        High-throughput Screening Strategy Used for Enhanced Production of Pigment by Monascus purpureus D39-4

        Most of the fermentation experiment designs were limited by the low-throughput of shake flask, especially for the medium optimization. A simple high-throughput screening system was developed for the determination of pigment in Monascus purpureus fermentation samples. This downscaled system was designed to optimize medium composition combined with statistical methods. The total 29 experiments designed by the Box–Behnken were used to study the 4 most important operating variables on pigment production. The analysis revealed that the optimum concentrations of glucose, peptone, NaNO3, and KH2PO4were 51.42, 4.91, 1.00, and 1.00 g/L, respectively. A production of 69.5 U/mL was achieved in agreement with the prediction (68.9 U/mL) fermented in 24-deep-well microtiterplates. Furthermore, the fermentation medium optimized in the high-throughput system was verified in shake flasks, and the pigment production could be enhanced from 206.5 U/mL in un-optimized medium to 265.8 U/mL,giving nearly 1.30-fold increase in production.

        Power control of CiADS core with the intensity of the proton beam

        This paper reports the control method for the core power of the China initiative Accelerator DrivenSystem (CiADS) facility. In the CiADS facility, an intense external neutron source provided by a protonaccelerator coupled to a spallation target is used to drive a sub-critical reactor. Without any control rodinside the sub-critical reactor, the core power is controlled by adjusting the proton beam intensity. Inorder to continuously change the beam intensity, an adjustable aperture is considered to be used at theLow Energy Beam Transport (LEBT) line of the accelerator. The aperture size is adjusted based on theProportional Integral Derivative (PID) controllers, by comparing either the setting beam intensity or thesetting core power with the measured value. To evaluate the proposed control method, a CiADS coremodel is built based on the point reactor kinetics model with six delayed neutron groups. The simulations based on the CiADS core model have indicated that the core power can be controlled stably byadjusting the aperture size. The response time in the adjustment of the core power depends mainly onthe adjustment time of the beam intensity

        Extracellular products-mediated interspecific interaction between Pseudomonas aeruginosa and Escherichia coli

        The Gram-negative pathogen Pseudomonas aeruginosa adopts several elaborate strategies to colonize a wide range of natural or clinical niches and to overcome the neighboring bacterial competitors in polymicrobial communities. However, the relationship and interaction mechanism of P. aeruginosa with other bacterial pathogens remains largely unexplored. Here we explore the interaction dynamics of P. aeruginosa and Escherichia coli, which frequently coinfect the lungs of immunocompromised hosts, by using a series of on-plate proximity assays and RNA-sequencing. We show that the extracellular products of P. aeruginosa can inhibit the growth of neighboring E. coli and induce a large-scale of transcriptional reprogramming of E. coli, especially in terms of cellular respiration- related primary metabolisms and membrane components. In contrast, the presence of E. coli has no significant effect on the growth of P. aeruginosa in short-term culture, but causes a dysregulated expression of genes positively controlled by the quorum-sensing (QS) system of P. aeruginosa during subsequent pairwise culture. We further demonstrate that the divergent QS-regulation of P. aeruginosa may be related to the function of the transcriptional regulator PqsR, which can be enhanced by E. coli culture supernatant to increase the pyocyanin production by P. aeruginosa in the absence of the central las-QS system. Moreover, the extracellular products of E. coli promote the proliferation and lethality of P. aeruginosa in infecting the Caenorhabditis elegans model. The current study provides a general characterization of the extracellular products-mediated interactions between P. aeruginosa and E. coli, and may facilitate the understanding of polymicrobial infections.

        Topological structure of Rubik’s cube mechanism and synthesis of the regular hexahedron Rubik’s cube mechanism

        This study investigates the factors affecting the topology of Rubik’s cube mechanism (RCM) and presents the synthesis theory of RCM. First, the revolute axis symmetry of the RCM is described and analyzed, and the general expression needed for the revolute axis to meet the symmetry is introduced. Next, the symmetry expression of the revolute pair contact surface (RPCS) of the RCM is given. The intersection between the RPCSs is analyzed via the adjacency matrix, and the adjacency matrix characteristics are given along with the motion characteristics of RCM. After determining the RPCS intersection, its topology is studied. Finally, the synthesis theory of RCM is presented, and the synthesis process and flowchart are proposed. The regular hexahedron RCMs with different topological structures are obtained after applying the synthesis theory. Hence, the theory proposed in this study provides strong theoretical support for the synthesis of other new RCMs.

        Electrosynthesis of Pure Poly(3,4-ethylenedioxythiophene) (PEDOT) in Chitosan-Based Liquid Crystal Phase

        Ordered poly(3,4-ethylenedioxythiophene) (PEDOT) was successfully prepared by electrochemical polymerization of 3,4-ethylenedioxythiophene in lyotropic liquid crystal phase formed by chitosan in an acetic acid solution. The structure, performance, and morphology of PEDOT were characterized by infrared absorption spectra, cyclic voltammograms, polarizing optical microscopy, and scanning electron microscopy. The ordered pure PEDOT film with good electrochemical behavior exhibited obvious birefringence.

        Alpha-Hemolysin from Staphylococcus aureus Obstructs Yeast-Hyphae Switching and Diminishes Pathogenicity in Candida albicans

        The use of antibiotics can disrupt the body’s natural balance and increase the susteptibility of patients towards fungal infections. Candida albicans is a dimorphic opportunistic fungal pathogen with niches similar to those of bacteria. Our aim was to study the interaction between this pathogen and bacteria to facilitate the control of C. albicans infection. Alpha-hemolysin (Hla), a protein secreted from Staphylococcus aureus, causes cell wall damage and impedes the yeast–hyphae transition in C. albicans. Mechanistically, Hla stimulation triggered the formation of reactive oxygen species that damaged the cell wall and mitochondria of C. albicans. The cell cycle was arrested in the G0/G1 phase, CDC42 was downregulated, and Ywp1 was upregulated, disrupting yeast hyphae switching. Subsequently, hyphae development was inhibited. In mouse models, C. albicans pretreated with Hla reduced the C. albicans burden in skin and vaginal mucosal infections, suggesting that S. aureus Hla can inhibit hyphal development and reduce the pathogenicity of candidiasis in vivo.
