        간세포암과 하대정맥 막성폐쇄의 치료 후 호전된 백혈구파괴혈관염 1예

        저자들은 하지 자색반과 고환 통증으로 내원한 알코올성 간경화증 환자에서 LCV의 원인 감별하는 과정에서 HCC와 하대정맥 막성폐쇄를 진단하고 이에 대한 치료 후 LCV의 호전된 증례를 경험하였기에 문헌고찰과 함께 보고하는 바이다. Vasculitis is a heterogeneous group of diseases that destroy blood vessel walls by inflammation. Approximately half of vasculitis cases are idiopathic, but sometimes associated with genetic factors, medicines, chronic infection, autoimmune diseases, and malignancies. Although the mechanism remains unclear, vasculitis secondary to malignancy, also known as paraneoplastic vasculitis, has been reported. It is generally associated with hematologic malignancies rather than solid malignancies and commonly presents as leukocytoclastic vasculitis or polyarteritis nodosa. We experienced a case of leukocytoclastic vasculitis in a patient with hepatocellular carcinoma and membranous obstruction of the inferior vena cava. Here, we report this case with a brief review of literature. (J Rheum Dis 2015;22:322-326)

        This study, changes in raw water quality is to indicate on the efficiency of ozone treatment of each pollutant as compared to derive the appropriate operating measures.         2010 해남, 평창, 원주지역 갈대 등 야초류의 이용실태와 사료가치 평가

        본 연구는 부존 조사료자원 이용으로 수입사료를 줄이고 사료비를 절감하고자 2010년도에 전남 해남(간척지), 강원 평창(강변), 원주(섬강변), 전남 영광(와탄강변) 등 4개 지역에서 갈대와 억새 등 야초 자생지를 탐색하고, 수확·이용실태 조사와 함께 사료가치를 분석하였다. 전남 해남 해남목장 내 갈대(Phragmites communis) 위주의 야초 자생지는 300 ha로 예년에는 2회 곤포작업하여 연간 사일리지로 2,300~2,500톤(ha당 8톤)을 생산하였으나 2010년도는 1회 곤포조제로 사일리지 2,000톤(ha당 7톤)을 생산하였다. 유통가격은 롤 당 53천원으로 ㎏당 110원이었다. 곤포는 전량 자가소비하며, 외부 작업단에 롤 당 26천원의 작업비를 지불하였다. 연간 조수입은 2억 2,260만원이며, 소득은 1억원 수준(345천원/ha)이었다. 초장 150 ㎝의 갈대는 조단백질 8.4%, 상대사료가치 71.9 (건초 5등급), 건물 소화율 60.1%로 사료가치는 볏짚에 비해 약간 나은 것으로 평가되었다. 갈대 사일리지의 사료가치와 유기산 함량은 동일 지역에서 동일한 재료로 사일리지를 조제하더라도 수확시기에 따라 차이가 컸으며(CP 4.7~6.4%, RFV 62.2~78.9, 건물 소화율 41.9~53.9%), 갈대 사일리지의 품질은 수단그라스에 비해 불량하였다. 강원 평창 강변 자생갈대의 사료가치는 초장이 79, 117, 121, 142㎝로 생육이 진행됨에 따라 낮아져, 조단백질은 각각 13.9%에서 7.6%로, 상대사료가치는 90.7 (건초 3등급)에서 76.1 (건초 4등급)로, 건물 소화율은 72.9%에서 54.7%로 감소하였다. 원주 섬강 주변 야초류는 곤포수확 작업에 어려움이 있었으며, 초장 130 cm의 갈대는 조단백질 8.5%, 상대사료가치 82.3 (건초 4등급), 건물 소화율 70.2%로 사료가치는 높았으며, 220㎝의 성숙한 갈대는 조단백질 10.2%, 상대사료가치 65.1 (건초 5등급), 건물 소화율 48.9%로 볏짚 대비 조금 나은 것으로 평가되었다. 전남영광지역에서는 와탄천 주변에 자생하는 갈대를 수거 이용하고자 시도하였으나 경제성이 없어 중단하였다. 이상의 결과를 종합하여볼 때, 갈대, 억새 등 야초류는 현 상태에서 최대한 수거·이용하되, 사료가치를 유지하고 가축 기호성이 양호한 시기에 수확하는 것이 중요하며, 최소한 볏짚과 비슷하거나 그 이상의 사료가치를 기대하기 위해서는 7월 중순 이전(늦어도 7월까지)에 수확·이용하는 것이 권장된다. This study was carried out in 2010 to investigate the utilization and forage quality of native grasses, such as Phragmites communis which might reduce the cost of feeding domestic cattle. The regions surveyed were the Haenam ranch in Haenam, Pyeongchang, Wonju, and Yeonggwang. In Haenam, yearly silage production harvested from 300 ha was 2,000 MT (7 MT/ha). All of those round bale silages were self-consumed in that region, and marketing price was 50,000 55,000 won per roll (110 won/㎏). Phragmites communis of 150 ㎝ in length contained 8.4% crude protein (CP) with relative feed value (RFV) 71.9 and 60.1% in vitro dry matter digestibility (IVDMD), which was somewhat more favorable than forage quality of rice straw. The silage quality of Phragmites communis varied greatly by the time of harvest (CP 4.7 6.4%, RFV 62.2 78.9, and IVDMD 41.9 53.9%), even with the same district and of the same original forage materials. Generally, the quality of Phragmites communis silage of was poorer than that of sorghum * sudangrass hybrid. In Pyeongchang, forage quality of Phragmites communis was decreased from 13.9% to 7.6% in CP, 90.7 to 76.1 in RFV, and 72.9% to 54.7% in IVDMD, as plant was getting mature, from 79 ㎝ to 117 ㎝, 121 ㎝ or to 142 ㎝ in length. In Wonju, the quality values of Phragmites communis of 130 ㎝ in length were 8.5% CP, 82.3 RFV and 70.2% IVDMD, while those of matured grasses of 220 ㎝ in length were lower (10.2% CP, 65.1 RFV and 48.9% IVDMD), but this was a little more favorable than quality of rice straw. In Yeonggwang, feeding Phragmites communis was tried in a Hanwoo feed, but stopped due to low profitability. In conclusion, the overall quality of most native grasses including Phragmites communis in this survey was poor. Therefore, we recommend that Phragmites communis and native grasses should be harvested on June or July to obtain richer forage quality in forage values than rice straw.

        Recovery Effects from Oxidative Cell Damage by So-Hap-Hyang-Won on Bovine Aortic Endothelial Cells (BAEC)

        So-Hap-Hyang-Won, a traditional oriental medicine used in the treatment of stroke patients, was examined for its ability to reverse the cell damage caused by lipid peroxidation products and oxidative stress in bovine aortic endothelial cells (BAEC). The effects of herbal medicine on cell proliferation and recovery of oxidative damaged situation were studied in BAEC, which was considered an appropriate in vitro model for stroke resulting from various vascular diseases prevalent in advanced age. In a clinical study of stroke patients, So-Hap-Hyang-Won appeared to improve considerably arm and leg movements as well as consciousness disturbance condition, compared with other traditional medicines used for stroke. When BAEC were treated with extracts of the lyophilized herbal medicines, only that of So-Hap-Hyang-Won stimulated cell proliferation and showed no toxicity even at high concentrations. In studies of BAEC treated with extracts of the lyophilized material of the 14 components of So-Hap-Hyang-Won, only the extract of Foeniculi Fructus stimulated cell growth at all concentrations tested. Moreover, when cells were treated with Foeniculi Fructus (10 and 100 mg/ml) extract after prior exposure to t-BHP ($l0\mu\textrm{M}$) or HNE ($0.2\mu\textrm{M}$), lipid peroxidation products which are known to be involved in aging and vascular diseases, or after the exposure to SIN-l ($500\mu\textrm{M}$), which generates nitric oxide (NO) and other reactive oxygen species, there was substantial recovery from the oxidative damage, presumably due to the radical-scavenging effect of Foeniculi Fructus extract. Foeniculi Fructus not only showed stimulatory effects on cell growth and cell damage repair in BAEC, but also appeared to show the most anti-aging activity among all the herbal components of So-Hap-Hyang-Won.

        Promiscuous methionyl-tRNA synthetase mediates adaptive mistranslation to protect cells against oxidative stress

        <P><B>ABSTRACT</B></P><P>Aminoacyl-tRNA synthetases (ARSs) acylate transfer (t)RNAs with amino acids. Charging tRNAs with the right amino acids is the first step in translation; therefore, the accurate and error-free functioning of ARSs is an essential prerequisite for translational fidelity. A recent study found that methionine (Met) can be incorporated into non-Met residues of proteins through methionylation of non-cognate tRNAs under conditions of oxidative stress. However, it was not understood how this mis-methionylation is achieved. Here, we report that methionyl-tRNA synthetase (MRS) is phosphorylated at Ser209 and Ser825 by extracellular signal-related kinase (ERK1/2) under conditions of stress caused by reactive oxygen species (ROS), and that this phosphorylated MRS shows increased affinity for non-cognate tRNAs with lower affinity for tRNA<SUP>Met</SUP>, leading to an increase in Met residues in cellular proteins. The expression of a mutant MRS containing the substitutions S209D and S825D, mimicking dual phosphorylation, reduced ROS levels and cell death. This controlled inaccuracy of MRS seems to serve as a defense mechanism against ROS-mediated damage at the cost of translational fidelity.</P>

      • Expression of an artificial gene encoding a repeated tripeptide lysyl-glutamyl-tryptophan in Tobacco plant

        식물 단백질의 영양가 향상을 위한 일환으로 필수아미노산의 조성이 풍부한 인공단백질을 암호화하는 인공유전자를 담배 식물체에서 발현을 시도하기 위하여, 식물에서 외래유전자의 발현에 널리 사용되는 Cauliflower mosaic virus (CaMV)의 35S promoter를 이중으로 중첩되도록 하고, (Lys-Glu-Trp)이 64번 반복되는 인공유전자 및 nopaline synthase (nos) terminator를 갖고 있는 binary vector pART4-4를 구성하였다. 이 재조합 플라스미드는 Agrobacterium tumefaciens를 이용한 형질전환에 의해 Nicotiana tabacum (Var. Xanthi)으로 도입되었다. Kanamycin이 포함된 신초 유도배지 및 뿌리 유도배지를 이용하여 정상적으로 재생된 담배 식물체로부터 도입된 인공유전자의 발현을 분석하였다. 추출한 genomic DNA를 EcoPI으로 자른 다음 Southern blot 분석에 의하면, 효소 절단 시 예상되는 1.1 kb에서 band를 형성하였으며 각각의 형질전환 식물체에 인공유전자가 1 또는 3 개씩 도입되어 있음을 확인하였다. Northern blot 분석에 의하면 약 1.2 kb 전사체가 비교적 안정하게 발현되었으며, 잎, 줄기, 뿌리로부터 RNA를 분리하여 promoter의 조직 특이성 발현을 분석한 결과, 잎에서 생성되는 RNA가 줄기나 뿌리 조직보다 안정하게 발현되었다. 형질전환 식물체에서 Western blot에 의한 단백질 분석 결과, 잎에서 추출한 단백질로부터 원하는 크기인 33 kDa의 인공단백질이 생성됨을 확인하였으며 발현 수준은 전체 세포 단백질의 0.1%로서 낮은 수준이었다. To investigate expression of the artificial gene encoding a repeated tripeptide lysyl-glutamyl-tryptophan in tobacco plant, the plant binary vector, pART404 has been constructed, which contains the duplicated CaMV 35S promoter, an artificial gene coding for repetitive polymer (Lys-Glu-Trp)_64, and nopaline synthase (nos) terminator. The recombinant expression vector was introduced in Nicotiana tabacum (var. Xanthi) via Agrobacterium tumefaciens-mediated transformation. The transgenic calli selected by kanamycin containing medium were then regenerated to whole plants. Southern blot analysis indicated that five transgenic plants (No. 1, 7, 9, 43, 45) showed the hybridizing signals at 1.1kb of the expected size on ExoRI digestion and each of the transgenic plants contained 1 or 3 copies of the artificial gene inserted into its genome. By northern blot analysis, the size of the hybridized total RNA was estimated to be approximately 1.2 kb and the RNA appeared generally to have the integrity. Western blot indicated that the protein was detected at the position of 33 kDa and the expression level of the polypeptide in the transgenic plant (No. 45) was measured to approximately 0.1% of the total protein.

        A New Double-Stranded RNA Mycovirus from Pleurotus ostreatus (ASI 2504)

        A new virus with a dsRNA genome was isolated and characterized from the Suhan-:neutari strain (ASI 2504) of Pleurotus ostreatus, which was characterized as long and slightly bent with small caps on the stipe of fruit body. Thirty nm isometric viruses with three dsRNA segments (approximately 2.0, 1.84 and 1.82 kb in sizes) were isolated by ultracentrifugation in sucrose gradients. Western analysis of protein extracted purified viruses with anti-virus polyclonal antibody confirmed that viruses have two specific proteins (36 and 68 kDa). Computer analysis of 2.0 kb segment shows that high. sequence identity with RNA-dependent RNA polymerase (RdRp) of partitiviruses, respectively. When compared to other dsRNA mycoviruses in a phylogenetic analysis, OMDV was most related to Pleurotus ostreatus virus 1.
