Pseudomonas syringae pv. actinidiae strains, the causal agents of bacterial canker on kiwifruit, were isolated from Korea and Italy in 2011. Among 87 isolates, a total of six representative strains, three from Korea and three from Italy, were identified on the basis of biochemical and physiological tests. Identities were confirmed by PCR using P. syringae pv. actinidiae-specific primers PsaF1/R2, which amplified a 280-bp DNA fragment. The strains isolated from Korea in this study displayed BOX-PCR patterns similar to those isolated from Italy but different from those isolated previously in Korea or the pathotype P. syringae pv. actinidiae strain. The effector hopA1 and hopH1 genes, which are known to be present in strains isolated recently from France and Italy, were also present in P. syringae pv. actinidiae strains, SYS1, SYS2 and SYS4, isolated from Korea in this work. However, no amplicons of the expected size were obtained from strains previously isolated from Korea and Japan. In addition, the Korean strains isolated in this work belonged to haplotype I for the cts gene identical to those strains isolated from recent outbreaks in Italy. These results suggest that P. syringae pv. actinidiae strains isolated from Korea and examined in this work are a new type of strain similar to those found from recent outbreaks in Italy. This is the first report on the occurrence of cts haplotype I strains of P. syringae pv. actinidiae affecting kiwifruit plants in Korea.

Background This study aimed to investigate the feasibility of circulating tumor cells (CTCs) and peripheral blood cells (PBCs) as predictive markers for immunotherapy in advanced non-small cell lung cancer (NSCLC). Methods This study included patients with advanced NSCLC receiving pembrolizumab or atezolizumab from July 2019 to June 2020. Blood was collected in K2-EDTA tube before each administration from cycle 1 to 4 (C1-4), and PBC counts (absolute neutrophil count[ANC], neutrophil-to-lymphocyte ratio[NLR], derived NLR[dNLR], platelet-to-lymphocyte ratio[PLR]) were calculated at each cycle. CTCs were enriched by using CD-PRIMETM system, the antibody-independent size-based isolation Method, and identified by single positive cells (EpCAM/CK+CD45-) in Bioview CCBS system. Results Among 89 response-evaluable patients, objective response rate was 21% and durable clinical benefit (DCB) were 35%. Baseline (C1) CTC and PBC count did not show a significant difference according to best response and DCB. However, patients with lower NLR, dNLR and PLR at C1 showed significant benefit in progression-free survival (PFS) and overall survival (OS) (p<0.05 for all). Patients with decreased CTC at C2 compared to C1 showed benefit in median PFS (6.2 vs 2.3, p=0.078) and OS (not reached [NR] vs 6.8, p=0.021) than those with increased CTC count. Patients with decreased ANC and NLR at C3 compared to C1 showed benefit in median PFS (6.3 vs 1.9, p=0.001 and 6.3 vs 1.9, p=0.007) and OS (NR vs 5.1, p=0.014; NR vs 6.8, p=0.045) than those with increased ANC and NLR. Low dNLR (≤2.0) at C1, decreased CTC at C2 and decreased ANC at C3 compared to C1 were the independently significant factors for survival (Fig. 1). Conclusion The CTC and PBC count at baseline and their changes during treatment could be potential biomarkers to predict the survival benefit and may help risk-group stratification in patients with advanced NSCLC who received anti-PD-1/PD-L1 immunotherapy.
Influence of Effective Microorganisms on Polluted Marine Sediment and Its Microbial Community

Lactobacillus sp., Acetobacter sp. and yeast were the most dominant organisms in the EM stock culture and subculture product. Lactic acid bacteria and yeast were able to grow in the fermentation process utilizing seawater. EM treatment of higher concentrations using EM stock culture and EM clay balls (1% or 4%) contributed to an early removal of malodor and an increase of DO in the polluted sediments, indicating an odor-removing activity of EM. The EM treatment of higher concentrations (1% or 4%) somewhat appeared to modify the microbial communities within the sediments, which was confirmed by existence of a few unique fragments from the stock culture based on PCR-DGGE. It still remains to be elucidated that EM cultures were directly involved in the malodor removal and potential sediment bioremediation.
Actuation of PVDF Cantilever with PEDOT/PSS (DMSO) Electrode Using Ion-Assisted-Reaction

Bimorph cantilevers were fabricated by using the piezoelectric polymer [poly(vinylidene uoride), PVDF, phase] for the active layers and the highly conducting polymer [poly(3,4-ethylenedioxy thiophene)/poly(4-styrenesulfonate), PEDOT/PSS] for the electrodes. In order to make the hydrophilic layer and diusion barrier on the polymers, the lms were modied by an ion-assistedreaction (IAR) method. The surface energy of the IAR-treated samples was calculated from the measured contact angles. The hydrophilic groups on the IAR-treated samples were veried by Xray photoelectron spectroscopy (XPS) analysis. In order to conrm the penetration of the epoxy adhesive into the PEDOT/PSS (DMSO as solvent) lm, the XPS and the scanning electron microscopy (SEM) images were analyzed. To fabricate the bimorph cantilevers, the two active PVDF layers both having the PEDOT/PSS (DMSO) electrodes were bonded by using epoxy adhesive. The displacement of the cantilevers was measured at the resonance frequency, and the actuation of the device made with the IAR-treated electrode was higher than that of the device made with the pristine electrode.

본 연구의 목적은 다이아지논(Diazinon; O, O-diethyl O-[6-methyl-2 (1-methylethyl)-4-pyrimidinyl] phosphorothioate)에 노출된 모델 생물체(송사리)의 행동변화와 관련된 분자생물학적 기전 규명을 통하여 비정상적 행동의 모니터링을 위한 생물지표(biomarker)를 개발하는데 있다. 이를 위해 우선 suppression subtractive hybridization (SSH) 및 DNA microarray 기법을 활용하여 다양한 유전자를 스크리닝하였다. 다이아지논에 노출시킨 송사리에서 발현의 차이가 나는 상향 조절된 유전자 97개 (알려지지 않은 유전자 27개 포함)와 하향 조절된 유전자 99개 (알려지지 않은 유전자 60개 포함)를 동정 하였고 이들 중 이상행동과 관련되는 것으로 보이는 유전자 10개 (상향조절 5개, 하향조절 5개)를 선발하였다. 이들 중에서 primer 제작이 잘된 beta-1, Orla C3-1, parvalbumin 및 apolipoprotein E을 선발하여 그 유전자 발현을 real-time PCR 기법을 사용하여 정량적으로 모니터링 하였다. Orla C3-1, parvalbumin 및 apolipoprotein E는 고농도의 다이아지논 처리(1000 ppb; 24 h)에서 그 발현이 억제됨이 관찰되었다. 다이아지논 처리 시 신경질환 (알츠하이머 병 및 다운신드롬)에 관련된 apolipoprotein E와 근육세포의 유연화에 작용하는 parvalbumin 등의 발현억제는 송사리의 인지능력 교란 및 근육세포의 경직 등을 각각 유도하여 송사리의 비정상적 행동을 야기하는 것으로 판단되었다. 따라서 이들 생물지표는 신경독성물질에 의한 송사리 및 기타 어류의 이상행동의 변화의 감지에 활용될 수 있을 것으로 사료된다.

The objective of this minireview is to examine how cometabolic biodegradation of polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs) might be affected by plant terpenes and lignins as natural substrates abundant in nature. The topics covered, hence, are environmental significance of PCBs and PAHs, nature and distribution of plant terpenes and lignin, structural and metabolic similarities of the natural compounds to PCBs and PAHs, and possible roles of the natural substrates in inducing the biodegradative pathways of PCBs and PAHs.
Neuroprotective Effects of Carpinus tschonoskii MAX on 6-Hydroxydopamine-Induced Death of PC12 Cells

The present study investigated the neuroprotective effect of Carpinus tschonoskii MAX and its intracellular protective mechanism on 6-hydroxydopamine (6-OHDA)-induced oxidative damage in PC12 cells. We found that pretreatment of PC12 cells with C. tschonoskii extract significantly inhibited the cell death induced by 6-OHDA in a dose dependent manner. C. tschonoskii extract decreased 6-OHDA-induced apoptotic events such as chromatin condensation, DNA fragmentation, the decrease of Bcl-2/Bax ratio, caspase-3 activation and PARP cleavage. C. tschonoskii extract also reduced generation of 6-OHDA-induced reactive oxygen species and nitric oxide. Furthermore, C. tschonoskii extract up-regulated the myocyte enhancer factor 2 D (MEF2D), a critical transcription factor for neuronal survival, and Akt activity, whereas it inhibited the activity of ERK1/2 and JNK. The results suggest that C. tschonoskii extract decreases 6-OHDA-induced oxidative stress and could prevent PC12 cell apoptosis induced by 6-OHDA via the up-regulation of MEF2D and Akt activity, and thus may have application in developing therapeutic agents for Parkinson`s disease.
