis based on data from the GEO database, we obtained a co-expressed gene list which included 725 genes. Gene Ontology analysis revealed that these genes are involved in actin filament-based processes, locomotion and cell morphogenesis. Further analysis of the gene list should provide important clues for developing new prognostic markers and therapeutic targets.
Shotgun analysis on the peritrophic membrane of the silkworm Bombyx mori

The insect midgut epithelium is generally lined with a unique chitin and protein structure, the peritrophic membrane (PM), which facilitates food digestion and protects the gut epithelium. We used gel electrophoresis and mass spectrometry to identify the extracted proteins from the silkworm PM to obtain an in-depth understanding of the biological function of the silkworm PM components. A total of 305 proteins, with molecular weights ranging from 8.02 kDa to 788.52 kDa and the isoelectric points ranging from 3.39 to 12.91, were successfully identified. We also found several major classes of PM proteins, i.e. PM chitin-binding protein, invertebrate intestinal mucin, and chitin deacetylase. The protein profile provides a basis for further study of the physiological events in the PM of Bombyx mori.

Transgenic research was preformed by transferring a cyanobacterial (Nostoc flagelliforme) iron superoxide dismutase gene (NfFeSOD) into heavy metal hyperaccumulator Sedum alfredii via Agrobacterium-mediated method. Beyond expectation, NfFeSOD-overexpressing S. alfredii plants exhibited profound impairments, including plant growth retardation, abnormal root architecture, and reduced leaf greenness, photosynthetic efficiency and metal accumulation efficiency. Although transgenic plants appeared physiologically sensitive to high temperature, a higher relative biomass growth was still observed under long-term high temperature and osmotic stresses. Further investigation found that reactive oxygen species (ROS) homeostasis of transgenic plants was significantly affected, being ~50 % reduction of H2O2 level relative to wild-type plants. Gene transcription including ROS responsive genes was overall attenuated in transgenic plants, being more significant at normal temperature than at high temperature. In addition, ascorbate peroxidase (APX) activity was increased nearly twofolds in transgenic plants as compared to wild-type control. It may be inferred that ectopic NfFeSOD overexpression gives rise to a substantial increase of APX activity and leads to a sharp reduction of H2O2 level, thus impairing basal ROS signaling and plant growth. Specific genetic background of S. alfredii may be responsible for this sharp reduction of H2O2 level induced by NfFeSOD overexpression. S. alfredii plant has acclimated to elevated levels of ROS induced by heavy metals in native habitats and should require high ROS levels for basal signaling. We thus suppose that a sustained disturbance of high basal ROS signaling in metal hyperaccumulators may instead incur very sensitive response and thus result in profound growth impairments.
