      • 인슐린 세포사멸 억제활성과 FAK/p130 Cas 신호전달과의 관계

        The focal adhesion kinase (pp125FAK) is a nonreceptor tyrosine kinase involved in integrin-mediated signal transduction pathway which includes the activation of' the Ras/mitogen-activated protein (MAP) kinase cascade. Tyrosine phosphorylation of FAK is essential to its activity, as interference with FAK phosphorylation results in apoptotic death in various cellular systems. Insulin receptor has the ability to suppress apoptotic death mediated by growth factor withrawal using signal pathways which is dependent on protein farnesylation. In the present study, we investigated the impact of FAK tyrosine phosphorylation in two responses of insulin: (1) Ras/MAP kinase activation, and (2) Prevention of apoptosis. To this purpose, we examined whethei@ insulin can mudulate FAK activity together with p130Cas, which is a downstream component of FAK signaling. It was also investigated whether insulin can protect cells from apoptosis in CHO IR/△SOS cells. lacking the intrinsic function of mSOS1-Ras activation. Unexpectedly, insulin decreased the degree of tyrosine phosphorylation of FAK as well as pl30Cas where it protected cells from apoptosis both in wild (CHO-IR) and mSOS-Ras-deficient (CHO-IR/△SOS) cells. These results suggest that neither the Ras activity nor FAK activity play role in antlapoptotic protection by insulin. We also tested the ability of other survival factor (DPI) or death factor (TNF-α) to affect insulin's function to dephosphorylate p130Cas or to protect cells from apoptosis. Neither of them could affect tyrosine phosphorylation of p130Cas without regards to their antiapoptotic- and proapoptotic activities, respectively. Taken together, it is suggested that FAK/p130Cas signaling does not play any significant role(s) in insulin's antiapoptotic protection.

      • 설치류 뇌에서 c-Fos를 이용하여 삼투조절과 관련된 신경회로의 규명

        본 연구는 생쥐의 중추신경계통에서 고장액에 반응하여 c-Fos에 대한 면역양성을 나타내는 지역을 규명하고자 수행하였다. c-Fos의 발현은 면역조직화적으로 검출하고 신경세포의 활성화에 대한 표식자로 이용하였다. 고장액은 생쥐 뇌의 supraoptic nuclus, median preoptic nucleus organum vasculosum lamina terminalis, sufornical organ, locus coeluleus, nucleus of solitary tract, ventrolateral medulla, parabrachial nucleus 등에서 발현되었다. 특히 retrochiasmatic area의 배쪽 부분에서 많은 c-Fos면역양성의 세포가 관철되었으며, 시상하부의 paraventricular nucleus나 두뇌의 area postrema에서는 c-Fos면역양성세포가 거의 관철되지 않았는데 이들 지역들은 흰쥐에서는 다수의 c-Fos면역양성 세포가 나타나는 지역이다. 이상의 결과들은 생쥐에서 고장액이에 의해 c-Fos면역양성 세포가 나타나는 지역은 흰쥐에서와는 일부 상이한 결과를 보여주는 것으로 나타났다. Experiments were carried out on adult mouse to investigate the regional localization of the cells inducing c-Fos immunoreactivity in central nervous system in response to hypertonic saline(HS). c-Fos induction, detected immunohistochemically, was regarded as a marker for neuronal activation. Intraperitoneal injection of HS resulted in dense c-Fos-like immunoreactivity (FLI) in several forebrain and brainstem area. These areas include the supraoptic nucleus(SON), median preoptic nucleus (MnPO), organum vasculosum lamina terminalis(OVLT), subfornical organ(SFO), locus coeruleus(LC), nucleus of solitary tract(NTS), ventrolateral medulla(VLM), and parabrachial nucleus(PBN). In particular, the ventral region of retrochiasmatic area showed numerous and very intensely labeled FLI cells. No FLI or few FLI cells were, however, detected in the hypothalamic paraventricular nucleus (PVN) and the area postrema(AP) respectively, the regions which revealed very intensive FLI in rat after HS treatment. These data suggest that FLI induced in response to osmotic stimuli in the mouse brawn has a different regional distribution from that of rat brain.

        This study rearches how graded maximal exercise has an effect on the university handball atheletes group and the general individuals group before exercise, after exercise and in the recovering two hours through cellic immunoreaction. Graded maximal excercise has been done by means of treadmill, applying Bruce protocol which was practised by male university students to see what difference there is between the athelete group and general individual group. The following is the results of comparing and analyzing the immunoreaction of the drawn blood before exercise, after exercise and in recovering two hours. 1. When graded maximal exercise is done, total white blood cells increase more after exercise for both the athlete group and the general individual group. And they are inclined to increase continously in the recovering two hours. Statistically there is no meaningful difference between them. 2. When graded maximal exercise is done, neutrophils increase more after exercise than before exercise for both the athlete group and the individual group. And they are inclined to increase continously in the recovering two hours. Statistically there is no meaningful difference between them. 3. When graded maximal exercise is done, lymphocytes increase more after exercise for both the athletes group and the general individuals group. For the athletes group, lymphocytes increase continously in the recovering two hours. But for the general individuals group, lymphocytes are inclined to come to stability as usual. Statistically there is no meaning difference between them. 4. When graded maximal exercise is done, monocytes increase more after exercise than before exercise. And they continue to increase in the recovering two hours for the athletes group. Monocytes decrease after exercise, but they are inclined to increase in the recovering two hours. Statistically there is no meaningful difference between them. 5. When graded maximal exercise is done, natural killer cells increase more after exercise than before exercise. And they continue increasing in the recovering two hours for both the athletes group and the general individuals group. Statistically there is no meaningful difference between them. All the above results being considered, the graded maximal exercise make immune cells change a lot. It is suggested that especially lymphcytes and natural killer cells can be a factor which enforces human body immunity function.

        노인틀니 보험급여를 위한 건강보험재정추계연구

        The purpose of this study is to estimate the National Health Insurance budget needed for the Korean elderly's denture service coverage. For more exact estimating the budget, denture need and demand were calculated by using 2000 National Oral Health Survey data. The results were summarized as follows ; 1. Over 65 years old, full denture and partial denture need per arch were each 8.5% and 16.8%. And over 70 years old, those were each 11.3% and 17.8%. 2. Over 65 years old, full denture and partial denture demand per arch were each 22.8% and 21.2%. And over 70 years old, those were each 27.%% and 21.1%. 3. Based on the denture need, the health insurance budget needed in the first operative year was about 955.4 billion won, over 65 years old and about 644.2 billion won, over 70 years old. 4. Based on the denture demand(wear), the health insurance budget needed in the first operative year was about 207.5 billion won, over 65 years old and was about 151.6 billion won, over 70 years old. If health insurance policy of elderly's denture service coverage is performed, actual denture wear rate is going to increase, and the budget needed is also increasing. Because current National Health Insurance budget status is poor, it is difficult to execute health insurance coverage for elderly's denture service. Therefore we suggest that health insurance coverage for preventive service is to be considered in order to reducing denture need in the long term view.

      • Prevalence and resistance patterns of extended-spectrum and AmpC &bgr;-lactamase in <i>Escherichia coli</i>, <i>Klebsiella pneumoniae</i>, <i>Proteus mirabilis,</i> and <i>Salmonella</i> serovar Stanley in a Korean tertiary hospital

Prevalence and resistance patterns of extended-spectrum and AmpC &bgr;-lactamase in <I>Escherichia coli</I>, <I>Klebsiella pneumoniae</I>, <I>Proteus mirabilis,</I> and <I>Salmonella</I> serovar Stanley in a Korean tertiary hospital. APMIS 2010; 118: 801–8.</P><P>A total of 100 clinical isolates of <I>Escherichia coli</I> (n = 35)<I>, Klebsiella pneumoniae</I> (n = 63)<I>, Proteus mirabilis</I> (n = 1), and Salmonella serovar Stanley (n = 1), showing resistance to cefoxitin, or returning positive in extended-spectrum &bgr;-lactamase (ESBL) by Clinical and Laboratory Standards Institute (CLSI) ESBL confirmatory method, were studied. The isolates were examined by the boronic acid (BA) disk test, polymerase chain reaction, and pulsed-field gel electrophoresis (PFGE) to investigate genetic similarities. The concurrence rates for ESBLs by the CLSI and the BA disk test were 97% for <I>E. coli</I> and 96.7% for <I>K. pneumoniae</I>. A total of 41 isolates showing cefoxitin resistance yielded all positive by the BA disk test. All the 33 <I>K. pneumoniae</I> isolates, which showed positive by the BA disk test, were carrying AmpC genes. The TEM and CTX-M types were predominant in <I>E. coli</I> and the SHV and the CIT and/or DHA types were predominant in <I>K. pneumoniae</I>. PFGE analysis showed almost 75% of genetic similarities among <I>K. pneumoniae</I> isolates producing ESBLs and/or AmpC &bgr;-lactamases (AmpCs) as each <I>K. pneumoniae</I> carried variable genes and showed variable antibiotic patterns. Clearly, the BA disk test was a useful method for the detection of ESBLs and AmpCs. In particular, cefoxitin resistance and BA-positive trait of <I>K. pneumoniae</I> do reflect the presence of AmpC genes in the organism.</P>

      • O-2 : Evaluation of PCR-reverse blot hybridization assay to identify pathogens and detect antimicrobial resistance genes in positive and negative blood samples

        Background: This study evaluated the diagnostic performance of newly developed polymerase chain reaction-reverse blot hybridization assay (PCR-REBA) in diagnosing bacteremia and detecting antimicrobial resistance genes. Methods: A total of 1,400 consecutive blood culture samples were selected from March to July 2013. The bottle included 300 that were positive and 1,100 that were negative by continuous monitoring blood culture system (CMBCS) with a delta neutrophil index of > 2.7%. The positive and negative bottles were tested by PCR-REBA and real-time PCR-REBA, respectively. Results: Of the 300 positive samples, 201 (67.0%) contained Gram-positive bacteria (GP), 73 (24.3%) contained Gram-negative bacteria (GN), 14 (4.7%) contained Candida species, and 12 (4.0%) contained polymicrobials. The correct agreement rates between blood culture and PCR-REBA for GP, GN, fungi, and polymicrobials were 94.5%, 97.3%, 100%, and 91.7%, respectively. Among the 92 methicillin-resistant Staphylococcus samples, the mecA gene was detected in 90 isolates (97.8%), and the vanA gene was correctly detected in 1 vancomycinresistant isolate. When the cycle threshold for real-time PCR was 30, 2.1% of negative samples (23/1,100) tested positive. Among the negative samples, the positive rate by real-time PCRREBA was low and the results of the conventional test, real-time PCR-REBA, and 16S rRNA sequence analysis were not concordant. Conclusion: The PCR-REBA system could be useful to simultaneously and rapidly detect causative agents and antimicrobial resistance genes, such as mecA and vanA or vanB, in positive blood samples.

        항공전자시스템 개발치구 개발

        ASDR은 군용 항공기의 주 장비인 ASC와 이에 탑재되는 임무 운용 소프트웨어인 OFP 개발중에는 개발도구로 사용되며 또한 ASC/OFP 개발 후에는 유지/보수용 도구로 사용 될 수 있다. 본 논문에서는 저속통제기용으로 개발된 ASDR의 전체 구조, 기능 및 특히 관성 및 공기 데이터 센서 시뮬레이션 부분에 대하여 기술한다. ASDR(Avionic System Development Rig) is used as a development tool during the design and development of the ASC/OFP and then can be used as a maintenance tool after developing them for a military aircraft. This paper describes the overall structure and the important features of KO-l ASDR, especially about the dynamic simultion of inertial and air data sensors.

        정밀유도 비행체 탑재용 전원공급장치 개발

        A precision guided kit is a GPS/INS guided range extension system to improve the accuracy and availability of existing dumb bombs. The PSD(power supply device) installed in the GPS guided kit supplies electrical power to all electrical loads in the kit. In this paper, design process and test results of the PSD have been presented. Additionally, the performance results of the PSD during actual ground and flight tests have been described.
