Overexpression of several aquaporins (AQPS) has been reported in different types of human cancer but roles in human carcinogenesis have yet to be clearly defined. Here, we up-regulated expression of the AQP8 gene in SiHa human cervical cancer cells with a lentivirus transfection system and investigated its effects as a potential therapeutic target for cervical cancer. Results showed AQP8 overexpression did not affect their substrate adherence and proliferation, but accelerated migration as assessed by transwell migration and wound healing assays. Moreover, AQP8 overexpression significantly enhanced local invasion of SiHa cells in nude mice. These findings altogether indicate that AQP8 overexpression increases migration of SiHa cells and probably participates in the process of tumor local invasion.

Overexpression of several aquaporins (AQPs) has been reported in different types of human cancer but their role in carcinogenesis, for example in the cervix, have yet to be clearly defined. In this study, expression of AQPs in cervical carcinomawas investigated by real-time PCR, immunofluorescent and immunohistochemical assays and evaluated for correlations with clinicopathologic variables. AQP1, 3, 8 exhibited differential expression in cervical carcinoma, corresponding CIN and mild cervicitis. AQP1 was predominantly localized in the microvascular endothelial cell in the stroma of mild cervicitis, CIN and cervical carcinoma. AQP3 and AQP8 were localized in the membrane of normal squamous epithelium and carcinoma cells, local signals being more common than diffuse staining. AQP1 and AQP3 expression was remarkably stronger in cervical cancer than in mild cervicitis and CIN2-3 (P<0.05). AQP8 expression was highest in CIN2-3 (91.7%), but levels in cervical carcinoma were also higher than in mild cervicitis. AQP1, AQP3, AQP8 expression significantly increased in advanced stage, deeper infiltration, metastatic lymph nodes and larger tumor volume (P<0.05). Our findings showed that AQPs might play important roles in cervical carcinogenesis and tumour progression in Uygur women.

The tributyltin (TBT)-resistant bacterium, Pseudomonas aeruginosa 25W, which was isolated in seawater from the Arabian Sea, was subjected to transcriptome analysis in the presence of high concentrations of TBT. Only slight effects were observed at TBT concentration of 50 μM, but exposure to 500 μM resulted in the upregulation of 6 genes and the downregulation of 75. Among the 75 downregulated genes, 53% (40 out of 75) were of hypothetical function, followed by 14 transcriptional regulation- and translationassociated genes. The results of this study indicated that although the 25W strain was highly resistant to TBT, high concentrations of TBT result in toxic effect on the transcriptional and translational levels. The target genes likely belong to a specific category of transcription- and translation-associated genes rather than to other gene categories.
Spin-torque Magnetic Resonance of Fe Nanoparticles in Fe/MgO/Fe Magnetic Tunnel Junctions

The magnetic resonance of Fe nanoparticles has been observed in single-crystal Fe/MgO/Fe magnetictunnel junctions (MTJs). In the MTJs, one magnetic layer is a continuous Fe film, whichacts as a magnetic reference layer, and the other is made up of noncontinuous nanoparticles of Fe,which act as a free layer. The magnetic resonance is electrically excited by the spin torque andis detected as the homodyne detection voltage, which is called the spin-torque diode effect. Fromthe magnetoresistance characteristics, the average diameter of the Fe nanoparticles is estimatedto be 6.8 nm; these nanoparticles exhibit superparamagnetic behavior at room temperature. Thisspin-torque magnetic resonance in magnetic nanoparticles enables us to study the magnetizationdynamics in small spin systems.

In this study, we investigate whether arsenite-induced DNA damage leads to p53-dependent premature senescence using human glioblastoma cells with p53-wild type (U87MG-neo) and p53 deficient (U87MG-E6). A dose dependent relationship between arsenite and reduced cell growth is demonstrated, as well as induced YH2AX foci formation in both U87MG-neo and U87MG-E6 cells at low concentrations of arsenite. Senescence was induced by arsenite with senescence-associated β-galactosidase staining. Dimethyl- and trimethyl-lysine 9 of histone H3 (H3DMK9 and H3TMK9) foci formation was accompanied by p21 accumulation only in U87MG-neo but not in U87MG-E6 cells. This suggests that arsenite induces premature senescence as a result of DNA damage with heterochromatin forming through a p53/p21 dependent pathway. p21 and p53 siRNA consistently decreased H3TMK9 foci formation in U87M G-neo but not in U87MG-E6 cells after arsenite treatment. Taken together, arsenite reduces cell growth independently of p53 and induces premature senescence via p53/p21-dependent pathway following DNA damage.

Two diastereoisomers of cyclo(Pro-Tyr) have been synthesized simultaneously. The crystal structures and conformations of both cyclo(L-Pro-L-Tyr) and a racemic mixture of cyclo(D-Pro-L-Tyr) and cyclo(L-Pro-DTyr), abbreviated as rac-cyclo(D-Pro-L-Tyr/L-Pro-D-Tyr), have been determined by a single-crystal X-ray diffraction study at low temperature. The crystals of rac-cyclo(D-Pro-L-Tyr/L-Pro-D-Tyr) belong to orthorhombic space group Pna21 with a = 10.755 (1), b = 12.699 (1), c = 9.600 (1) Å and Z = 4. The tyrosine side chain is folded towards the diketopiperazine (DKP) ring. The DKP ring adopts a twist boat conformation with pseudo symmetry C2v. The pyrrolidine ring has an envelope conformation with the N5, C4, C7 and C8 atoms in a plane. The crystal of rac-cyclo(D-Pro-L-Tyr/L-Pro-D-Tyr) is stabilized by hydrogen bonds between amide N2-H2 and carbonyl oxygen O2 in the neighbor. The hydroxyl group of tyrosine residue is also hydrogen bonded to the oxygen of the carbonyl group of the DKP ring in the next molecule. The spectroscopic properties of both isomers are also described.
Peptides from Bombyx Fibroin Counter D-Galactose-induced Hair Aging in Mice

Using proteases, we produced a peptide mixture from fibroin of the silkworm $Bombyx$ $mori$. Mice received D-galactose by subcutaneous injection for 8 weeks to accelerate aging, and then received the peptide mixture orally for 7 weeks. In the mice aged with D-galactose, the coefficient of friction of hair increased significantly up to 1.6-fold, but in the mice subsequently given the peptide mixture, it was normal. Scanning electron microscopy showed improved hair cuticles in the latter mice too. These results indicate that oral administration with peptides from $Bombyx$ fibroin counters the aging of hair cuticles.
