        Comparative Evaluation of Three Purification Methods for the NucleocapsidProtein of Newcastle Disease Virus from Escherichia coli Homogenates

        In the present study, the performances of conventional purification methods, packed bed adsorption (PBA), and expanded bed adsorption (EBA) for the purification of the nucleocapsid protein (NP) of Newcastle disease virus (NDV) from Escherichia coli homogenates were evaluated. The conventional methods for the recovery of NP proteins involved multiple steps, such as centrifugation, precipitation, dialysis, and sucrose gradient ultracentrifugation. For the PBA, clarified feedstock was used for column loading, while in EBA, unclarified feedstock was used. Streamline chelating immobilized with Ni2+ ion was used as an affinity ligand for both PBA and EBA. The final protein yield obtained in conventional and PBA methods was 1.26% and 5.56%, respectively. It was emonstrated that EBA achieved the highest final protein yield of 9.6% with a purification factor of 7. Additionally, the total processing time of the EBA process has been shortened by 8 times compared to that of the conventional method.

        Real-Time Tumor Gene Expression Profiling to Direct Gastric Cancer Chemotherapy: Proof-of-Concept “3G” Trial

        <P><B>Purpose:</B> The oxaliplatin plus S-1 and cisplatin plus S-1 regimens are interchangeably used in the management of advanced gastric cancer. The previously reported G-intestinal (G1) and G-diffuse (G2) intrinsic gene expression signatures showed promise for stratifying patients according to their tumor sensitivity to oxaliplatin or cisplatin.</P><P><B>Experimental Design:</B> The proof-of-concept, multicenter, open-label phase II “3G” trial was done to prospectively evaluate the feasibility and efficacy of using genomic classifiers to tailor treatment in gastric cancer. Patients’ tumors were classified as “G1” or “G2” using a nearest-prediction template method, or “G3” (unclear assignment) when FDR ≥ 0.05. The first 30 patients in the “G1” cohort were assigned oxaliplatin plus S-1 (SOX) chemotherapy; thereafter, subsequently recruited “G1” patients were treated with cisplatin plus S-1 (SP) chemotherapy. “G2” patients and “G3” patients were treated with SP and SOX chemotherapy, respectively.</P><P><B>Results:</B> A total of 48, 21, and 12 patients, respectively, were given “G1,” “G2,” and “G3” genomic assignments. Median turnaround time was 7 days (IQR, 5–9). Response rates were 44.8%, 8.3%, 26.7%, and 55.6% for the “G1-SOX,” “G1-SP,” “G2,” “G3” cohorts, respectively; and was higher in G1 patients treated with SOX compared with SP (<I>P</I> = 0.033). Exploratory analyses using the genomic classifier of Lei and colleagues validated the utility of the metabolic signature as a biomarker for predicting benefit from chemotherapy (log-rank <I>P</I> = 0.004 for PFS), whereas the Asian Cancer Research Group classifier did not demonstrate any predictive value.</P><P><B>Conclusions:</B> This bench-to-bedside effort establishes a reasonable turnaround time for gene expression profiling and possible utility of genomic classifiers in gastric cancer treatment stratification. <I>Clin Cancer Res; 24(21); 5272–81. ©2018 AACR</I>.</P>

        Effect of Annealing Atmosphere on Photoluminescence and Gas Sensing of Solution-Combustion-Synthesized Al, Pd Co-Doped ZnO Nanoparticles

        Al, Pd co-doped ZnO nanoparticles (NPs) synthesized using a solution combustion method and subsequent annealing process under various atmospheres, including air, nitrogen, and hydrogen, were characterized using x-ray diffraction, energy-dispersive x-ray spectroscopy, fieldemission scanning electron microscopy, transmission electron microscopy, and photoluminescence spectroscopy. The gas-sensing properties of the sensors based on the NPs were also examined. The results indicated that the Al, Pd co-doped ZnO NPs, with an average crystallite size of 10 nm, exhibited enhanced gas-sensing performance compared with that of pure ZnO and Al-doped ZnO. The response of the Al, Pd co-doped ZnO NPs annealed in N2 to ethanol (49.22) was nearly 5.7 times higher than that to acetone (8.61) and approximately 20 - 27 times higher than that to benzene (2.38), carbon monoxide (2.23), and methane (1.78), which demonstrates their excellent selectivity to ethanol versus other gases. This high ethanol response can be attributed to the combined effects of the small size, Schottky barrier, lattice defects, and catalysis.

        Panax ginseng and its ginsenosides: potential candidates for the prevention and treatment of chemotherapy-induced side effects

        Chemotherapy-induced side effects affect the quality of life and efficacy of treatment of cancer patients. Current approaches for treating the side effects of chemotherapy are poorly effective and may causenumerous harmful side effects. Therefore, developing new and effective drugs derived from natural nontoxiccompounds for the treatment of chemotherapy-induced side effects is necessary. Experimentsin vivo and in vitro indicate that Panax ginseng (PG) and its ginsenosides are undoubtedly non-toxic andeffective options for the treatment of chemotherapy-induced side effects, such as nephrotoxicity, hepatotoxicity,cardiotoxicity, immunotoxicity, and hematopoietic inhibition. The mechanism focus on antioxidation,anti-inflammation, and anti-apoptosis, as well as the modulation of signaling pathways, suchas nuclear factor erythroid-2 related factor 2 (Nrf2)/heme oxygenase-1 (HO-1), P62/keap1/Nrf2, c-jun Nterminalkinase (JNK)/P53/caspase 3, mitogen-activated protein kinase (MEK)/extracellular signalregulatedkinases (ERK), AMP-activated protein kinase (AMPK)/mammalian target of rapamycin(mTOR), mitogen-activated protein kinase kinase 4 (MKK4)/JNK, and phosphatidylinositol 3-kinase(PI3K)/AKT. Since a systemic review of the effect and mechanism of PG and its ginsenosides onchemotherapy-induced side effects has not yet been published, we provide a comprehensive summarizationwith this aim and shed light on the future research of PG.

        A Sb2Se3/Palygorskite Nanocomposite Catalyst for p-Nitrophenol Reduction

        A nanocomposite consisting of rod-like Sb2Se3 and fiberous palygorskite (Pal) was prepared using cetyltrimethyl ammonium bromide (CTAB) as a pillaring agent to change Pal microstructure. CTAB optimal dosage was 0.2 g (versus 0.388 g of Pal). Sb2Se3/Pal has a high catalytic activity towards the catalytic reduction of p-nitrophenol to p-aminophenol while in the presence of NaBH4 at room temperature. The excellent performance is attributed to dispersion of rod-like Sb2Se3 in fiberous Pal which promoted more active sites exposure. The results provide new ideas for preparing new catalysts.

        Efficacy and Safety of Peroral Endoscopic Myotomy in Achalasia Patients with Failed Previous Intervention: A Systematic Review and Meta-analysis

        Peroral endoscopic myotomy (POEM) has emerged as a rescue treatment for recurrent or persistent achalasia after failed initial management. Therefore, we aimed to investigate the efficacy and safety of POEM in achalasia patients with failed previous intervention. We searched the MEDLINE, Embase, Cochrane, and PubMed databases using the queries “achalasia,” “peroral endoscopic myotomy,” and related terms in March 2019. Data on technical and clinical success, adverse events, Eckardt score and lower esophageal sphincter (LES) pressure were collected. The pooled event rates, mean differences (MDs) and risk ratios (RR) were calculated. A total of 15 studies with 2,276 achalasia patients were included. Overall, the pooled technical success, clinical success and adverse events rate of rescue POEM were 98.0% (95% confidence interval [CI], 96.6% to 98.8%), 90.8% (95% CI, 88.8% to 92.4%) and 10.3% (95% CI, 6.6% to 15.8%), respectively. Seven studies compared the clinical outcomes of POEM between previous failed treatment and the treatment naïve patients. The RR for technical success, clinical success, and adverse events were 1.00 (95% CI, 0.98 to 1.01), 0.98 (95% CI, 0.92 to 1.04), and 1.17 (95% CI, 0.78 to 1.76), respectively. Overall, there was significant reduction in the pre- and post-Eckardt score (MD, 5.77; p<0.001) and LES pressure (MD, 18.3 mm Hg; p<0.001) for achalasia patients with failed previous intervention after POEM. POEM appears to be a safe, effective and feasible treatment for individuals who have undergone previous failed intervention. It has similar outcomes in previously treated and treatment-naïve achalasia patients. (Gut Liver 2021;15:153-167)

        Protective Effects of Silibinin and Its Possible Mechanism of Action in Mice Exposed to Chronic Unpredictable Mild Stress

        Silibinin, a natural flavonoid antioxidant isolated from extracts of the milk thistle herb, has recently been identified as having antihepatotoxic and anticancer properties. In this paper, we investigated the effects of silibinin on behavior and neuroplasticity in mice subjected to chronic unpredictable mild stress (CUMS). After 5 consecutive weeks of CUMS, the mice were treated with silibinin (100 mg/kg, 200 mg/kg and 400 mg/kg by oral gavage) for 3 consecutive weeks. The results showed that silibinin administration significantly alleviated the CUMS-induced depressive-like behavior, including the total number of squares crossed and the frequency of rearing in the open field test, the immobility time in the tail suspension test and the forced swimming test. Furthermore, silibinin treatment increased the levels of brain-derived neurotrophic factor (BDNF), serotonin (5-HT) and norepinephrine (NE) in the prefrontal cortex and hippocampus. Our study provides new insight into the protective effects of silibinin on the depressive status of CUMS mice, specifically by improving neuroplasticity and neurotransmission.

        Optimal Conditions for Hepatitis B Core Antigen Production in Shaked Flask Fermentation

        The effects of various environmental factors such as pH (5, 6, 7, 8 and 9), temperature (30, 37 and 40C) and rotational speed (150, 200 and 250 rpm) on the growth and the hepatitis B core antigen (HBcAg) production of Escherichia coli W3110IQ were examined in the present study. The highest growth rate is achieved at pH 7, 37C and at a rotational speed of 250 rpm which is 0.927 h-1. The effect of pH on cell growth is more substantial compared to other parameters; it recorded a 123% different between the highest growth rate (0.927 h-1) at pH 7 and lowest growth at pH 5. The highest protein yield is achieved at pH 9, rotational speed of 250 rpm and 40C. The yield of protein at pH 7 is 154% higher compared to the lowest yield achieved at pH 5. There is about 28% different of the protein yield for the E. coli cultivated at 250 rpm compared to that at 150 rpm which has the lowest HBcAg yield. The yield of protein at 40C is 38% higher compared to the lowest yield achieved at 30C.

        Next-generation sequencing of the mitochondrial genome of Dolichovespula panda (Hymenoptera: Vespidae) with a phylogenetic analysis of Vespidae

        For the first time the mitochondrial genome of a Dolichovespula species, D. panda Archer (Hymenoptera: Vespidae), was sequenced with a next-generation sequencing approach. The sequenced mitochondrial genome is 17137 bp long and consists of 13 protein-coding, 22 tRNA and two rRNA genes, as well as a partial A +T-rich region. Twenty-two of the genes are encoded on the majority strand and 15 genes on the minority strand. All protein-coding genes start with ATN codons and have a TAA termination codon, except for one with a TA codon. Compared with the putative ancestral arrangement of insects, the D. panda mitochondrial genome shows the shuffling of trnN and trnE, and of trnQ and trnM, the translocation of trnY to upstream of trnI, and of trnL1 to the region between trnS2 and nad1 and a reversal of trnS1. A phylogenetic tree within the Vespidae was reconstructed using the 13 protein-coding mitochondrial genes. This shows a sister group relationship between Dolichovespula and a clade formed by Vespa and Vespula. It also corroborated the position of Eumeninae as sister group of the clade Polistinae +Vespinae.
