        [10]-Gingerol from Zingiber Officinale Reduces Oxidative Stress via the ERK/Nrf2/HO-1 Signaling Pathway in Human Keratinocytes

        생강(ginger, Zingiber officinale Roscoe)은 생강과(Zingiberaceae)에 속하는 다년생 초본(草本)으로 열대 아시아가 원산지이며, 아시아, 아프리카, 아메리카 지역에서 재배되고 있다. 지하에서 성장하는 덩이줄기인 생강은 색이 진한 노란색부터 황색 또는 붉은색을 띄는 다육질이며 향신료 및 기능성 소재로 꾸준히 활용되고 있다. 생강에는 폴리페놀 등 400여 종류의 화합물이 밝혀진 바 있으며, [6]-shogaol, [6]-gingerol, [8]-gingerol 및 [10]-gingerol 등의 향신 화합물이 주요한 성분으로, 항암, 항산화, 혈소판응고억제, 항균, 혈당저하, 관절염 개선, 탈모 억제, 항염증, 체중 감소, 해열 및 멜라닌 생성 억제 효과 등이 보고되었다. 본 연구는 생강의 주요성분이 함유된 대용차 소재개발을 목적으로, 국내산 생강으로부터 [10]-gingerol을 분리하여 동정한 후, 인체 피부세포(human keratinocytes)에서 생성되는 산화적 스트레스에 의한 [10]-gingerol의 효능을 분자수준에서 관찰하였다. 실험 결과, [10]-gingerol은 1, 3, 5 μM 농도에서 인체 피부세포의 heme oxygenase-1 (HO-1) mRNA levels을 농도 의존적으로 증가시켰으며, nuclear factor erythroid 2-related factor 2 (Nrf2)의 증가 및 kelch-like ECH-associated protein 1에 의해 세포질 안정성이 저하되는 것을 확인하였다. 결론적으로, mild oxidative stress 조건에서 [10]-gingerol이 ERK/Nrf2/HO-1 signaling pathway을 통해 antioxidant enzyme의 작용을 증가시키는 것을 확인하였다. The present study was designed to investigate the molecular mechanisms of bioactive components against mild oxidative stress in human keratinocytes (HaCaTs). Solvent extraction and fractionation of the rhizomes of Zingiber officinale resulted in the isolation of [10]-gingerol. At concentrations of 1, 3, and 5 µM [10]-gingerol dose-dependently increased HO-1 (heme oxygenase-1) mRNA and protein levels, with a significant effect at 5 µM. This augmentation of HO-1 by [10]-gingerol was attributed to the upregulation of Nrf2 (nuclear factor erythroid 2-related factor 2), which markedly reduced the cytoplasmic stability of Keap1 (kelch-like ECH-associated protein 1). Furthermore, our findings indicate that the upregulation of HO-1 by [10]-gingerol increased resistance to mild oxidative stress, suggesting that [10]-gingerol might enhance the activities of antioxidant enzymes by stimulating Nrf2. Our experimental findings indicate that the mild attenuating effect of [10]-gingerol against oxidative stress involves the ERK/Nrf2/HO-1 signaling pathway.

        [10]-Gingerol Induces Intrinsic Apoptosis in A2058 Human Melanoma Cells

        The objective of the present study was to investigate the molecular mechanisms involved in the activity of [10]-gingerol using A2058 human melanoma cells. [10]-Gingerol inhibited the proliferation of A2058 cells by 50% at a concentration of 52 μM. Such inhibition was dose-dependent accompanied by morphological change indicative of apoptosis. Furthermore, flow cytometric analysis by Annexin V and PI double staining showed that [10]-gingerol increased the extent of apoptosis. Analysis of the mechanism of these events indicated that [10]-gingerol increased the ratio of Bax to Bcl-2, resulting in the activation of caspase-9, caspase-3, and poly-ADP-ribose polymerase in a dose-dependent manner.

        In Vitro Antiproliferative and Apoptosis Inducing Activity of Bibimbap on HT-29 Human Colorectal Adenocarcinoma Cells

        The present study was designed to investigate the antiproliferative activity and molecular mechanisms of Bibimbap in HT-29 human colorectal adenocarcinoma cells. Bibimbap extract inhibited the proliferation of HT-29 cells by 50% at a concentration of 10.1±0.17 mg/mL for 48 h. The population of live cells decreased slightly, and the morphology changed with a reduction in cell volume (pyknosis) with Bibimbap. Treatment with 5 mg/mL of Bibimbap resulted in slight cell shrinkage. Furthermore, as the Bibimbap dose increased to 10 mg/mL, these characteristics were more evident, and HT-29 cells exhibited partial detachment by staining with the DNA-binding dye Hoechst 33342. Flow cytometric analysis by Annexin V and PI double staining showed that Bibimbap increased the levels of apoptosis. Analysis of the mechanism of these events showed that Bibimbap-treated cells exhibited a mitochondria-dependent apoptotic pathway through the modulation of caspase-3, caspase-8, caspase-9, and poly-ADP ribose polymerase, as well as Bax and Bcl-2 expression in dose- and time-dependent manners. Consequently, Bibimbap exerts a significant antiproliferative effect on HT-29 human colorectal adenocarcinoma cells.         녹차(대작)를 첨가한 기능성 젤리의 품질 특성과 항산화 효능

        본 연구는 건강효능이 우수한 대작의 소비촉진을 목적으로 기능성 젤리를 제조하여 품질평가를 시행하였다. 실험 결과, 대작의 첨가비율이 20, 40, 60%로 증가할수록 젤리의 pH가 5.89, 5.58과 5.35로 감소하였다(p<0.05). 젤리의 색도를 측정한 결과, L 값도 동일하게 43.31, 40.56과 39.88로 유의적으로 감소하였다(p<0.05). 또한, 적색도(redness)를 나타내는 a값과 황색도(yellowness)를 나타내는 b값은 각각 -0.11, -0.05과 -0.01 및 2.77, 3.08과 3.42로 유의적으로 증가하였다(p<0.05). 젤리 L값의 감소는 찻잎에 함유된 당과 아미노산이 제조과정 중고온처리(hot-air drying)로 인한 비효소적 갈변(Maillard reaction)에 의한 것으로 판단되며, a값과 b값의 증가는찻잎의 폴리페놀 화합물에 의한 산화적 갈변반응에 의한 것으로 판단된다. 젤리의 경도는 0.93, 0.84, 0.79 N으로감소하였으며, 이는 젤리의 유기산이 gel 단백질 분자의 결합을 저해하여 강도를 감소시킨 것으로 판단된다. 대작 젤리의 총 페놀성 화합물 함량은 각각 16.98, 26.75, 32.17 mg GAE/g으로 유의적으로 증가하였으며(p<0.05), DPPH free radical 및 ABTS radical 소거능은 각각 64.84%, 75.46%, 79.85% 및 19.01%, 39.01%, 54.59%로 유의적으로 증가하였다(p<0.05). 이번 실험결과는 소화가 용이하고 항산화 효능이 우수한 기능성 젤리 소재개발에 기여할 수 있을 것으로 판단된다. This study examined the quality characteristics and antioxidant properties of jellies with Daejak. Daejak jellies were prepared with 0, 20, 40 and 60% of brewed Daejak (1 g/100 mL hot water), sugar, and gelatin. As a result, the pH of brewed Daejak jellies contents of 20, 40, and 60% were 5.89, 5.58, and 5.35, respectively (p<0.05). The L-value and hardness decreased significantly with increasing levels of brewed Daejak (p<0.05), but the a-value and b-value increased significantly with increasing levels of brewed Daejak in jellies (p<0.05). The total phenolic compounds and DPPH, ABTS activities increased with increasing concentration of brewed Daejak (p<0.05). Overall, Daejak can be useful in the production of high-quality jelly.

        대용차 소재 아프리카 약용식물의 항산화 및 HCT116 인체 대장암세포 성장저해 효과

        본 연구에서는 천연에 존재하는 약용작물을 활용한 대용 차 소재 발굴을 목적으로, 아프리카의 대표적 약용작물인 K. africana 및 M. oleifera 잎 추출물의 항산화 효과 및 인체 대장암 세포주인 HCT116 증식억제에 미치는 효과를 규명 하였다. 실험 결과, 건조된 시료 추출물의 총 페놀성 화합물 함량은 K. africana의 경우 1.308 mg/g, M. oleifera는 1.241 mg/g으로 나타났다. DPPH radical scavenging 효과는 K. africana 잎 추출물의 경우, 0.1 mg/mL 농도에서 83.22%, 0.2 mg/mL에서 99.78%로 나타났으며, M. oleifera는 0.1mg/mL 농도에서 40.82%, 0.2 mg/mL와 0.8 mg/mL에서는 각각 65.12%와 88.48%로 나타나 시료 농도와 비례하여 유 의적으로 증가하였다. 또한 인체 대장암 세포주인 HCT116 에 K. africana 및 M. oleifera 잎 추출물을 농도별로 처리하 였을 때, 성장저해가 유의적(p < 0.05)으로 증가하였으며, 특히 M. oleifera 잎 추출물이 성장저해 효과가 높은 것으로 확인되었다. 이어서 HCT116 세포 생존을 조절하는 것으로 알려진 P53과 Bcl-2 family 발현에 미치는 K. africana 및 M.oleifera 효과를 관찰한 결과, 암 억제 유전자인 p53과 Bax의 발현은 증가하였고, 반대로 Bcl-2의 발현은 감소하여, K.africana 및 M. oleifera 잎 추출물이 HCT116 세포사멸에 중요한 역할을 하는 것으로 관찰되었다. 이상의 실험결과를 통해 K. africana 및 M. oleifera 잎 추출물이 항산화 및 HCT116 세포사멸에 효과가 있음이 확인되었으며, 이를 활용한 대용차 개발 및 건강기능 소재 발굴을 위한 기초자료 로 활용될 수 있으리라 기대된다. The present study was performed to investigate the antioxidant and anti-proliferative activities of African traditional plants, Kigelia africana and Moringa oleifera leaves, on HCT116 human colon cancer cells. This study examined the antioxidants and apoptotic analysis with the 2,2-diphenyl-1-picrylhydrazyl free radical scavenging, 3-[4,5-dimethylthiazol- 2-yl]-2,5-diphenyltetrazolium bromide assay and western blotting experiments. The total phenolic contents of the 70% ethanolic extracts of K. africana and M. oleifera leaves were 1.308±0.0071 mg GAE/g dry weight and 1.241±0.9205 mg GAE/g dry weight, respectively. The effects of K. africana and M. oleifera leaves on the apoptotic pathway were also investigated. The Bcl-2 level decreased, but the Bax and p53 levels increased in a dose-dependent manner. These results suggest that K. africana and M. oleifera leaves might be promising candidates for natural ingredients to produce healthy food and alternative tea resources.

        보이차(Pu-erh tea)의 HaCaT 세포보호 및 HCT116 세포사멸 효과

        The present study was performed to investigate the biological activity of fermented Pu-erh (FP) and raw Pu-erh (RP) tea extractives. In this study, we examined the antioxidants with the DPPH and ABTS assays and conducted an apoptotic analysis with the MTT assay and western blotting experiments. The total phenolic contents in FP and RP were 38.59 ± 15.23 and 36.83 ± 6.60 mg GAE/g dry weight, respectively. The effects of FP and RP on the apoptotic pathway were also investigated. It was found that the levels of Bcl-2 decreased, whereas those of Bax and cleaved caspase-9, caspase-3 and PARP increased in a dose-dependent manner. In addition, we investigated the protective effects of FP and RP against ultraviolet B-induced oxidative damage in human keratinocytes (HaCaT cells). To induce oxidative stress in the HaCaT cells, they were irradiated with UVB at 30 mJ/㎠. FP and RP was found to be reduced phosphorylation of IkBα and IKKα/β, block nuclear factor-kB p65 nuclear translocation, and strongly suppress the pro-inflammatory mediators.

        우전 추출물의 항산화 및 HCT116 인체대장암 세포사멸 효과

        Woojeon is a green tea plucked in early to mid-April, which has a beautiful, smooth and concentrated flavor that lingers in the mouth and evaporates on the mid-tongue. Polyphenols, the main components of green tea, are antioxidant phytochemicals and are known to have protective effects against various diseases. The objective of this study is to investigate the antioxidant and anticancer activities of Woojeon on HCT116 human colon cancer cell lines. It was found that the total polyphenol contents of Woojeon was 30.09 mg GAE/g, and antioxidant activities were 73.85%, 33.26% and 48.52 mg TEAC/g, measured by DPPH, ABTS and FRAP assay, respectively. The treatment of HCT116 human colon cancer cell lines with Woojeon at various concentrations resulted in a sequence of changed characteristic of apoptosis, including the loss of cell viability and morphological changes. Further, annexin V-fluorescein isothiocyanate and propidium iodide double staining through flow cytometry analysis demonstrated that the degree of apoptosis was increased by Woojeon. In addition, the Bax and Bcl-2 expression levels were increased in a dose-dependent manner, resulting in the activation of caspase-9, caspase-3 and cleaved poly-ADP-ribose polymerase; these observations are hallmarks of apoptotic events. Moreover, c-Jun N-terminal kinase, p38 MAPK and extracellular signal-regulated kinase was indicated Woojeon-induced apoptosis, and was accompanied by phosphorylation of the mitogen-activated protein kinase family.
