      • Molecular and Cytogenetic Analysis of Transgenic Plants of Rice(Oryza sativa L.) Produced by Agrobacterium-mediated Transformation

        To demonstrate the importance of transformation efficiency in independent event, molecular and cytogenetic analysis were conducted with genomic DNA and chromosome of transgenic plants produced by Agrobacterium tumefeciens LBA4404 (pSBM-PPGN: gusA and bar). Selection ratios of putative transgenic calli were similar in independent experiments, however, transformation efficiencies were critically influenced by the type of regeneration media. MSRK5SS-Pr regeneration mediun, which contains 5 mgL$^{-1}$ 수식 이미지 kinetin, 2% (w/v) sucrose in combination with 3% (w/v) sorbitol, and 500 mgL$^{-1}$ 수식 이미지 proline, was efficient to produce transgenic plant of rice from putative transgenic callus in the presence of L-phosphinotricin (PPT). With MSRK5SS-Pr medium, transformation efficincies of Nagdongbyeo were significantly enhanced from 3.7% to 6.3% in independent callus lines arid from 7.3% to 19.7% in plants produced, respectively. Stable integration and expression of bar gene were confirmed by basta herbicide assay, PCR amplification and Southern blotting of bar gene, and fluorescence in situ hybridization (FISH) analysis using pSBM-PPGN as a probe. In Southern blot analysis, diverse band patterns were observed in total 44 transgenic plants regenerated from 20 independent PPT resistant calli showing from one to five copies of T-DNA segments, however, the transformants obtained from one callus line showed the same copy numbers with the same fractionized band patterns.

      • 생물학적 폐수처리 공정의 공정제어를 위한 on-line 전문가 시스템

        The study on process simulations, in the basis of a hybrid intelligent expert system which can support operations for the biological wastewater treatment processes, was practically performed. This expert system provides (ⅰ) tracking the cause of problems resulted from inexperienced process operations, such as an emergency shut-down or start-up, (ⅱ) process system and trend analysis and (ⅲ) real-time optimal process controls. Knowledge-base consists of about 100 production-rules for the activated sludge treatment process, including bio-reactor and final clarifier. It was designed to capture some of the knowledge in any case of the absence of experienced operators. The system provides stable process control and management and effectively helps inexperienced operators with advanced and standard technologies. It was implemented to be a hybrid-typed intelligent expert system by mutually connecting with the process control systems which has the capability of process diagnosis, analysis and control.

      • The Process Modelling and Control System Simulation for Optimal Control of Rotary Kiln Incinerator

        본 연구에서는 로타리 킬른식 소각로를 대상으로 공정 모델링 및 제어 시스템 모사를 수행하였다. 공정 모델링은 제어 시스템 모사에 적용되는 전문가 제어 시스템(RKEXPERT)의 정량적 공정조업 규칙(production rules)을 도출하기 위한 방편으로써 3차원 열모델을 적용했다. 3차원 열모델은 로타리 킬른의 각 축방향에서 상부층, 하부층, 기상 그리고 내화벽에서의 열 전달을 고려하여 온도 분포를 예측할 수 있다. 3차원 열모델을 이용함으로써 공정조업 규칙은 제어변수와 조작변수간의 상관관계를 통해서 얻을 수 있다. 제어 시스템 모사를 통해서 개발된 혼합형(hybrid) 지능형 공정조업 시스템은 공정조업을 감시하고 조업 데이터를 분석하는 공정 모니터링 및 제어 시스템과 공정변수의 변화가 발생될 때 즉, 설정된 정상 범위에서 벗어난 이상 상태일 때 그에 따른 적절한 조치사항을 추론(rule-compiling)을 통하여 제시하는 전문가 제어 시스템을 상호 인터페이스 시켜 비정상조업과 공정내 발생 가능한 이상 및 문제점에 대해 진단, 분석, 제어 기능을 동시에 수행할 수 있게 하였다.

      • 간암과 간세포이형성 부위의 철침착 저항성에 대한 조직학적 연구

        Liver cell dysplasia(LCD) is commonly associated with some liver diseases such as liver cirrhosis or hepatocellular carcinoma and has been regarded as a premaligant condition. In order to clarify the question of whether or not LCD is a premalignant liver lesion, iron special staining, Prussian blue, was done. The specimens were collected at the department of pathology, Chungnam National University College of Medicine from January 1983 to September 1992. The results were as follows: 1. Of observed 48 cases, 34 were hepatocellular carcinoma(70.8%), 11 were metastatic carcinoma(22.9%), and 3 were hepatoblastoma(6.3%). 2. LCD was present in 26 of 34 cases(76.5%) of hepatocellular carcinoma, 5 of 11 cases(45.5%) of metastatic carcinoma, and 1 of 3 case(33.3%) of hepatoblastoma. 3. The carcinoma cells were free of iron deposition(iron excluding foci) in contrast to the neighboring dysplastic or nonneoplastic normal hepatocytes. 4. The presence of iron deposition in LCD cases was similar to or more severe than that of normal hepatocytes. In summary, the results obtained from the present study indicate that LCD reflects neither a regenerating condition nor a premalignant lesion but suggests a reactive change or accompaning lession.

        하악 대구치 근심근관에 대한 전자근관장 측정기의 정확도

        근단부위 형성이 완료되고 우식이 없는 하악 대구치 근심치근 50개를 대상으로 주파수 의존형 전자근관장 측정기를 이용하여 측정한 근관장의 정확도를 평가하기 위해 치수강 개방 후 동일한 치아에서 방사선 사진으로 확인한 실측 근관장을 대조군, 각종 전자근관장 측정기로 측정한 근관장을 실험군 (Ⅰ군: Root-ZX, Ⅱ군: Bingo, Ⅲ군: Propex, Ⅳ군: Diagnostic)으로 분류하여 근심협측과 설측의 근관장을 근관의 입구 측으로부터 치근단 1/3까지 확대여부 및 서로 다른 측정시기에 비교 평가한 결과 다음과 같은 결론을 얻었다. 하악 대구치 근심치근의 협측과 전자근관장은 측정기의 종류, 근관의 입구 측으로부터 치근단 1/3까지의 확대여부 및 측정시기에 상관없이 0.5 ㎜ 이내의 오차범위를 나타내었으며, 근관치료시 오차한계인 ± 1 ㎜ 에서 정확성은 근관의 입구 측으로부터 치근단 1/3까지 미리 확대한 후에 근관장을 측정할 경우와 측정시기가 반복될 경우에 증가되는 경향을 보여 임상에 유용하게 활용할 수 있을 것으로 사료된다. The aim of this study was to compare the length between the mesio-buccal and mesio-lingual canal of the mandibular molars before and after early coronal flaring at the different measuring time using several electronic apex locators. Fifty mandibular molars with complete apical formation and patent foramens were selected. After establishing the initial working length of the buccal and lingual canal of the mesial root using a surgical microscope (Carl Zeiss Co. Germany) at 25X with #15 K-file tip just visible at the foramen, radiographs were taken for the working length. After measuring the length of mesio-buccal and mesio-lingual canal (control group), the electronic lengths were measured at different times using several electronic apex locators (experimental groups; Ⅰ-Root ZX, Ⅱ-Bingo, Ⅲ-Propex, Ⅳ- Diagnostic). After early coronal flaring using the K^(3) file, the additional electronic lengths were measured using the same manner. The results were as follows: One canal has a correct working length for the mesial root of the mandibular molar, it can be used effectively for measuring the electronic working length of another canal when the files are superimposed or encountered at the apex. In addition, the accuracy of the electronic apex locators was increased as the measurement was accomplished after the early coronal flaring of the root canal and the measuring time was repeated.

      • ECH 유도체와 수지성 폴리머를 함유한 Brine 폐수의 전처리를 위한 응집 및 침전 반응의 최적화 연구

        Epoxy resins are produced by the dehydro condensated reaction with ECH (Epichlorohydrin) and BPA(Bisphenoi-A) as raw reactants under sodium hydroxide(NaOH) as a catalyst, and from this reaction, salted water named of brine, which contains ECH derivatives in condition of emulsion as likely as glycidol and polymer resins, is produced as an undesired side product. This brire water is alkaline wastewater and causes process fouling problems by plugging and chemically depositing polymer particles on the surface of inner wall of reactors and pipes. and decreases the biodegradable efficiency in the wastewater process. In this study, the optimization of coagulation and sediment reactions, using inorganic and organic polymer coagulants, were performed to remove the causes occurring the process fouling phenomena. And also, based on this study, the methodologies applicable to the commercial processes including economical analysis were presented.

      • 화옹호 유역의 비점오염부하량 산정에 관한 연구

        In this study, the estimation of non-point source pollutant loads in accordance with the utilization of lands in the area of Hwa-Wong lake was carried out in order to provide basic engineering data for building up the treatment or management facilities applicable to this corresponding area. On the basis of water Qualify and flow measurements at the on-sites around three main streams (Nam-yang, Ja-an and Eo-eun) located in that area, the yearly discharging amount of non-point source pollutants and its contribution rate to the total amount of water pollutants were calculated by applying the effluent coefficients and reasoning equation method incited from references. It was found that the contribution rates of water pollutants resulted from the non-point sources were 79.2%, 81.4%, and 81.1% for BOD, T-N, and T-P respectively. Because of insufficient measuring data including rainfalls and inaccurate local statistics, in the period of this study, the accuracy on estimations needs to be more studied.

      • 고도 하수처리 공정의 최적 제어 알고리즘에 관한 연구

        Because of the limitation of controllable operation variables for the wastewater treatment process with variable hydrodynamic flow patterns, it may preclude the use of this type of nutrient removal activated sludge process. Only temperature and dissolved oxygen (DO) as the operation variables have been used to operate the process. From this point, this study starts to improve treatment efficiency and operability of the process, mentioned above, by the following methodologies: 1) the process and operation data analysis using the process simulation, 2) the determination of optimal control logic or algorithm using the pilot-scaled experimental apparatus and its operations, 3) the applications of experimental and simulation results to find the optimal process operation modes. In this study, it was found that the optimal operation mode, named 'save mode', in the basis of process variables such as the ammonia nitrogen concentration of inlet flow, temperature and flow rate, can reduce the operation cost, comparing with the present normal operation mode. And also, the stable conditions in nitrification were shown by the proportional control of DO with the inlet air flow rate of blower and the mixing rate of mechanical aeration. This study has shown to be considerably feasible for the applications of its results to the commercial scaled wastewater treatment process.

        In this study, fiber-optic sensor is fabricated to measure PDD of CLINAC. As a scintillating light generated by a high energy X-ray from the medical LINAC is measured using photo diode amp system, It is expected that a high-resolution, real-time fiber-optic dosimeter for medical LINAC can be developed.
