        ROOM-TEMPERATURE SOLID-STATE REACTION TO TREE-LIKE FRACTAL NANO-CdS AND THIRD-ORDER NONLINEAR OPTICAL PROPERTIES

        A novel tree-like nano-cadmium sulfide (CdS) with the fractal feature is synthesized by solid-state reaction at room temperature from complex precursor with aminotrimethylenephosphonic acid (ATMP) as ligand. The obtained sample is the crystalline cubic beta cadmium sulfide. The tree is composed of nanorods with an average diameter of ca. 95 nm and a length of up to 100–650 nm. The nanorods grow in the asymmetrical "Y" shape. The amount of ATMP plays an important role in the formation of fractal structure. Nonlinear optical (NLO) measurements by the Z-scan technique exhibit that the tree-like fractal nano-CdS has the third-order nonlinear optical properties of both NLO absorption and NLO refraction with self-focusing effect and the optical limiting performance.

      • Long-term Efficacy of Microwave Hyperthermia Combined with Chemoradiotherapy in Treatment of Nasopharyngeal Carcinoma with Cervical Lymph Node Metastases

        Objective: The long-term efficacy of microwave hyperthermia combined with chemoradiotherapy in treating nasopharyngeal carcinoma (NPC) with metastatic foci in cervical lymph nodes was evaluated. Methods: A total of 154 cases of N2 or N3 stage NPC were randomized into two groups: hyperthermia group (76 cases) and control group (78 cases). Both received cisplatin chemotherapy and radiotherapy. In addition, the hyperthermia group further received microwave hyperthermia to the metastatic cervical nodes with different patterns (before or after radiotherapy), heating temperatures (T90< $43^{\circ}C$ and $T90{\geq}43^{\circ}C$) and hyperthermia episodes (< 4 times, 4-10 times and > 10 times). Results: The 3-month and 5-year complete response (CR) rates of cervical lymph nodes in the hyperthermia group were significantly higher than those in the control group. The 5-year disease-free survival (DFS) rate and the 3-year / 5-year overall survival rate in the hyperthermia group were also significantly higher. There was no significant difference in 5-year metastatic rates. In the hyperthermia group, the 3-month and 5-year CR rates of T90< $43^{\circ}C$ treatment were significantly lower than with $T90{\geq}43^{\circ}C$ treatment. The CR rate was highest when the hyperthermia was performed 4-10 times. There were no significant differences in 3-month and 5-year CR rates between hyperthermia before or after radiotherapy treatment. Conclusion: Microwave hyperthermia combined with chemoradiotherapy can increase local control, DFS and 3, 5-year overall survival rates of patients with N2 ~ N3 stage NPC. The heating temperature should be over $43^{\circ}C$ with hyperthermia repeated 4-10 times.

      • Study on knee joint injury and rehabilitation training of traditional Chen style Tai Chi practicers

        Background The purpose of this study was to further investigate the causes of knee injuries in tai jiquan athletes and to propose a rehabilitation training program based on the characteristics of traditional Chen Tai Chi and to summarize the application experience. Subjects and Methods Seventy patients with knee injuries were included in a screening study using the single-leg squat test (SLS test) and the McMurray test. They were randomly divided into a training group and a control group. The training group underwent rehabilitation training (Chen-style Tai Chi) under the guidance of a Tai Chi instructor for three months, and the control group did not receive any training instruction. The general characteristics of the patients before and after training were evaluated for knee function using the Knee Injury Rating Scale, which confirmed the good therapeutic effect of Chen-style Tai Chi on patients with osteoarthritis of the knee. RESULTS Before the rehabilitation training, the severity of injury was basically the same in both groups. At the end of the training, the training group scored significantly higher than the control group (P<0.05). The knee function score in the training group improved to excellent. which indicates the Tai Chi training played a significant role in restoring and improving knee joint function. Conclusion Attention to rehabilitation training of Tai Chi not only will not damage the knee joint, but also can reduce the chance and severity of knee injury in traditional Chen-style Tai Chi and improve the function of the knee joint. Specific recommendations are as follows: pay attention to warm-up exercises, pay attention to functional training of the knee joint, pay attention to rehabilitation factors such as relaxation and massage.

        Effect of L-carnitine on sperm quality during liquid storage of boar semen

        Objective: This study was conducted to investigate the effect of L-carnitine on the pig semen characteristics during storage. Methods: Spermatozoa samples were examined for spermatozoa quality and then randomly divided into 5 groups: 0 (control), 12.5, 25, 50, and 100 mM L-carnitine. Sperm motility, plasma membrane integrity and antioxidant parameters (total reactive oxygen species, total antioxidant capacity, and malondialdehyde) were evaluated after 0, 3, 5, and 10 day cooled-storage at 17°C. Moreover, ATP content, mitochondria activity as well as sperm-binding and in vitro fertilizing ability of preserved boar sperm were also investigated. Results: Supplementation with 50 mM L-carnitine could effectively maintain boar sperm quality parameters such as sperm motility and membrane integrity. Besides, we found that L-carnitine had positive effects on boar sperm quality mainly through improving antioxidant capacities and enhancing ATP content and mitochondria activity. Interestingly, by assessing the effect of L-carnitine on sperm fertility and developmental potential, we discovered that the extender containing L-carnitine could improve sperm quality and increase the number of sperms bounding to zona pellucida, without improving in vitro fertility and development potential. Conclusion: These findings suggested that the proper addition of L-carnitine to the semen extender improved boar sperm quality during liquid storage at 17°C.

        Online to offline is a new business mode combining the online shopping and the front line transactions. No previous studies have simultaneously examined these variables as antecedents of e-loyalty intention or the possible relationships among them within O2O platform. Our paper has examined the effects of inertia; perceived ease of use, customization and quality of offering on the consumer’s switching costs. These findings provide a theoretical foundation for academics and also practical guidelines for service providers in dealing with the promotion of loyalty at O2O platforms.

        Sliding mode and predictive current control strategy of the three-phase Vienna rectifier

        This study involves the design and implementation of a novel double closed-loop control of the three-phase Vienna rectifier. To achieve the unit power factor operation of the rectifier and reduce the harmonic content of the grid side, the double closed-loop control strategy is proposed on the basis of the voltage control of the outer loop and the current control of the inner loop. The voltage outer loop adopts the sliding-mode variable structure control strategy based on the improved reaching law; this strategy can effectively suppress switching jitter. The current inner loop adopts an improved predictive current control strategy that combines the traditional finite control set model predictive control (FCS-MPC) with the space vector modulation method (SVPWM), such that the optimal switching state obtained by the predictive controller is modulated to generate a switching signal. Moreover, the switching frequency can be controlled by adjusting the frequency of the carrier, and the switching frequency is fixed while preserving the advantages of model prediction control. The primary advantages of the proposed double closed-loop control strategy are as follows: (1) it does not require proportional-integral controllers in the current control loops, and (2) it keeps the output voltage stable. The Vienna rectifier simulation is performed on MATLAB. Then an experimental prototype based on SiC devices is developed to verify the excellent performance of the proposed strategy in steady and transient states.

        Pan-cancer Analysis of Tumor Mutational Burden and Homologous Recombination DNA Damage Repair Using Targeted Next-Generation Sequencing

        Purpose Current variability in methods for tumor mutational burden (TMB) estimation and reporting demonstrates the urgent need for a homogeneous TMB assessment approach. Here, we compared TMB distributions in different cancer types using two customized targeted panels commonly used in clinical practice. Materials and Methods TMB spectra of 295- and 1021-gene panels in multiple cancer types were compared using targeted next-generation sequencing (NGS). The TMB distributions across a diverse cohort of 2,332 cancer cases were then investigated for their associations with clinical features. Treatment response data were collected for 222 patients who received immune-checkpoint inhibitors (ICIs) and their homologous recombination DNA damage repair (HR-DDR) and programmed death-ligand 1 (PD-L1) expression were additionally assessed and compared with the TMB and response rate. Results The median TMB between gene panels was similar despite a wide range in TMB values. The highest TMB was eight and 10 in patients with squamous cell carcinoma and esophageal carcinoma according to the classification of histopathology and cancer types, respectively. Twenty-three out of 103 patients (22.3%) were HR-DDR–positive and could benefit from ICI therapy; out of those 23 patients, seven patients had high TMB (p=0.004). Additionally, PD-L1 expression was not associated with TMB or treatment response among patients receiving ICIs. Conclusion Targeted NGS assays demonstrated the ability to evaluate TMB in pan-cancer samples as a tool to predict response to ICIs. In addition, TMB integrated with HR-DDR–positive status could be a significant biomarker for predicting ICI response in patients.

        Ginsenoside Rb1 and compound K improve insulin signaling and inhibit ER stress-associated NLRP3 inflammasome activation in adipose tissue

        Background: This study was designed to investigate whether ginsenoside Rb1 (Rb1) and compound K (CK) ameliorated insulin resistance by suppressing endoplasmic reticulum (ER) stress-induced inflammation in adipose tissue. Methods: To induce ER stress, epididymal adipose tissue from mice or differentiated 3T3 adipocytes were exposed to high glucose. The effects of Rb1 and CK on reactive oxygen species production, ER stress, TXNIP/NLRP3 inflammasome activation, inflammation, insulin signaling activation, and glucose uptake were detected by western blot, emzyme-linked immunosorbent assay, or fluorometry. Results: Rb1 and CK suppressed ER stress by dephosphorylation of $IRE1{\alpha}$ and PERK, thereby reducing TXNIP-associated NLRP3 inflammasome activation in adipose tissue. As a result, Rb1 and CK inhibited IL-$1{\beta}$ maturation and downstream inflammatory factor IL-6 secretion. Inflammatory molecules induced insulin resistance by upregulating phosphorylation of insulin receptor substrate-1 at serine residues and impairing insulin PI3K/Akt signaling, leading to decreased glucose uptake by adipocytes. Rb1 and CK reversed these changes by inhibiting ER stress-induced inflammation and ameliorating insulin resistance, thereby improving the insulin IRS-1/PI3K/Akt-signaling pathway in adipose tissue. Conclusion: Rb1 and CK inhibited inflammation and improved insulin signaling in adipose tissue by suppressing ER stress-associated NLRP3 inflammation activation. These findings offered novel insight into the mechanism by which Rb1 and CK ameliorate insulin resistance in adipose tissue.

      • A Novel Approach to Cloning and Expression of Human Thymidylate Synthase

        Thymidylate synthase (TS) catalyzes the transfer of a methyl group from methylenetetrahydrofolate to dUMP to form dTMP. It is a primary target in the chemotherapy of colorectal cancers and some other neoplasms. In order to obtain pure protein for analysis of structure and biological function, an expression vector TS-pET28b (+) was constructed by inserting wild-type human thymidylate synthase (hTS) cDNA into pET28b (+). Then an expression strain was selected after transformation of the recombined plasmid into Rosetta (DE3). Fusion protein with His-tag was efficiently expressed in the form of inclusion bodies after IPTG induction and the content was approximately 40.0% of total bacteria proteins after optimizing expression conditions. When inclusion bodies were washed, dissolved and purified by Ni-NTA under denatured conditions, the purity was up to 90%. On SDS-PAGE and West-blotting, the protein band was found to match well with the predicted relative molecular mass-36kDa. Bioactivity was 0.1 U/mg. The results indicated that high-level expression of wild-type hTS cDNA can be achieved in prokaryotes with our novel method, facilitating research into related chemotherapy.
