Streptavidin has applied to many areas including detection, purification, labeling, crosslinking and immobilization resulting in a high demand on its production. In this study, we report a method for preparation of recombinant core streptavidin (cSAV) protein using highperformance hydrophobic interaction chromatography (HPHIC). Firstly the cSAV was successfully cloned and expressed in Escherichia coli as inclusion bodies. A bifunctional stationary phase mainly working as HIC mode accompanied by weak anion exchange chromatography (WAX) was prepared using β-phenylethylamine (PEA) as a ligand. The denatured cSAV was then refolded and simultaneously purified by PEA hydrophobic interaction chromatography (PEA-HIC). The mass recovery and purity of cSAV by single-step were 30.2% and 98%, respectively. The bioactivity was determined to be 13.2 U/mg by biotin binding capacity assay. This method provides a new possibility for fast separation with simultaneous renaturation of cSAV.
Eco-friendly Strategies for the Material and Fabrication of Wearable Sensors
Yan Liu,Siyao Shang,Shuting Mo,Peng Wang,Hai Wang 한국정밀공학회 2021 International Journal of Precision Engineering and Vol.8 No.4
Wearable sensors are attracting great attentions due to their potential applications in human health monitoring and caring systems. The wide utilization of wearable electronics may cause great burden to the environment, due to the vast contaminants generated in their fabrication and after their usage. Consequently, great eff orts are devoted to the eco-friendly strategies for the material and fabrication in wearable sensors. Herein, recent advantages in developing wearable sensors with eco-friendly materials and green manufacturing approaches are reviewed. The functional materials with accessibility, recoverability and degradability have participated in the sensors as substrates, sensing elements and conductors. The fabrication strategies, from facile manual schemes to low-emission automated techniques are also introduced with their merits and demerits. Finally, the existing challenges and future opportunities in this field are summarized.

Background: Warm acupuncture, a combination of the mechanical stimulation of acupuncture and thermal stimulation of moxibustion, is commonly used in treating acute low back pain (LBP). This trial aimed to compare the efficacy of stronger (above 43°C) and weaker (above 40°C) heat stimulation in warm acupuncture on the function and pain in patients with acute LBP due to lumbosacral disc degeneration (LDD). Methods: One hundred and fifty-nine adults were randomly assigned to receive warm acupuncture treatment with silver needle (SvN) or with stainless steel needle (SSN) (1:1). Both groups received a 3-week therapy with 3 sessions per week. The primary outcome was the modified Oswestry Disability Index at week 4. The secondary outcomes included average pain, three physical sign tests and adverse events. Participants were followed up at week 16 and week 28 after randomization. Results: The LBP related disability and pain intensity significantly relieved more in the SvN warm acupuncture group than in the SSN group, in both the short and long term (p<0.001). The between-groups difference in physical signs showed statistical significance only in the short term (p = 0.024), but not in long term (p = 0.081; p = 0.069). Conclusion: Compared with warm acupuncture with stainless-steel needle at above 40°C, warm acupuncture with silver needle at above 43°C relieved more disability and pain in patients with acute LBP due to LDD. Effects of subsequent curing on chloride resistance and microstructure of steam-cured mortar
Hu, Yuquan,Hu, Shaowei,Yang, Bokai,Wang, Siyao Techno-Press 2020 Advances in concrete construction Vol.9 No.5
The influence of subsequent curing on the performance of fly ash contained mortar under steam curing was studied. Mortar samples incorporated with different content (0%, 20%, 50% and 70%) of Class F fly ash under five typical subsequent curing conditions, including standard curing (ZS), water curing(ZW) under 25℃, oven-dry curing (ZD) under 60℃, frozen curing (ZF) under -10℃, and nature curing (ZN) exposed to outdoor environment were implemented. The unsteady chloride diffusion coefficient was measured by rapid chloride migration test (RCM) to analyze the influence of subsequent curing condition on the resistance to chloride penetration of fly ash contained mortar under steam curing. The compressive strength was measured to analyze the mechanical properties. Furthermore, the open porosity, mercury intrusion porosimetry (MIP), x-ray diffraction (XRD) and thermogravimetric analysis (TGA) were examined to investigate the pore characteristics and phase composition of mortar. The results indicate that the resistance to chloride ingress and compressive strength of steam-cured mortar decline with the increase of fly ash incorporated, regardless of the subsequent curing condition. Compared to ZS, ZD and ZF lead to poor resistance to chloride penetration, while ZW and ZN show better performance. Interestingly, under different fly ash contents, the declining order of compressive strength remains ZS>ZW>ZN>ZD>ZF. When the fly ash content is blow 50%, the open porosity grows with increase of fly ash, regardless of the curing conditions are diverse. However, if the replacement amount of fly ash exceeds a certain high proportion (70%), the value of open porosity tends to decrease. Moreover, the main phase composition of the mortar hydration products is similar under different curing conditions, but the declining order of the C-S-H gels and ettringite content is ZS>ZD>ZF. The addition of fly ash could increase the amount of harmless pores at early age.
Improved Space Vector Modulation Strategy for AC-DC Matrix Converters
Xiao Liu,Qingfan Zhang,Dianli Hou,Siyao Wang 전력전자학회 2013 JOURNAL OF POWER ELECTRONICS Vol.13 No.4
In this paper, an approach to reduce the common-mode voltage and to eliminate narrow pulse for implemented AC-DC matrix converters is presented. An improved space vector modulation (SVM) strategy is developed by replacing the zero space vectors with suitable pairs of active ones. Further, while considering the commutation time, the probability of narrow pulse in the conventional and proposed SVM methods are derived and compared. The advantages of the proposed scheme include: a 50% reduction in the peak value of the common-mode voltage; improved input and output performances; a reduction in the switching loss by a reduced number of switching commutations and a simplified implementation via software. Experimental results are presented to demonstrate the correctness of the theoretical analysis, as well as the feasibility of the proposed strategy.
