        Postthrombolytic Antiplatelet Use for Patients with Intercerebral Hemorrhage without Extensive Parenchymal Involvement Does Not Worsen Outcome

        Background and Purpose It is unclear whether postthrombolytic antiplatelet (AP) therapy after thrombolytic-related hemorrhage without extensive parenchymal involvement (THEPI) afects the clinical outcome. Tis study explored whether AP administration in patients with THEPI afects short- and long-term outcomes. Methods All of the data for this study were collected from the Trombolysis Implementation and Monitor of Acute Ischemic Stroke in China (TIMS-China) registry. Patients with THEPI were assigned to either the AP (AP therapy should be commenced 24 h afer intravenous thrombolysis) or AP-naïve groups. THEPI was defned according to European-Australasian Acute Stroke Study II criteria. Te 90-day functional outcome, 7-day National Institutes of Health Stroke Scale (NIHSS) score, and 7-day and 90-day mortalities were compared between the AP and AP-naïve groups. Logistic regression analysis was used to evaluate the effects of AP therapy on the short- and long-term clinical outcomes. Results Of the 928 patients enrolled from those in the TIMS-China registry (n=1,440), 89 (9.6%) had nonsymptomatic intracerebral hemorrhage (ICH) within 24–36 h afer thrombolysis; 33 (37%) of these patients were given AP therapy (AP group) and 56 (63%) were not (APnaïve group). No significant differences were found for the risk of 7-day aggravated ICH (p=0.998), 7-day NIHSS score (p=0.5491), 7-day mortality [odds ratio (OR)=3.427; 95% confdence interval (95% CI)=0.344–34.160; p=0.294], 90-day mortality (OR=0.788, 95% CI=0.154– 4.040, p=0.775), or modifed Rankin score 5 or 6 at 90-days (OR=1.108, 95% CI=0.249–4.928, p=0.893) between the AP and AP-naïve groups afer THEPI. Conclusions Early administration of postthrombolytic AP therapy afer THEPI does not worsen either the short- or long-term outcome. AP therapy may be a reasonable treatment option for patients with THEPI to reduce the risk of ischemic stroke recurrence.

        Effect of 20 mg/day Atorvastatin: Recurrent Stroke Survey in Chinese Ischemic Stroke Patients with Prior Intracranial Hemorrhage

        Background and Purpose Treatment with atorvastatin (80 mg) in stroke secondary prevention for patients with prior intracranial hemorrhage (ICH) has been associated with a higher frequency of ICH. The aim of this study was to determine whether 20 mg/day atorvastatin is linked to stroke recurrence in Chinese ischemic stroke patients with prior ICH. Methods A single-center retrospective cohort study was conducted, involving 354 cases from 395 Chinese in-patients who had ischemic stroke with prior ICH history in Beijing Chaoyang hospital from May 1, 2005 to October 31, 2010. Survivors were followed by telephone interviews for 12-60 months. Cox regression and Kaplan-Meier plot analysis were used to evaluate the effect of 20 mg/day atorvastatin on cerebral infarction and ICH recurrence. Results The overall rate of stroke recurrence was lower in the 20 mg/day atorvastatin group(χ2=6.687, p=0.022) than in the control group. The incidence of cerebral hemorrhage was increased by 20 mg/day atorvastatin for ischemic stroke cases with a history of ICH compared tothose not receiving the drug, but the difference was not significant [hazard ratio (HR)=1.097,95% confidence interval (CI)=0.800-1.243, p=0.980]. The incidence of ischemic stroke recurrence was significantly reduced in subjects receiving atorvastatin (HR=0.723, 95% CI=0.578-0.862, p=0.028), and the mean duration of all stroke recurrences was significantly prolonged,compared with those not exposed to the drug (χ2=5.351, p=0.021). The mean duration of ICH recurrence appeared to have shortened with atorvastatin, but the difference was not significant (χ2=0.680, p=0.480), and the mean duration of cerebral infarction recurrence was significantly prolonged (χ2=8.312, p=0.004). Conclusions Medication with 20 mg/day atorvastatin may be beneficial in reducing ischemic stroke recurrence in ischemic stroke patients with a history of ICH and is not associated with an increased risk of ICH recurrence.

        Black rice anthocyanidins prevent retinal photochemical damage via involvement of the AP-1/NF-κB/Caspase-1 pathway in Sprague-Dawley Rats

        The effects of black rice anthocyanidins (BRACs) on retinal damage induced by photochemical stress are not well known. In the present study, Sprague-Dawley rats were fed AIN-93M for 1 week, after which 80 rats were randomly divided into two groups and treated with (n = 40) or without BRACs (n =40) for 15 days, respectively. After treatment, both groups were exposed to fluorescent light (3,000 ± 200 lux; 25oC), and the protective effect of dietary BRACs were evaluated afterwards. Our results showed that dietary BRACs effectively prevented retinal photochemical damage and inhibited the retinal cells apoptosis induced by fluorescent light (p < 0.05). Moreover, dietary BRACs inhibited expression of AP-1 (c-fos/c-jun subunits), up-regulated NF-κB (p65) expression and phosphorylation of IκB-α, and decreased Caspase-1 expression (p < 0.05). These results suggest that BRACs improve retinal damage produced by photochemical stress in rats via AP-1/NF-κB/Caspase-1apoptotic mechanisms.

        MiR-17-5p modulates osteoblastic differentiation and cell proliferation by targeting SMAD7 in non-traumatic osteonecrosis

        MicroRNAs (miRNAs) have recently been recognized to have a role in human orthopedic disorders. The objective of our study was to explore the expression profile and biological function of miRNA-17-5p (miR-17-5p), which is well known to be related to cancer cell proliferation and invasion, in osteoblastic differentiation and in cell proliferation. The expression levels of miR-17- 5p in the femoral head mesenchymal stem cells of 20 patients with non-traumatic osteonecrosis (ON) and 10 patients with osteoarthritis (OA) were examined by quantitative reverse transcription-PCR (qRT–PCR). Furthermore, the interaction between miR-17-5p and SMAD7 was observed. We found that in non-traumatic ON samples the level of mature miR-17-5p wassignificantly lower than that of OA samples (P¼0.0002). By targeting SMAD7, miR-17-5p promoted nuclear translocation of b-catenin, enhanced expression of COL1A1 and finally facilitated the proliferation and differentiation of HMSC-bm cells. We also demonstrated that restoring expression of SMAD7 in HMSC-bm cells partially reversed the function of miR-17-5p. Together, our data suggested a theory that dysfunction of a network containing miR-17-5p, SMAD7 and b-catenin could contribute to ON pathogenesis. The present study prompts the potential clinical value of miR-17-5p in non-traumatic ON.

        Characterization of Bacteriostatic Sausage Casing: A Composite of Bacterial Cellulose Embedded with ε-Polylysine

        The feasibility of a novel bacteriostatic sausage casing made of bacterial cellulose (BC) embedded with ε-polylysine (ε-PL) was evaluated. The ε-PL/BC composite was prepared by immersing BC tubes in ε-PL solution and its characteristics were analyzed with Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD),and atomic force microscopy (AFM) techniques, respectively. The results suggested that ε-PL molecules were incorporated into the cellulose fiber networks and the ε-PL/BC composite might have a novel unique structure. No significant loss of antimicrobial activity was observed even after autoclaving at 121℃ for 30 min and the oxygen permeability was far below than that of polyethylene (PE) and polyvinyl alcohol (PVA) membrane. Its tensile strength was 51.8 MPa. The ε-PL/BC composite exhibits bacteriostatic and/or bacteriocidal activities against a broad spectrum of Gram-positive and Gram-negative bacteria; as a result, an extended shelf life than controls was observed for sausage packaged with the ε-PL/BC composite.

        Performance of Ca-Based Oxygen Carriers Decorated by K<sub>2</sub>CO<sub>3</sub> or Fe<sub>2</sub>O<sub>3</sub> for Coal Chemical Looping Combustion

        <P>Chemical looping combustion (CLC) of coal is an attractive technology with inherent CO<SUB>2</SUB> separation and high energy utilization efficiency. The large-scale preparation of a cheap oxygen carrier with high attrition resistance challenges the scale-up step of CLC reactor systems. To improve the reactivity between the CaSO<SUB>4</SUB>/bentonite (CaBen) oxygen carrier and coal, CaSO<SUB>4</SUB>–Fe<SUB>2</SUB>O<SUB>3</SUB>/bentonite (CaFeBen) and CaSO<SUB>4</SUB>–K<SUB>2</SUB>CO<SUB>3</SUB>/bentonite (CaKBen), decorated by Fe<SUB>2</SUB>O<SUB>3</SUB> and K<SUB>2</SUB>CO<SUB>3</SUB>, respectively, were prepared in this work. The active component content, multi-cycle reactivity, and enhancement mechanism of two decorated oxygen carriers were investigated in a fluidized bed with steam as the gasification–fluidization medium. Finally, three types of coals, including lignite, bitumite, and anthracite, were used as fuel. The addition of Fe<SUB>2</SUB>O<SUB>3</SUB> and K<SUB>2</SUB>CO<SUB>3</SUB> can improve the reactivity of the CaBen oxygen carrier but degrade the attrition resistance slightly. The multiple-cycle experiments indicated that Fe<SUB>2</SUB>O<SUB>3</SUB> itself is the oxygen carrier for coal CLC with high reactivity, while K<SUB>2</SUB>CO<SUB>3</SUB> acts as the catalysis for coal gasification. The carbon conversion rate of the three coals that reacted with the CaKBen oxygen carrier was higher than that with CaFeBen as the oxygen carrier because of catalysis of potassium on the coal gasification reaction. However, the CaKBen oxygen carrier particles were seriously sintered, and the potassium content in the oxygen carrier reduced with the increasing redox cycles. The coals with high volatile and ash contents have a high instantaneous rate of carbon conversion reacted with two decorated oxygen carriers.</P>

        The Up-Regulation of miR-199b-5p in Erythroid Differentiation Is Associated with GATA-1 and NF-E2

        MicroRNAs (miRNAs) represent a class of small non-co-ding regulatory RNAs that play important roles in normal hematopoiesis, including erythropoiesis. Although studies have identified several miRNAs that regulate erythroid commitment and differentiation, we do not understand the mechanism by which the crucial erythroid transcription factors, GATA-1and NF-E2 directly regulate and control differentiation via miRNA pathways. In this study, we identified miR-199b-5p as a key regulator of human erythropoiesis, and its expression was up-regulated during the erythroid differentiation of K562 cells. Furthermore, the increase of miR-199b-5p in erythroid cells occurred in a GATA-1- and NF-E2-dependent manner during erythrocyte maturation. Both GATA-1 and NF-E2 bound upstream of the miR-199b gene locus and activated its transcription. Forced expression of miRNA-199b-5p in K562 cells affected erythroid cell proliferation and maturation. Moreover, we identified c-Kit as a direct target of miR-199b-5p in erythroid cells. Taken together, our results establish a functional link among the erythroid transcription factors GATA-1/NF-E2, miR-199b-5p and c-Kit, and provide new insights into the coupling of transcription and post-transcription regulation in erythroid differentiation.

      • Coal Chemical Looping Gasification for Syngas Generation Using an Iron-Based Oxygen Carrier

        <P>The chemical-looping gasification (CLG) of coal is a clean and effective technology for syngas generation. Sharing principles with chemical-looping combustion (CLC), CLG also uses oxygen carriers to transfer lattice oxygen to the fuel. Investigations into CLG with different O/C ratios are carried out in a fluidized bed reactor with steam used as the gasification–fluidization medium. The effect of the active component content of the oxygen carrier on the gas selectivity is performed, and reaction mechanisms between the Fe<SUB>2</SUB>O<SUB>3</SUB> oxygen carrier and coal with steam as the gasification agent are discussed. Moreover, we also assessed the reactivity of the CaO-decorated iron-based oxygen carrier particles in multicycle reactions. The carbon conversion efficiency is increased from 55.74 to 81% with increasing O/C ratio, whereas the content of H<SUB>2</SUB> first decreases and then increases. The addition of CaO can increase the carbon conversion efficiency and the gasification rate substantially and reduce the generation rate of H<SUB>2</SUB>S from 1.89 × 10<SUP>–3</SUP> to 0.156 × 10<SUP>–3</SUP> min<SUP>–1</SUP>. Furthermore, X-ray diffraction (XRD) images indicate that the CaO-decorated iron-based oxygen carrier particles were completely regenerated after six redox cycles. Finally, the peak fitting of gasification reaction rate curves is used to explore the reaction mechanism between coal char and the CaO-decorated iron-based oxygen carrier, indicating that the reactions in the CLG include three stages: the complex reactions involved an oxygen carrier, coal char, and steam; the gasification of coal char; and the reduction of Fe<SUB>3</SUB>O<SUB>4</SUB> to FeO. The two-segment modified random pore model (MRPM) fits the experiment data well.</P>

        The Detection of Osthole and Application of Cell Imaging Based on Nitrogen-Doped Carbon Dots

        A fluorescence probe has been synthesized for the detection of osthole using the nitrogen-doped carbon dots (NCDs) as shown in Fig. 1. The NCDs were fully characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD), x-ray photoelectron spectroscopy (XPS), Fourier transform infrared spectroscopy (FTIR). Under the optimal experimental conditions, the NCDs fluorescence probe was highly selective and sensitive to osthole. The linear response range for osthole was 5.0–7 5μM with a detection limit of 38 nM. The mechanism of the interaction of osthole and NCDs was discussed. The fluorescence probe has been applied to the analysis of biological samples. The as-synthesized NCDs with high fluorescence intensity, low toxicity and good biocompatibility were applied to cell imaging.
