Bitter taste is crucial for the survival of organisms because it helps to avoid the ingestion of potentially harmful substances mostly found in plants. It is mediated by the bitter taste receptors that reside on the surface of the taste cells in the tongue. Genetic variations of these receptors have shown different responses to bitter taste compounds and the frequencies of these variations were different within and between populations. In this study, we investigated the diversity and frequencies of functional variants of human bitter taste receptor (hTAS2R) genes by direct sequencing of 25 hTAS2R genes in Koreans. A total of 59 cSNPs with an average of two cSNPs per gene were identified and ~80% of cSNPs are nonsynonymous substitutions resulting in amino acid replacement. Also, 85 different haplotypes ranging from 2 to 18 haplotypes in each gene were identified in this study. One or two haplotypes of each hTAS2R gene were generally common, but the repertoire of haplotypes was different from other populations since different cSNPs were observed in the Koreans. The results of this study were similar to those of other populations supporting the fact that human bitter taste receptor genes have more genetic variation within and between populations than do most other genes. The repertoire and frequencies of cSNPs and haplo-types in the hTAS2R genes will provide information to identify a functional receptor for a ligand which is common in our environment.
Description of Temperature Dependence of Critical Micelle Concentration
Kim, Hong-Un,Lim, Kyung-Hee Korean Chemical Society 2003 Bulletin of the Korean Chemical Society Vol.24 No.10
A new equation has been derived on the basis of ${\delta}G^o$ = -RT lnK, linear behavior of the enthalpy of micellization with temperature, and the Gibbs-Helmholtz relation. It describes correctly the dependence of critical micelle concentration $(X_{CMC})$ on temperature and has yielded excellent fitting results for various surfactant systems. The new equation results in the linear behavior of the entropy of micellization with temperature and accounts for the compensation phenomena observed for the micellization in aqueous solutions, along with the linear dependence of the enthalpy of micellization on temperature. These results imply that the new equation of $X_{CMC}(T)$ accounts for the temperature dependence of CMC correctly.
The neuronal activity changes by oxidative injury in OHSC using optical imaging system

Oxidative stress produces neurotoxicity often related with various CNS disorders. Ascorbic acid (AA) have been shown to have protective effects as anti-oxidants in experimental neurological disorder models such as stroke, ischemia, and epileptic seizures. The present study was conducted to examine the protective effects of AA on kainic acid (KA) neurotoxicity using organotypic hippocampal slice cultures. AA may enhance the protection effect and benefit to improve neuronal plasticity in post-injury. Thus, we investigated that the antioxidant affects neuroprotective signaling and neuroplastic changes in hippocampus after oxidative injury. Electrophysiological and biochemical assays were used to observe changes in synaptic efficacy following electrical and/or pharmacological manipulation of synaptic function. Neuronal cell death, as assessed by propidium iodide (PI) uptake, was reduced by AA treatment compared with KA treatment. AA significantly prevented cell death and inhibited reactive oxygen species (ROS) level, and mitochondrial dysfunction. Optical imaging revealed that KA treatment tended to delay the latency of electrical stimulation and decrease the amplitude of optical signals of synaptic activity. These results suggest that AA may protect hippocampal neurons against oxidative stress and the survived neurons may functional to synaptic plasticity changes. Insect plasma protein is abundant in the hemolymph of holometabolous insect larvae and is used as a source of amino acids and energy for construction of adult structures during metamorphosis. In order to understand the mechanism of decomposition of larval plasma proteins by hemocyte protease, we tried to purify a cysteine protease from the hemocyte lysate by using Carbobenzoxy-L-Phenylalanyl-L-Arginine-4-Methyl-Coumaryl-7-Amide (Z-Phe-Arg-MCA) as substrate and to identify plasma proteins that are selectively susceptible to the purified protease. Here, we describe the purification and characterization of a cysteine protease that specifically hydrolyzes the plasma protein of the coleopteran insect, Tenebrio molitor, larvae. The molecular mass of this enzyme was 25 kDa, as determined by SDS-PAGE under reducing conditions. The amino acids sequence of its NH₂-terminus was determined to be Leu-Pro-Gly-Gln-Ile-Asp-Trp-Arg-Asp-Lys-Gly. This sequence contained Pro, Asp, and Arg residues, conserved in many papain superfamily enzymes. The specific cysteine protease inhibitors, such as E-64 and leupetin, inhibited its hydrolytic activity. One plasma protein with a molecular mass of 48 kDa was selectively hydrolyzed within 3 h when the purified enzyme and plasma proteins were incubated in vitro. However, the 48 kDa protein was not hydrolyzed by the purified 25 kDa protease in the presence of E-64. Western blotting analysis at various developmental stages showed that the purified enzyme was detected at larvae, pupae, and adult stages, but not the embryo stage.

<P>Novel triangular ring resonators combining extremely small multimode-interference (MMI) coupler, low loss total internal reflection (TIR) mirrors, and semiconductor optical amplifiers are reported for the first time. The MMI length of 90 microm is among the shortest reported. The incidence angle of the TIR mirror inside the resonator is 22 degrees. A free-spectral range of approximately 2 nm is observed near 1550 nm along with an on-off ratio of 17 dB. The triangular resonators with a sharp angle are very attractive components due to their promise of compact size and high levels of integration. Therefore, large numbers of resonators can be integrated on a chip to increase functionality in future optical wavelength division multiplexing system.</P>
Prevention of Precipitation in Sand Lance Fish Sauce by Chelating Agents
Kyung Whan Moon,Seong Yeong Kim,Jin Man Kim,Un Jae Chang,Song Hwan Bae,Hyung Joo Suh 한국식품과학회 2008 Food Science and Biotechnology Vol.17 No.1
Chelating agents to sand lance fish sauce for the prevention of precipitate formation were applied. The precipitates consisted of crude protein (74.4%), ash (18.7%), and other components (6.9%). Sand lance sauce was mainly composed of glutamic acid (3.69 mg/g), alanine (2.96 mg/g), and lysine (2.64 mg/g). However, there was an increase in the amount of hydrophobic amino acids, phenylalanine, isoleucine, and leucine, in the precipitates. Sodium ions were not detected in the precipitate; rather, the main elements were Mg (1.98×10⁴ μg/g), K (1.36×10⁴ μg/g), and Ca (6.66×10² μg/g). In HPLC analysis, fish sauce was composed of 2 main peaks with molecular weights of 85.5 and 528.4 Da, respectively. However, the precipitate contained one peak with a molecular weight of 1,513.5 Da. The addition of 0.2% malic acid and citric acid caused 55 and 70% prevention of the precipitate, respectively. Citric acid was the most effective chelating agent and efficiently prevented precipitation in the fish sauce.
