<P>Abstract: </P><P>Tanshinone IIA, a major component extracted from the traditional herbal medicine, <I>Salvia miltiorrhiza</I> Bunge, improves blood circulation and treats chronic hepatitis and hepatic fibrosis. Activation of hepatic stellate cells (HSCs) is the predominant event in liver fibrosis. The therapeutic goal in liver fibrosis is the reversal of fibrosis and selective clearance of activated HSCs. We used rat HSCs transformed by Simian virus 40 (t-HSC/Cl-6) to overcome the limitations inherent in studying subcultures of HSCs. Treatment of t-HSC/Cl-6 cells with tanshinone IIA inhibited cell viability in a dose- and time-dependent manner. Tanshinone IIA induced apoptosis as demonstrated by DNA fragmentation, poly(ADP-ribose) polymerase and caspase-3 cleavage, increased Bax/Bcl-2 protein ratio, and depolarization of mitochondrial membranes to facilitate cytochrome <I>c</I> release into the cytosol. Furthermore, this compound markedly induced S phase cell cycle arrest, and down-regulated cyclins A and E, and cdk2. Thus, tanshinone IIA induces apoptosis and S phase cell cycle arrest in rat HSCs <I>in vitro</I>.</P>
Apoptotic Effect of Propyl Gallate in Activated Rat Hepatic Stellate Cells

Hepatic stellate cells (HSCs) play a central role in liver fibrosis. Inhibition of HSC growth and induction of apoptosis have been proposed as therapeutic strategies for the treatment and prevention of liver fibrosis. Propyl gallate (PG) is an antioxidant widely used in processed foods, cosmetics and medicinal preparations. However, the anti-fibrotic effect of PG in liver injury is unclear. In this study, we investigated whether PG could induce apoptosis in activated HSCs. Treatment of activated HSCs with PG inhibited cell viability in a dose- and time-dependent manner. PG induced apoptosis as demonstrated by morphological changes, poly(ADP-ribose) polymerase (PARP) cleavage, caspase-3 cleavage, increased Bad expression, and decreased Bcl-2 protein expression. Through stimulation of the activation of c-Jun NH2-terminal protein kinase (JNK) and p38 mitogen-activated protein kinases (MAPK) by PG treatment, we demonstrated that JNK and p38 MPAK are not involved in PG-induced apoptosis using their specific inhibitors. Taken together, these findings indicate that PG induces apoptosis in activated HSCs. The potential anti-fibrotic effect of PG warrants further evaluation.

본 연구에서는 쪽방촌에 거주하는 장애인들의 쪽방 유입 과정과 현재 생활에 대한 생애사 분석을 하였다. 특히 장애가 이들의 쪽방 유입과정 및 현재의 생활에 어떤 영향을 미쳤는지에 관하여 분석하였는데, 장애는 빈곤을 심화시켜 참여자들을 쪽방으로 유입시킨 주요원인이었으며, 탈빈곤할 수단인 노동능력의 감소를 야기한 조건이었고, 결국 수급자로서 만성적 빈곤상태에서 살아가도록 만든 요인이었다. 또한, 한편으로는 장애가 최저생계의 생활을 할 수 있는 수급권 보장의 도구로 기능하여 쪽방촌에서 안정적으로 생활하는데 도움이 되기도 했다. 쪽방촌 거주 현실에 대한 분석 결과, 쪽방 거주 장애인은 “빈곤과 불편의 생활화 및 장애와 갈등의 일상화”로 표현되는 힘든 생활을 하고 있었고, 이웃과 갈등 또는 협력을 하고 있었다. 본 연구는 “현실에서의 감사와 이웃에서 찾은 희망”으로 요약되는 긍정적빈곤문화를 창출해내고 있는 연구 참여자들의 삶을 통해서 사회복지 정책적 시사점을 제시한다. 쪽방거주 장애인의 탈빈곤과 자립을 돕는 정책 마련이 필요하며, 특히 지역사회 자조활동을 지원하는 지역복지 대응체계가 필요하다. 빈곤연구 및 장애인복지 연구의 영역에서 쪽방거주 장애인을 고려한 후속연구가 또한 필요하다. The purpose of this study is to understand persons with disabilities living alone in a single room occupancy community and to suggest social work strategies for them. For data analyses, the life history methods (LHM) which focuses on the personal life story was employed. A total of 6 participants were selected from the original data collected by a research team at Korea University for a research grant funded by the Korea Health Promotion Foundation. Their open coded data were analyzed for this study. As a result, disability led the participants to live in a single room occupancy of poor people villege, jokbang-chon. The persons with disabilities experience long lasting poverty, physical pain, and environmental limitations. They also experiences ‘ambivalent culture of poverty`` representing conflicts or cooperations among neighbors. Based on trusts and cooperations in this poor village, more supports for their self-help groups are necessary. The persons with disabilities living alone in a single room occupancy should be also considered in the on-going discussion of exit-poverty and disability policies.
