        Discovery of novel glycoside hydrolases from C-glycoside-degrading bacteria using sequence similarity network analysis

        C-Glycosides are an important type of natural product with significant bioactivities, and the C-glycosidic bonds of C-glycosides can be cleaved by several intestinal bacteria, as exemplified by the human faeces-derived puerarin-degrading bacterium Dorea strain PUE. However, glycoside hydrolases in these bacteria, which may be involved in the C-glycosidic bond cleavage of C-glycosides, remain largely unknown. In this study, the genomes of the closest phylogenetic neighbours of five puerarin-degrading intestinal bacteria (including Dorea strain PUE) were retrieved, and the protein-coding genes in the genomes were subjected to sequence similarity network (SSN) analysis. Only four clusters of genes were annotated as glycoside hydrolases and observed in the genome of D. longicatena DSM 13814T (the closest phylogenetic neighbour of Dorea strain PUE); therefore, genes from D. longicatena DSM 13814T belonging to these clusters were selected to overexpress recombinant proteins (CG1, CG2, CG3, and CG4) in Escherichia coli BL21(DE3). In vitro assays indicated that CG4 efficiently cleaved the O-glycosidic bond of daidzin and showed moderate β-D-glucosidase and β-D-xylosidase activity. CG2 showed weak activity in hydrolyzing daidzin and pNP- β-D-fucopyranoside, while CG3 was identified as a highly selective and efficient α-glycosidase. Interestingly, CG3 and CG4 could be selectively inhibited by daidzein, explaining their different performance in kinetic studies. Molecular docking studies predicted the molecular determinants of CG2, CG3, and CG4 in substrate selectivity and inhibition propensity. The present study identified three novel and distinctive glycoside hydrolases, highlighting the potential of SSN in the discovery of novel enzymes from genomic data.

        Clinical Significance of Serum IgG4 in the Diagnosis and Treatment Response of IgG4-Related Disease in Adults of Southwest China: A Retrospective Study

        Background: There is no standard cut-off value of serum IgG4 concentration and serum IgG4/total IgG ratio for the diagnosis of IgG4-related disease (IgG4-RD) or as a marker of treatment responses. We aimed to explore this issue through a retrospective cohort analysis of adults in southwest China. Methods: The diagnostic performance of serum IgG4 concentration and IgG4/IgG ratio for IgG4-RD was evaluated in a retrospective analysis of 177 adults newly diagnosed as having IgG4-RD and 877 adults without IgG4-RD. Dynamic analysis was performed to evaluate the significance of serum IgG4 concentration on IgG4-RD treatment responses. Results: The serum IgG4 concentration differed according to sex. The optimal cut-off values of serum IgG4 concentration and IgG4/IgG ratio for IgG4-RD diagnosis were 1.92 g/L and 0.12 in males and 1.83 g/L and 0.11 in females, respectively. For patients with serum IgG4 concentration >2.01 g/L, the cut-off values in the total population were >3.00 g/L and 0.19, respectively. The median serum IgG4 concentration decreased over time, and the decrease rate increased over time. The serum IgG4 concentration significantly decreased at >1 week post-treatment (P=0.004), and the median decrease rate was close to 50% at >4 weeks post-treatment. Conclusions: Serum IgG4 can be a good indicator for IgG4-RD diagnosis; however, different diagnostic cut-off values should be determined according to sex. The decreasing rate is more conducive than the serum IgG4 concentration to monitor treatment efficacy. The IgG4/IgG ratio did not improve the diagnostic efficacy for IgG4-RD.

      • The Study of Ultrasonic Nonlinearity in Heat Treatment Material

        Nonlinear ultarsonic is very senstive to material property change. This paper attempts to study the correlation between heat treatment and ultrasonic nonlinearity by taking the nonlinearity factors into account. The modified ultrasonic velocity formula was deriven. It turns out that nonlinearity varies with ultrasonic velocity change in the material heat treatment, the experimental results show that relative nonlinearity parameters obtained from the modified ultrasonic velocity and the rai\tio of amplitudes(second harmonic and fundamental wave)have a good agreement. Both of them prove that the heat treatment can change the material nonlinearity. Furthermore, then relative nonlinearity calculated from the modified velocity is much more sensitive, representing the relative nonlinearity change based on the modified ultrasonic velocity in this case.

        Influence of Microstructure on Corrosion Properties of Fe–V Alloy from Vanadium Slag

        Four types of multiphase Fe–V alloys were examined, comprising ferrite, pearlite, and bainite microstructures. Electrochemicalimpedance spectroscopy and potentiodynamic polarization were employed to study the effect of microstructureon electrochemical properties of the alloys in 3.5% NaCl solution. The cathode reaction was found to be the main stepcontrolling the corrosion dissolution process. The lowest corrosion current density (2.104 μA/cm2) for the Fe–V alloy withbainite indicated that it had outstanding corrosion resistance. Although an increase in pearlite content had a negative effecton corrosion resistance of the alloy, refinement of the lamellar spacing had the opposite effect. Surface corrosion morphologieswere observed by scanning electron microscopy. The results indicate that fine cementite uniformly distributed in thebainite promoted slight uniform dissolution of ferrite. Corrosion and dissolution that occurred at the junctions of the pearlitecolonies with different directions was more severe. The increase of content of V effectively improved the corrosion resistanceof Fe–V alloy. Compared with the matrix, the VC precipitated phase was cathodically protected in the electrochemicalcorrosion process by virtue of its positive corrosion potential.

        열처리된 재료의 초음파 비선형성에 관한 연구

        비선형 초음파는 선형 초음파와 비교하였을 때 재료의 물성 변화에 민감하게 반응한다. 이 연구의 목표는 비선형 요소를 포함하여 열처리 조건과 초음파 비선형성의 상관관계를 알아내는 것이다. 본 연구에서 기존의 방법과는 다른 수정된 초음파속도계산 식이 새롭게 제안되었다. 이것은 열처리된 재질에서 초음파 속도변화가 재료의 비선형성 때문에 발생하는 것을 찾아내었다. 실험을 통하여 수정된 초음파 속도와 1차 조화 및 2차 조화파의 진폭 비율이 서로 동일한 경향을 보이는 것을 알아냈다. 그리고 수정된 초음파속도식을 통해 계산된 상대적인 비선형계수는 기존의 방법을 이용하였을 때보다 더 민감함을 알아냈다. Ultrasonic nonlinearity is very sensitive to changes in material properties. This paper describes the study of the correlation between heat treatment and ultrasonic nonlinearity by taking nonlinear factors into consideration. A modified formula was proposed for ultrasonic velocity. This formula indicated that the changes occurring in nonlinearity during heat treatments cause changes in the ultrasonic velocity. The experimental results show that the relative nonlinearity parameters calculated from the modified ultrasonic velocities and the ratio of amplitudes of the second harmonic and fundamental wave are in good agreement. The experimental results prove that heat treatment can result in changes in material nonlinearity. Moreover, the relative nonlinearity parameter calculated from the modified velocity formula is has a large value. Since this parameter has high sensitivity to changes in nonlinearity, it can be used to represent the relative nonlinearity change calculated in this study by using the modified formula for ultrasonic velocity.

        Self-Supported Bi2MoO6 Nanosheet Arrays as Advanced Integrated Electrodes for Li-Ion Batteries with Super High Capacity and Long Cycle Life

        Hierarchical Bi2MoO6 nanosheet arrays (BNAs) growing on three-dimensional (3D) Ni foam are synthesized by one-step template-free route. The obtained BNAs are used as binder-free integrated electrode for Li-ion batteries (LIBs) directly. The electrode exhibits a super high reversible discharge capacity of 2311.7 μAh·cm-2 (1741.4 mAhg-1), and an excellent cycle stability. The outstanding electrochemical properties are reasonable from the self-supported integrated electrode in which the electrolyte is easy to infiltrate active materials, electrons and ions are readily transported along the 3D conductive substrate and the stable electrode structure.

        Evaluation of surface cracking in micron and sub-micron scale scratch tests for optical glass BK7

        In order to obtain the fundamental information on the deformation and fracture behavior of brittle materials during precision and ultraprecision grinding, micron and sub-micron scale scratch tests were conducted on optical glass BK7 using Vickers indenters. Three types of surface cracking were observed around the scratch grooves. They are lateral cracking, radial cracking and cracking in front of the moving indenter. It is found that lateral cracking is the main damage type due to its large damage size and low crack initiation load. The effect of surface cracking on the relationship between the normal load and the square of scratch depth was studied. The plastic zone size as well as the sliding blister field strength was expressed as a function of the contact zone size of the indenter. A prediction model for the size of damage zone induced by lateral cracking was established and was compared with experimental results.

        Third Harmonic Generation of Shear Horizontal Guided Waves Propagation in Plate-like Structures

        The use of nonlinear ultrasonics wave has been accepted as a promising tool for monitoring material states related to microstructural changes, as it has improved sensitivity compared to conventional non-destructive testing approaches. In this paper, third harmonic generation of shear horizontal guided waves propagating in an isotropic plate is investigated using the perturbation method and modal analysis approach. An experimental procedure is proposed to detect the third harmonics of shear horizontal guided waves by electromagnetic transducers. The strongly nonlinear response of shear horizontal guided waves is measured. The accumulative growth of relative acoustic nonlinear response with an increase of propagation distance is detected in this investigation. The experimental results agree with the theoretical prediction, and thus providing another indication of the feasibility of using higher harmonic generation of electromagnetic shear horizontal guided waves for material characterization.

        Characteristics of Ultrasonic Nonlinearity by Thermal Fatigue

        The deterioration conditions of nuclear power plants and chemical plants have raised deep concerns about the safety of high-risk structures. The safety of a structure can be secured by evaluations of breakages and lifespan in the early stage. Therefore, the material condition of a structure needs to be estimated. A nonlinear ultrasonic evaluation method is reportedly more sensitive on microscopic changes of a material than a linear evaluation method. Therefore, the nonlinear ultrasonic technique based on ultrasonic nonlinearity is considered a promising method of evaluation for breakages and lifespan. This study measured the changes in nonlinear parameters by using a nonlinear ultrasonic method, observed the microstructure of a heated material, and analyzed the relation between the size of the microstructure and the changes in the nonlinear parameters.

        A prevalence survey of Wolbachia in Polytremis fukia (Lepidoptera: Hesperiidae)

        Wolbachia are endosymbiotic bacteria that infect arthropod and nematode hosts. Only a few studies have been made on Wolbachia in butterflies. In this study, we identified and determined the molecular characteristics of Wolbachia strains in Polytremis fukia (Lepidoptera: Hesperiidae), from seven locations in China. We also sequenced three portions of the mitochondrial DNA from the individuals of P. fukia to infer the effect of Wolbachia on host mitochondrial variation. The results show that 47% (15/32) are Wolbachia positive among all specimens. The infection rates in female and male are 69% (11/16) and 25% (4/16). Phylogenetic analysis of variation in the ftsZ gene from the two strains of Wolbachia (wFuk1 and wFuk2) showed they belonged to supergroup A. On the phylogeny of the mitochondrialDNA fromP. fukia, the sequences are split into three clades. Clade I and II are consistent with the distribution of geographical population. Clade III consists of eight females invariably infected with wFuk1which showed aweak association existed between mitochondrial DNA haplotypes andwFuk1 infection status.
